1.Experimental study in microwave tissue coagulation of hepatic VX_2 carcinoma with power Doppler and contrast-enhanced ultrasonography

Objective To evaluate the value of power Doppler and contrast-enhanced ultrasonography in the detection of tumor after microwave tissue coagulation (MTC). Methods Ultrasound Microwave Theraphy-1 (UMT-1) equipment was used,and a 9 mm length antenna was inserted directly into the tumor at laparotomy in 10 VX 2-bearing rabbits. The energy output was 60 W?120 s per bout. Two-dimensional, power and contrast-enhanced (Levovist, 300 mg/ml, 0.3 ml/kg) imaging was performed pre- and post-treatment. Pathology examination was applied in contrast. Results Along the needle being narrow hyper echoic imaging, broad-equable hypo echoic imaging was observed around the center after MTC immediately and the edge of tumor was unclear. Intratumoral blood flow was observed in 4 VX 2 tumors with power ultrasonography while there was enhanced-signal in 10 VX 2 tumors using contrast-enhanced ultrasonography. These tumors were confirmed incomplete necrosis by pathology. Conclusions Contrast-enhanced power Doppler ultrasonography shows more sensitive in assessing the therapeutic effect of microwave on VX 2 tumor. Residual tumor can be seen as persistent color enhancement by using contrast-enhanced ultrasonography.
2.Experimental study of acetic acid injection and microwave coagulation in treatment of liver VX_2 tumors of rabbit

Objective To compare the effectiveness of acetic acid injection(AI) and microwave(MW) coagulation in the treatment of rabbits hepatic VX 2 tumors. Methods Eighteen rabbits with VX 2 tumor were divided into two groups randomly,one was treated by precutaneous acetic acid injection,and the other by microwave at laparotomy. Contrast-enhanced ultrasonography (Levovist) was used to assess the therapeutic effect after treatment. Pathologic changes were observed chronologically. Results Anomalo-necrosis areas including tumors and surrounding normal liver tissue were observed after AI,multiple point or sheet necrosis was confirmed by pathology. Long oval-shap coagulated areas were observed after MW,incomplete necrosis after treatment was immediately confirmed by pathology and changed to complete necrosis in two weeks. Conclusions AI is more effective in the treatment of liver tumors,but diffuses easily to normal liver and the necrosis area is not stable. MW thermal field is steady and necrosis area is reliable.
3.Research progress of bone microarchitecture and microdamage detection

BACKGROUND:Bone fragility and poor bone quality due to osteoporosis are a major and increasing concern. Bone microarchitecture and microdamage, the important factors of bone quality, their detection technology and instrument have experienced a long development process. OBJECTIVE:To give a brief introduction of the concept of the bone microarchitecture and microdamage, then to summarize the research progress of their detective methods. METHODS:PubMed and CNKI databases were retrieved for reviews and articles related to bone microarchitecture and microdamage published from January 1990 to June 2016 using the keywords of“bone microarchitecture, bone microdamage and detect/detective/detecting”in Chinese and English, respectively. Finaly a total of 65 articles were selected for overview. RESULTS AND CONCLUISON:(1) Bulk staining is a quick and useful way to confirm and assess linear microcracks and diffuse damage. Micro-CT and confocal microscopy al ow visualization at the micron scale, and are useful tools to understand the three-dimentional nature of bone microdamage. Scanning electron microscope lacks the ability to investigate large regions of microdamage, but al ows users to probe in extensive details at the nano scale. (2) Ultimately, we recommend the use of multiple imaging modalities according to the experimental needs to obtain useful information about bone quality and microdamage formation, across the scales of hierarchy in bone.
4.Effects of four kinds of ceramic casting alloys on P70S6k expression of human gingival fibroblasts

BACKGROUND: It has been confirmed that some casting alloy can inhibit the activity of human gingival fibroblasts in vitro, which can be used for inducing apoptosis of human gingival fibroblasts (HGFs). However, the mechanism of this effect is poorly understood.OBJECTIVE: To investigate the effect of Ni-Cr, Co-Cr, Au and pure Ti ceramic alloys on the expression of P70S6k by HGFs in vitro.DESIGN, TIME AND SETTING: Grouping controlled experiments with the expression of P70s6k by HGFs in each group as observation objects. Experiments were performed at the Institute of Immunohistochemistry and Western blotting, Technology Building of Liaoning Medical University from July to October 2009.MATERIALS: Ni-Cr, Co-Cr, Au and pure Ti ceramic alloys were processed by Shenzhen Mecodent Dental Laboratory Co., Ltd. The gingival from gingival neck of the first premolar (which goes to be extracted due to the need of orthodontic treatment) of teenager who was15 years old without any clinical disease was obtained, followed by the primary culture of HGFs. The fifth passage of cells in logarithmic phase were used to measure indexes.METHODS: Ni-Cr, Co-Cr, Au and pure Ti ceramic alloys were incubated in DMEM to prepare alloy leaching liquor, which were then added in HGF medium at 10% concentration. DMEM containing 10% fetal bovine serum was used in negative controls. MAIN OUTCOME MEASURES: The expression of P70S6k in each group was measured by Western Blotting following 72 hours of intervention with alloy leaching liquor.RESULTS: Western-blot results showed that there was no significant difference in the average gray value of P70S6k expression in Au, pure Ti ceramic alloys and control groups (P > 0.05). There was also no significant difference between Au ceramic alloys and pure Ti ceramic alloys groups or between Ni-Cr ceramic alloys and Co-Cr ceramic alloys groups. (P > 0.05), but Ni-Cr ceramic alloys group or Co-Cr ceramic alloys group showed significant difference compared with the Au, pure Ti ceramic alloys and control groups (P < 0.01).CONCLUSION: Au ceramic alloys and pure Ti ceramic alloys show that they have no obvious effect on the proliferation activity of HGFs. Ni-Cr and Co-Cr ceramic alloys showed an inhibitory effect on the proliferation activity of HGFs.
5.Effect of Faecalibacterium prausnitzii on Treg Cells and Cytokines in Colitis Mice with LFA-1 Knockout

It has been widely accepted that Faecalibacterium prausnitzii (Fp) induces the differentiation of Treg cells.Lymphocyte function-associated antigen-1 (LFA-1) is also involved in the differentiation of Treg cells.Aims: To investigate the effect of Fp on Treg cells and cytokines in colitis mice with LFA-1 knockout (LFA-1-/-).Methods: Twenty wild type mice and twenty LFA-1-/-mice with same genetic background were randomly divided into wild type control group, wild type treatment group, LFA-1-/-control group and LFA-1-/-treatment group.Colitis model was induced by drinking DSS solution.Mice in the two treatment groups were intragastrically administrated with Fp.General status and histopathological score were assessed.Percentages of Treg cells in spleen and mesenteric lymph nodes were measured by flow cytometry.Serum levels of IL-10 and TGF-β1 were measured by ELISA.mRNA expressions of IL-10 and TGF-β1 in colonic tissue were detected by real time PCR.Results: Compared with corresponding control groups, histopathological score was significantly decreased in wild type treatment group (P<0.05);percentages of Treg cells in spleen and mesenteric lymph nodes were significantly increased (P<0.05), serum levels of IL-10 and TGF-β1 were significantly increased (P<0.01), expression of TGF-β1 mRNA was significantly increased in wild type treatment group and LFA-1-/-treatment group (P<0.05);expression of IL-10 mRNA was significantly decreased in LFA-1-/-treatment group (P<0.01).Compared with wild type treatment group, serum level of TGF-β1 was significantly decreased (P<0.05), and mRNA expressions of IL-10 and TGF-β1 were significantly decreased in LFA-1-/-treatment group (P<0.05).Conclusions: Fp can up-regulate the percentages of Treg cells and enhance the secretion of anti-inflammatory cytokines IL-10 and TGF-β1 in LFA-1-/-mice.The therapeutic efficacy for colitis in wild type mice is superior to that in LFA-1-/-mice, which may be related to the inhibition of function of Treg cells and secretion of cytokines due to LFA-1 knockout.
6.Screening of host proteins interacting with GP, VP30, NP,and L proteins of Ebola virus by yeast two-hybrid system

Objective To study the biological function of GP,VP30,NP and L proteins in Ebola virus(EBOV) infectious diseases,and to screen the host proteins which interact with GP,VP30,NP,and L proteins of EBOV from human liver cDNA library using yeast two-hybrid technique.Methods Recombinant PCR was used to construct bait plasmids pGBKT7-GP,pGBKT7-VP30,pGBKT7-NP and pGBKT7-L.Bait strains were combined with human liver cDNA library strains.The positive clones were analyzed by DNA sequencing and bioinformatics.A yeast recovery experiment was performed to further verify and exclude false positive results.Results We constructed bait plasmids pGBKT7-GP,pGBKT7-VP30,pGBKT7-NP and pGBKT7-L with the recombinant PCR method.Six host proteins which could interact with GP,VP30,NP,and L proteins were screened,including COMMD1,ALB,PSMD8,APOA2,CYP2E1,and HP.The yeast recovery experiment proved that COMMD1 and APOA2 might interact with NP protein.Conclusion A number of prey proteins which interact with GP,VP30,NP,and L proteins of EBOV are screened,which may provide reference for the research of EBOV.
7.Diagnostic value of endoscopic ultrasonography for large gastric folds

Objective To evaluate endoscopic ultrasonography (EUS) for diagnosis of large gastric folds. Methods EUS was performed in 66 patients with possible large gastric folds which could not be diagnosed by conventional gastroscopy. The characteristics of EUS findings were analyzed, and the EUS results were compared with pathological findings to evaluate the sensitivity, specificity and accuracy of EUS. Results The diagnostic sensitivity, specificity and accuracy of EUS for diffused infiltrated gastric cancer were 92.3%(24/26), 95.1% (39/41) and 95.5% (63/66), respectively, and those for gastric lymphoma were 92. 3% ( 12/13), 96. 1% (49/51) and 92. 4% (61/66), respectively. The accuracy in differential diagnosis of benign and malignant disease was 93.9% (62/66). There were statistical differences between benign and malignant large gastric folds in characteristics of EUS findings, including width of gastric wall, enlargement of muscularis propria and preservation of gastric layer ( P < 0. 05 ). Conclusion EUS is of high accuracy for diagnosis of large gastric folds, especially for diffused infiltrated gastric cancer and gastric lymphoma. Such features as width of gastric wall, the enlargement of muscularis propria and the preservation of gastric layer are important features for differential diagnosis of benign and malignant lesions.
8.Value of a diagnostic criterion of capsule endoscopy for small bowel Crohn's disease

Objective To investigate the value of a diagnostic criterion of capsule endoscopy (CE)for small bowel Crohn's disease (CD). Methods The clinical data of 50 patients with suspected CD who underwent CE from March 2003 to October 2008 were analyzed retrospectively. Diagnoses were made based on a criterion proposed by De Bona et al. Patients were grouped and followed up for 1 to 5 years. Results A total of 50 patients were enrolled and divided into 3 groups according to the results of CE. Group A included 25 patients as having small intestine CD on CE, in which 3 received surgery and was confirmed by pathology, and the other 22 got improved after treatments for CD. Group B was comprised of 15 patients who were clinically diagnosed as small intestinal CD and received experimental treatments, in which 13 patients improved and 2 others received surgery and were confirmed to be small intestine diverticula. Patients in group C ( n = 10) were diagnosed as non-specific enteritis and received medication of Bifico and metronidazole for at least 3 months. All patients were followed up for 1 to 5 years and no recurrence was observed. Conclusion This diagnostic criterion of CE for small bowel CD enables early diagnosis of the disease.
9.Quantitative echo-tracking technique for evaluating carotid arterial elasticity function in patients with systemic lupus erythematosus

Objective To explore the clinical value of early diagnosis of atherosclerosis in patients with systemic lupus erythematosus (SLE) using vascular echo-tracking technique and to detect changes of elastieity of carotid artery quantitatively in SLE patients.Methods Fifry patients with SLE were divided into SLE1 group(duration≤1 year),and SLE2 group(duration>1 year)based on different course.An ultrasonic echo-tracking method was used to measure patients'pressure strain elastic modulus (Ep),the stiffness constant(β),arterial compliance(AC),augmentation index(AI),pulse wave velocity (PWVβ) and intimamedia thickness(IMT)of the common carotid arteries in 50 patients with SLE and in 25 healthy control subjects.Results Among carotid artery elasticity indicators of three groups,there was no significant difference in AI(P>0.05).Ep,β,PWVβ parameters of SLE1 group and SLE2 group were statistically higher than that of the control group[Ep of SLE1 group,SLE2 group,control group was (69±20),(103±40),(48±18)kPa respectively;β was 5.2±1.9,8.0±3.1,4.2±1.3 respectively;PWVβ was 5.2±0.7,6.3±1.1,4.5±0.7]respectively,but AC(AC of SLE1 group,SLE2 group,control group was(1.1±0.3),(0.8±0.3),(1.2±0.6)mm2/k respectively]lower than the controls(P<0.01).Ep,β,PWVβ in SLE2 group was significantly increased compared with SLE1 group,but AC was decreased (P<0.01).Conclusion The application of echo-tracking technology can be used to diagnose early atherosclerosis.Complications of cardiovascular disease in SLE have high clinical value.
10.In vitro studies of glypican-3 targeting pretargeting technology for molecular MRI of hepatocellular carcinoma

Objective To explore the value of pretargeting technology in vitro MRI of L5 peptide guided streptavidin-conjugated and polyethylene glycol modification protected ultra-small superparamagnetic iron oxide(SA-PEG-USPIO) to hepatocellular carcinoma(HCC) via glypican-3(GPC3) receptor.Methods Direct immumofluorescence assay with carboxyfluorescein(FAM) labeled L5 and competitive inhibition was performed in HepG2 and HL-7702 cells.Imaging was obtained from fluorescent microscope.Immunoassay fluorescence images were carried out to determine the expression of GPC3 in HepG2 cell.PEG-USPIO conjugated with streptavidin was made by carbodiimide reaction,and the hydrodynamic diameters,Zeta potential and magnetic relaxivity of SA-PEG-USPIO and PEG-USPIO were measured.HL7702 cells were used for evaluate cells viability of SA-PEG-USPIO and PEG-USPIO.HepG2 and HL-7702 cells were used as experimental and control group respectively.Each of the two cell lines were further divided into three groups:L5-BT united SA-PEG-USPIO group,SA-PEG-USPIO group and control group.Prussian blue staining and MRI was preformed to observe the targeting efficacy of SA-PEG-USPIO respectively,and normalized T2 signal was recorded.The significant changes of normalized T2 signal intensity among groups was deterumine by using One-way analysis of variance.Results There were much more fluorescences on the membrane and cytoplasm of HepG2 cells than those on HL-7702 cells and cells of competition group.And indirect immunofluorescence images show the obvious expression of GPC3 in HepG2 cell.The SA-PEG-USPIO and PEG-USPIO nanoparticles had hydrodynamic diameters of (22.73 ± 3.31) and (35.97±5.19)nm,Zeta potential of them were (4.22±0.53) and (-7.91± 1.22)mV and magnetic relaxivity were 0.139 4× 103 and 0.103 9 × 103 mM-1s1.Although the highest concentration of SA-PEG-USPIO and PEG-USPIO was 2.4 mmol/L,cells viability was greater than 80%.The most iron particle was observed in L5-BT united SA-PEG-USPIO group of HepG2 cells.In vitro MR,the normalized T2 signal intensity of HepG2 cells in L5-BT united SA-PEG-USPIO group,SA-PEG-USPIO group and control group were 39±7,77 ± 12 and 93 ± 4.There was significant difference among those three groups (F=23.96,P<0.01).The normalized T2 signal intensity of HL-7702 cells in each of three groups were 69± 11,78±8 and 95±5.There was no significant difference among those three groups (F=2.86,P>0.05).Conclusion By the pretargeting method,L5 peptide guided SA-PEG-USPIO has effective targeting ability to HepG2 cells in vitro.
