1.The curative effect of recombinant human interferon α-2b gel on flooding prevention in loop electrosurgical excision procedure

Objective To evaluate the efficacy of recombinant human interferon α-2b gel onflooding prevention in loop electrosurgical excision procedure (LEEP) during and after the operation.Method The 152 patients with cervical disease were randomly divided into three groups: group A(recombinant human interferon α-2b gel treatment group), group B (hypophysin treatment group)and groupC(control group). Results The amount of intraoperative bleeding and the amount of post-operative bleedingof the group A was (2.57±1.19) ml and (2.24±1.75) ml, the group B was (3.00±1.49) ml and (45.86±26.14) hal, the group C was (45.62±39.57) ml and (56.90±41.90) ml, respectively. There was no differencebetween group A and group B in the amount of intraoperative bleeding (P>0.05), but significantly less thangroup C (P<0.01). There was no difference between group B and group C in the amount of post-operativebleeding (P>0.05), but significantly much more than group A (P<0.01). Conclusion Recombinanthuman interferon α-2b gel is effective on reducing intraoperative and post-operative bleeding in LEEP.

4.Study on inhibitory effect of EGCG on Calcium oxalate nephrolithiasis in rats and its related mechanism.

In the study, the inhibitory effect of epigallocatechin gallate (EGCG) on Calcium oxalate nephrolithiasis and its possible mechanism were investigated. The rat Calcium oxalate nephrolithiasis model was induced through the combined oral administration of ethylene glycol and ammonium chloride, which was intervened with EGCG. Rat blood samples were collected to detect blood creatinine (Cr), blood urea nitrogen (BUN) and blood calcium. Rat urine samples were collected to observe and compare 24-hour urine volume, oxalic acid (Ox) and calcium in urine. Renal samples were collected to prepare tissue slices and observe the pathological changes in Calcium oxalate nephrolithiasis. The expression of osteopontin (OPN) in renal tissues was evaluated by Real-time PCR and Western blot. According to the results, compared with normal rats, rats in the nephrolithiasis model showed significant increases in Cr, BUN, urine Calcium, urine Ox and renal OPN expression (P < 0.05), but obvious decrease in 24-hour urine volume (P < 0.05); Compared with rats with nephrolithiasis, those processed with EGCG revealed remarkable declines in Cr, BUN, urine Calcium and urine Ox (P < 0.05), with significant rise in 24-hour urine volume (P < 0.05) in a concentration-dependent manner. Additionally, compared with the control group, nephrolithiasis rats showed significant pathological changes in Calcium oxalate calculus. After ECCG treatment, the renal pathological changes and OPN expression attenuated significantly in a concentration-dependent manner. The results showed that EGCG inhibits the formation of calcium oxalate nephrolithiasis in rats and shows a notable protective effect on renal functions.

5.Treatment of murine hypoparathyroidism with transgenetic hemopoietic stem cell transplantation

Objective To investigate the gene therapy of hypoparathyroidism (HPT) with hemopoietic stem cell s(HSCs) integrated with parathyroid hormone (PTH) gene.Methods Human PTH gene was amplified by polymerase chain reaction from pcDNA 3.1 -PTH templates and inserted into murine stem cell virus (MSCV) vectors. The plasmids recombinated with PTH gene were transfected into PA317 packaging cells and screened for positive clones with Geneticin. The condensed recombinant retroviruses were obtained and used to infect HSCs coming from umbilical cord blood. The infected HSCs were injected into the mice suffering from HPT. The change of symptoms and serum level of PTH and calcium in each group of mice was observed.Results Human PTH gene was proved to be inserted into MSCV vectors and the titres were up to 2?10 7 colony forming unit (CFU)/ml in condensed retroviral solution. The secretion of PTH reached 15 ng per 10 6 cells per 48 h. The wild type viruses were not detected by PCR amplification. The mice suffering from HPT recovered quickly and the serum level of calcium and PTH remained normal for about 3 months after the HSCs recombinated with PTH gene were injected into them. In the groups of mice subject to the injection of the recombinant retroviruses containing PTH gene, the serum level of PTH and Calcium was significantly increased in a short time, and then declined slowly. And the mice took on the manifestation of hypoparathyroidism again. The mice only subject to the implantation of HSCs were all dead within 20 days.Conclusion A long term therapeutic effect could be obtained by infusing the HSCs which were integrated with PTH gene into the mice suffered from hypoparathyroidism.
6.Histological study on remodeling and maturation of hamstring autograft in postoperative patients

Objective To probe into the remodeling and maturity process along with extension of time in anterior cruciate ligament (ACL) reconstruction with hamstring autograft in order to confirm the date of maturity postoperatively. Methods Thirty-three patients after arthroscopic ACL reconstruction with hamstring autograft, were undergone second-look arthroscopical surgery. Meantime, biopsy specimens were obtained from the mid-zone of the hamstring graft. As a control group, specimens of normal ACL were obtained from total knee replacement of four cases, and specimens of semitendinous tendons obtained from ACL reconstruction of the four cases. The interval from initial reconstruction to second-look arthroscopy ranged from 2 to 36 months(mean 11.9 months). Patients were divided into different groups by postoperative time such as 1-, 4-, 7-, 10-, 13-, 18- and 25- month group. Thirty-three specimens were ordinarily sliced up into longitudinal sections. The sections were stained with hematoxylin and eosin so that cell and collagen fibrils were observed under light microscope, then compared the results with those of control group. Results Under microscopical observation, it was showed that tissue of hamstring graft was remodeled from more blood vessels and cellular amount to normal one, and from an irregular orientated crimp pattern of collagen to regular orientated crimp pattern of collagen in a time-dependent manner. Decreasing cellular amount, changing cell type, and a regular orientated crimp pattern of collagen of the hamstring graft were observed in 7- month group primarily, comparing with the original ACL. However, there existed a slower process in the other groups that are older than that of 7- month. Conclusion There are well histological features in the process of remodeling and maturation of the hamstring autograft after operation following extension of time, such as better survival of early period, and remodel of vessels and collagen. The process of remodeling and maturation in the hamstring autograft is similar to BPB autograft. The maturation period of the hamstring autograft after ACL reconstruction in human patients appears between 7 month and 9 month postoperatively.
7.Effects of Microwave Radiation Exposure of Pregnant Mice on Offspring

Objective To study the relationship between microwave radiation exposure during pregnancy and the offspring health. Methods The pregnant Kunming mice were divided into 3 groups and exposed to microwave radiation of 0, 570 and 1 400 ?W/cm2 respectively. The bone marrow cells of the offspring were collected for the determination of chromosome aberration (CA), sister chromatid exchange (SCE) and micronucleus frequency (MNF). Results SCE showed a significant change in high intensity group, as for CA and MNF, no significant change was seen compared with the control. Conclusion Microwave radiation exposure during pregnancy may induce germ plasm damages in the offspring.
8.Arthroscopic reshaping for the treatment of discoid meniscus

Objective To explore the therapeutic method and efficacy of discoid meniscus urder arthroscope. Methods 37 patients with 37 discoid menisci underwent arthroscopic lateral partial or total meniscectomy. 33 cases were operated on by reshaping (partial meniscectomy) and 4 cases with extensive laceration in the joint capsule rim of hastring tendon underwent total meniscectomy under arthroscope. Results Bases on Ikeuchi's grading, 19 cases were excellent (51.4%), 13 cases good (35.1%) and 5 cases fair(13.5%). Conclusions Arthroscopic reshaping for discoid meniscus can obtain excellent and good efficacy, so it is recommended that patients with discoid menisci should be treated by reshaping.
9.Clinical research on the arthroscopic hamstring tendon autograft for reconstruction of the anterior cruciate ligament.

Objective To explore the surgical technique and clinical effect of arthroscopic hamstring tendon autograft on reconstruction anterior cruciate ligament (ACL). Methods 22 patients underwent ACL reconstruction with hamstring tendon autograft under arthroscopy. The mean age was 30 7 years (17years~50 years). Results The motion range of knee joint of all cases was normal or nearly normal. The postoperative Lachman test was ≤1+ in 19 patients, 2+ in 2 patients and 3+ in 1 patient. 20 patients also showed an absent pivot shift.
