1.Expression of cholecystokinin receptor B in a variety of human tumor tissues and cells

Objective To investigate the expression of cholecystokinin receptor B(CCK-BR)in a variety of human tumor tissues and cells.Methods RT-PCR and Western Blot were used to detect the expression of CCK-BR mRNA and protein in human gastric cancer AGS and SGC-7901cells,human hepatoma cancer HepG-2 cells,human neuroblastoma SH-SY5Y cells and human lung canc-er A549 and NCI-H446 cells,human prostate cancer DU145 cells,human bladder cancer T24 cells.CCK-BR expression was detected in the clinical cancer tissues including the stomach,breast,cervical,bladder,lung and liver and AGS and SGC-7901 cells by immuno-histochemical and immunocytochemical methods.Results CCK-BR mRNA and protein were express in the cytoplasm and cell mem-brane of the above 8 cell lines.Positive expression of CCK-BR was discovered in the above cancer tissues.Conclusion CCK-BR are expressed in a variety of human tumor tissues and cells.
2.Minimally invasive treatment of extrahepatic bile duct stones

With the continuous improvement of endoscopic technology and equipment,minimally invasive treatment of bile duct stones be-comes increasingly diversified.In this paper,the concept of minimally invasive treatment of bile duct stones,surgical indications and contra-indications,surgical methods,and the advantages and disadvantages of surgical methods are introduced.It is shown that the application of laparoscope has laid the foundation for the minimally invasive treatment of bile duct stones,and the combined application of choledochoscope and endoscope has brought new ideas for minimally invasive treatment of bile duct stones.The combination of three endoscopes avoids the disadvantages of traditional open surgery:large trauma and long operation time,and reduces the patient′s pain and complications.In the near future,the combination of three endoscopes still has much room for growth.As long as the indications for several surgeries are mastered and the safety and efficacy are evaluated objectively,the combination of three endoscopes will play the biggest role.
3.Cytokeratin 19 and pancreas stem cells

BACKGROUND: To synthetically analyze the correlation between cytokeratin 19 and pancreas stem cells.DATA SOURCES:References about the correlation between cytokeratin 19 and pancreas stem cells were retrieved in Medline and Ovid database with the key words of "cytokeratin, pancreas, stem cells, marker, differentiation" in English from January 2000 to May 2006.STUDY SELECTION:Literatures about the correlation between cytokeratin 19 and pancreas stem cells were retrieved and those characterized by strong pertinency, published in the near future and selected from authoritative journals were selected in this paper.DATA EXTRACTION: Among 86 articles, 35 of them met the inclusion criteria and 51 were excluded.DATA SYNTHESIS:Pancreas stem cells are characterized by multi-directional differentiation.Cytokeratin 19 has a positive expression in pancreas stem cells and a molecular sign of pancreas stem cells. This provides a new way to further understand bionomics of pancreas stem cells,study separation,purification,transconformation and direction differentiation to islet cells, and treat diabetes mellitus with transplantation of pancreas stem cells.CONCLUSION: Expression of cytokeratin 19 is observed during the differentiation of pancreas stem cells and cytokeratin19 is a sign of pancreas stem cells.
4.PPARγ and the invasion and metastasis of hepatocellular carcinoma

Recent studies indicate that the activation of peroxisome proliferator-activated receptor gamma (PPARγ) can inhibit the invasion and metastasis of hepatocellular carcinoma (HCC) by inhibiting proliferation and adhesion of HCC cells,degradation of extracellular matrix and angiogenesis.Therefore,the mechanisms that PPARγsignaling pathway inhibits the invasion and metastasis of HCC may provide help for the clinical treatment,and is expected to improve the survival rate of HCC patient.
5.Expression of 3-hydroxyacyl-CoA dehydrogenase in E.coli and preparation of its antiserum

Objective:To clone and express the HCDH gene fragment in E.coli,and then to prepare its antiserum.Methods:The HCDH gene fragment was amplified by PCR,and ligated to the expression vector pMAL-P2X after DNA sequencing analysis.Recombinant plasmid was transformed into the E.coli.Then its expression was induced by IPTG and analyzed by SDS-PAGE.The otter rabbit was immunized with maltose resin affinity purified antigen.Antiserum valence was detected using ELISA and AGP test.The specificity was detected using Western blot.Results:The objective products of 786 bp were obtained,then the prokaryotic expression vector of HCDH gene was established and the corresponding antiserum was prepared successfully.Conclusion:It lays solid foundation for the further researches on detecting the changes of HCDH protein on the expression level or the expression regulation of HCDH gene.
6.Clinical Observation of Applying DCF Regimen Induction Chemotherapy before Ⅲ-Ⅳ Stage Oral Squa-mous Cell Carcinoma Resection

OBJECTIVE:To observe therapeutic efficacy and safety of applying dexamethasone+docetaxel+cisplatin+fluoroura-cil(DCF)regimen induction chemotherapy before Ⅲ-Ⅳ stage oral squamous cell carcinoma resection. METHODS:A total of 120 patients with Ⅲ-Ⅳ stage oral squamous cell carcinoma were randomly divided into control group(60 cases)and observation group (60 cases). Both groups were given oral cancer resection,control group received radiotherapy 4-6 weeks after surgery,observation group was additionally given Dexamethasone acetate tablets 0.75 mg orally once a day,d1-7+Docetaxel injection 80 mg/m2 added in-to 0.9% Sodium chloride solution 250 mL intravenously for 2 h,d2+Cisplatin injection 80 mg/m2 added into 0.9% Sodium chloride solution 250 mL intravenously for 1 h,d2+Fluorouracil injection 750 mg/m2 added into 0.9% Sodium chloride solution 500 mL in-travenously,d2-5. Treatment course of 2 groups lasted for 7 d. Clinical efficacies of 2 groups were observed as well as EORTC QLQ-C30 score before and after treatment,recurrence rate 12,24,36 months and the occurrence of ADR. RESULTS:The short-term total response rate of observation group was significantly higher than that of control group;12,24,36 months fol-low-up recurrence rate of observation group were significant lower than those of control group;the incidence ofⅠ-Ⅱ degree nausea and vomiting,leucopenia in observation group were significantly higher than control group,with statistical significance (P<0.05);there was no statistical significance in the incidence of Ⅲ-Ⅳ degree nausea and vomiting,leucopenia andⅠ-Ⅱ,Ⅲ-Ⅳ de-gree diarrhea between 2 groups (P>0.05). Before treatment,there was no statistical significance in EORTC QLQ-C30 score be-tween 2 groups(P>0.05). After treatment,EORTC QLQ-C30 score of 2 groups were significantly lower than before,with statisti-cal significance (P<0.05),but there was no statistical significance between 2 groups (P>0.05). CONCLUSIONS:The applying of DCF regimen induction chemotherapy can improve short-term efficacy before Ⅲ-Ⅳ stage oral squamous cell carcinoma resec-tion,reduce reoccurrence risk but induce high incidence ofⅠ-Ⅱ degree nausea and vomiting,leucopenia.
7.Interventive effect of Angongniuhuang injection on inflammatory response after brain injury in rats

Objective To observe the influence of Angongniuhuang injection on the level of in- terleukin-1?(IL-1?),intercellular adhesion molecule-1 (ICAM-1),serum protein S100B and neuron specific enolase (NSE) after brain injury to explore its protective effect on the injured brain tissues. Methods Brain contusion model was made in rats by Feeney's method.Then,the levels of IL-1?and ICAM-1 in the brain tissues and the levels of serum protein S100B and NSE in serum were measured by ELISA method at different time points.Results The level of IL-1?and ICAM-1 in brain tissues and that of S100B protein and NSE in serum in treatment group were lower than that in control group 6-48 hours after injury (P<0.05).Conclusion Angongniuhuang injection can alleviate inflammatory re- sponse after brain injury and protect effectively brain tissues.
8.Effect of exogenous interleukin-10 on liver regeneration in rats with biliary obstruction

Objective To investigate the effect of exogenous interleukin-10 (IL-10) on liver regeneration in rats with biliary obstruction.Methods Male Wistar rats were divided randomly into either a sham operation (SO) group,obstructive jaundice (OJ) group,or IL-10 treated group.Rats in OJ and IL-10 treated groups underwent ligation and division of the common bile duct,while only the SO group had division of the common bile duct.Rats in the IL-10 treated group received daily intraperitoneal injection of IL-10 at a dose of 4 μg/kg after the operation.Quantitative fluorescence real-time PCR was performed to detect hepatic TGF-β1 mRNA expression.Immunohistochemistry for hepatic proliferating cell nuclear antigen (PCNA) labeling index was used to evaluate and epitope of IL-10.The serum levels of total bilirubin (T Bil),direct bilirubin (D Bil),alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were also measured.Results Hepatic TGF-β1 mRNA,PCNA labeling index and serum levels of ALT and AST in OJ group rats were significantly higher than SO group rats at days 3 and 7 after the operation (P<0.05).Compared to the OJ group,the IL-10 group had significantly lower hepatic TGF-β1 mRNA and serum levels of ALT and AST.Conversely the IL10 group's hepatic PCNA labeling index was significantly increased as compared with those in OJ group 7 days after operation (P<0.05).Conclusion Exogenous IL-10 could enhance liver regeneration and alleviate hepatic dysfunction by down-regulating hepatic TGF-β1 expression in rats with biliary obstruction.
9.Advances in intraductal therapy for breast cancer prevention and treatment

The understanding of the biological behavior of breast cancer has deepened, hence, local treatments for breast cancer have changed from resection to minimally invasive surgery. For patients with early breast cancer, constructing a minimally invasive interven-tion with low systemic toxicity is a problem, especially for cases involving precancerous lesions. Intraductal therapy for breast cancer, which is performed by inserting reagents through breastfeeding openings using suitable carriers, is a promising, accurate, and minimal-ly invasive method for breast cancer prevention and treatment. The combination of intraductal therapy with new therapeutic strate-gies, such as targeted therapy, endocrine therapy, and immunotherapy, might improve the therapeutic effect. Moreover, the mecha-nisms of intraductal therapy for breast cancer incorporate nanotechnology, molecular imaging, and gene sequencing. Intraductal thera-py is based on clinical and pathologic characteristics of Chinese breast cancer patients, and such characteristics must be determined prior to clinical application. This article mainly discusses the research progress of breast intraductal intervention.
10.Clinical detection on anti-Toxoplasma gondii antibody,antisperm antibody and interferon-? in infertile females

Objective To determine the levels of anti-Toxoplasma gondii antibody (ATAb) and antisperm antibody (AsAb) in serum samples and interferon-?(IFN-?) in supernatants of peripheral blood cell culture in females with infertility, and to analyze their correlation. Methods ELISA was applied to determine and compare the ATAb and AsAb in serum samples of 182 women with infertility and those of 94 women with normal pregnant history. The level of IFN-? in supernatants of peripheral blood cell culture was also measured and compared by ELISA. Results The positive rates of serum ATAb and AsAb in the cases of infertility were 15.38% and 18.13% respectively, which were significantly higher than those of women with normal pregnant history( 6.38% and 5.32%).
