Objective In order to understand the mechanism of solute transperitoneal transport, we studied the relationship between IL-8 level and protein transperitoneal transport. Method 12 New Zealand White rabbits were randomly divided into experimental and control groups. 2 5% glucose peritoneal dialysis solution (40ml/kg)contained 3?10 9 CFU staphylococcus aureus 1ml was injected into the abdominal cavity of experimental NZW rabbits and 2 5% glucose peritoneal dialysis solution (40ml/kg)contained 0.9%NaCl solution 1ml was injected into the abdominal cavity of controlling NZW rabbits. The plasma and effluent concentrations of blood creatinine ,glucose, total proteins and albumine were determined respectively, the D/P or D/Do values of creatinine , total proteins ,albumine or glucose were calculated respectively. Results The D/P ratios of creatinine, total proteins and albumin were increased significantly, while the D/Do of glucose was decreased in experimental group on different dwelling time points ,there was a significantly difference,as compared with the control group (P0 05),while there was significantly correlation after 60 mins(P
4. Polymorphism and pharmacokinetic research of ribavirin
Chinese Pharmaceutical Journal 2013;48(8):621-628
OBJECTIVE: To establish the preparation method and detection technology of ribavirin polymorphism, and to estimate the bioavailability differences among the ribavirin polymorphs. METHODS: Through polymorphism screening, ribavirin crystals of the form A, B, C, D were obtained. The crystal forms of ribavirin were characterized by different analysis methods, such as single crystal X-ray diffraction method (SXRD), powder X-ray diffraction method (PXRD), differential scanning calorimetry method (DCS), infrared spectrum method (IR) and melting point method (MP). solid ribavirin in different forms were orally administered to rats, and an HPLC-MS method was established to determine the plasma levels of ribavirin, and the bioavailability was analyzed. RESULTS: Of the four polymorphs of ribavirin, form C and form D were reported for the first time. The four polymorphic forms all could be identified by PXRD, DSC, IR and MP. The ρmax and AUC0-t of form A were superior to those of the other forms. CONCLUSION: Characteristic data for ribavirin polymorphism are obtained; and form A of ribavirin is a suitable medicinal crystal. This study has provided the basis for choice of medicinal crystal and the improvement of quality standards.

NDUFS3 is an essential subunit of mitochondrial NADH:ubiquinone oxidoreductase (complex Ⅰ ) and plays a critical role in the mitochondrial typeⅠ respiration chain.Mutations in this gene are shown to cause neurodegenerative disease such as Leigh syndrome (subacute necrotizing encephalopathy). In recent years, many evidences show that the expression of NDUFS3 proteins are lower in many cancerous cells compared to the corresponding normal cells. It comes to the conclusion that NDUFS3 may play a role in the tumorigenesis.

Thioredoxin-related protein 14 (TRP14),a new member of Trx family,is a novel disulfide reductase with conservative CPDC motif. Its structure and function are comparable to Trx,which is the representative member of Trx family. However, there are many differences. When tumor necrosis factor-α (TNF-α) induced cells to produce the active oxygen,TRP14 can change oxidatin state of dynein light chain(its substrate),act as a sensor of the intracellular redox state to regulate NF-κB signaling pathways induced by TNF-α,MAPK and apoptosis signaling pathways.

Objective To investigate the inhibitory effects and related mechanism of S100A4 gene silencing on oxygen-induced retinal neovascularization. Methods 7-day-old C57BL/6J mice were randomly divided into 5 groups including normal group,normal-S100A4 group, oxygen induced retinopathy (OIR) group,OIR-S100A4 group,OIR-green fluorescent protein (GFP) group. To establish the OIR model, mice from all groups except normal one were exposed to (75±2) % oxygen for 5 days and then to room air.In the OIR-S100A4 group and OIR-GFP group, the OIR mice were given an intravitreal injection of 1 μl of 1.0 × 109 PFU/ml adenovirus of Ad-S100A4-RNAi or Ad-GFP at P12,and then returned to normoxia for the next 5 days.In the OIR group,OIR was induced in C57bl/6J mice from P7 to P17.In the normal-S100A4 group, the normal P12 mice were give an intravitreal injection of 1 μl of Ad-S100A4-RNAi adenovirus, and maintained in room air from P12 to P17.In normal group, newborn mouse litters were maintained in room air from P0 to P17 without any treatment.Mice in all five groups were euthanized at P17,and retinas were collected for biochemical assays and morphological study.Retinal neovascularization (RNV) was evaluated by counting the number of pre-retinal neovascular cells and the whole mount immunofluorescent staining of the mouse retina.Protein and mRNA expression levels of S100A4,cAMP responsive element binding protein (CREB),B cell lymphoma-2 (bcl-2),Caspase-3 were determined with western blot and real-time PCR.Results The number of pre-retinal neovascular cell nuclei in retinas from OIR-S100A4 group were obviously lower than those in the retinas from OIR group and OIR-GFP group (t=13.61,14.64;P<0.05).In OIR-S100A4 group, the retinal neovascular tufts area and the vaso-oblitertion area were both significantly smaller than those in OIR group and OIR-GFP group (P<0.05).Protein level of CREB and bcl-2 were significantly down-regulated in OIR-S100A4 group than those in OIR and OIR-GFP group (P<0.05).On the contrary, protein levels of Caspase-3 were up-regulated in OIR-S100A4 group than those in OIR and OIR-GFP group (P<0.05).Conclusion Ad-S100A4-RNAi transfer ameliorates RNV in mouse model of OIR maybe through down-regulating the expression of bcl-2 and CREB,and up-regulating the Caspase-3.

Objective To analyze the diagnostic and therapeutic process of 3 cases of relapsing polychondritis(RP) and explore strategies for early diagnosis and effective therapy. Methods With retrospective study on three relapsing polychondritis patients in our hospital,we observed their major manifestations at admissions and summarized therapeutic experiences. Results All three patients had symptoms of otalgia, hoarse voice and nasal bridge pit, while their entry causes at admission were anemia, renal function failure and chest pain respectively.Only one of them was insensitive to prednisone but sensitive to methotrexate. Conclusions Because PR being easily misdiagnosed, clinical doctors should improve their caution of diagnostic criteria of RP.Prednisone is the best choice for therapy, but for those insensitive ones methotrexate is also alternative.

0.05). In 30 eyes of diabetics with phacoemulsification, 11 eyes had new macula edema after surgery and 3 eyes had significant retinal thickening. In 6 eyes with macular edema before surgery, the macular edema were aggravated in 3 eyes after surgery. The macular structural changes were not found in two control groups. Conclusion The thickness of retina is inreased after phacoemulsification in deabetics, and morbidity and its severity of postopevative macular edema are increased as well.

Objective To evaluate the effect of fluvastatin on the tubule function of the patients with the nephrotic syndrome. Methods Fifty two patients were divided into the experimental group and the control group at random ,all patients were treated with the standard therapies and fluvastatin was added to the experimental group.Plasma albumin, serum cretinine, serum lipid profiles,24hr urinal protein and urinary RBP were examined before treatment and after 8 weeks in all patients. Results The levels of TG,LDL,CH,urinary RBP,urinary protein after 8 weeks of the treatment were significant lowered than those before the treatment (P
