2.hMLH1 promoter hypermethylation correlates with platinum-resistance in non-small cell lung cancer cells

Background and purpose:hMLH1 promoter methylation is one of the factors in the development of lung cancer.However,little is known about hMLH1 promoter methylation in association with chemotherapy resistance in lung cancer cells.This study aimed to investigate whether resistance to DDP can be overcame by restoring the expression of hMLH1 in A549/DDP cells after treatment with a demethylating agent,5-Aza-2'-deoxycytidine (5-Aza-CdR).Methods:hMLH1 mRNA was determined by RT-PCR in A549,A549/DDP and A549/DDP treated with 5-Aza-CdR;MSP was carried out to determine the methylation status of hMLH1 in A549 and A549/DDP,A549/DDP treated with 5-Aza-CdR.MTT test,Hoechst 33258 staining and flow cytometry were used to determine the change of cell proliferation,apoptotic morphology and the cell apoptosis index,respectively.Results:hMLH1 mRNA was expressed higher in A549 than in A549/DDP.The unmethylation status of hMLH1 was observed in A549 cells whereas partial methylation status of hMLH1 in A549/DDP cells.After treatment with 5-Aza-CdR,the A549/DDP cells displayed an unmethylation of the hMLH1 gene.After the treatment with cisplatin,the IC50 of A549,A549/DDP and A549/DDP treated by 5-Aza-CdR were (4.7?0.7),(30.1?1.8) and (6.9?0.6)?mol/L,respectively.Apoptotic bodies were less in A549/DDP cells than in A549/DDP cells treated with the 5-Aza-CdR.The rate of apoptosis of A549/DDP treated by 5-Aza-CdR was higher than that of A549/DDP.Conclusion:The methylation of hMLH1 may be relevant to the resistance to platinum-based chemotherapy in the A549/DDP cell line.5-Aza-CdR can inhibit hMLH1 methylation,restore the expression of hMLH1,and enhance cell apoptosis as well as the inhibition of cell proliferation of A549/DDP cells after the treatment of cisplatin.

Biliary atresia(BA) is a serious digestive system disease caused by the obliteration of intrahepatic and extrahepatic biliary system of infancy.Without prompt treatment,BA can cause biliary cirrhosis,and ultimately cause liver failure,leading to death.The early diagnosis of BA is crucial to the early treatment.At present,liver biopsy is considered to be the gold standard of diagnosis of BA,but this method has some shortcomings such as invasive,repeated sampling and subjectivity.In contrast,ultrasonography (US),magnetic resonance imaging (MRI) and hepatobiliary scintigraphy(HBS) gradually play important roles in clinical diagnoses of BA and become the hotspot of BA diagnoses.This paper focuses on reviewing the clinical applications of BA imaging diagnoses and the latest research progress.
5.Expression and clinical significance of FGF-2 and hTERT in non-small cell lung cancer

Objective To investigate the expression and clinical significance of fibroblast growth factor-2 (FGF-2) and human teclomerase reverse transcriptase (hTERT) in non-small cell lung cancer (NSCLC) tissues and analyze the correlation between them.Methods The expression of FGF-2 and hTERT were detected by immunohistochemistry in 90 NSCLC tissues and 20 non-lung carcinoma tissues.The correlation between the expression and clinicopathological features was also analyzed.Results The positive expressions of FGF-2(66.7% ) and hTERT (75.6%) in NSCLC tissues were significantly higher than that in nonlung carcinoma tissues (15% and 20%,respectively),P <0.01.The expression of FGF-2 and hTERT were significantly related to TNM stages and lymph node metastasis (P < 0.01 ),but not to histological types,sex and age of NSCLC patients ( P > 0.05 ).A positive correlation was found between the expressions of FGF-2 and hTERT in NSCLC(P < 0.01 ).Conclusion The expression of FGF-2 and hTERT in NSCLC,which can be regarded as important references for prognosis of NSCLC,play important roles in the initiation and development of NSCLC.
6.Effects of Pinacidil preconditioning on hsp70 and cPLA2 expression in hemorrh agic shock rats

Objective To investigate the mechanisms of Pinacidil prec onditioning protection of hemorrhagic shock rats. Methods The rats were divided into 3 groups: a normal group ( n=10), a control group (n=40) and a preconditioning group (n=40). Pinacidil preconditio ning was processed 24 h before making the hemorrhagic shock model. The cPLA 2 expression in myocardium was observed at different time points with western blot; Pinacidil preconditioning was processed 24, 48 and 72 h before making th e hemorrhagic shock model to observe hsp70 expression in myocardium and liver tissue with western blot. Results Pinacidil preconditioning could increase hsp70 expres sion and decrease cPLA2 expression. Conclusions Pinacidil preconditioning protects "shock cell" by inducing hsp70 overexpression and inhibiting cPLA2 expression, which is re sponsible for protecting myocardial and liver tissues of the hemorrhagic shock rats.
7.A meta-analysis of diffusion tensor imaging for diagnosis of early radiation-induced brain injury

Objective To analyze the diagnostic value of diffusion tensor imaging (DTI) for early radiation-induced brain injury.Methods We searched CNKI and PubMed and included articles according to the PRISMA statement.FA value, λ||, and λ丄 were pooled, and Forest plot generation, cumulative analysis, sensitivity analysis, and regression analysis were performed.We also used the funnel plot to analyze publication bias.Results A total 11 studies were included for meta-analysis.The results of FA value heterogeneity test were as follows:P=0.000, I2=93.6% in China;P=0.004, I2=82.2% in other countries and regions;P=0.000, I2=91.5% in all regions.The regression analysis of time-dose dependence and study design showed significant and non-significant associations, respectively, with the heterogeneity of these studies (adjusted P=0.026, P=0.005;adjusted P=0.859, P=0.908).The results of λ||and λ丄 heterogeneity tests were as follows:P=0.908, I2=0.0%;P=0.100, I2=56.6%.There was a sign of publication bias by Egger's test (coefficient:-6.26, 95%CI:-9.31 to-3.20, P=0.001) and Begg's test (P=0.004).Conclusions DTI-derived metrics are sensitive and reliable in the diagnosis of early radiation-induced brain injury.
8.Evaluation of the value of resting state BOLD-fMRI in radiation-induced brain injury after nasopharyngeal carcinoma radiotherapy

Objective To analyze the value of resting state BOLD-fMRI in the early diagnosis of brain injury after nasopharyngeal carcinoma radiotherapy.Methods Four groups of nasopharyngeal carcinoma patients,including before radiotherapy group (G0),0-3 months after radiotherapy group (G1), 3-6 months after radiotherapy group (G2) and 6-9 months after radiotherapy group (G3),were underwent a resting state BOLD-fMRI scan, and Matlab software DPARSF tool was used to analyze and postprocess the data based on G1-G0,G2-G0,G3-G0,G2-G1,G3-G2 and G3-G1.Results Compared with the G0 group,the activity of bilateral hippocampus and temporal lobes was decreased in G1 and G2 groups,and the decreased degree in G2 group was smaller than that in G1 group.Compared with G1 group,brain metabolic activity increased in G2 and G3 group,but did not return to the initial level before radiotherapy.Conclusion Abnormal changes of resting state BOLD-fMRI have an important clinical significance in early diagnosis of radiation-induced brain injury after nasopharyngeal carcinoma radiotherapy.

Atherosclerosis is a chronic inflammatory vascular disease developing from early fatty streaks to highly rupture-prone unstable plaques.Many cellular and molecular events are involved in each step.With the development of targeting probe technology,molecular magnetic resonance imaging techniques are expected to revolutionize the treatment strategies of atherosclerosis in the near future.Many desirable molecular probes targeting various components of plaque have emerged in recent years.This article reviews the molecular magnetic resonance imaging techniques of atherosclerosis and their application.
10.Application research of coronary CT angiography using low tube voltage in patients with a normal body mass index

Objective To assess the image quality of coronary CT angiography(CCTA) by using high-pitch spiral CT under 80 kV in patients with a normal body mass index(BMI), and to investigate the feasibility of low radiation dose.Methods 60 patients,who suspicious coronary artery diseases and BMI between 18.6-25.0 kg/m2 underwent CCTA in our hospital.All patients were randomly divided into group A and group B(each with 30).Patients in group A were given 100 kV CCTA and filter back-projection algorithm (FBP), and patients in group B were given 80 kV CCTA and iterative reconstruction(IR).CT values of aortic root, right coronary artery (RCA), left anterior descending artery (LAD), left circumflex (LCX) and standard deviation (SD) were measured.Then signal-to-noise ratio (SNR) and contrast-to-noise ratio (CNR) were calculated.All images were given quality subjective scoring by using 3-score method.The CT dose index was recorded and effective radiation dose (ED) was calculated.Results ED of group A and group B was 0.8 mSv and 0.41 mSv, respectively.Statistically significant difference was found between these two groups.In the study, SD was (28±6) HU in group A and (46±9) HU in group B, and the difference was statistically significant between the two groups.The CT values of aortic root, RCA, LAD and LCX under 80 kV were significantly increased, and the difference was statistically significant between the two groups.There was no statistically significant difference between two groups in RCA-SNR, LAD-SNR and RCA-CNR.But in LCX-SNR, LAD-CNR, LCX-CNR, there was statistically significant difference between two groups.There was no significant difference between the two groups in coronary artery segments which could be used for diagnosis.Conclusion For patients with a normal BMI, CCTA using Flash spiral CT under 80 kV could diagnose coronary artery disease and significantly reduce the radiation dose.
