1.Construction of Knowledge Base Model of Syndromes and Prescriptions in Shanghan Lun Based on Artificial Neural Network

This study was aimed to explore the nonlinear relationship between syndromes and prescriptions by using the artificial neural network (ANN) algorithm. The clauses composed of syndromes and prescriptions in Shanghan Lun (Treatise on Cold Damage and Miscellaneous Diseases) were sorted out. A total of 245 clauses were trained with ANN to establish the knowledge base model of syndromes and prescriptions. After that, 100 clauses were tested. And then, the symptoms of Gui-Zhi-Tang (GZT) syndrome, Ma-Huang-Tang (MHT) syndrome, Xiao-Chai-Hu-Tang (XCHT) syndrome and Bai-Hu-Tang (BHT) syndrome were input into the model to predict the Chinese herbs. The results showed that the test accuracy of the model was 79%. For the herbs to GZT and MHT syndrome, the test error was less than 0.1. For the main herbs to XCHT and BHT syndrome, the test error was less than 0.1. While for other herbs, the test error was less than 0.3. It was concluded that the ANN algorithm can simulate the nonlinear relationship between syndromes and prescriptions, which can be applied to study the syndrome differentiation, in order to contribute to the syndromes and prescriptions standardization and the traditional Chinese medicine (TCM) information.

[Objective]To discuss professor Wu Kaopan’s academic thoughts by studying plain questions(Su Wen), Miraculous Pivot(Ling Shu), Medical Prob-lems. [Methods]Through the study on Wu’s papers and works of plain questions(Su Wen), Miraculous Pivot, using literature research method, sort and study these papers and works. [Results]This paper introduces Wu’s study on classics, using methods not only col ating, exegetics, annotation and modern translation, but also classification, analysis and exposition, the result is theoretical essence and academic ideas in such works are explored. [Conclusion]For researchers, the study of classics of Chinese medicine requires basic theory of it, what’s more, broad and profound literature knowledge is more important.
3.A method for improving acute cumulative death rates in animals by intravenous administration

AIM: To improve on the acute cumulative death rates method in animal by intravenous (iv) administration. METHODS: A mathematic model was established to simulate the pharmacodynamic and pharmacokinetic process by the program written by Visual Basic.net. This program evaluated the acute cumulative death rates (ACD method) for use of pharmacokinetics of traditional Chinese medicine (TCM). RESULTS: The ACD method related on its first dosage, and an unsuitable dosage could lead to a wrong results. It also had been proved that, by adjusting the 2nd dosage it was a more suitable to wider range of dosage. CONCLUSION: By suitable 2nd dosage and enough animal number, ACD method can be used in the pharmacokinetics of TCM.

Objective: To investigate the effects of vitamin B 1, C and orange juice on human lung cancer cell proliferation in vitro. Methods: Vitamin C degradation in culture medium was evaluated. 5.DETERMINATION OF VITAMIN C IN SERUM BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

Objective: To assay the content and stability of vitamin C(VC) in serum by a simple, specific method. Methods: Serum samples were extracted by metaphosphoric acid, and analyzed on a column C 18 of HPLC with DAD detector at 234 nm. Mobile phase was 100 mmol/L KH 2PO 4, pH3.00. The flow rate was 1 ml/min. Results: Under this condition, the retention time of VC was 5.76 min. The within run and between run reproducibilities tests were 0.42% and 2.11% respectively, and the recovery rate ranged from 82% to 88.5%. Stored at -70 ℃ and compared to that of its metaphosphoric acid extract, serum VC content was stable only for two days, and then decreased rapidly, whereas it did not decrease in metaphosphoric acid extract until the fifth day. Conclusion: A simple, sensitive and reliable method of HPLC with DAD detector is established for measuring VC in either serum or fruit, such as orange juice. VC should be determined immediately or should be extracted with metaphosphoric acid as soon as possible, then the extracts are kept at -70 ℃, and analysed within 5 d.
6.The effect of PTEN expression downregulation on biological characteristics of breast cancer cell line

Objective To investigate expression of the phosphatase and tensin homolog (PTEN) gene in breast cancer cell line and its effect on biologic characteristics.Methods The normal PTEN expression cell line MDA-MB-231 (M231) was used in this study.PTEN-shRNA plasmid was transected into M231 breast cancer cells to knock down the expression of PTEN.The changes of PTEN expression,proliferation,invasion and metastasis of PTEN knocked down cell were tested by RT-PCR,Western blot,CCK-8,scratch and Transwell.Results PTEN-shRNA was successfully transected into M231 cells.PTEN mRNA and protein expression was efficiently inhibited in M231-3001 cell lines than that in control group M231-scr(P < 0.01),M231-3001 cell lines showed a greater capability of colony formation,migration and invasion than that in control group M231-scr (all P < 0.05).Conclusions PTEN,as a suppression gene,its low expression can promote the proliferation,migration and invasion of breast cancer cells.
7.A meta-analysis of the efficacy of combined hepatectomy and splenectomy in treating hepatocellular carcinoma complicated with hepatic cirrhosis and hypersplenism

Objective To systematically evaluate the efficacy and safety of combined hepatectomy and splenectomy in hepatocellular carcinoma complicated with hepatic cirrhosis and hypersplenism.Methods Medline (1966-August 2009), Embase (1974-August 2009), Cochrane Library, CBMdisc (1978-August 2009), and Wanfang Database were searched without language limitation. All relevant studies were screened and the data were extracted by two independent reviewers, and the methodological qualities of the included studies were evaluated by the Minors scale. The data were analyzed with the RevMan5 software. Results Five non-randomized comparative studies (NRCs) involving 476 patients (232 in HS group, 244 in control group) were enrolled into the analysis. There was no significant difference in the operative mortalities (OR=0. 57, 95%CI 0. 12-2. 66, P=0. 47) and postoperative morbidities (OR= 0. 93, 95 % CI 0.59- 1.46, P = 0.75) between the two groups. Compared with hepatectomy only, CD4+ T cell (WMD=7.90, 95%CI 7.01-8.79, P<0.01), CD4+ T cell/CD8+ T cell ratio (WMD=0. 75, 95%CI 0. 70-0.80, P<0.01), white blood cell count (WMD=5.47, 95%CI 5.13-5.82, P<0.01) and platelet count (WMD=174.89, 95%CI 116.61-233.18,P<0.01) were significantly higher, but CD8+ T cell (WMD = - 7.66, 95%CI - 8. 53~ - 6. 79,P<0. 01) was lower compared with combined hepatectomy and splenectomy. There was no significant difference in the 5-year survival rates (OR= 1.37, 95%CI 0.86-2.18, P=0. 18). Conclusion Combined hepatectomy and splenectomy did not increase the operative mortalities and postoperative morbidities in hepatocellular carcinoma complicated with hepatic cirrhosis and hypersplenism. The white blood cell and platelet counts markedly increased after surgery. There was no evidence to show any improvement in the 5-year survival.
8.The correlation between serum concentrations of apolipoprotein A5 and severity of coronary atherosclerosis

Objective To study the relationship between severity of coronary artery lesion and serum level of apolipoprotein A5 (ApoA5). Methods The level of serum total cholesterol (TC), triglyceride (TG), high-den-sity lipoprotein cholesterol (HDL-C ), low density lipoprotein cholesterol (LDL-C ), apolipoprotein A1 ( APOA1), apolipoprotein B (APOB) ,Lipoprotein(a) and uric acid(UA) were examined in 114 patients with coronary heart disease(CHD) and 40 healthy control subjects;Enzyme-linked immunosorbent assay (ELISA) methods was used to determine APOA5. The eoronay heart disease patients were divided into tree groups by the severity of coronary artery lesion: that is one, two and three vessel lesion. Results Compared with control group, APOA5, ApoA1, HDL-C lev-el of CHD groups were lower(P <0.01 or P<0.05) ,TG ,LP(a)and UA were higher(P <0.01 or P <0.05) ,the difference of TC, LDL-C and APOB were not significant (P>0.05 ). In the subgroups of CHD patients, The serum APOA5 concentrations were signficant different between the CHD patients and control group( F=18.605 ,P<0.01 ). Along with the severity of coronary artery,the level of ApoA5 concentrations had a lower trend. The level of ApoA5 was negatively correlated with serum TG level ( r=-0.208, P=0.040) and LP (a) ( r=-0.088, P <0.001). The level of APOA5 had a positive correlation with the serum HDL-C (r= 0.241, P = 0.016). Conclusion There is negative correlation between severity of coronary artery lesion and serum level of ApoA5. The decrease of ApoA5 maybe a risk factor.
9.A method based on cumulative toxicity for evaluating pharmacokinetics of oral traditional Chinese medicine in animals

AIM: To improve the acute cumulative death rates method (ACD method) in animal by oral administration. METHODS: A mathematic model was established to evaluate the dose-toxic effect relationship by twice oral administration and the experimental steps were improved too. The methodological quality was explored using the simulation data from computer program and the real experimental date from the reference paper. RESULTS: The results showed that the experimental data could be fitted to its theoretical data from LD_5/2 to LD_ 95/2. Concentration-time curve after po ordinary powder of Semen Strychni in mice were fitted to a one-compartment with T_ 1/2(ka)= 1.136 h,T_ 1/2(ke)= 7.100 h,and T_ max= 3.576 h. CONCLUSION: The improved ACD method can be used in the pharmacokinetics of TCM by oral administration.
10.Effect of compound salivia miltorrhiza injection on LPS-induced renal microvascular thrombosis

AIM: To evaluate the effects of compound salivia miltorrhiza injection on an experimental model of kidney thrombus induced by lipopolysaccharide(LPS).METHODS: The model of microvascular thrombosis in the rabbits' kidney was performed by the method of Hermida,which was induced by infusing LPS.Treatments were begun simultaneously with LPS infusion,through the contralateral marginal ear vein.Six different groups were established: NS 10(ml?h~(-1)) was infused as the negative control group,compound salivia miltorrhiza injection was infused with the dosage of(0.1)(Low-dose),(0.2)(medium-dose),and 0.4(high-dose)(ml?kg~(-1)?h~(-1)),heparin 600,000(IU?kg~(-1)?h~(-1)) as positive control group.The further rabbits, which were given neither LPS nor compound salivia miltorrhiza injection,were infused with saline solution through both marginal ear veins.The measurement of fibrinogen concentrations and platelet counts were used to assess the degradation of microvascular thrombosis.Kinney sections were examined for the presence of fibrin microthrombi.RESULTS: Compound salivia miltorrhiza injection was infused with the dosage of(0.1)(Low-dose),(0.2)(medium-dose),and(0.4)(high-dose)(ml?kg~(-1)?h~(-1)),and the fibrinogen concentrations and blood platelet counts were improved,and the fibrin deposition was degraded.CONCLUSION: Compound salivia miltorrhiza injection can inhibit effectively LPS-induced renal microvascular thrombosis.
