1.The relationship between high-sensitivity C-reactive protein level and carotid plaque in patients with acute cerebral infarction

Objective To study the relationship between high-sensitivity C-reactive protein level and carotid plaque in patients with acute cerebral infarction. Methods 73 patients with acute cerebral infarction were selected as research subject,then all the patients were tested with color doppler ultrasound.The patients were divided into unstable plaque group(A group)23 cases,stable plaque group(B group)25 cases and no plaque group(C group)25 cases,at the same time,25 healthy persons were selected as the control group(D group).The high-sensitivity C-reactive protein level and carotid plaque of the four groups were studied. Results The rate of carotid plaque with acute cerebral infarction was higher than that of healthy person(all P<0.05),and the high-sensitivity C-reactive protein level of D group was lower than that of A group,B group and C group,and the level of A group was higher than that of B group and C group(all P<0.05). Conclusion The detection rate of carotid plaque and high-sensitivity C-reactive protein level with acute cerebral infarction were both higher than those of healthy persons.It indicated there was cloce relationship between high-sensitivity C-reactive protein level and carotid plaque.
2.Effect of Alcohol Concentration of Shangtong Tincture on Percutaneous Absorption of Paeonol

Objective: To select the optimum alcohol concentration of Shangtong Tincture. Methods: The percutaneous absorption test was performed on excised rats skin in improved Franz diffusion cell. The penetrative amount of paeonol was determined quantitatively by HPLC and the content of paeonol as a marker. Results: The penetrative amount of paeonol of Shangtong Tincture with 60%~80% alcohol concentration have a optimal value, the average rate is 0.010mg/cm 2?h. Conclusion: Shangtong Tincture with 70% alcohol concentration is the best.
3.The clinical study on valsartan combined with indapamide in the treatment of hypertension

Objective: To explore the clinical effect of valsartan combined with indapamide in the treatment of hypertension. Methods: In our hospital in 2012 August ~ 2013 August treatment of 280 cases of hypertensive patients, all patients had drug withdrawal for 2 weeks. Were randomly divided into observation group 100 cases and control group 1, control group 2 of the 90 cases, control group 1 given valsartan, control group 2 given indapamide, the observation group were given valsartan combined with indapamide, were treated for 8 weeks. Results:three groups of patients before treatment, the blood pressure were not significantly different, after the treatment, the observation group decreased blood pressure was significantly higher than that in control group, the total efficiency had significant difference (x2=6.472, P<0.05). Conclusion:Valsartan combined with indapamide in the treatment of hypertension has fast acting, high curative effect, long duration, few adverse reactions, is the ideal choice for the treatment of hypertension, is worth the clinical promotion and application.
4.Surgical treatment of uremia caused secondary hyperparathyroidism

Uremia caused secondary hyperparathyroidism is one of the common and serious complications caused by uremia. Surgery treatment is effective. Total parathyroidectomy plus forearm transplantation is more commonly used because of exact outcomes and easy monitoring. It is necessary to be fully prepared be-fore operation,particularly to clarify the location of parathyroid gland. Yet each surgical method has its re-lapse rate.
5.Isolation, culture and CM-Dil labeling of rat mesenchymal stem cells in vitro

BACKGROUND:Currently, there is no uniform, standardized approach to isolate, purify and proliferate bone marrow mesenchymal stem cells. Chlormethylbenzamido-1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine (CM-Dil) is a stable, reliable, high marking and simple marker.
OBJECTIVE:To develop the methods for isolation, culture and identification of rat bone marrow mesenchymal stem cells in vitro.
METHODS:Two male Sprague-Dawley rats, weighing 50-100 g were taken to col ect the bilateral femur and tibia bone marrow under sterile conditions, and then, primary bone marrow mesenchymal stem cells were isolated and cultured using bone marrow adherent separation and density gradient centrifugation. cells were amplified and purified through timely and repeated passage, and labeled at the third generation with fluorescent dyes CM-Dil in vitro as a source of donor cells.
RESULTS AND CONCLUSION:Bone marrow mesenchymal stem cells were cultured successful y in vitro using bone marrow adherent separation and density gradient centrifugation separation methods, but the former was superior to the latter in the number of cultured cells significantly, while the two methods were not different significantly in terms of cellviability and proliferation. Flow cytometry results showed that the positive rates of cultured cells were 17.5%for CD34, 97.9%for CD44, and 91%for CD90. CM-Dil can label bone marrow mesenchymal stem cells successful y, which is a stable, reliable, high marking and simple marker.
6.Effects of enalapril on expression of MMP-2 and MMP-9 in renal hypertension rats with focal cerebral ischemic reperfusion

Objective To observe the effects of ACEI( enalapril) on expression of MMP-2 and MMP-9 in renal hypertension rats with focal cerebral ischemia-reperfusion injury. Methods Twenty-eight Wistar male rats were randomly divided into two groups: hypertension group and normaltension group. The former which formed model of renal hypertension rats by constricted renovascular were randomly divided into enalapril group (Y) and hypertension ischemia and reperfusion group(HIR),which fed with enalapril 2mg/kg and equal volum saline respectively;The latter were divided into sham-operation group ( N) and normaltension ischemia and reperfusion group (IR). The focal cerebral ischemia model was established in Wistar rats by using the method of thread inserting left middle cerebral artery occlusion(MCAO) for 2h. After ischemic 24h,the expression of MMP-2 and MMP-9 were measured by immunohisto-chemistry,and the gray scale value was measured by imaging analysis. Results Compared with N,the gray scale values of MMP-2 and MMP-9 in IR were higher(P <0. 01); Compared with IR, the gray scale values of MMP-2 and MMP-9 in HIR were higher(P<0. 05);Compared with HIR,the gray scale values of MMP-2 and MMP-9 in Y were lower(P<0.01). Conclusion Hypertension can increase the expression of MMP-2 and MMP-9 in rats with focal cerebral ischemia-reperfusion. ACEI (Enalapril) could inhibit the expression of MMP-2 and MMP-9 in renal hypertension rats with focal cerebral ischemia-reperfusion.
7.Analysis of distribution and the risk factors of arterial stenosis in patients with ischemic cerebrovascular dis-ease

Objective To investigate the distribution of arterial stenosis in patients with ischemic cerebrovas-cular disease and its risk factors .Methods 224 patients with ischemic cerebrovascular disease were divided into four groups according to DSA results .Patients showed no stenosis or mild stenosis were selected as control group ( 43 ca-ses),the other patients were divided into pure extracranial stenosis group (41 cases),simple intracranial stenosis group (93 cases) and extracranial stenosis group (47 cases).The results of laboratory test were analyzed .The ques-tionnaire was designed and the indicators including name ,age,long-term smoking,drinking,hypertension and diabetes were collected .Results Of all the subjects , there were 181 cases with artery stenosis .Single factor results showed that gender,age,long-term smoking,long-term drinking,hypertension,diabetes,high homocysteine,high level of lyso-phosphatidic acid were the independent risk factors of cerebral artery stenosis (χ2/t =8.744, 5.562, 10.736, 11.032,9.812,10.002,9.083,2.576,all P<0.05).Multivariate analysis showed that high homocysteine ,long-term smoking and drinking were the risk factors of simple intracranial artery stenosis .High homocysteine and high fibrino-gen were risk factors of simple extracranial stenosis .Age,hypertension,long-term smoking and drinking ,high homocys-teine and fibrinogen were risk factors of intracranial and extracranial stenosis .Conclusion In patients with ischemic cerebrovascular diseases ,the simple intracranial artery stenosis is most common .The incidence of cerebral artery ste-nosis has age characteristic ,with the increase of age ,the incidence rate of intracranial and extracranial artery stenosis is rising.High homocysteine,long-term smoking and drinking,hypertension,diabetes,high lysophosphatidic acid are independent risk factors of cerebral artery stenosis .
8.Serum PDGF-BB, sFlt-1 and U-Ⅱ in patients with preeclampsia

Objective To explore the clinical diagnostic value of joint detection of serum platelet-derived growth factor BB (PDGF-BB), soluble vascular endothelial growth factor receptor -1 (sFlt-1) and urotensinⅡ( U-Ⅱ) in preeclampsia disease.Methods The cases of obstetric patients suffering from preeclampsia in the Third People′s Hospital of Liaocheng , Shandong Province between October 2012 and April 2014 were enrolled , including 96 cases of mild preeclampsia and 81 cases of severe preeclampsia.Totally 68 cases of normal pregnant women with similar age and gestational age were selected as control group.A case-control study was applied for the following investigations.The concentrations of serum PDGF-BB, sFlt-1 and U-Ⅱwere measured using ELISA.The diagnostic value of PDGF-BB, sFlt-1 and U-Ⅱalone or in combination for preeclampsia was analyzed and evaluated with receiver operating characteristic curve ( ROC) and Logistic regression analysis.Results Serum concentrations of PDGF-BB, sFlt-1 and U-Ⅱin mild preeclampsia group were (80.45 ±21.87)ng/L,(23.03 ±6.67)μg/L and(4.54 ± 1.02)ng/L, and those in severe preeclampsia group were (124.91 ±47.54)ng/L,(35.65 ±12.45)μg/L and(6.29 ±2.31) ng/L, while those in control group were (60.89 ±19.38) ng/L,(17.19 ±7.867)μg/L and ( 3.81 ±1.01 ) ng/L, respectively.The three parameters in mild preeclampsia group and severe preeclampsia group were significantly higher than those in control group ( P<0.01; F value was 79.43, 79.28 and 50.72 respectively ).In the same situation , these three indicators in severe preeclampsia group were significantly higher than those in mild group (P<0.01).Moreover, the AUC of serum PDGF-BB, sFlt-1 and U-Ⅱalone or in combination were 0.821, 0.786, 0.772 and 0.933, respectively.The differences between joint detection and three individual detections were statistically significant ( P<0.05 ) , as combined detection having a sensitivity of 95.7% and a specificity of 86.6%.Conclusion The combined detection of Serum PDGF-BB, sFlt-1 and U-Ⅱ had an important value in early assessment and treatment of preeclampsia.
9.Perioperative nutritional support in patients with liver transplantation

Objective: To investigate the effective methods of nutritional support, to improve the endurance for the operation and to promote postoperative recovery in patients undergoing liver transplantation. Methods: The perioperative nutritional condition and nutritional-support methods were reviewed in 33 patients with liver transplantation. In the first 3 postoperative days, parenteral nutrition (PN) was used in combination with infusion of human albumin and plasma. From the 4th to 5th day, enteral nutrition (EN) was used in combination with PN. Finally, the complete oral intake of food was applied. Results: Of 33 patients, there were 30 patients whose serum levels of total protein (TP) and albumin (ALB) were elevated, and glutamic-pyruvic transminase (GPT), total-bilirubin (T-BIL) and combined-bilirubin (D-BIL) were descended after nutrition support. Conclusion: The operative endurance and postoperative recovery were improved effectively by proper nutritional support in liver transplantation patients.
10.Expression of fused P-glycoprotein in E. coli DH5?

Objective To study the expression of the extracellular fragment of P glycoprotein in the bacteria. Methods According to the P glycoprotein antigen analyses and mdr1 gene structure, a couple of primers were designed to amplify the extracellular DNA fragment of the protein with PCR. The product was inserted into pGEM T vector followed by sequencing. The sequencing verified vectors were cloned into pGEX 2T to get a highly expression recombinant vector pGEX Pgp. The recombinant was transferred into E. coli DH5?, and its expression, identification and purification were studied. Results The highest level of recombinant expression was achieved at 4 h after IPTG induction. SDS PAGE analysis indicated that the expressed protein was about 30?10 3. Conclusion The target gene fragment expression at high level is established for the preparation of its antibody and biospanning of its specific peptide by using random phage library, founding a basis for the targeting treatment of multidrug resistance.
