1.Expression and clinicopathological significance of NF? B p65 and PTEN in laryngeal squamous cell carcinoma

Objective:To investigate the expression of NF?B p65 and PTEN in laryngeal squamous cell carcinoma(LSCC) and their clinical significance.Methods:Immunohistochemical staining was used to examine the expression of NF?B p65 and PTEN in 52 cases of LSCC and 30 cases of adjacent normal tissues.Results:The expression positive rates of NF?B p65 in LSCC were obviously higher than those in adjacent normal tissues(P0.05).The Spearman analysis indicated that the expression level of NF?B p65 was negatively correlated with that of PTEN significantly(rs=-0.580,P
2.Indwelling Catheter and Urinary Infection:Monitoring Analysis and Nursing

OBJECTIVE To find out risk factors of urinary infection caused by indwelling catheter. METHODS The data of indwelling catheter patients were recorded according to the pre-planned table by adopting survey techniques of prefigure and analyzed retrospectively. The infection cases were diagnosed by rule of Diagnostic Standard of Hospital Infection issued by Ministry of Health. RESULTS A total of 734 indwelling catheter patients were surveyed from Jan to Dec in 2006. Of the 100 patients,the infection rate was 13.62%. The main reasons were the duration of indwelling catheter and improper usage of antibiotics. The main pathogeny was G-bacteria,of which Escherichia coli accounted for 28.95%,fungi for 31.58%. CONCLUSIONS Enhancing nursing care for indwelling catheter patients; reducing the duration of indwelling as soon as possible and using antibaotics properly according to germiculture and sensibility.
3.Serum levels of soluble interleukin-2 receptor in patients with head and neck cancer

Objective:To study the levels of soluble interleukin-2 receptor(sIL-2R) in patients with head and neck cancer.Methods:Using the method of sandwich ELISA, the sIL-2R in 60 cases of patients with head and neck cancer, 60 with benign tumor and 50 of healthy subjects were measured.Results:The concentration(U/ml) of serum sIL-2R in patients with head and neck cancer was significantly higher than that in those with benign tumor(571.9?294.2 vs 305.9?104.8, P0.05).Conclusion:sIL-2R may be related to head and neck cancer.
4.Expression of cytokeratin-19 as a stem cell marker in different-purity islet samples of rats

BACKGROUND: Pancreatic stem cells can be differentiated into endocrine functioning cells under a suitable cultivation condition, suggesting that pancreatic stem cells can be used as a new source of islet in treating diabetes mellitus.OBJECTIVE: To discuss the expression of stem cell in different purities of pancreatic islets.DESING: A controlled observation based on cells.SETTING: Third People's Hospital of Mianyang City.MATERIALS: The experiment was conducted in the Center Laboratory (Key Laboratory of Chongqing City), the First Affiliated Hospital of Chongqing Medical University from March 2005 to March 2006. Thirty healthy male SD rats, of clean grade, aged 50 days were used in this study. During the experiment, the disposal of animals corresponded to Animal Ethical Standard. Cytokeratin (CK)-19 and β-actin were designed by Shanghai Invitrogen Biological Co., Ltd.Brdu reagent, mouse anti-rat Brdu monoclonal antibody, rabbit anti-mouse (RAM) CK-19 polyclonal antibody and Brdu immunohistochemical kit were from Boster Co., Ltd. (Wuhan); HistostainTM -DS double immunohistochemical staining kit was purchased from Beijing Zhongshan Company.METHODS: Rats were intraperitoneally injected with Brdu for labeling, and subjected to perfusion with Ⅴ-type collagenase via pancreatic ducts, with pancreas being excised, ripped, beaten up, digested and centrifuged to obtain pancreatic islet sediments. After being precipitated, the sediments were divided into 3 groups. Group A: The isolated sediments of islets were not purified; Group B: The sediments were added slowly with 25% Ficoll-400 and Hanks solution for purification; Group C: The sediments were slowly added with 25% Ficoll-400, 11% Ficoll-400 for purification in order.MAIN OUTCOME MEASURES: Expressions of Brdu and CK-19-positive cells in the islet samples were detected by immunohistochemical method, and mRNA expression of CK-19 in the different purities of islets was detected by reverse transcription-polymerase chain reaction.RESULTS: ① The purity of islets in the group A was the lowest. The purity of islets obtained by different purification techniques showed significances among the three groups (F =89.42, P < 0.05). ②Positive expressions of Brdu and CK-19 in the group A were significantly higher than those in the group B and group C (F =18.64, 22.12, 38.61. P <0.01). ③Expression of CK-19 mRNA in the group A was significantly higher than that in other groups, with group C showing the lowest.CONCLUSION: Expressions of Brdu and CK-19-positive cells exist in different purities of islets, suggesting the existence of more stem cells in the low purify of islets.
5.Comparison and observation of A-scan ultrasound biometry measurements before and after cataract surgery

Objective Cataract surgery is also considered as a type of refractive surgery , but there is few research on the change of preoperative and postoperative ocular biometry measurements .The aim of the study was to compare the A-scan ultrasound biom-etry measurements before and after phacoemulsification with intraocular lens implantation , followed by the analysis on its clinical signifi-cance . Methods Dynamic observation was conducted in 188 eyes of 155 cataract patients who received cataract operation from January 2011 to January 2013 in the department of ophthalmology in Nanjing Gernal Hospital .Measurements were made before surgery and 14 days after surgery by Ultrascan Digital 2000 contact ultraound A-scan (Alcon), including anterior chamber depth (ACD), vitreous cham-ber depth ( V) and axial length ( AL) .Simultaneously , a prospective comparison of measurements was made by A-scan ultrasound in sit-ting and decubitus position .Measurements were also conducted in preoperative and postoperative visual acuity and intraocular pressure of the patients. Results visual acuity and intraocular pressure: The difference between preoperative and postoperative visual acuity [(0.17 ±0.19) vs (0.61 ±0.27)] and intraocular pressure [(15.09 ±8.50) mmHg vs (12.99 ±4.44) mmHg] was of statistical sig-nificance ( P<0.05) .ACD:The difference between peroperative and postoperative ACDs measured by A-scan ultrasound was of statisti-cal significance (P<0.05).No significant difference existed between ACDs in sitting and decubitus positions before and after the opera -tion (P>0.05).V:The difference between peroperative and postoperative Vs was of significant difference (P<0.05).No significant difference existed between Vs in sitting and decubitus position before the operation [(16.568 ±2.406) mm vs (16.524 ±5.544) mm,with intraocular lens implantation can get better operation result. In addition, different measuring positions have no influence on A-scan ultrasound measurements except the postoperative vitreous cavity depth .
6.CT-guided 125I seed implantation for retroperitoneal lymph node metastases:its technical points

Objective To introduce several commonly used puncture techniques for retroperitoneal lymph node metastases in order to provide technical guidance in performing 125I seed implantation therapy. Methods The puncture methods used at authors’ interventional department in performing 125I seed implantation therapy for retroperitoneal lymph node metastases were summarized. Results The commonly used puncture methods included parallel needle puncturing technique , trans-viscera puncturing technique , broadening path puncturing technique and coaxial needle puncturing technique. Conclusion An effective combination use of various puncturing techniques to uniformly implant the radioactive 125I seeds into the lesion can improve the therapeutic result and reduce the occurrence of complications.
7.Clinical value of CT-guided percutaneous biopsy of peritoneum in diagnosis of patients with unknown ascites

Objective To investigate the diagnostic value of CT-guided percutaneous biopsy of peritoneum for unknown ascites.Methods The clinical data of 106 cases of unknown peritoneum in our hospital were collected retrospectively over the past five years (May 23,2011 to August 2,2016).The sensitivity and specificity of the percutaneous biopsy of peritoneum in diagnosis of the patients with unknown etiological ascites were evaluated.Complications were collected to evaluate the safety.Results The 106 patients with unknown ascites were performed with CT-guided percutaneous biopsy of peritoneum.The diagnoses of 91 patients were confirmed pathologically,accounted for 85.85% of the total.Among the 91 patients,there were 60 cases (56.60%) of malignant and 31 cases (29.25%) of benign.The other 15 patients had no clinical diagnosis,which accounted for 14.15% of the total.The diagnostic sensitivity and specificity was 88.35% and 100% respectively,which were significantly superior to cytological examination,biochemical markers and other routine analysis.No severe complications were observed in all patients.Conclusion CT-guided percutaneous biopsy of peritoneum for unknown ascites plays an important instructive role in diagnosis with high accuracy,and it is a safe and effective method.
8. The expression of thrombospondin 1 and its gene hypermethylation in laryngeal squamous cell cancer

Objective: To explore the mRNA and protein expressions of thrombospondin 1 (THBS1) gene and the DNA methylation of its promoter in laryngeal squamous cell cancer (LSCC), and to analyze their relationship to clinicopathological features of LSCC. Methods: Sixty-six pairs of fresh LSCC tissues and the matching adjacent normal tissues were taken from 66 cases of LSCC. The mRNA and protein expressions of THBS1 gene were detected by revere transcription PCR and Western blotting assay, respectively. Then their relationship to clinicopathological features of LSCC was analyzed by one-way ANOVA and t test. The methylation of THBS1 gene promoter was detected by the DNA methylation specific PCR, and its relationship to clinicopathological features of LSCC was analyzed by ?2 test. Results: The mRNA and protein expressions of THBS1 gene in LSCC tissues were significantly decreased as compared with those in the adjacent normal tissues (P < 0.05), and these expressions were also associated with lymph node involvement and TNM staging (P < 0.05). The positive rate of the hypermethylation of THBS1 gene in the LSCC tissues (48.50%±6.15%) was significantly higher than that in the matching adjacent normal tissues (6.06%±2.92%) (P < 0.05). The mRNA and protein expressions of THBS1 gene were associated with the hypermethylation of its gene promoter (P < 0.05). THBS1 gene hypermethylation status in LSCC tissues was associated with lymph node involvement and TNM staging (P < 0.05). Conclusion: The decreased expressions of THBS1 mRNA and protein and the high methylation of its gene promoter may play crucial roles in carcinogenesis and development of LSCC. 9.Correlation between serum levels of neuron specific enolase and inflammatory factors and recovery of neurological function in patients with severe traumatic brain injury

Objective:To correlate serum levels of neuron specific enolase (NSE) and inflammatory factors with recovery of neurological function in patients with severe traumatic brain injury.Methods:Ninety-six patients with severe traumatic brain injury who received treatment from January 2018 to January 2020 in Taizhou Hospital were included in this study. These patients were divided into a mild-to-moderate group ( n = 51) and a severe group ( n = 45). Additional 60 healthy controls who concurrently received health examination were included in the healthy control group. Serum NSE level was detected by enzyme-linked immunosorbent assay, serum C-reactive protein (CRP) level by immunoturbidimetry, serum procalcitonin (PCT) level by chemiluminescent assay, and serum interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) levels by enzyme-linked immunosorbent assay. All patients were followed up for 3 months. Recovery of neurological function was evaluated by modified Rankin Scale. Results:Serum NSE level was (50.42 ± 13.25) μg/L and (36.79 ± 10.28) μg/L in the severe and mild-to-moderate groups, respectively, which was significantly higher than that in the healthy control group [(6.13 ± 1.78) μg/L, t = 25.641, 22.688, both P < 0.05). Serum NSE level in the severe group was significantly higher than that in the mild-to- moderate group ( t = 5.576, P < 0.05). Serum CRP, PCT, IL-6 and TNF-α levels were (78.95 ± 15.46) mg/L, (3.46 ± 0.75) μg/L, (432.15 ± 78.29) μg/L and (36.57 ± 8.98) μg/L] respectively in the severe group, (34.65 ± 7.48) mg/L, (1.68 ± 0.51) μg/L, (285.41 ± 36.75) μg/L and (17.54 ± 5.26) μg/L] respectively in the mild-to-moderate group and (3.25 ± 0.86) mg/L, (0.08 ± 0.02) μg/L, (73.52 ± 13.89) μg/L and (1.64 ± 0.50) μg/L, respectively in the healthy control group. Serum CRP, PCT, IL-6 and TNF-α levels in the severe and mild-to-moderate groups were significantly higher than those in the healthy control group ( t = 37.890, 34.922, 34.870, 30.099, 32.284, 24.315, 40.980, 23.312, all P < 0.05). Serum levels of these indicators in the severe group were significantly higher than those in the mild-to-moderate group ( t = 17.493, 13.414, 11.500, 12.451, all P < 0.05). In the severe group, neurological function recovered well in 34 patients and poorly in 17 patients. Serum NSE level in patients with poor neurological function recovery was significantly higher than that in patients with good recovery [(68.93 ± 14.25) μg/L vs. (34.61 ± 12.36) μg/L, t = 8.457, P < 0.05). Serum CRP [(113.24 ± 27.39) mg/L], PCT [(4.57 ± 0.87) μg/L], IL-6 [(598.90 ± 43.52) μg/L] and TNF-α [(58.78 ± 12.13) μg/L] levels in patients with poor recovery were significantly higher than those in patients with good recovery [(32.19 ± 6.90) mg/L, (2.23 ± 0.65) μg/L, (261.39 ± 26.56) μg/L and (14.53 ± 4.26) μg/L, t = 11.956, 9.788, 29.280 and 14.537, all P < 0.05). Serum NSE, CRP, PCT, IL-6 and TNF-α were positively correlated with poor prognosis ( r = 0.849, 0.743, 0.795, 0.683, 0.701, all P < 0.05). Conclusion:In patients with severe traumatic brain injury, serum NSE, CRP, PCT, IL-6 and TNF-α levels increase, which are positively correlated with poor prognosis.

Nuclear receptor transcription factors are ligand-activated proteins that control various biological events from cell growth and development to lipid metabolism, and energy and glucose homeostasis. Nuclear receptors are important drug targets for metabolic diseases. Liver X receptors (LXRs) are nuclear receptor transcription factors that play essential roles in regulation of cholesterol, triglyceride, fatty acid, and glucose homeostasis. LXR-deficient mice have shown the association of LXR-signaling pathway dysfunction with several human pathologies including atherosclerosis, hyperlipidemia, Alzheimer's disease and cancer. Thus, LXRs are promising pharmacological targets for these diseases. Synthetic LXR agonists may lower cholesterol, but increase triglyceride and induce fatty liver. The naturally occurring LXR ligands, with moderate activity, may serve as nutraceuticals for prevention or treatment of the disorders, while minimizing potential side effects. In this review, recent advances in natural LXR modulators are summarized including agonist, antagonist and the modulator of LXR pathway.
