1.Clinico-pathological and biological significance of human kallikrein gene 6 mRNA expression in breast cancer

objective To quantify the expression of kallikrein gene 6 (KLK6) in malignant and benign breast tissues and to statistically analyze the relationship between KLK6 expression levels and the clinico-pathological variables in patients with breast cancer. Method Paired breast tissue samples of cancerous and corresponding non-cancerous tissues were obtained from 48 patients with breast cancer who underwent surgical resection. Quantitative analysis of KLK6 mRNA expression was done using real-time quantitative reverse transcription-PCR (RT-PCR). The relationship between KLK6 expression intensity and various clinico-pathological variables was analyzed. In addition, the technique of small interfering RNA-mediated gene silencing was used to suppress the KLK6 expression in MCF-7, and to investigate the effects of KLK6 on cell proliferation rate and cell cycle. Results KLK6 mRNA over expression was found in cancerous tissues in 35 out of 48 patients (73%) as compared with normal breast tissue. Objective To analyze the effects of Xiaogan capsule, Xiaoyao pill combined with Shu er capsule on hyperplasia of mammary gland and its effect on vascular endothelial growth factor ( VEGF ) and basic fibroblast growth factor ( bFGF ) .Methods 106 cases of patients with breast hyperplasia treated in our hospital from April 2013 to August 2016 were randomly divided into experimental group and control group according to the order of admission, with 53 cases in each group.The control group was treated with Xiaojin Capsule and Xiaoyao Pills.The experimental group was treated with Xiaojin Capsule, Xiaoyao Pill and Shuerjing Capsule.Analyze the total effective rate of clinical treatment, incidence of adverse reactions, visual analogue pain score and the influence on the expression of VEGF and bFGF in the two groups.Results The total effective rate was 96.23%(51/53) in the experimental group and 86.79%(46/53) in the control group,the total effective rate in the experimental group was much higher than that in the control group (P<0.05).The incidence of adverse reactions was 11.32%(6/53) in the experimental group and 13.21%(7/53) in the control group,there was no significant difference in adverse reactions between the two groups.The visual analogue pain score of the experimental group was (2.85 ±0.41) points, the visual analogue pain score of the control group was (5.25 ±1.84),the visual analogue pain score of the experimental group was significantly lower than that of the control group ( P <0.05 ) .After treatment, the expression of VEGF and bFGF in the patients with hyperplasia of mammary gland decreased, and the levels of VEGF and bFGF in the patients with hyperplasia of mammary glands were significantly higher than those in the untreated group (P<0.05),the expression of VEGF and bFGF in the control group was enhanced.Conclusion Xiaojin capsule,Xiaoyao pill and Shu er jing capsule have higher application value in the patients with hyperplasia of mammary gland , and significantly lower the expression of VEGF and bFGF .
3.Role of nitric oxide in hemorrhagic shock/reperfusion and the effect of taurine

AIM: To investigate the effect of nitric oxid e (NO) and taurin on hemorrhagic shock /reperfusion injury. METHODS: Twenty-four rabbit s were divided randomly into 3 groups ( n= 8): control group and shock group and taurine group. The model of hemorrhagic shock/reperfusion was used. The activities of ni tric oxide synthase (NOS),superoxide dismutase(SOD),lactate dehydrogenase (LDH)and the contents of malondialdehyde(MDA),nitric oxide pvoducts( NO - 2/NO - 3) in plasma were observed before shock and shock 1.5 h,reper fusion 1 h ,2 h and reperfusion 3 h. RESULTS: ①During shock/reperfusion, the ac tivities of NOS,LDH and the contents of MDA,NO - 2/NO - 3 were significan tly highe r, but the activity of SOD was significantly lower in plasma of shock group than that of before shock and shock 1.5 h. ②At 3 h reperfusion, the activity of NOS and the contents of MDA, NO - 2/NO - 3 were significantly higher,bu t the act ivity of SOD was significantly lower in the lung and heart of shock group than t hat of control group. ③ Taurine(40 mg?kg -1 ,iv) attenuated all the c hanges above mentioned at total time points of reperfusion. ④ A close correlation was shown between MDA content and NO - 2/NO - 3 content in plasma, lung and i n heart. CONCLUSION: These results suggeste that NO may be involved in oxida nt-mediated shock/reperfusion, antagonistic effect of taurine on hemorrhagic shock/reperfusi on injury may be relate d to decreasing the generation of NO and anti-lipoperoxidation.
4.Clinical study on human kallikrein gene 5 messenger RNA expression in breast cancer

Objective The purpose of the current study was to develop a real-time quantitative reverse transcription-PCR method for detection of human kallikrein gene 5(KLK5) mRNA expression,quantify the expression of KLK5 mRNA in malignant and benign breast tissues,and statistically analyze whether KLK5 expression levels correlate with clinicopathologic variables in the patients with breast cancer.Methods Paired breast tissue samples from cancerous and corresponding noncancerous tissues were obtained from 48 patients with breast cancer who underwent surgical resection.The relationship of KLK5 mRNA expression status with various clinicopathologic variables were quantitativly analyzed by real-time RT-PCR.Results KLK5 mRNA expression in cancerous tissues was decreased in 38 of 48(79%) breast cancer patients compared with normal counterparts.The mean expression level of KLK5 mRNA in cancerous tissues was significantly lower than that in noncancerous tissues(P0.05).Conclusion The results of this study indicated that KLK5 mRNA expression was significantly lower in cancerous tissues than that in noncancerous breast tissues,and low expression of KLK5 mRNA correlated with TMN stage,metastasis and ER status.
5.Association of dietary niacin intake with metabolic syndromeamong adults in Zhejiang Province

Objective :
To explore the association of dietary niacin intake and metabolic syndrome ( MS ) and its components among adults in Zhejiang Province, so as to provide the reference for dietary intervention of MS.
Methods:
Using the multi-stage cluster random sampling method, the permanent residents aged 18 years and above were selected and investigated by a questionnaire developed by China Center for Disease Control and Prevention. Their waist circumference, blood pressure, glucose and lipid were measured. The daily dietary niacin intake of each person were calculated by "24-hour dietary review for 3 consecutive days", and divided into Q1, Q2, Q3 and Q4 groups according to quartiles. The multivariate logistic regression model was used to analyze the association of niacin intake with the risks of MS and its components.
Results:
Among 2 438 participants, 871 cases with MS were detected, with a detection rate of 35.73%. The multivariate logistic regression analysis showed that compared with niacin intake Q1 group, Q2 ( OR=0.741, 95%CI: 0.561-0.978 ) and Q4 group ( OR=0.679, 95%CI: 0.487-0.947 ) had a lower risk of MS, Q2 ( OR=0.688, 95%CI: 0.516-0.919 ) and Q4 group ( OR=0.678, 95%CI: 0.479-0.960 ) had a lower risk of abdominal obesity, Q4 group ( OR=0.721, 95%CI: 0.536-0.969 ) had a lower risk of hyperglycemia. Further stratificating by gender, compared with niacin intake Q1 group, Q2 ( OR=0.664, 95%CI: 0.453-0.972 ) and Q3 group ( OR=0.646, 95%CI: 0.432-0.965 ) in women had a lower risk of MS, Q2 (OR=0.667, 95%CI: 0.460-0.967) and Q3 group ( OR=0.607, 95%CI: 0.408-0.902 ) had a lower risk of abdominal obesity; Q2 group ( OR=1.836, 95%CI: 1.202-2.805 ) in men had a higher risk of low high-density lipoprotein cholesterol.
Conclusion
Niacin intake is associated with an increased risk of MS, abdominal obesity and hyperglycemia.
