1.Effect of C-phycocyanin from Spirulina platensis on erythropoiesis in mice

To investigate the effects of C-phycocyanin(C-PC) from Spirulina platensis on erythropoiesis in mice.The colony forming uniterythroid(CFU-E) and burst forming unit-erythroid(BFU-E) were determined using micro-methycellulose culture method in vitro.The erythropoietin(EPO)-like activity of C-PC was examined using the technique of CFU-E culture of fetal liver cells in mice in vitro.Anemic mice models were established by ~(60)Co ?-ray irradiation(5Gy) and intra-peritoneal injected with benzohyarazine hydrochloride.After normal mice were intra-peritoneal injected with C-PC (50 mg?kg~(-1)) for 5d,C-PC provided the only increase in the numbers of CFU-E and BFU-E-derived colonies.C-PC exhibited higher EPO-like activity.12.5 ng of C-PC could match with EPO 1.06U.The specific activity of C-PC was 84,800 U?mg~(-1) C-PC.When anemic mice were intra-peritoneal injected with C-PC for 5d,on d10 the leukocyte,erythrocyte and hemoglobin were significant increased.
2.Leukocyte Chemotaxis in Filter Assay and Its Affected Factors

In this paper, polymorphonticlear leukocyte chemotaxis filter assay and its affected factors have been studied in accordance with the practical conditions.The results indicated that leukocyte chemotaxis may be detected by using Boycen chemotaxischamber prepared in our laboratory and filters made of mixed esters of cellulose athome.The appropriate experimental conditions are as follows:Single filter methodselected the filter of 3?m pore size.The cells were suspended in Eagle medium at aconcentration of 1?1.5/ml.The chamber was incubated at 37℃ in humidified airfor 120 min.Optimal concentration of bacterial culture filtration,zamsan activatedserum and lymphocyte derived chemotactic factor had eqeal cytotactic potency. Leukocyte chemotaxis is quantitated by counting the number of migrating cells atvarious depths within the filter.The mean number of migrating cells toward chemo-tactic factor is usually about twice as many as that of medium of control.Themodified method which may be acted as an index for investigating leukocyte functionin the experimental immunology and clinical research.
3.EFFECT OF C - PHYCOCYANIN ON GRANULOPOIESIS AND MONOCYTOPOIESIS IN MICE

C - phycocyanin (C - PC) was isolated and purified from blue - green alga Spirulina platensis. By using the thechnique of culture of hemopoietic progenitor cell in vitro, we studied the effect of C -PC on granulopoiesis and monocytopoiesis in mice. The results indicate that C -PC can enhance the proliferation of CFU -GM, promote the granulopoiesis and monocytopoiesis in mice. The C -PC can markedly increase the colony stimulating activity of serum in normal mice. The prepared serum -free spleen cell conditioned media stimulated by C -PC in vivo and in vitro provided the only increase in CFU - GM colony formation. After in -vitro addition of C -PC in the presence of GM -CSF, there was a pronounced increase in the number of CFU -GM colonies.
4.THE EFFECTS OF EXOPOLYSACCHARIDES FROM APHANOTHECE HALOPHYTICA (EAH ) ON IMMUNO-LOGICAL FUNCTION OF MICE

The effects of exopolysaccharides from Aphanothece halophytica(.EAH) on immunological function were studied by immunological techniques. The results showed that EAH could not only enhance the mitogenic response of mice's thymus cells, murine bone marrow cells proliferation,murine mixed lymphocyte reaction and the activity of NK cells in mice,but promote the interleukinl(IL-1)production and increase the activity of 1L-2 as well. The study suggests that EAH can improve the immunological function of mice.
5.Effect of Preoperative Regional Intra-Arterial Infusion Chemo therapy on the Prognosis of Advanced Gastric Cancer

Objective To evaluate the effect of p reoperative regional intra-arterial infusion chemotherapy on the prognosis of a dvanced gastric cancer. Methods The clinical data of 80 patients who u nderwent curative resection for advanced gastric cancers were summarized.Among t hem,33 patients carried out preoperative regional intra-arterial infusion chemo therapy were as the interventional chemotherapy group,and the remaining 47 patie nts were the control group.Eleven factors including clinical and pathological da ta,treatment procedures and molecular biological makers that contributed to the long-term survival rate were analyzed by Cox multivariate regression analysis.Results The 5-year survival rate of the interv entional group was 59.3%,and the control group 47.6% .There was significant difference between two groups ( P
6.Endoscopic diagnosis and therapy for patients with relapsing pancreatitis after cholecystectomy

Objective To investigate the endoscopic diagnosis and therapy for patient with relapsing pancreatitis after cholecystectomy. Methods The clinical data of 21 patients with relapsing pancreatitis after cholecystectomy underwent endoscopic retrograde cholangiopancreatography (ERCP) and endoscopic sphincterotomy (EST) were analyzed. Results Nineteen out of 21 patients were diagnosed as sphincter of Oddi dysfunction (SOD), and remaining 2 patients as choledocholith iasis. The treatment outcome of EST for the 21 patients in short-term after EST was satisfactory, and there was no complication of EST. Conclusions ERCP has a great value in the diagnosis of the cause of relapsing pancreatitis after cholecystectomy.The treatment of EST for patients with relapsing pancreatitis after cholecystectomy is safe and effective.
7.The protective effect of tetramethylpyrazine on rat myocytes with transient oxygen-glucose deprivation followed by reoxygenation injury and mechanism study

Objective This study explored the protective effect of tetramethylpyrazine on myocardial ischemia/reperfusion injury via inhibiting oxidative stress. Methods Primary cultured neonatal myocytes were applied to explore the anti-ischemia/reperfusion injury property in vitro. The survival viability of myocytes was determined by MTT; enzyme activities such as lactate dehydrogenase, creatine kinase, superoxide dismutase, malondialdehyde, and nitric oxide were analyzed with assay kits; inducible nitricoxide synthase and endothelial nitric oxide synthase expressions were determined by Westernblot. Results Tetramethylpyrazine significantly improved the beating frequencies of myocytes after oxygen-glucose deprivation/reoxygenation procedure, decreased lactate dehydrogenase, creatine kinase, and malondialdehyde levels and enhanced superoxide dismutase activity.Tetramethylpyrazine also inhibited excessive production of nitric oxide through downregulating inducible nitric oxide synthase as well as upregulating endothelial nitric oxide synthase during ischemia/reperfusion injury. Conclusion Tetramethylpyrazine could significantly improve the oxidative-stress tolerance of myocytes to keep cell membrane integrity and protect the myocardial tissue of normal physiological function via an antioxidant effect and by restoring the balance between inducible nitric oxide synthase and endothelial nitric oxide synthase, while inhibiting the generation of cytotoxic concentrations of NO.

Objective To observe the effects of selenium extracellular polysaccharide of Porphyridium sp(Se-PSP) on the growth and apoptosis of BEL-7402 tumor cell line.Methods The Se-PSP was obtained from a sort of culture medium with some selenous acid solution.In vitro,human tumor cells were cultured in sorts of PSP or Se-PSP liquors with different concentrations,and the result of which was determined by MTT,Focus apoptosis assay and DNA fragmentation.Results and Conclusion Se-PSP(80mg?L-1) displayed strong growth inhibitory effects against BEL-7402 human cancer cell line,and induced the apoptosis of BEL-7402 cells.

10.Pathological Change in Hydatid Cysts of Echinococcus granulosus Treated with High Intensity Focused Ultrasound

Objective To investigate the pathological change in Echinococcus granulosus hydatid cysts treated with high intensity focus ultrasound (HIFU). Methods Thirty cysts with thinner wall and proper elasticity, taken from livers of infected sheep, were randomly divided into three groups. By cyclical multilayer radiation around the cyst wall,two experiment groups were treated with HIFU under 150 W and 250 W sound power respectively. The control group was treated by ordinary ultrasound for 2 min. Results The inner cyst wall of hydatid treated with HIFU became curved,thicker, stiffer, white and less transparent. The germinal layer was detached mostly from the laminated layers of hydatid in the experiment groups. Images from the transmission electron microscopy showed that in the experiment groups fabric texture of hydatid changed significantly and germinal cells were broken. Conclusion HIFU in a model of cyclical multilayer radiation causes pathological damage of the E. granulosus hydatid.
