Objective To investigate the correlation between p21-actived kinase 1(PAK1) gene polymorphism and prognosis of endocrine therapy for breast cancer. Methods Polymerase chain reaction-restriction fragment length polymorphism was employed to analyze the genotype of PAK1 gene rs2298793 T→C polymorphism and rs2844327 C→T polymorphism of 105 patients with breast cancer accepting endocrine therapy. Prognostic factors such as age, state of menses, magnitude of tumor, pathological type, condition of operation, clinical stage, estrogen receptor, progesteron receptor, Her-2 and endocrine therapy drug were comprehensively considered, and Cox model was employed to analyze the relation of PAK1 genotype with the time-to-tumor progression (TTP). AIM: To compare the peripapillary retinal nerve fiber layer ( RNFL ) thickness and peripapillary topographic map's parameters between amblyopic eyes and non -amblyopic eyes in adolescent with anisometropic amblyopia by optical coherence tomography(OCT).
METHODS: Thirty - four juveniles with anisometropic amblyopia were selected. Peripapillary RNFL thickness and peripapillary topographic map were measured by frequency domain OCT with both eyes in all participants, and the differences between amblyopic eyes and non -amblyopic eyes were compared.
RESULTS: There was no significant difference in average thickness of peripapillary RNFL and in any other region of peripapillary RNFL. The disc area in amblyopic eyes was bigger than that in non - amblyopic eyes ( t =2. 8054,P= 0. 0263). The disc area in amblyopic eyes were significantly related to the thickness of nasal RNFL and the rim area(r= 0. 7592,0. 7501;P= 0. 0289,0. 0321).
CONCLUSION: There existed some difference in peripapillary structure between amblyopic eyes and non-amblyopic eyes in adolescent with anisometropic amblyopia.

6.Effects of imatinib mesylate on rat hepatic fibrogenesis and the expression of transforming growth factor-?1

Objective To investigate the anti-fibrogenesis property of imatinib mesylate in a rat model of liver fibrosis induced by carbon tetrachloride/olive oil and its effect on the expression of trans- forming growth factor(TGF)-?1.Methods Rat liver fibrosis was induced by intraperitoneal administra- tion of carbon tetrachloride and olive oil mixture twice a week for eight weeks.Imatinib mesylate was given 20 mg/kg daily by oral lavage.The control rats received saline by oral iavage.Liver collagen depo- sition was evaluated by immunohistochemistry with Masson staining.The activation of hepatic stellate cells was detemined by the immunohistoehemistry staining of?-smooth muscle actin.The mRNA expres- sions of TGF-?1,c-Abl and TIMP-1 were measured by RT-PCR.While protein expressions of TGF-?1, phosphorylated platelet-derived growth factor receptor and c-Abl were detected by Western blot and im- munohistochemical staining.Hepatic hydroxyproline content was also quantified.Results The collagen deposition[(16.23?1.01)%vs(25.61?0.92)%]and the number of activated HSCs(10.52?1.33vs 13.10?1.21)were reduced in the imatinib mesylate treatment group compared with the control group by 35% and 20%,respectively(P

10.Baicalein induced Nrf2 activation and its protection against hepatotoxicity

Aim To study the activation of Nrf2 in-duced by baicalein ( BAI ) , and its protection against carbon tetrachloride ( CCl4 ) , ethanol and acetamino-phen ( APAP )-induced hepatotoxicity. Methods A reporter gene assay was conducted in human normal liver L-02 cells to detect the activation of transcription factor Nrf2 induced by baicalein. APAP ( 10 mmol · L-1 ) , CCl4 (10 mmol·L-1 ) and Ethanol (100 mmol · L-1 ) were used to induce hepatotoxicity in L-02 cells. After the pre-incubation with Baicalein (0, 1, 10, 25, 50, 100 μmol·L-1 ) for 15 min, cells were administrated with or without those above hepatotoxins. 48 h later, cell viability was detected by 3-(4, 5-dim-ethylthiazol-2-yl ) 2 , 5-diphenyltetrazolium bromide (MTT) method. Results Baicalein (25, 50 μmol· L-1 ) induced the activation of Nrf2 ( P <0. 01 , P <0. 05) in the reporter gene assay. As compared with control, three hepatotoxins ( APAP, CCl4 , Ethanol ) all decreased cell viability ( P<0. 01 ) , and baicalein significantly reversed such decreases in a concentra-tion-dependent manner ( P<0. 01 ) . Conclusion Ba-icalein can induce the activation of transcription factor Nrf2 , which is probably one of the mechanisms con-tributing to the protection of baicalein against hepato-toxins (APAP, CCl4, Ethanol)-induced hepatotoxici-ty.
