1.Expression and significance of apoptosis-related protein Fas/FasL and BcL-2 in autoimmune thyropathy

2.Investigation and reflection about cognition and attitude of clinical nnrses for carrying out holisticnursing

Objective To investigate the attitude of clinical nurses to the practice of holistic nursing and provide basis for establishing harmonious relationships between patients and nurses.Methods primary nurses were randomly selected and underwent investigations on quality check of nursing work quarterly by serf-designed inventory.Results 24 nurses considered it necessary to carry out holistic nursing but 70 of them were reluctant to undertake it.They thought it boring because of the high requirement of holistic nursing and much literal document in practice.Conclusion Nursing services calls out seeking truth and dealing with concrete matters at present.The holistic nursing is not equipped with professional title since serious shortage of nursing staff.The emaphasis should be focused on the health education according to different situations in hospitals.Besides we should practice holistic nursing step by step instead of by the modality.
3.Effect of sodium selenite on Treg cells in peripheral blood mononuclear cells in patients with Graves disease

Objective:Recently,selenium was used for the treatment of Graves disease. However,there has been no reports that selenium affected the immune function of Graves disease at the cellular level so far. It is reported the effect of sodium selenite on Treg cells in peripheral blood mononuclear cells in patients with Graves disease in the present study. Methods:AFS were used to detect the level of serum selenium;PBMC( Peripheral blood mononuclear cells) were extracted,then,cultured with sodium selenite. Foxp3 mRNA expression was tested by Real-time fluorescence PCR,and ELISA was used to detect the secretion of IL-35. Results:The serum sodium selenite levels of GD group are significantly lower than the HC group;The expression of Foxp3 mRNA and secretion of IL-35 are both increased. Conclusion:It shows sodium selenite could improve the state of immune disorders by recovering the function of Treg cells.

5.Effect of IL-21 on Treg cells in peripheral blood mononuclear cells in pathogenesis of Graves disease

Objective:To explore the role of IL-21 in regulating the expression of peripheral blood Treg cells in the pathogenesis of Graves disease ( GD ) . Methods: Electrochemical luminescence detection was used to determine levels of thyroid function indexes and autoantibodies. Peripheral blood mononuclear cells (PBMCs) were extracted,then,cultured in the presence or absence of IL-21 in vitro. The level of IL-10 protein was measured by enzyme-linked immunosorbent assay ( ELISA) ,and expressions of Foxp3 and IL-10 mRNA were examined by Real-time PCR. Results: Compared to eGD and control group, there were significant differences in the levels of thyroid function indexes in GD group (P<0. 05),whereas there was no difference between control group and eGD group (P>0. 05). Before IL-21 stimulation,compared to eGD and control groups,expressions of Foxp3 and IL-10 mRNA and level of IL-10 protein were significantly higher in GD group ( P<0. 05 );but there were no significant differences between eGD group and control group (P>0. 05). After IL-21 stimulation,expressions of Foxp3,IL-10 mRNA and IL-10 protein levels were decreased markedly in all three groups,among which GD group showed the greatest change(P<0. 05). Conclusion:IL-21 may inhibit the differentiation of Treg cells and the production of IL-10,therefore decreasing the number of Treg cells and the ability of suppressing effector T cells ,thus should be involved in the pathogenesis of GD.
6.Bacterial Endotoxins Test of Chlorphenamine Maleate Injection

OBJECTIVE:To establish a method for the bacterial endotoxin test of chlorphenamine maleate injection. METHODS: The interference test and bacterial endotoxin test of 28 batches of chlorphenamine maleate injections from 14 manufacturers were carried out in accordance with the bacterial endotoxin test approved by Chinese Pharmacopeia (2005 edition). RESULTS: Diluted to a concentration of below 0.025 mg?mL-1,the samples showed no interference action on bacterial endotoxin test. The limit value of the bacterial endotoxin was 5 EU?mg-1. The results of the bacterial endotoxin test of the 28 batches of samples were all accorded with the standard of the established quality control. CONCLUSION: The established TAL method is applicable for bacterial endotoxin test of chlorphenamine maleate injection.
7.Detection of Lead Ions in Water Based on Surface Energy Transfer between Gold Nanoparticles and Fluorescenct Dyes

We established a sensitive method for the detection of lead ions in aqueous solution based on the surface energy transfer between gold nanoparticles and fluorescent dyes. The fluorescein-modified thrombin aptamer (5′-FAM-GGT TGG TGT GGT TGG-3′) can be selectively transformed to G-quadruplex induced by lead ions. The conformation changes of the aptamer can alter the distance between the energy donor of fluorescent dyes and the energy receptor of gold nanoparticles, and further enhance the fluorescence intensity. The fluorescence recovery efficiency ( F/F0 ) is proportional to the concentration of lead ions in the range of 12. 5-100 nmol/L following the linear regression equation of y=0. 910﹢0. 007c(R2=0. 997)and the limit of determination is about 10 nmol/L. The proposed method has been applied to the determination of lead ions in tap water with satisfied results.
8.Cytotoxicity evaluation of nano-Ag-SiO2urethral catheter for external urethral mucosa cells

BACKGROUND:Nano-Ag is increasingly used as antibacterial materials in medical products due to its excelent antibacterial activity. Antibacterial activated nano-Ag-SiO2 OBJECTIVE: To evaluate the biosecurity of nano-Ag-SiO urethral catheter can reduce the incidence of catheter-associated urinarytract infections obviously. As more and more nano-Ag medical products have been developed, their biological security is becoming a focused issue. 2 urethral catheter through comparing the active influence of nano-Ag-SiO2 METHODS: Mechanical separation and enzymatic digestion were used to isolate and culture urothelial mucosa of benign prostatic hyperplasia patients to make cellsuspension after proliferationin vitro. The medium was replaced by nano-Ag-SiO urethral catheter and ordinary urethral catheter extracts onin vitro cels of human prostatic urethra epithelial cels in different periods. 2 and general catheter tube extracts, respectively. MTT assay was performed to quantify thein vitro cytotoxicity of two extracts on human urethra epithelial cels which were cultured for 2, 5 and 7 days. Absorbance values of nano-Ag-SiO2 RESULTS AND CONCLUSION:The cytotoxicity of extract of nano-Ag-SiO and general urinary catheter groups were measured by enzyme-linked immunosorbent assay to calculate the relative growth rate, and toxicity evaluation was compared and evaluated. Under an inverted microscope, cellmorphology, growth and proliferation were observed. 2 catheter was 1 grade, and that of general medical catheter extract was 0 to 1 grade. There was no statistical difference in group effect of relative proliferation rate (F=0.544,P=0.475); there was statistical difference in time effect of relative proliferation rate (F=3.031,P=0.086); no statistical difference was found in the interaction between group and time (F=0.130, P=0.879). These findings indicate that nano-Ag-SiO2 catheter has no or low cytotoxicity effect on cellgrowth and proliferation, which is consistent with the biological evaluation standards of medical devices.

The co-metabolic carbon sources,dynamics and influence factors of atrazine biotransformation by genetically engineered microor- ganism(GEM)were investigated in the paper.The results showed that glucose was superior to acetate as co-metabolic carbon sources.The concentration of carbon sources affected little on atrazine transformation but much on GEM growth.The specific transformation rate(STR)of atrazine was influenced not by cell density of GEM but by atrazine concentration.Monod model was used to simulate atrazine transformation dynamics and the parameters were v_(max)=0.168/h and Ks=30.49 mg/L.When temperature reduced,STR decreased markedly.The trans- formation was efficient in weak alkaline condition and was inhibited in acidic condition.To some extent,salinity did not affect transformation activity of GEM.Co~(2+),Fe~(2+),Fe~(3+),and Zn~(2+) improved atrazine transformation,but Mn~(2+),Ni~(2+) and Cu~(2+) inhibited atrazine transforma- tion.The results also showed that there existed a direct correlation between adsorption and transformation of atrazine by GEM.
