1.The True Academic Essence of Pro. Wu Kaopan's Studying and Disabusing the Classics

[Objective]To discuss professor Wu Kaopan’s academic thoughts by studying plain questions(Su Wen), Miraculous Pivot(Ling Shu), Medical Prob-lems. [Methods]Through the study on Wu’s papers and works of plain questions(Su Wen), Miraculous Pivot, using literature research method, sort and study these papers and works. [Results]This paper introduces Wu’s study on classics, using methods not only col ating, exegetics, annotation and modern translation, but also classification, analysis and exposition, the result is theoretical essence and academic ideas in such works are explored. [Conclusion]For researchers, the study of classics of Chinese medicine requires basic theory of it, what’s more, broad and profound literature knowledge is more important.
2.Comparison of four methods of fixation for intertrochanteric fractures in elderly patients

Objective To compare the internal fixation methods for treatment of intertrochanteric fractures in elderly patients.Methods One hundred and sixty four elderly patients with intertrochanteric fractures were treated from June 2006 to September 2011.The fractures were fixed with dynamic hip screws (DHS,n =42),locking proximal femur plate (LPFP,n =40),Gamma nails (n =36) or proximal femoral nail anti-rotation (PFNA,n =46),respectively.The clinical data were collected and the operative time,blood loss,time of clinical healing,postoperative complications and Harris hip scores of 6 months after surgery were compared among groups.Results All patients were followed-up for 6 to 36 months.The operative times of DHS,LPFP,Gamma nails and PFNA groups were (103 ± 15),(90 ± 13),(79 ± 11)and (65 ±9)min,respectively(F =2.87,P <0.05).The blood loss of 4 groups was (202 ±23),(181 ±23),(98 ± 13) and (87 ± 11) ml,respectively (F =3.21,P < 0.05).The times of clinical fracture healing were (16.1±1.9),(14.6±1.8),(12.9±1.7) and (11.5±1.4) weeks,respectively(F=2.66,P<0.05).The postoperative complications of 4 groups were 4/42,3/40,2/36 and 1/46,respectively (x2 =5.67,P <0.05).The Harris hip scores of 6 months after surgery of 4 groups were 81% (34/42),85% (34/40),89% (32/36)and 93% (43/46),respectively(H =20.03,P <0.05).Conclusions In treatment for intertrochanteric fractures in elderly patients,the efficacy of intramedullary fixation (Gamma nails and PFNA) is better than extramedullary fixation DHS and LPFP).PFNA is more effective than other 3 methods and should be preferentially chosen.
3.Changes in neural stem cell proliferation after rat cerebral ischemia/reperfusion injury is treated with mild hypothermia

Objective To study changes in neural stem cell proliferation in ischemic brain tissue after cerebral ischemia/reperfusion injury treated with mild hypothermia. Methods The middle cerebral arteries (MCA) of Sprague-Dawley rats were occluded for 2 hours, then reperfused for 3, 7, 11, 14, 18 and 28 days. Using immunohisto-chemical staining, the bromodeoxyur-idine ( BrdU)-positive cells in the brain tissue of the operationally intervened side were examined in a sham-operation group, a control group, and in rats treated using mild hypothermia. Results There were a few BrdU-positive cells in the sham-operated rats, but there were obviously more in the mild hypothermia group than in the control group. The peak period for proliferation of neural stem cells in the ischemic brain tissue was longer in the mild hypothermia group than that in the control group. Conclusion Mild hypothermia may promote proliferation of neural stem cells in ischemic brain tissue after cerebral ischemia/reperfusion injury.

Professor Wu Kaopan is a famous contemporary Chinese medicine scientist,Chinese medicine literature scientist,and Chinese medicine educator.This paper summarizes Professor Wu Kaopan's academic thoughts.Professor Wu Kaopan not only re-searched classics such as Basic Questions,The Spiritual Pivot,The Classic of Difficult Issues,Treatise on Cold Damage,and Essentials from the Golden Cabinet,but also proposed that we should rationally understand the role and value of the five elements theory;that the way to be a doctor should be familiar with the meridian theory;that we should know the constant and adapt to changes,and oppose the blind adherence to the five movements and six qi like looking for a noble steed to correspond with the one drawn;that the medicine should be effective and the method should be flexible.Professor Wu Kaopan has devoted his life to the inheritance and development of Chinese medicine,and has contributed his life to the fields of Chinese medicine teaching,scientific research,and clinical practice.
5.Effect of NPM1 mutant A on TGF-β1-induced K562 cell proliferation and AKT phos-phorylation

Objective: To investigate the effect of nucleophosmin 1 (NPM1) mutant A on TGF-β1-induced K562 cell proliferation and AKT phosphorylation. Methods: K562 cells were infected with Ad5-NPM1 to create an NPM1 over-expression cell model. NPM1 levels were determined by ELISA and Western blot analysis. The levels of AKT and P-AKT were determined by Western blot. MTT was used to measure the proliferation of K562 cells. Results: NPM1 protein levels in K562 cells increased in an Ad5-NPM1-MOI-dependent manner. Cell proliferation and NPM1 levels in the supernatant were significantly increased in K562 cells infected with Ad5-NPM1-30 and Ad5-NPM1-100 compared to those infected with Ad5-vector-100 (P<0.01). Treatment with (10 ng/mL) TGF-β1 increased P-AKT levels, but not total AKT levels in K562 cells. TGF-β1-induced phosphorylation of AKT was significantly increased by infection of K562 cells with Ad5-NPM1-100. No significant differences were found in total AKT levels among all groups. TGF-β1 (10 ng/mL) treatment also in-creased the proliferation of K562 cells. TGF-β1-induced K562 cell proliferation was significantly increased by infection with Ad5-NPM1-100 (P<0.01). Conclusions: NPM1 improves TGF-β1-induced cell proliferation by up-regulating AKT phosphorylation levels.
6.Research on the storage-expelling characteristic of kidney in visceral manifestation and the theory of "retaining essence and removing turbidity"

The theories of "absence of kidney excess syndrome" and "treating kidney disease only by the tonifying method without the purgative method" have been highly praised by medical practitioners throughout history. However,the syndrome of intermingled deficiency and excess of kidney in clinical practice is not uncommon,and there are many cases that treating kidney disease with the method of expelling kidney turbidity. However,its theoretical basis has not been systematically organized and studied. This article,based on the consideration of the "three purging" herbs of Liuwei Dihuang Pill,finds that "kidney" in traditional Chinese medicine not only stores essence,but also removes turbidity. From the theory of storage-expelling of five zang viscera,the physiological characteristic of kidney is integrated with storing and expelling functions. In addition to storing essence and qi to maintain normal life activities in the human body,it also exercises the function of excretion through the bladder and sanjiao. Under pathological conditions,both the function of storing essence and excretion of turbidity in kidney can be affected,which manifests as lack of essence and accumulation of turbidity. The pathological features of this disease are characterized by kidney qi deficiency as the root cause,accumulation of turbidity as the result,and intertwined with each other. Based on the physiology and pathology of visceral manifestation of kidney in traditional Chinese medicine,the theory of "retaining essence and removing turbidity" can be used in the clinical diagnosis and treatment of kidney disease. When treating kidney disease,the method of invigorating the kidney can also be combined with the method of expelling kidney turbidity,such as promoting diuresis,dissipating phlegm,promoting blood circulation,or removing toxin. Flexible selection of the above treatment methods can improve the therapeutic effect.
7.Summary of Literature Research and Utilization of Unknown Prescriptions

Nameless prescriptions are an important component of traditional Chinese medicine literature,with a large number and wide distribution in various types of literature such as Chinese medical literature,excavated medical literature,stone inscriptions,Dun-huang Turpan medical literature,and overseas returned medical literature.The research on the literature organization of unnamed pre-scriptions involves multiple perspectives and levels of research,including the screening of the sources of unnamed prescriptions,the study of citation and loss,the study of unnamed prescription drugs,the study of disease names and treatment standards,the study of the evolution of unnamed and famous prescriptions,the study of dosage forms and usage,and the identification and application of au-thentic and fake literature.Organizing literature on anonymous prescriptions throughout history,tracing the academic development of a-nonymous prescriptions,and summarizing the characteristics and characteristics of anonymous prescriptions are of great significance for promoting research on prescription literature and further utilization of anonymous prescriptions.
8.Radiation dosimetry and biodistribution of 68Ga-FAPI-04 PET in patients with hepatobiliary tumor

Objective:To investigate the radiation dosimetry and biodistribution of 68Ga-FAPI-04 PET/CT in patients with hepatobiliary tumor. Methods:A total of six patients with hepatic lesions who underwent PET/CT examination in Peking Union Medical College Hospital were enrolled. After intravenous injection of radiotracer 68Ga-FAPI-04 at (170.57 ± 14.43) MBq, whole-body imaging were performed at the time points of 3, 10, 15, 20, 30 and 60 min, respectively. Biodistribution pattern was observed. Regions of interest were manually delineated. Radiation dosimetry of all target organs were calculated by Olinda/EXM software. Results:The radioactive uptake dissipated gradually in liver whereas it was relatively stable in tumor lesions. The average SUV max of tumor lesions reached the maximum value (13.87± 2.55) at 20 min after injection. The target-to-background ratio increased with time, reaching the maximum value (10.09 ± 8.17) at 30 min after injection. The average effective dose in total body was (0.020 ± 0.002) mSv/MBq and organ with the highest effective dose was bladder wall at (0.146 ± 0.035) mSv/MBq. Conclusions:The effective dose in total body of 68Ga-FAPI-04 was similar to that of 18F-FDG. 68Ga-FAPI-04 is expected to be a PET/CT radiotracer for hepatobiliary tumors in consideration of rapid tumor uptake, low accumulation of liver background, and no influence of blood sugar levels.
9.Construction of mouse intestinal organoid inflammation model

Objective To establish in vitro the small intestinal organoid culture system and to investigate the effect of lipopolysaccharide(LPS)on the growth of small intestinal organoids and the secretion of inflammatory factors.Methods In vitro,the small intestinal crypt cell mass of C57BL/6 mice was aseptically isolated,collected and embedded in organoid matrix.Under the support of complete medium,the small intestinal organoids with three-dimensional multi-leaf structure with small intestinal epithelioid structure were formed.The small intestinal organoids were subcultured after 5-7 d culture.On the third day after passage,the small intestinal organoids were randomly divided into different mass concentrations of LPS groups(0,150,175,200,225,250,275 and 300 mg/L).After 24 h and 48 h of LPS induction,morphological changes of small intestinal organoid growth and differentiation were observed.CCK-8 method was used to detect the effect of different time points and mass concentrations of LPS on the proliferative activity of small intestinal organoids after induction of inflammation.The effects of four different mass concentrations of LPS(0,175,200 and 225 mg/L)on expression levels of granulocyte-macrophage colony stimulating factor(GM-CSF),interleukin(IL)-1α,IL-6 and IL-10 in organoid culture supernatant at different times were detected by enzyme-linked immunosorbent assay(ELISA).Results The mouse small intestinal organoid culture system was preliminarily constructed.After different time and mass concentration of LPS induced inflammation of small intestinal organoids,it was observed by morphology that small intestinal organoids would have different degrees of expansion and apoptosis in lumen.The proliferation,differentiation and budding of damaged intestinal epithelial crypts or intestinal stem cells were also inhibited to varying degrees,indicating that the growth of small intestinal organoids would be limited to varying degrees after induced inflammation.The proliferation activity of small intestinal organoids decreased to varying degrees after 24 h and 48 h of LPS induction at 175-225 mg/L(P<0.05),but the cell viability was still greater than 50%.The levels of IL-1α,IL-6 and GM-CSF partially increased after induction with 200 mg/L and 225 mg/L LPS for 24 h and 48 h(P<0.05).The level of IL-10 decreased after induction with 200 mg/L LPS for 24 h and 48 h(P<0.05).Conclusion In this study,a model of intestinal inflammatory injury in vitro induced by LPS with different mass concentrations and time points is preliminarily constructed,which provides a more reliable research platform for the mechanism research of intestinal diseases and the screening of effective drugs in the future.
