1.Effects of lemur tyrosine kinase 3 on the cytobiological function of human lung cancer A549 cells

Objective To investigate the effects of down-regulation of lemur tyrosine kinase 3 (LMTK3) on the cytobiological behaviors of human lung cancer A549 cells.Methods The expression of LMTK3 in A549 cells was interfered with small hair RNA (shR-NA),and the expression level of LMTK3 was determined by RT-PCR.The effects of LMTK3 on the proliferation,migration,cell cycle and apoptosis of A549 cells were determined by the CCK-8 assay,scratch assay,Transwell assay and flow cytometry,respectively.Results The shLMTK3 cells with stably low expression of LMTK3 and control cells (Scramble) with normal expression of LMTK3 were successfully obtained.The relative proliferation rates of shLMTK3 cells cultured for 24,48 and 72 hours were significantly lower than those in the control cells (0.305 ±0.018 vs 0.354 ±0.011,t =5.24,P<0.01;0.461 ±0.044 vs 0.551 ±0.027,t =3.91,P <0.01;0.74 ± 0.029 vs 0.881 ± 0.028,t =7.70,P < 0.01).The relative migration rate of shLMTK3 cells 24 hours after scratching was significantly lower than that of control cells (0.51 ±0.096 vs 1.00 ± 0.029,t =4.81,P < 0.01).Transwell assay showed that the number of migration cells in shLMTK3 group was significantly less than that in Scramble group (161 ±9.29 vs 308.66 ± 17.60,t =7.42,P < 0.05).The results of flow cytometry showed that shLMTK3 cells were blocked at G1 phase (t =4.35,P < 0.05),and that the inhibition of LMTK3 had no influence on the apoptosis of A549 cells.Conclusion Down-regulation of LMTK3 expression in human lung cancer A549 cells may inhibit the proliferation and migration of A549 cells significantly,indicating that the abnormal expression of LMTK3 in lung carcinoma cells may regulate the biological behaviors and progression of tumors.
2.Performance evaluation and study on open assembly line system in medical laboratory

Objective To investigate the advantages and continuous optimization of laboratory automation system through analysis and assessment of the core data and performance after the application of open assembly line.Methods Collect the data of biochemical and immunoassay in Shuguang Hospital attached to Shanghai University of Traditional Chinese Medicine from April to October 2017.( 1 ) Cost analysis of the assembly line schemes;(2) Analysis of workflow before and after the application of assembly line;(3) Analysis of the volume of samples collecting before and after the application of assembly line ;(4) Analysis of TAT data before and after the application of assembly line; ( 5 ) Analysis of staffs allocation before and after the application of assembly line; (6) Analysis of samples rechecking before and after the application of assembly line .Results (1) Open assembly line costs least on hardware (8 million) and site among various projects;(2) Inspection process is greatly simplified after the application of assembly line;(3) The samples′volume of biochemical and immunoassay inspection were reduced by 31.85%;(4) The items′test cycle decreases after the application of assembly line , the average TAT is reduced by 32 minutes;(5) Staffs for samples pretreatment can be reduced by 50%after the application of assembly line , and the quantity of operators does not change;(6) The number of re-check samples increase except the gray zone and critical values , which ensures the reliability of the results .Conclusion To analyze the core data and to evaluate the performance , the laboratory improve on detection cycle ,staffs,and test efficiency.
3.The effect of salpingectomy with preservation of the mesosalpinx on ovarian reserve function

Objective To investigate the impact of salpingectomy with preservation of fallopian tube mesentery on ovarian reserve function.Methods A total of 281 patients with tubal disease who were admitted from January 2020 to March 2021 were collected as research objects.According to the different treatment methods,they were divided into traditional bilateral resection group(n=53,con-ventional bilateral salpingectomy),traditional unilateral resection group(n=56,conventional unilat-eral salpingectomy),preservation of bilateral resection group(n=60,bilateral salpingectomy with preservation of fallopian tube mesentery),preservation of unilateral resection group(n=54,unilater-al salpingectomy with preservation of fallopian tube mesentery),and control group[n=58,patients with in vitro fertilization-embryo transfer(IVF-ET)but no history of salpingectomy].The operation time,intraoperative blood loss,and postoperative exhaust time were compared among the groups.Two months after the operation,IVF-ET was performed,and the number of follicles and oocytes,pregnan-cy rate,and fertilization rate were detected.The levels of anti-mullerian hormone(AMH),follicle-stimulating hormone(FSH),estradiol(E2),and luteinizing hormone(LH)in peripheral blood were detected after IVF-ET.Follow-up was conducted until March 2023,and pregnancy success rates were recorded for the five groups.Results There were no significant differences in perioperative opera-tion time,intraoperative blood loss,and postoperative exhaust time among traditional bilateral resec-tion group,traditional unilateral resection group,preservation of bilateral resection group,preserva-tion of unilateral resection group(P>0.05).After IVF-ET,there were significant differences in AMH,FSH,E2,and LH levels among the five groups(P<0.05).The levels of AMH and E2 in a rising sequence were traditional bilateral resection group,traditional unilateral resection group,pres-ervation of bilateral resection group,preservation of unilateral resection group and control group,while the levels of FSH and LH in descending sequencing were traditional bilateral resection group,traditional unilateral resection group,preservation of bilateral resection group,preservation of unilat-eral resection group and control group.Follow-up was conducted until March 2023,and the preg-nancy success rates after IVF-ET were 45.28%,50.00%,53.33%,59.26%,and 58.62%in the traditional bilateral resection group,traditional unilateral resection group,preservation of bilater-al resection group,preservation of unilateral resection group,and control group,respectively.There was no significant difference in pregnancy success rate after IVF-ET among the five groups(x2=3.044,P=0.551).Conclusion Compared with traditional salpingectomy,salpingectomy with preservation of fallopian tube mesentery has less impact on ovarian reserve function.
