1.AICAR Blocks the Proliferation of Pulmonary Artery Smooth Muscle Cells by Inhibition of Cell Cycle in G0/G1

Objective To establish the PDGF-BB-induced proliferation of rat pulmonary artery smooth muscle cells(PASMCs), and investigate the effects of the AMPK agonist AICAR on the cell cycle of PASMCs, in order to search new drugs for prevention of pulmo-nary vascular remodeling. Methods 20ng/ml PDGF-BB was used to induced the proliferation of PASMCs, and the effect of 0. 5mmol/L AICAR on the proliferation of PASMCs was observed. Western blot was used to detect the total and phosphorylated AMPK. The prolifer-ation of PASMCs was determined by CCK-8. The mRNA expression of cyclinD1, cyclinE and CDK2/4/6 were detected by flow cytometry analysis cell cycle,quantitative real-time PCR. Results Western blot Results indicated AICAR could promote the activation of AMPK. CCK-8 test Results showed that AICAR blocked the proliferation of PASMCs induced by PDGF-BB. Flow cytometry analysis indicated that AICAR arrested the cell cycle in G0/G1 to S phase. RT-PCR Results demonstrated that AICAR inhibited the mRNA expression of cy-clinD1, cyclinE and CDK2/4/6. Conclusion The AMPK agonist AICAR can block the proliferation of PASMCs induced through arrest cell cycle in G0/G1-S phase by regulation the mRNA expression of cyclin D1, cyclinE, CDK2/4/6, and AICAR has a potential applica-tion in preventing pulmonary vascular remodeling.
2.Differentiation of bone marrow and umbilical cord blood mesenchymal stem cells into osteoblasts in vitro:comparison of their osteogenic potentials

BACKGROUND:Mesenchymal stem cels isolated from cord blood and bone marrow have multi-directional differentiation ability under a certain condition of induction. OBJECTIVE:To compare the difference of differentiation of umbilical cord blood and bone marrow mesenchymal stem cels into osteoblasts. METHODS:Human umbilical cord blood and bone marrow mesenchymal stem cels were isolated and cultured by density gradient method. When reached 90% confluency, mesenchymal stem cels were digested by trypsin for subculture. At the third passage, umbilical cord blood mesenchymal stem cels and bone marrow mesenchymal stem cels at 8×104/wel were incubated. When reached 80% confluency, cels were treated with low-glucose DMEM supplemented with 10% fetal bovine serum, 0.1 μmol/L dexamethasone, 50 μmol/L vitamin C and 10 mmol/L β-sodium glycerophosphate. RESULTS AND CONCLUSION:There was no significant difference in morphology and biological properties of the two kinds of mesenchymal stem cels. Cels were highly expressed CD44, CD29, but did not express CD34. They had the ability to differentiate into osteoblasts, which had a positive staining for known markers: alkaline phospatase and calciumin vitro mineralization. There was no significant difference in the activity of osteoblasts of two kinds of cels. Results verify that umbilical cord blood and adult bone marrow mesenchymal stem cels can be induced into osteoblasts with a similar ability, and they can be used as seed cels for bone tissue engineering.

4.Sensitivity regulatory mechanism of ziprasidone capsules on PRL and FINS in male adolescents with episode schizophrenia

Objective To explore the sensitivity regulatory mechanism of ziprasidone capsules on prolactin( PRL) and fast insulin( FINS) in male adolescents with episode schizophrenia.Methods According to inclusion and exclusion criteria, 52 adolescent males selected from January 2015 to March 2016 were confirmed the psychiatric treatment of schizophrenia in our hospital, their ages, ziprasidone capsule usage and suffering schizophrenia stage of disease, height, waist circumference, body weight, hip circumference, and in the morning fasting blood glucose, FINS and PRL content were recorded.Mathematical model were used to calculate QUICKIFINS sensitivity index, and in accordance with the classification criteria BMI, 52 male adolescents with schizophrenia were divided into normal group and overweight group.The correlation detection, and anthropometric parameters, PRL and FINS sensitivity parameters were analysed.Results PRL and FINS, QUICKI indicators were not correlated(r=0.153, P >0.05; r=-0.101, P >0.05) statistically significant; ziprasidone capsule dose has a positive correlation with FINS(r =0.376,P <0.05), and QUICKI negative indicators (r=-0.362,P<0.05); waist-hip ratio or fat content ratio has no correlation with FINS, QUICKI indicators, but BMI and FINS has a positive correlation (r=0.389,P<0.05) and QUICKI indicators with negative correlation(r=-0.413,P<0.05);PRL level between normal group and overweight group was not statistically significant, and levels of FINS in overweight group(10.89 ±8.23) mU/L was higher than the normal group(5.79 ±3.18) mU/L, the difference was statistically significant(P <0.05).QUICKI index between the two groups was statistically significant, and the overweight of FINS was sensitivity less than the normal group.Conclusion Ziprasidone capsule has no relationship with PRL and FINS resistance in the treatment of adolescent patients with schizophrenia, while body mass index correlated with FINS resistance, and ziprasidone capsule dose correlated with FINS resistance.
5.Autologous CD34+cell transplantation promotes angiogenesis in older adult patients with atherosclerotic ischemia:study protocol for a prospective, single-center, open-label, randomized controlled clinical trial

BACKGROUND:The main clinical manifestation of senile arteriosclerosis obliterans is lower limb ischemia, which is currently difficult to treat. One method is by autologous stem cell transplantation into the muscles of ischemic limbs to improve the formation of new capillaries and restore lower limb blood flow. Endothelial progenitor cell marker CD34+ cell transplantation has been shown to promote angiogenesis in ischemic limbs. Therefore, we propose that peripheral blood autologous CD34+ cell transplantation in older adult patients with atherosclerotic ischemia could effectively promote angiogenesis.OBJECTIVE:To assume that peripheral blood autologous CD34+ cell transplantation in the elderly with atherosclerotic ischemia could effectively promote angiogenesis.METHODS:This is a prospective, single-center, open-label, randomized, and controlled clinical trial that will be completed at the Qingdao No. 9 People's Hospital, China. Twenty older adult patients with atherosclerotic lower limb ischemia will be randomized into two groups. In the cell transplantation group (n=10), peripheral blood CD34+ cells transfected with vascular endothelial growth factor 165 (VEGF165) gene will be intramuscularly transplanted into the ischemic limbs in older adult patients with atherosclerotic lower limb ischemia. In the control group (n=10), normal saline will be intramuscularly injected into the ischemic limbs. All patients will be followed up for 6 months. The primary outcome will be ankle-brachial indices before and 6 months after transplantation to assess lower limb ischemia in both groups.The secondary outcomes will be the number of microvessels in the lower limb muscles before and 6 months after transplantation, the morphology of new blood vessels revealed by CT angiography, the number of VEGF-immunoreactive cells 6 months after transplantation and the incidence of adverse reactions. The trial was registered at the ClinicalTrials.gov (identifier:NCT03098771), and the study protocol was approved by the Ethics Committee of Qingdao No. 9 People's Hospital of China. All protocols will be in accordance with Declaration of Helsinki,formulated by the World Medical Association. All patients will be informed of study protocols and provide a written informed consent prior to the beginning of the trial.DISCUSSION:This trial will begin in January 2018 and finish in December 2019. We aim to quantify the effects of VEGF165 gene-modified CD34+ cell transplantation in the treatment of older adult patients with atherosclerotic ischemia to develop a new effective treatment of lower limb ischemia.
6.Analysis on budget execution of infrastructure projects in 13 hospitals

Objective:To analyze the budget execution of infrastructure projects in public hospitals, and to analyze the problems existing in the construction process of infrastructure projects.Methods:The budget execution of infrastructure projects in 13 large-scale public hospitals in a province from 2016 to 2019 was investigated, and the problems in budget execution and the reasons for low budget execution rate were analyzed.Results:Seventeen hospital infrastructure projects in 13 hospitals were generally beyond the planned construction period, and the budget execution rate was generally low. The reasons included the problems of project owner units, project construction units and other external problems.Conclusions:Public hospitals should actively improve the management ability of infrastructure construction, strengthen the whole process management of infrastructure projects, so as to ensure the smooth construction of infrastructure projects and improve the efficiency of capital utilization.
7.Relationship between polymorphism of N-acetyltransferase 2 and genetic susceptibility to laryngeal carcinoma.

OBJECTIVETo investigate the relationship between polymorphism of N-acetyltransferase (NAT2) gene and genetic susceptibility to laryngeal carcinoma.
METHODSA case-control study on 62 laryngeal carcinoma patients and 56 controls was conducted. NAT2 alleles were differentiated by polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP) methods using originally created PCR primers and genomic DNA extracted from peripheral white blood cells. Genetic risk for NAT2 genotype was analyzed by smoking index (SI, cigarettes smoked per day x years of smoking).
RESULTSThe frequency of NAT2 slow genotype was 80.6% in patients with laryngeal carcinoma and 60.7% in the controls, the difference of which was statistically significant (chi(2) = 5.70, P = 0.017). The odds ratios were 2.70 (95% CI 1.19 approximately 6.11). Among the individuals with NAT2 slow genotype at high level of cigarette smoking, there was a significantly higher risk of 5.64 (95% CI 1.77 approximately 17.92), while those at low level were considered the reference group (OR 1.38, 95% CI 0.42 approximately 4.52).
CONCLUSIONNAT2 slow genotype increases the risk of susceptibility to laryngeal carcinoma. The combined effect of NAT2 slow genotype and exposure to smoking is observed during the development of laryngeal cancer.

Objective To improvethe control of infection source by improving the continuous quality of sputum disposal management. The cross-sectional survey of tuberculosis ward in 32 cases of hospital patients, as control group.PDCA first round of "sputum cup distribution and use of work flow" for quality improvement,the reason leading the work flow of workers to nurse led work flow,the improved cross-sectional survey in 36 cases of hospital patients as experimental group 1.Aiming at the problem of low utilization rate of special sputum cup in the improvement of the first round of PDCA,the improvement of the sputum cup was carried out,On this basis, the "special sputum disposal Cup" was designed to obtain the national patent authorization. After the second round of improvement, the cross-sectional investigation in 35 patients in the hospital,as the experimental group 2.Results After two rounds of PDCA improvement,experimental group 2 groups compared with control,bedside sputum cup configuration was not in place rate decreased from 53.1% (17/32)to 0,the difference was statistically significant(Χ2=24.916,P<0.05);configuration after the sputum cup unused rate from 53.1%(17/32)down to 8.6%(3/35),the difference was statistically significant(Χ2=15.846,P<0.05).Conclusion PDCA method can improve the quality of management standard sputum disposal. Ward sputum cup ration is standardized sputum disinfection ward-based nurse intervention and leading sputum cup release, ward can improve the sputum cup ration, improved sputum containers (sputum cup) can improve patient compliance standard of spitting.
