1.The exploration of establishing the health care resource pool of Chinese medicine

With the improvement of health care consciousness,it is very urgent to accelerate the dissemination and learning of health care to prevent disease by using Chinese medicine.The establishment of the health care resource pool of Chinese Medicine is to provide convenience for people to learn the knowledge about health care.The essay is aimed to explore the necessity and feasibility of building the health care resource pool of Chinese Medicine from the aspects of discussing the meaning,predominance,essentials and contents of resources.
2.Effect of Herba Hedyotis Diffusae on Living Murine Ascites Hepatoma H22 Cells and T Lymphocytes

【Objective】To observe the effect of Herba Hedyotis Diffusae(HHD)on the histological features of living murine ascites hepatoma H22 cells as well as CD+4 and CD+8 expression in T lymphocytes.【Methods】Special pathogen free(SPF)Kunming mice were randomized into 4 groups:model group,HHD(25 g/kg)group,Radix Codonopsis(RC,25 g/kg)group,heat-shock group.The murine ascites hepatoma H22 model was established by abdominal transplantation of H22 cells.The histological features of H22 cells from living murine ascites were examined through living mice ascites smear and giemsa staining.The CD+4 and CD+8 expression in ascites T lymphocytes was detected by immunohistochemical method.【Results】H22 cell deformation such as cell membrane incomplete and vesicant,some nucleus being migration and pycnosis was obvious in HHD and heat-shock groups.The expression of both CD+4 and CD+8 expression in T lymphocytes of HHD groups was increased,but only CD+4 expression in RC group and only CD+8 expression in heat-shock group was increased.【Conclusion】HHD exerts some inhibitory-killing effect on living murine H22 cells,and its mechanism may be related to the increase of immune function and to the cytotoxic effect.
3.Effect of Heat-clearing and Detoxifying Herbs and Tonifying Herbs on Mice Hepatoma Cells Line H_(22) and Their Immunologic Mechanism

[Objective] To compare the effects of heat-clearing and detoxifying herbs with that of tonifying herbs on mice hepatoma cells line H22, and to explore their immunologic mechanisms. [Methods] Herba Rabdosiae Rubescentis (HRR) and Herba Scutellariae Barbatae (HSB) were selected as the heat-clearing and detoxifying herbs, and Radix Astragali (RA) as the representative of tonifying herbs. Fifty KM mice were inoculated with hepatoma cells line H22 and then were randomized into five groups: HRR group (30 g?kg-1?d-1 by gavage for 12 days), HSB group (15 g?kg-1?d-1 by gavage for 12 days), RA group (15 g?kg-1?d-1 by gavage for 12 days) , heat-shock group (treated 20 min at 43℃, once every 4 days and three times in all) and model group. Twelve days later, the mice were executed and the ascites smears were prepared. The cell feature in ascites was observed under light microscope and the expression of CD4+ and CD8+ on the cellular membrane of the T lymphocyte in ascites was detected with immunohistochemical method. [Results] A large amount of damaged and apoptotic H22 cells were found in HRR, HSB and heat-shock groups, and the changes of cell feature in RA group were not obvious. The expressions of CD4+ and CD8+ on the membrane of T cells in ascites was increased in HRR and HRB groups, but the expression of CD4+ was increased in RA group, and CD8+ expression was increased in heat-shock group.[Conclusion] Heat-clearing and detoxifying herbs can induce the necrosis and apoptosis of mice hepatoma cells line H22 and the mechanism may be related to the increase of cellular immunity, thus enhancing the body immunity to kill the tumor cells. RA can not kill the tumor cells directly and its antitumor action may be related to the increase of CD4+ expression.
4.Influence of assisted reproduction technologies on genomic imprinting of embryos and offspring.

Assisted reproduction technologies (ART) include controlled ovarian hyperstimulation, in vitro fertilization-embryo transfer, intracytoplasmic sperm injection, in vitro maturation of oocytes, pre-implantation genetic diagnosis, etc. They have been used for the treatment of impaired fertility but may damage the health of offspring. The ART procedures may alter the epigenetic status of these offspring and DNA methylation may be a crucial mechanism. This paper summarizes epigenetic alterations in ART embryos and offspring, and discusses the risks.

5.In vitro culture of isolated primary hepatocytes and stem cell-derived hepatocyte-like cells for liver regeneration.

Various liver diseases result in terminal hepatic failure, and liver transplantation, cell transplantation and artificial liver support systems are emerging as effective therapies for severe hepatic disease. However, all of these treatments are limited by organ or cell resources, so developing a sufficient number of functional hepatocytes for liver regeneration is a priority. Liver regeneration is a complex process regulated by growth factors (GFs), cytokines, transcription factors (TFs), hormones, oxidative stress products, metabolic networks, and microRNA. It is well-known that the function of isolated primary hepatocytes is hard to maintain; when cultured in vitro, these cells readily undergo dedifferentiation, causing them to lose hepatocyte function. For this reason, most studies focus on inducing stem cells, such as embryonic stem cells (ESCs), induced pluripotent stem cells (iPSCs), hepatic progenitor cells (HPCs), and mesenchymal stem cells (MSCs), to differentiate into hepatocyte-like cells (HLCs) in vitro. In this review, we mainly focus on the nature of the liver regeneration process and discuss how to maintain and enhance in vitro hepatic function of isolated primary hepatocytes or stem cell-derived HLCs for liver regeneration. In this way, hepatocytes or HLCs may be applied for clinical use for the treatment of terminal liver diseases and may prolong the survival time of patients in the near future.

6.Mangiferin inhibits proliferation,migration and inflammatory factor expression of fibroblast-like synoviocytes in rheumatoid arthritis

BACKGROUND:Mangiferin is a biphenylpyridone compound extracted from mango leaves,bark and roots.Previous studies have shown that mangiferin can exert anti-systemic inflammatory effects through the activation of transcription factors such as NF-κB and JAK/STAT. OBJECTIVE:To investigate the effects and mechanisms of mangiferin on proliferation,migration and inflammatory factor release of rheumatoid arthritis fibroblast-like synovial cells(RA-FLS). METHODS:RA-FLS were divided into blank group,R848(TLR7/8 agonists)stimulated group,mangiferin low-,medium-,high-dose groups(2,4 and 8 μg/mL)and positive control group(Cu-CPT8,TLR8 pathway inhibitor).The cytotoxic effect of different mass concentrations of mangiferin was detected using cell counting kit-8 method and the final cellular dosing mass concentration was screened.The proliferation ability of RA-FLS was detected by cell clone formation assay,the migration ability of RA-FLS was detected by scratch assay and Transwell migration assay,and the expression of interleukin 1β,interleukin 6 and tumor necrosis factor α mRNA in RA-FLS was detected by qRT-PCR. RESULTS AND CONCLUSION:Compared with the blank group,the viability of RA-FLS was inhibited after treatment with mangiferin at 2-10 μg/mL,but there was no significant difference among groups(P>0.05),indicating that the toxic effect on RA-FLS was minimal.Compared with the R848-stimulated group,mangiferin decreased the number of cell clones,the scratch healing rate and the number of migrating cells in all dosing groups(P<0.01);and the expression of interleukin 1β,interleukin 6 and tumor necrosis factor α mRNA was also reduced in the mangostin medium-and high-dose groups(P<0.01).Compared with the R848-stimulated group,the number of cell clones,the scratch healing rate and the number of migrating cells as well as the expression levels of interleukin 6 and tumor necrosis factor α mRNA were significantly reduced in the positive control group(P<0.05,P<0.01).But there was no significant difference in the expression level of interleukin 1β.To conclude,mangiferin may exert its anti-rheumatoid arthritis effects through the TLR7/8 signaling pathway by inhibiting RA-FLS proliferation,migration,and inflammatory factor release.
7.Application of an artificial intelligence-assisted endoscopic diagnosis system to the detection of focal gastric lesions (with video)

Objective:To construct a real-time artificial intelligence (AI)-assisted endoscepic diagnosis system based on YOLO v3 algorithm, and to evaluate its ability of detecting focal gastric lesions in gastroscopy.Methods:A total of 5 488 white light gastroscopic images (2 733 images with gastric focal lesions and 2 755 images without gastric focal lesions) from June to November 2019 and videos of 92 cases (288 168 clear stomach frames) from May to June 2020 at the Digestive Endoscopy Center of Renmin Hospital of Wuhan University were retrospectively collected for AI System test. A total of 3 997 prospective consecutive patients undergoing gastroscopy at the Digestive Endoscopy Center of Renmin Hospital of Wuhan University from July 6, 2020 to November 27, 2020 and May 6, 2021 to August 2, 2021 were enrolled to assess the clinical applicability of AI System. When AI System recognized an abnormal lesion, it marked the lesion with a blue box as a warning. The ability to identify focal gastric lesions and the frequency and causes of false positives and false negatives of AI System were statistically analyzed.Results:In the image test set, the accuracy, the sensitivity, the specificity, the positive predictive value and the negative predictive value of AI System were 92.3% (5 064/5 488), 95.0% (2 597/2 733), 89.5% (2 467/ 2 755), 90.0% (2 597/2 885) and 94.8% (2 467/2 603), respectively. In the video test set, the accuracy, the sensitivity, the specificity, the positive predictive value and the negative predictive value of AI System were 95.4% (274 792/288 168), 95.2% (109 727/115 287), 95.5% (165 065/172 881), 93.4% (109 727/117 543) and 96.7% (165 065/170 625), respectively. In clinical application, the detection rate of local gastric lesions by AI System was 93.0% (6 830/7 344). A total of 514 focal gastric lesions were missed by AI System. The main reasons were punctate erosions (48.8%, 251/514), diminutive xanthomas (22.8%, 117/514) and diminutive polyps (21.4%, 110/514). The mean number of false positives per gastroscopy was 2 (1, 4), most of which were due to normal mucosa folds (50.2%, 5 635/11 225), bubbles and mucus (35.0%, 3 928/11 225), and liquid deposited in the fundus (9.1%, 1 021/11 225).Conclusion:The application of AI System can increase the detection rate of focal gastric lesions.
