1. Preparation of X-ray developable alginate-barium sulfate microspheres as material for embolic agent

Objective: To prepare an X-ray developable alginate-barium sulfate microspheres as material for embolic agent. Methods: The emulsification-internal gelation technique was used to prepare alginate microspheres and the orthogonal design was adopted to optimize the technique. The diameter, shape, dispersibility, stability, suspension property, coating ratio and developability under X-ray of the microspheres were examined. Results: The average diameter of the microspheres was (53. 08± 32. 72) μm; they were well distributed and had a round shape. The ratio of the destroyed microspheres was (2. 0±1, 1) % after heated at 100°C for 2 h,(86. 0± 19. 2)% after frozen at - 4°C for 24 h, (39. 0± 14. 7)% after vibrated at 37°C for 24 h, and (10. 3±3. 2)% after irradiated with 60Co(10kGy) for 0. 5 h. The ratio of sedimentation volume in 0. 25 % suspension of sodium alginate microspheres was (0. 92±0. 018). The coating rate was (69. 2±13.2) % when the feeding amount of barium sulfate was 2. 0 g, and the result of X-ray development was good. Conclusion: Alginate-chitosan can be used for the preparation of barium sulfate microspheres; and the emulsification-internal gelation technique can prepare the microspheres with good dispersibility and shape.

This study was aimed to investigate active ingredients from A mp e lop s is me galop hylla of protein expression by using two-dimensional gel electrophoresis (2-DE). Methods: isn this study, proteinsfrom the tender leaves of A mpelopsis megalophylla were precipitated by Tris-phenol extraction and ammonium acetate methanol precipitation. Then using 2-DE technology to isolate protein.Finally, 2-DE was analyzed by the scanner, and got protein characteristic maps. Result:According to the differences in ratio of gray value, the seven differential protein points were chosen to identify the mass spectrum. At present, two proteins were identified, namely the hypothetical protein and the unnamed protein product. Conclusion: the application of 2-DE technology for A mpelopsis megalophyllathe active ingredient of protein expression at the level of molecular research laid a good foundation.
3.Changes in serum concentrations of angiogenic factors in patients with unstable angina pectoris

Objective To detect the changes in serum concentrations of angiogenic factors in patients with unstable angina pectoris.Methods A total of 57 patients with unstable angina pectoris were eligible for study and divided into diabetes group (n =23) and non-diabetes group (n =34).Exclusion criteria included complicated diseases,symptomatic peripheral vascular diseases,renal or liver diseases,cerebral embolism and evidence of ongoing infection.Another 36 age-matched healthy subjects from medical examination center were enrolled as control group.Serum samples were collected,and serum concentrations of vascular endothelial growth factor (VEGF),angiopoietin-1,angiopoietin-2,angiogenin,angiostatin,basic fibroblast growth factor (bFGF) and platelet-derived growth factor-BB (PDGF-BB) were measured by using cytokine array technology.The data were analyzed by independent-samples t test with the SPSS 13.0statistic software package.Results Compared with the control group,the serum concentrations of VEGF and angiopoietin-2 were significantly increased in patients with unstable angina pectoris (both P < 0.01).Whereas,no significant differences in serum concentrations of angiogenin,angiopoietin-1,angiostatin,bFGF,and PDGF-BB were detected between control group and patient groups.There were no significant differences in serum concentrations of above all 7 biomarkers between diabetes group and non-diabetes group.Conclusions Serum concentrations of VEGF and angiopoietin-2 were increased in patients with unstable angina pectoris,and diabetes didn' t affect the increases in serum concentrations of VEGF and angiopoietin2 caused by unstable angina pectoris.
4.Changes in serum concentrations of angiogenic factors in patients with type 2 diabetes mellitus

Forty-two patients with type 2 diabetes (DM group),23 type 2 diabetic patients with unstable angina pectoris (DM + UAP group),and 36 healthy subjects (control group) were enrolled in this study.Serum samples were collected for determining serum concentrations of vascular endothelial growth factor (VEGF),Angiopoietin (Ang)-1,Ang-2,angiogenin,angiostatin,basic fibroblast growth factor,and platelet-derived growth factor-BB using protein array technology.The results showed that there were no significant differences in serum concentrations of all 7 angiogenic factors between control group and DM group.Whereas,serum concentrations of V EGF and Ang-2 were significantly increased in DM + UAP group compared with control group [(3 532.10 ± 1 813.72vs 2 444.50 ± 1 152.21) pg/ml,(286.90-± 217.01 vs 171.92 ± 106.63) pg/ml,both P<0.01].Pearson's correlation analysis revealed that serum concentrations of all 7 angiogenic factors were not related to HbA1C,fasting and 2 h postprandial plasma glucose,triglycerides,high density lipoprotein cholesterol,as well as low density lipoprotein cholesterol.
5.Extraction and Separation of Carotenoids of Hydrogen-Producing Rhodobacter sp.

Carotenoids play an important role in regulating the hydrogen production of hydrogen-producing Rhodobacter sp. The carotenoids of hydrogen-producing Rhodobacter sp. grown in acetate medium were extracted by using acetone-methanol (7∶2,V/V) solvent and were separated by using thin-layer chromatography on silica-gel plate. The qualitative and quantitative of the carotenoids were analyzed by spectrometry. The results showed that the carotenoids were completely extracted three times with acetone-methanol (7∶2,V/V) in two hours. The ultrasonication had little effect on yield of carotenoids. The yield of carotenoids was 2.81mg/g wet cell. There were 4 spots on the silica-gel plate in the order of yellow, red, light red and light yellow. Yellow spot and red spot were the dominant composition of carotenoid in Rhodobacter sp. The spectrometry data showed that the yellow and red component might be the spheroidene and spirilloanthin respectively.

7.Correlation between ultrasonic characteristics of carotid artery plagues and risk factors of diabetes mellitus type 2

Objective To evaluate the relationship between carotid artery plagues and the clinical risk factors in patients with diabetes mellitus type 2 by observing the ultrasonic characteristics of carotid atherosclerosis plagues.Methods The ultrasound results of carotid arteries in 665 patients with diabetes mellitus type 2 from January 2009 to December 2011 in Beijing Amery General Hospital were retrospectively analyzed.The location and quantity of carotid atherosclerotic plaques were observed .The relationship between the occurrence and risk factors of diabetes such as hypertension ,family history of diabetes and age were analyzed.Single factor and multi-factor analysis were applied for the related risk factors .Results The ultrasonography showed 405 cases of carotid artery plaques.Most carotid artery plaques were located in the posterior wall of carotid sinus and were hyperechoic ,hypoechoic or isoechoic.The single factor analysis showed hypertension,family history of diabetes and age are risk factors of carotid atherosclerosis (χ2 =42.322,9.682 and 140.658,all P<0.01).Multi-factor logistic regression analysis showed age was an independent risk factor of carotid atherosclerosis in patients with diabetes mellitus type 2.The risk factors scores of patients who were less than 45 years,45-65 years and over 65 years of age were (3.50 ±1.16)scores,(3.46 ±0. 92)scores and (3.21 ±0.88)scores,and the difference was statistically significant (F=5.781,P<0.001). There were significant differences between risk factors scores of different age groups ( t =2.084,2.002, 3.786,3.474,2.877 and 2.504,all P<0.05).In patients more than 65 years of age,the risk of atherosclerosis plaques is 28.732 times higher than that of patients less than 45 years of age.After controlling the age factor,the possibility of atherosclerosis plaque increased 1.201 times for each additional one point of score. Conclusions Most carotid artery plaques in patients with diabetes mellitus type 2 are located in the posterior wall of carotid sinus and are hyperechoic ,hypoechoic or isoechoic.Age is an independent risk factor for the formation of carotid plaque.Patients more than 45 years of age should undergo early intervention to avoid the occurrence of major vascular complications .
8.Expression of angiogenic factors in myocardial tissue of diabetic rats

AIM:To observe the expression of angiogenesis factors in the myocardial tissue of streptozotocin-induced diabetic rats .METHODS:The diabetic rat model was induced by intraperitoneal injection of streptozotocin .After 12 weeks, the cardiac function was measured by MPA cardiac function analysis system .The myocardial collagen volume fraction ( CVF) was assessed by Masson staining .The capillary vessels was quantified as the ratio of capillary to myocyte (C/M) using CD31 immunostaining.The expression levels of vascular endothelial growth factor (VEGF), angiopoietin ( Ang)-1, endostatin and Ang-2 were observed by Western blotting .RESULTS:Compared with normal control group , the left ventricular end-diastolic pressure (LVEDP) was evidently increased (P<0.01), but left ventricular pressure rise maximum rate (+dp/dtmax), left ventricular pressure decrease maximum rate (-dp/dtmax) and the ratio of capillary/myo-cyte (C/M) were significantly decreased (P<0.05).The CVF and the expression level of endostatin were significantly increased, whereas the expression levels of VEGF and Ang-1 evidently decreased (both P<0.05) in diabetic rats.Howev-er, no marked difference in the expression of Ang-2 between the 2 groups was observed (P>0.05).CONCLUSION:Im-balances between the angiogenic factors (VEGF and Ang-1) and anti-angiogenic factors (endostatin) may play an impor-tant role in the pathogenesis of diabetic cardiomyopathy .
9.Construction of adenovirus-mediated anti-sense RNA targeting K-ras exon 1 in SW1990 cell line

Objective:To construct an adenovirus-mediated anti-sense RNA targeting K-ras exon 1 of SW1990 cell line and observe its effect on ceil proliferation and apoptosis after transferred into SW1990 cell line.Methods:K-ras exon 1 cDNA was cloned into shuttle vector pShuttle-CMV and the resultant plasmid was confirmed by enzyme digestion and PCR.Clones with inverted insertion were selected and co-transferred into E.coli BJ5183 with an adenoviral backbone plasmid pAdEasy-1 to produce recombinant plasmid by homologous recombination.Recombinants were then selected and transfected into 293 cell line to produce recombinant adenovirus.Recombinant adenovirus production was confirmed by PCR analysis and was amplified and purified;the virus titer was determined.Ad-LacZ was used to infect SW1990 cells and the infection efficiency was observed by X-gal staining.SW1990 cells was infected with the recombinant adenovirus and their proliferation and apoptosis were determined by MTT and annexin V/PI FCM assay.Results:A 282 bp target gene fragment was acquired by PCR;the titer of recombinant adenovirus was 7.6?10~8 pfu/ml before purification by CsCl_2 gradient centrifugation and 5.0?10~(10)pfu/ml after CsCl_2 gradient centrifugation.When the recombinant adenovirus was at 100 MOI,the infection efficiency of SW1990 cells nearly reached 100%.The transfection of recombinant adenovirus significantly inhibited SW1990 cell proliferation(P
10.Advances of synthetic biology of flavonoids

Flavonoids is one of the biggest families of the plant-derived secondary metabolites with structural diversity. Until now, over 10 000 kinds of flavonoids with distinct structures have been purified and identified from plants, and some of them possess a range of important pharmacological effects, such as anticancer, anti-inflammatory and so on. So far, a number of genes and enzymes responsible for the biosynthesis of flavonoids have been reported, especially, a great of progress has been achieved in the synthetic biology of flavonoids in the recent years. Herein, based upon a brief introduction on the biosynthesis of flavonoids, this review summarizes the research advances in synthetic biology of flavonoids in the past two decades (2001-2021), highlighting the cell factories construction of the representative flavonoids. And, a brief discussion and prospects of the relevant metabolic bottlenecks and optimizing strategies are proposed.
