Objectives To observe influences of two functionally important polymorphisms (5'-UTR and 3 -UTR) in thymidylate synthase (TS) gene and their combinative effects with drinking on the susceptibility of colorectal cancer (CRC). Methods A case-control study was designed, which included 140 CRC patients and 343 control subjects. Risk of CRC was estimated by unconditional Logistic model. Results The genotype distributions of the polymorphisms did not show significant differences between the cases and the controls. However, in those with -6 bp/-6 bp genotypes, the 2R carriers had a decreased risk (OR = 0. 60, 95%CI, 0. 23-1. 52) , but in +6 bp carriers, the 2R carriers had an elevated risk (OR=1. 87, 95%CI, 0. 94-3. 72). The likelihood test revealed the interaction existed between these two polymorphisms (P for interaction = 0. 05). Moreover, the combinative effect between the 3 -UTR polymorphism and alcohol drinking showed a significant association with the susceptibility of CRC (P for interaction =0. 01). In non-drinkers, the-6 bp allele carriers had a significant decreased risk (OR = 0. 57, 95%CI, 0. 32-0. 99) , but in ever and current drinkers, the +6 bp allele (6-bp insertion) carriers had an elevated risk (OR=1. 88, 95%CI, 0. 80-4. 41) , in which OR values ascended with the prolongation of drink-time. Conclusions The polymorphisms of thymidylate synthase at 5'-UTR and 3'-UTR may not be regarded as independent predictive factors for the susceptibility of CRC. However, interactions exist in these two polymorphisms and between the 3'-UTR polymorphism and drinking. Further studies with larger sample size and in different ethnic groups are needed.

Objective To analyse the necessity and feasibility of HCV RNA detection for donor screening.Methods Plasma from 50 donors was mixed into a minipool, the nucleic acid was extracted by NucliSens Extractor and amplified by NASBA, the amplification products were detected by ECL. A performance control and a positive control were coupled with the samples through storage, nucleic acid extraction, amplification and detection.Results Two specimens were found HCV RNA positive in the 10000 anti HCV negative samples.Conclusion HCV RNA screening of plasma minipools by NASBA may prevent hepatitis C virus infection.

Objective To investigate the clinical features of acute coronary syndrome (ACS) combined with low T3 syndrome and its prognosis.Method Eighty-seven ACS patients with low free triiodothyronine (FT3) level (low FT3 group) and additional 64 ACS patients with normal FT3 level(control group) were selected in parallel; all the patients underwent coronary angiography.The contents of FT3,N-terminal pro-brain natriuretic peptide (NT-proBNP),cardiac troponin I (cTnI) and high sensitivity-C reactive protein (hs-CRP) were recorded,and the related factors were evaluated.The number of stenotic vessels was calculated.Results The contents of NT-proBNP,cTnI and hs-CRP in low FT3 group were elevated significantly compared with those in control group [(1242.14 ± 226.32) ng/L vs.(1126.36 ±195.55) ng/L,(0.92 ± 0.23) ng/L vs.(0.84 ± 0.19) ng/L,(6.46 ± 2.25) mg/L vs.(5.71 ± 1.78) mg/L],and there were significant differences (P ＜ 0.01 or ＜ 0.05).The contents of FT3 were negatively correlated with NT-proBNP,cTnI and hs-CRP in two groups (P ＜ 0.05).There was no significant difference in the number of stenotic vessels between two groups (x2 =4.512,P =0.105).Conclusion In patients with ACS,there is a negative correlation between contents of FT3 and the prognosis,but there is no correlation between contents of FT3 and the number of stenotic vessels.

ObjectiveTo explore the value of 64-slice CT perfusion imaging(CTPI) in the early diagnosis of radiation-induced lung injury ( RILI ).MethodsForty-eight patients with upper esophageal cancer resection underwent CTPI before and after radiotherapy,and the TNF-α and TGF-β1 were measured from patient's peripheral blood.Serum cytokine,conventional CT appearances and CTPI parameters (rrBF,rrBV,rrPS) in patients with RILI (Group A) and non-RILI (Group B) were compared and analyzed.A randomized block design t-test was used for comparison of serum cytokines and perfusion values between the two groups.The Chi-square ( x2 ) test was used for comparison of detection rate between conventional CT and CTPI.ResultsRILI occurred in 18 of 48 cases ( 18/48,Group A).In Group A,TNF-α and TGF-β1 preand pos-radiation were (36.1 ± 15.0),(30.4 ±t 14.9) ng/L and (17.5 ±9.8),(14.3 ±7.6) μg/L,respectively,and there were no statistically significant differences (t =1.14,1.I0,P =0.264,0.279).At half-dose time point of radiation,there were no significant differences for TNF-α and TGF-β1 pre- and posradiation [ ( 30.4 ± 14.9),( 28.9 ± 14.7 ) ng/L and ( 14.3 ± 7.6),( 14.4 ± 6.0 ) μg/L,respectively ]between Group A and B ( t =0.33,1.23 ; P =0.746,0.227).The rrBF,rrBV and rrPS of post-radiation from Group A were significantly higher than those of pre-radiation ( t =5.67,5.97,6.11,P =0.000,0.000,0.000),the rrBF and rrBV of post-irradiation from Group B were significantly higher than those of pre-irradiation (t =6.52,7.84,P =0.000,0.000).There was no significant difference for rrPS in Group B pre- and post-radiation (t =1.36,P =0.178 ).There were significant differences for all perfusion values detected from radiation lung fields between Group A and B ( t =2.32,2.18,6.04,P =0.025,0.034,0.000).Taking rrPS =1.28 as a threshold value on ROC,the sensitivity and specificity of CTPI for diagnosis of RILI were 77.8％,93.3％,respectively,which were much higher than those ( 11.1％,90.0％,respectively) of conventional CT (x2=13.61,P=0.000).ConclusionCTPI parameters may reflect the hemodynamic changes of post-radiation lung and have potential values for the early diagnosis of RILI.

Objective: To investigate whether bcl-2 directly contributes to the development of drug resistance and apoptosis in o- varian cancer cell lines cells OC3. Methods: Retrovirus expression cector pLXSN-bcl-2 was constructed and was transfected to ovarian cancer cell line cells OC3 using Lipofectin,with empty vector pLXSN as a control. bcl-2 expession of transfected cells was analyzed by FACS and Western blot and Adr-induced cytotoxicity was measured by MTT assay. Apoptosis was also detected by PI DNA staining and DNA Ladder analysis.Results:pLXSN/bcl-2 transfected cells OC3/bcl-2 overexpressed bcl-2 and were resistant to Adr-induced cytotoxicity.The ability against Adr-induced apoptosis was increased. Conclusion: bcl-2 may play an important role in the resistance to Adr-inducing apoptosis, thereby increasing resistance of OC3/bcl-2 cells to chemotherapy.

Objective To evaluate the value of three dimensional reconstruction images of rotational DSA on measuring aneurysmal necks and make a comparison with traditional DSA so as to provide more abundant and accurate informations for the enbolization of aneurysm. Methods A comparison was made between the measurement of aneurismal necks from 14 cases with traditional DSA examination and a measurement made on three dimensional reconstruction images of the same patient. Results There was a difference shown in the measurment of the aneurysmal necks between three dimensional reconstruction images of rotational DSA and those of traditional DSA, outcoming with more angles and data on three dimensional reconstruction images. Conclusions There are more angles of aneurysmal neck can be shown on rotational 3D DSA especially for the demonstration of the largest aneurysmal neck with a directional value for the intervention. Objective To evaluate the diagnostic value of MRI for the mammographic sign of focal asymmetry of the breast (FAB) .Methods The morphological features of MRI and Time‐signal intensity curve (TIC) types of 46 cases of mammographic signs of FAB were analyzed retrospectively .Results In all 46 patients ,MRI detected 46 lesions in the corresponding areas of FAB on mammography ,in which 35 cases of benign lesions and 11 cases of malignant lesions were confirmed by pathology .The accuracy , positive predictive value and negative predictive value of the sub‐leaf shape and edge burr of the breast lesions were 82 .61% ,66 .67% , 86 .49% and 86 .96% ,85 .71% ,87 .18% respectively .TIC showed:in type Ⅰ ,all 17 cases were benign;In type Ⅱ ,4 cases were be‐nign and 2 cases were malignant ;In type Ⅲ ,9 cases were all malignant .The diagnostic accuracy of MRI for the benign and malignant breast lesions was 86 .96% .The positive predictive value and negative predictive values were 81 .82% and 96 .88% respectively .Con‐clusion MRI can definitely estimate the potential character of lesion in FAB ,and more accurately discriminate malignant and benign breast lesions .

BACKGROUND:As a novel noninvasive method, high frequency ultrasound has been used for the detection of skeletal muscle widely. OBJECTIVE:To detect the thickness of the dorsalLisfrancligament between the first (medial) cuneiform (C1) and second metatarsal (M2) using high frequency ultrasound to evaluate the feasibility and clinical application value of high frequency ultrasound for the detection of theLisfrancjoint. METHODS:Both feet of 100 healthy volunteers were included and the thickness of dorsalLisfranc ligament and the distance between C1 and M2wereexamined using high frequency ultrasound. RESULTS AND CONCLUSION:The display rate of high frequency ultrasound for the dorsalLisfranc ligament in the 100 healthy volunteers was 100%. There was no significant difference in the thickness of the dorsalLisfrancligament between both feet or sexes in adults(P> 0.05); as did the distance between C1 and M2 (P> 0.05). These findings suggest that visual dorsalLisfrancligament is detected by high frequency ultrasound. The measurement of the distance between C1 and M2by high frequency ultrasound may be a feasible, noninvasive, low-cost, and early diagnostic method forLisfrancinjuries.
