2.“Metal Family”Analysis Based on Family Resemblance Theory

This study was aimed to analyze the similarities of characteristics between members of“metal family” in the five elements. Wittgenstein’s family resemblance theory was used in the analysis of members assigned to the category of“metal family”, which included metal, autumn, west, white, pungent and lung. The results showed that there was family resemblance between members of“metal family”. It was concluded that members were classified into the category of“metal family” through analogical reasoning of family resemblance. Any member to be classified as a member of“metal family” showed similarity with at least one of other family members. Certain member in the“metal family” had similarities in different aspects with multiple members in the family. Any two members of“metal family” may not always have any similarities. It can be deduced that certain physiological functions of lung were not owned by the lung itself, but by the characteristics of“metal family”. The range of“metal family” can be expanded and multiplied. The“metal family” can build new members in different areas.
4.Culture of vascular smooth muscle cells from coronary arteries of diabetic rats

Aim To investigate the method of culturing vascular smooth muscle cells (VSMC) of coronary arteries derived from experimental diabetic rats and to establish the model of VSMC for the study of chronic cardiovascular diseases of diabetes. Method After the establishment of the model of diabetic rats, the coronary arteries of rats were separated carefully to culture VSMC using the method of explanting culture. Results The primary culture of VSMC was performed successfully by the method mentioned above. The cells could be isolated by different kinds of digestive enzyme in three steps. 7 ~10 days afterward, the cells overlapped in layers and displayed the typical ‘hill and valley’ pattern and positive immunochemical staining for smooth muscle actin. Conclusion VSMC from experimental diabetes proliferates faster than normal VSMC and its cultivating condition is strictly limited. The morphology of diabetic VSMC is as same as that of normal rats.
5.CT Diagnostic Value of Right-sided Colon Cancer

Objective To evaluate the diagnostic value of CT by analyzing CT features of right-sided colon cancer.Methods CT findings of 35 cases with surgically proved right-sided colon cancer were analyzed retrospectively with regard to their pathologic features.Results 34 of 35(97.1%) were diagnosed accurately by CT . Of them , 29 cases (82.9%)were polypoid colon cancer , 6 cases were infiltrative colon cancer, 2 cases with typical “target sign”. 9 cases(25.7%) were found with pericolic infiltration during operation, 7 of 9 (77.8%) were detected by CT scan . Distal ileal involvement of 3 cases ( 8.6% ) were all detected by CT . Lymphadenopathy was showed in 11 (31.4%), 7 of 11(63.6%)were found by CT.Conclusion CT scan plays important role in diagnosis of right-sided colon cancer as well as in finding pericolic infiltration, distal ileal involvement and lymphadenopathy.
6.Effect of berberine on apoptosis and expression of Bax and Bcl-2 proteins in HeLa cells

Objective To investigate the effect of berberine on growth and apoptosis of human cervical cancer cell line HeLa and its possible mechanism.Methods The effect of berberine on growth of HeLa cells was studied by MTT assay.Apoptosis of HeLa cells exposed to berberine was observed by flow cytometry and DNA gel electrophoresis.The expression of Bax and Bcl-2 was studied by Western blotting analysis.Results Berberine markedly inhibited the proliferation of HeLa cells in a time-and dose-dependent manner.After incubation of HeLa cells with 20 and 40 mg/L berberine for 48 h,DNA Ladder can be observed.A typical "sub-G1 peak" was checked by flow cytometry.There was a very low rate of natural apoptosis(1.9?0.6)%,while in 5 mg/L berberine group,the apoptosis rate was(2.3?0.8)%.After exposing HeLa cells for 48 h to 20 and 40 mg/L berberine,the apoptosis rate reached(16.7?2.8)%(P
7.Effects of lumboperitoneal shunt and ventriculoperitoneal shunt in the treatment of post?traumatic hydrocephalus

Objective To compare the effects and complications of lumboperitoneal shunt ( LP ) and ventriculoperitoneal shunt ( VP ) in the treatment of post?traumatic hydrocephalus. Methods Eighty patients with post?traumatic hydrocephalus treated in the neurosurgery department of Zhangjiagang Hospital of Traditional Chinese Medicine from March 2014 to March 2010 were retrospectively analyzed. The patients were randomly divided into two groups,40 patients treated with LP were assigned into the LP group and 40 patients undergone VP treatment were seen as the VP group. All patients were followed up for 1 year to investigate and compare the symptom improvement rate,neurological deficit score and the incidence of complication of the two groups. Results The total effective rate of hydrocephalus disappearance in the LP group was 41. 50% ( 37/40) and 87. 50%( 35/40) in the VP group. The difference between the two groups was not statistically significant (χ2=0. 556,P=0. 456);the neurological deficit scores before treatment in the LP group and VP group were (28. 35±8. 64) points and (29. 13±7. 98) points,there was no significant difference between the two groups (t=0. 419,P=0. 676) . The difference in the scores of neurological deficit after treatment in the LP and VP group was not statistically significant ( (19. 32±5. 34) points vs. (21. 62±4. 86) points,t=1. 480,P=0. 143),the average scores of neurological deficits in the two groups were significantly better than those before treatment ( t=5. 623, 5. 084,P<0. 001 ) . There was no significant difference in the average score of improvement of urinary incontinence between the two groups before and after treatment ( t=0. 376,1. 265,P>0. 05) ,the average score of urinary incontinence improvement after treatment in both groups were better than those before treatment ( t=4. 891,5. 370,P<0. 001) . In the LP group,the incidence of bleeding,infection,shunt related complications and shunt abnormality were all 2. 50% ( 1/40 ) , the overall complication rate was 10. 00%, the incidence of bleeding,infection,shunt related complications and shunt abnormality in the VP group were 7. 50% (3/40), 10% ( 4/40 ) , 5% ( 2/40 ) 10%, ( 4/40 ) , the overall complication rate was 32. 50%, there was significant difference between the two groups (χ2=6. 050,P=0. 014) . Conclusion LP and VP have significant curative effect on the treatment of post?traumatic hydrocephalus,but the overall incidence of LP complication after 1 year is significantly lower than that of VP,and is worth popularizing widely in clinical practice
8.Reperfusion Therapy and White Blood Cell Count in Patients with Acute Myocardial Infarction

Objective To study the relationship between blood cell count and cardiac events in acute ST elevation myocardial infarction patients treated with reperfusion in the early stage. Methods In this study, we assigned 151 patients to whom reperfusion therapy had been delivered within 3 hours of STEMI symptom into two groups:primary percutaneous cor-onary intervention group and thrombolysis therapy group. Differences of cardiac events and white blood cell count in these two groups were analyzed in the first 4 days. Results In the second day and the forth day, cardiac events rate and white blood cell count were significantly lower in the primary percutaneous coronary intervention group than those in the thromboly-sis therapy group (P<0.05). In the second day, white blood cell count has a positive correlation with cardiac events rate (r=0.226, P<0.05). Conclusion Primary percutaneous coronary intervention decreased blood cell count and cardiac events rate. In patients with acute myocardial infarction, increasing white blood cell was related to cardiac events in the near future.
9.Comparison of blood high mobility group box-1 versus N-terminal prohormone of brain natriuretic peptide concentrations for assessment of cardiac dysfunction inpatients with severe sepsis

Objective To compare the blood high mobility group box-1 (HMGB-l) versus N-terminal prohormone of brain natriuretic peptide (NT-proBNP) for assessment of cardiac dysfunction in the patients with severe sepsis.Methods Ninety-eight patients of both sexes,aged 18-75 yr,were divided into 3 groups according to left ventricular ejection fraction (LVEF):non-cardiac dysfunction group (LVEF ≥ 50 %,n =43),mild cardiac dysfunction group (30% ≤ LVEF < 50%,n =41),and severe cardiac dysfunction group (LVEF < 30%,n =14).Acute physiology and chronic health evaluation Ⅱ (APACHE Ⅱ) score was performed.Venous blood samples were obtained for determination of the concentrations of HMGB-1 and NT-proBNP.Days of hospitalization and the survival rates within 28 days after admission to hospital were recorded.Pearson correlation between blood HMGB-l,NT-proBNP concentrations and LVEF was analyzed.Results Compared with non-cardiac dysfunction group,LVEF was significantly decreased,APACHE Ⅱ score and blood HMGB-1 and NT-proBNP concentrations were increased,days of hospitalization were prolonged,and the survival rates were decreased in mild and severe cardiac dysfunction groups,and the changes in the parameters mentioned above were more obvious in severe cardiac dysfunction group.The correlation coefficient between blood HMGB-1 concentration and LVEF was -0.639 and between blood NT-proBNP concentration and LVEF was-0.521,showing significant difference.Conclusion Blood HMGB-1 concentration provides higher reliability than blood NT-proBNP concentration in assessing the degree of cardiac dysfunction in the patients with severe sepsis.
10. AKT modulates PTEN expression in hepatocarcinoma HepG2 cells via C-MYC pathway

Objective: To investigate whether C-myc is involved in the modulation of protein kinase B (PKB, also known as AKT) on the expression of phosphate and tension homology deleted on chromsome ten (PTEN) in hepatoma HepG2 cells. Methods: HepG2 cells were treated with different concentrations of AKT inhibitor Capivasertib (5, 10 and 20 nmol/L). The expression levels of AKT, phosphorylated (p)-AKT, Bad, p-Bad, C-myc and p-C-myc proteins were detected by Western blotting. The transcription levels of C-myc downstream genes eukaryotic initiation factor-4E (eIF-4E), branched‐chain amino acid aminotransferase 1 (BCAT1) and WW domain-containing E3 ubiquitin protein ligase 1 (WWP1) were detected by real-time fluorescent quantitative PCR. The expressions of eIF-4E, BCAT1 and WWP1 genes were silenced by siRNA, then the transcription and expression of PTEN were detected by real-time fluorescent quantitative PCR and Western blotting, respectively. The effects of Capivasertib treatment or WWP1 gene silence on the expression and localization of PTEN protein in HepG2 cells were detected by cell immunofluorescence technique. Results: In HepG2 cells treated with different concentrations of Capivasertib for 8 h, the expression levels of p-AKT, p-C-myc and p-Bad were significantly downregulated as compared with the control group (all P < 0.01), and the expression levels of eIF-4E, BCAT1 and WWP1 mRNAs were also downregulated (all P < 0.01). Silencing eIF-4E, BCAT1 and WWP1 gene expressions had no significant effect on the transcription of PTEN (all P 0.01) but the silence of WWP1 gene could significantly enhance the expression of PTEN protein (P < 0.01). Both Capivasertib treatment and WWP1 gene silence could increase the expression level of PTEN and induce its aggregation on the cell membrane. Conclusion: AKT is able to affect the transcription of WWP 1 gene by C-myc pathwaty, and ultimately participates in the modulation of PTEN expression in HepG2 cells.
