Objective To investigate the effects of cyclooxygenase-2 (COX-2) on the expressions of vascular endothelial growth factor (VEGF) and prostaglandin E_2 (PGE_2) in pancreatic carcinoma both in vitro and in vivo, and to clarify the possible mechanism of PGE_2 in mediating COX-2 inducing angiogenesis of pancreatic carcinoma. Methods In vitro study, the inhibitory effects of Celebrex, a selective cyclooxygenase-2 inhibitor, on the expression of VEGF and PGE_2 in pancreatic carcinoma cell lines PC-3 were determined using either enzyme-linked immuno-absorbent assay (ELISA) or radioimmunoassay (RIA). Effect of exogenous PGE_2 on the down-regulation of VEGF by Celebrex was also assessed. In vivo study, PC-3 cell line xenograft nude mice model was established. Changes of VEGF expression and PGE_2 of tumor tissues after the treatment of Celebrex were investigated using Western blotting or RIA. Results Celebrex suppressed the expressions of VEGF and PGE_2 in cultured PC-3 cell line with a manner of dose- and time-dependence. Exogenous PGE_2 up-regulated the expression of VEGF, which was suppressed by Celebrex in a dose-dependent fashion. In vivo study, administration of Celebrex into xenograft nude mice inhibited expressions of VEGF and PGE_2 significantly. Conclusion COX-2 is involved in angiogenesis in pancreatic carcinoma probably through the inhibition of the production of angiogenic factors such as VEGF. PGE_2 is likely to act as an important mediator in this process.

AIM: To investigate the effects of the dominant-negative I?B? plasmid on the expression of NF-?B and cyclooxygenase-2 (COX-2) after its being transfected into pancreatic carcinoma PC-3 cell line. METHODS: The expression of NF-?B and COX-2 in PC-3 cell line was confirmed by immunohistochemistry. The effects of dominant-negative I?B? plasmid transfection on the expression of NF-?B and COX-2 were studied by reverse transcription polymerase chain reaction (RT-PCR) and Western blotting. RESULTS: Both NF-?B and COX-2 were expressed in pancreatic carcinoma PC-3 cell line, and the expression of NF-?B and COX-2 were down-regulated in a certain time-independent manner after dominant-negative I?B? plasmid transfection. CONCLUSIONS: The NF-?B and COX-2 are expressed in pancreatic carcinoma PC-3 cell lines. The expression of NF-?B and COX-2 are inhibited by dominant-negative I?B? plasmid, while NF-?B is likely to play an important role in regulating the expression of COX-2.

Objective To study the performance of children rhabdomyosarcoma (RMS) in CT examation. Methods Twenty-four cases clinical and CT data of RMS which were confirmed by pathology were analyzed retrospectively. Results In head and neck RMS (3 cases), the performance were well-circumscribed soft tissue with uniformity density which were obviously improved after enhancement and showed no invasion to the adjacent bone. In trunk limbs RMS (8 cases), the performance were well-circumscribed soft tissue of many different sizes with uniformity density and mild to moderate uniform(or non-uniform) enhancemen, and the invasion to the adjacent bone was not obvious. In peritoneum and retroperitoneum RMS (6 cases), 5 cases showed non-uniformity isodensity or slightly low density with non-uniformly enhancement and without retroperitoneal lymph node metastasis. The mesenteric (1 case) showed low uniformity density mass and non-uniformly bar enhancement. In urogenital system RMS (7 cases), 2 cases located in the bladder, and 4 cases respectively 1ocated in the vagina, scrotum, testicles and perineum. The performances were uniformity density soft tissue mass like nodular with moderate uniform enhancement. One case located in posterior urethral, whose performance was cystic and solid mass. The cystic part had no enhancement, and the solid part had obviously uniform enhancement. Conclusion The characteristic performance about children RMS is different along with the different parts and age in CT, which has important value for early diagnosis.

Objective To investigate the biological effects of 12C heavy ions with different LETs,and to construct the dose-effect curves of chromosomal aberrations and micronucleus. Methods Human peripheral blood lymphocytes were irradiated with 0.5 - 5.0 Gy of 60Co γ-rays,29 and 148 keV/μm 12C with a dose rate of 0.5 Gy/min,respectively.The colchicine method and cytokinesis-blocking method were applied to measure chromosome aberrations and micronuclei formation, respectively. Results The chromosomal aberrations induced by 29 and 148 keV/μm of 12C ion had a linear dose response, while the dose-response of micronucleus induction followed to a linear-quadratic model.Chromosome aberration and micronucleus frequency increased with the LET of 12C ions. However, the micronucleus frequency increased slowly when the radiation doses exceeded 3.0 Gy..Chromosomal aberrations induced by 148 keV/μm 12C ions increased significantly when the time of colchicine treatment was prolonged. Conclusions Chromosome aberration and micronucleus induced by 12C ions increased with the LET,and the micronucleus frequency increased with dose less than 3 Gy.The biological effects of 12C ions were stronger than those of low LET irradiation of γ-rays.

Objective To establish the dose-effect curves of chromosome aberrations in human peripheral blood lymphocytes induced by different LET rays, including 60Co γ-rays,252Cf neutrons,14MeV neutrons and 12C heavy ions at low doses, respectively,in order to assess radiation-induced damages after occupational and accidental exposure. Methods Heparinized whole blood samples were irradiated with the various radiation devices and doses ranged from 0 to 1 Gy with the interval of 0.25 Gy.Conventional chromosome culture method was applied with adding colchicines at the beginning and chromosome specimens were prepared after 48 h incubation. The Metafer scanning system was used for automatical finder of chromosome metaphases. The mathematic models were fitted according to aberration data obtained. Results At the dose range of 0 - 1 Gy,the math models of dic + r were linear for 60 Co γrays and linear-quadratics for 252 Cf neutrons,14 MeV neutrons and 12 C heavy ions. The models of ace were linear-quadratic for 60Co γ-rays and 12C heavy ion beams, and linear for 252Cf and 14 MeV neutrons. The models of total aberrations were linear-quadratic for all types of radiation. Conclusions High LET rays have higher biological effects in inducing chromosome aberrations yields compared with low LET rays. Moreover, the severity of damage is 252Cf > 14 MeV neutrons > 12C heavy ions > 60Co γ-rays in turn. Therefore,in the range of low doses, the dic + r may be a better target of radiation damage for high LET radiation.

Objective To discuss the therapeutic effect of transcatheter arterial embolization using hardening agent combined with oral propranolol in treating giant hemangioma at maxillofacial region in infants. Methods During the period from October 2013 to December 2014 at Imaging Center of Anhui Provincial Children's Hospital, transcatheter arterial embolization using hardening agent combined with oral propranolol was employed in a total of 27 infants with giant hemangioma at maxillofacial region. The age of the infants ranged from 2 months to 22 months (mean 5.9 months) and the body weight was 4.5-10 kg with a mean of 6.32 kg. Angiography via femoral artery was performed, which was followed by super-selective catheterization of hemangioma-feeding artery, and then pingyangmycin lipiodol emulsion was injected into the hemangiomas with subsequent injection of PVA particles to obstruct the hemangioma-feeding artery. After the embolization treatment, the patient received oral propranolol for 3-6 months. Results All the infant patients were followed up for 3-6 months. Clinical examination and ultrasonography indicated that the hemangioma was cured in 20 infants (75%) and the therapeutic result was effective in 7 infants (25%). Skin necrosis at hemangiomas site was observed in 2 infants (7.5%), which was cured after symptomatic treatment. No serious complications such as pulmonary embolism, cerebral embolism occurred, and no recurrence was observed. Conclusion For the treatment of giant hemangioma at maxillofacial region in infants, transcatheter arterial embolization using hardening agent combined with oral propranolol is minimally invasive, quickly effective and highly safe;and this treatment leaves no scar formation in most cases. Therefore, this technique is worthy of clinical application.
