1.Development and application of Luminex liquichip

Liquichip is a new type of biochip emerging in the 1990s, which is integrated flow cytometry, laser technology, digital signal processing and traditional chemical technology with the greatest features of high-throughput and high-flexibility. Luminex has promoted new products from Luminex 100/200 to Flexmap 3D. This is an introduction to the development, principle, and application of Luminex liquichip.
2.Research on development of wearable electrotherapy instrument for chronic soft tissue lesion

Objective:To develop a wearable electro-therapeutic apparatus for chronic soft tissue lesion, which used low frequency electric pulse to treatment the shoulder, back and waist of human body.Methods: The fabric electrodes are fixed on specific position of the elastic vest. The vest elasticity ensures that the electrode and the users are in close contact. MSP430 microcontroller is used as the core of control board. Low frequency pulses for treatment are generated by PWM mode of MSP430 Timer1, then pass through the power amplifier and are added to human body by fabric electrodes. Meanwhile the impedance between electrodes is monitored to avoid the striking fire burns the skin of the human body and ensure user’s safety. The wireless data communication and control between intelligent mobile phone and MSP430 microcontroller are achieved by Bluetooth technology.Results:The developed wearable electrotherapy instrument is easy to operate, safe and reliable, convenient for the daily busy office workers treatment and rehabilitation in working time.Conclusion: It is very fitful for long-term therapy and save the time.
3.Identification of smooth muscle cells purified from rabbit bladder after in vitro culture

Objective To establish a method of obtaining the highly purified smooth muscle cells (SMCs) isolated from rabbit bladder and then to identify them after in vitro culture using methods of morphology and immunofluorescence. Methods Primary cell culture of SMCs was set up by enzymic method in total of 6 adult male New Zealand-White rabbits. The serosa and mucosa of bladder were carefully removed by micro-surgical instruments under the naked eyes. The morphology characteristics of cells and their pattern of cell growth were observed using inverted microscope. The cells were identified using methods of H-E staining, transmission electron microscope and immunofluorescence staining of α-actin. The curves of cell growth and proliferation were drawn after cell counting. Results The capability of adherence was observed in these isolated SMCs after in vitro culture for 24 hours, and passage was then conducted after culture for 7 days. The cells kept growing well and fast from the 1~(st) to 10~(th) generation. But starting from the 12~(th) generation, the deformed cells were found and then become those without typical morphological feature of SMCs at the 15~(th) generation, and finally died at the 20~(th) generation. The isolated cells after in vitro culture at 4~(th) passage were identified to be SMCs successfully using the methods of H-E staining, transmission electron microscope and immunofluorescence. Conclusions The highly purified bladder SMCs of rabbit were successfully obtained after isolated by enzymic method and in vitro culture within a short time. That would provide us the basis for future studies of pathophysiology and molecular biology of bladder SMCs.
4.Catheter needle technigue for Budd-Chiari syndrome

Objective The reseach of hepatic vein (HV) and inferior vein cavity(IVC) obstruction recanalized with catheter needle were evaluated. Methods 16 cases of BCS diagnosed with angiography in recent 5 years were all undergone interventional treatment. Five with HV web occlusion principally and 9 with IVC complete occlusion were treated with introducing a catheter needle to penetrate the block through jugular vein or femoral vein. Stenosis or needle track was dilated with balloon. Stent was planted to IVC in 10 cases. Results All cases were successful in technic including a segmental obstrution of IVC and with a lower IVC atresia. Venography demonstrated the patency of blood flow. HV pressure of 5 cases was reduced 1.85kPa, and IVC pressure of 11 cases was reduced 1.21kPa on an average.Conclusions The use of catheter needle technique increases the successful rate in BCS treatment.
5.Hair Lead Level of Children Living in Lead Polluted Area

Objective To understand the environmental lead pollution status in Tongzhou and the biological burden of lead in children living in the lead polluted area.Methods The 353 pupils aged 7-14 years were randomly selected in Zhangzhishan town central elementary school that was near to steel wire factories,283 pupils were selected as the control by using the same methods in an elementary school around where there is no any steel wire factory.Every students were investigated by questionnaires,meanwhile their hair were collected to determine lead with ASS method.Results In the polluted area group,hair lead of 77 pupils exceeded the normal level,the median was 4.437 ?g/g,the rate of exceeding the normal level(≤10 ?g/g)was 21.81%,and 65 pupils with lead poisoning related symptoms(18.41%).In the control group,the median was 3.965 ?g/g,the rate of exceeding the normal level was 11.31%,38 pupil with lead poisoning related symptoms(13.43%).After controlled the passive smoking confounder,the difference of hair lead level between these two groups was still significant(P
6.Perceptions on the risk communication strategy during the 2013 avian influenza A/H7N9 outbreak in humans in China: a focus group study

Objective: To identify the general public’s perceptions of the overall risk communication strategy carried out by Chinese public health agencies during the first wave of avian influenza A(H7N9) outbreak in humans in 2013.
Methods: Participants were recruited from communities in Beijing, Lanzhou and Hangzhou, China in May and June 2013 by convenience sampling. Demographics and other relevant information were collected using a self-administered questionnaire. Focus group interviews were conducted using a set of nine pre-developed questions and a tested moderator guide. The interviews were audio recorded and were transcribed verbatim. The constant comparative method was used to identify trends and themes.
Results: A total of nine focus group interviews, with 94 participants recruited from nine communities, were conducted. Most participants received H7N9 information via television and the Internet. A majority of the participants appreciated the transparency and timeliness of the information released by the government. They expressed a sense of trust in the recommended public health advice and followed most of them. The participants suggested that the government release more information about clinical treatment outcomes, have more specific health recommendations that are practical to their settings and expand the use of new media channels for risk communication.
Conclusion: The public perceived the overall risk communication strategy by the Chinese public health agencies as effective, though the moderator had a governmental agency title that might have biased the results. There is a need to expand the use of social media for risk communication in the future.
7.Arthroplasty versus joint preservation for displaced 3-and 4-part proximal humeral fractures:a meta-analysis

BACKGROUND:Currently, the treatment of proximal humeral fractures mainly contains joint preservation (conservative treatment, open reduction and internal fixation) and arthroplasty, but how to choose the treatment is stil controversial. OBJECTIVE:To evaluate and compare the clinical outcomes of joint preservation versus arthroplasty in the treatment of displaced 3-or 4-part humeral fractures in randomized control ed trials using meta-analysis. METHODS:Medline (January 1966 to December 2013), PubMed (January 1980 to December 2013), Embase (January 1990 to December 2013), Science (January 1990 to December 2013), Springer (January 1990 to December 2013), China National Knowledge Infrastructure (1994 to 2013), and Wanfang database (1982 to 2013) were searched for randomized control ed trials addressing joint preservation and arthroplasty for 3-or 4-part proximal humeral fractures. Articles meeting the inclusion criteria were included. The related data were extracted and loaded onto Comprehensive Meta-Analysis Software for meta-analysis. RESULTS AND CONCLUSION:Seven articles with 320 patients (165 patients undergoing joint preservation and 155 patients receiving arthroplasty) were accepted in this mete-analysis and al of them were high-quality English researches by modified Jadad Scale. Meta-analysis results displayed that the random-effects mean Constant score across al types was 55.9 (95%CI:50.7-61.1;P<0.001). Constant score was higher in the joint preservation group than in the arthroplasty group (P<0.01). The study displayed significant heterogeneity (I2 0.001). In the meta-analysis, Constant scores were decreased with increasing rate of male to female and rate of tuberosity resorption. These results suggested that 3-or 4-part proximal humeral fractures demonstrate improved Constant scores when treated with joint-preserving options compared with arthroplasty. Moreover, age, fracture pattern, gender ratio and complication rate are significant predictors of the Constant score. Given the observed heterogeneity and variance in treatment techniques in the included studies, more randomized control ed trial studies are needed to definitively recommend joint-preserving techniques versus arthroplasty for treatment of 3-or 4-part proximal humeral fractures.=88%, Q statistic=107.6, Q=13;P<0.001). 8.Value of multi-biomarkers plus PSA in the diagnosis of prostate cancer

Objective To evaluate the clinic value of multiple tumor markers plus PSA in diagnosing prostate cancer.Methods We collected serum samples of 140 prostate cancer patients with average age of 68 (48-82) years,104 benign prostate hyperplasia patients with average age of 70 (52-87) years,and 162 healthy people with average age of 38 (23-49) years.We had detected PSA levels and also the protein expressions of XAGE-1b,SSX-2,AM ACR and AKAP4.In healthy people,the ranges of the normal values with 95% data range were determined.Multiple tumor markers and PSA were calculated their positive rate,specificity and sensitivity in diagnosing prostate cancer.We had randomly chosen one positive serum,and analyzed the 4 protein expressions by Western bolt.We detected the serum with the four markers plus PSA by Luminex medthod,then drew ROC curve and calculated AUC area according to the results.Results Among all those samples,PSA levels of 266 samples were under 4tμg/L,86 samples were between 4-10 μg/L,and 54 samples were above 10 μg/L.The positive rates of XAGE-1b,SSX-2,AMACR and AKAP4 in prostate cancer patients were 53.6% (75/140),34.3% (48/140),27.9% (39/140),44.3% (62/140) respectively.The AUC of XAGE-1b,SSX-2,AMACR,AKAP4 and PSA were 0.666,0.615,0.551,0.768,0.675 respectively.The AUC of their combination was 0.887; The specificity and sensitivity of single PSA detection and combination detection were 60.0%,46.2% and 80.0%,82.2%.Among those whose PSA value was between 4-10 μg/L,the AUC of single PSA detection was 0.505,the specificity and sensitivity of single PSA detection were 43.2% and 31.8%; the AUC of the combination detection was 0.803; their specificity and sensitivity were 83.7% and 73.2%.Conclusions Compared with the single PSA detection,the combination of XAGE-1b,SSX-2,AMACR,AKAP4 and PSA has been greatly improved the specificity and sensitivity in prostate cancer detection.This tool still has significant value even in patients with PSA value between 4-10 μg/L.
9.Incidence of Metallo-beta-lactamase-producing Pseudomonas aeruginosa and their Antibiotic Resistance

OBJECTIVE To study incidence of metallo-beta-lactamase(MBL)-producing Pseudomonas aeruginosa and their drug sensitivity.METHODS A total of 323 isolates of P.aeruginosa from Nanshan District of Shenzhen between Jun 2007 and Jun 2008 were subjected to susceptibility testing against various antibiotics and identified by ATB-Expression.Imipenem and meropenem resistant isolates were selected for the detection of MBL production by disc synergia test.Enhancement of inhibition zone around imipenem and meropenem discs impregnated with EDTA was found to confirme MBL production.RESULTS Totally 323 P.aeruginosa were isolated and MBL-producing accounted for 46(14.3%),drug resistance rate to imipenem and meropenem was respectively 45.4% and 26.3%.Colistin was the most sensitive to P.aeruginosa(96.9%).CONCLUSIONS Our findings show it needs to detect MBL producers,judiciously use carbapenems to prevent their spread.
10.Two-stage delayed surgical treatment for type Ⅱ and type Ⅲ Pilon fractures

Objective To explore the therapeutic effect of two-stage delayed open reduction and internal fixation on Pilon fractures.Methods The clinical data of 38 patients with type Ⅱ and type Ⅲ Pilon fracture were analyzed retrospectively.According to the Ruedi-Allgower classification,there were 18 cases with type Ⅱand 20 cases with type Ⅲ fracture.All patients underwent temporary fixation with calcaneal traction or external fixation for 7 to 17 days,and then they were treated with delayed surgery when the soft tissue injuries subsided.Results Follow-up was performed for 10 to 18 months.Based on the Mazur classification,the ankle function in 12 cases was excellent,18 cases as good,6 cases as fair and 2 cases as poor,with a satisfactory rate of 78.9%.Conclusion Two-stage delayed open reduction and internal fixation is a reasonable and effective procedure in treating type Ⅱ and type Ⅲ Pilon fractures with advantages of few complications and satisfactory ankle function.
