Objective:To investigate the changes of humoral and cellular immunity markers in various liver disease by ABO blood group,the reliable evidences were provided.Methods:The cases of 500 health men (NC) as contrals,various acute and chronic liver disease with the positive index of hepatitis B which include acute hepatitis (ASC) 140 cases,chronic hepatitis (CH) 192 cases,post-hepatitis cirrhosis (LC) 160 cases,primary hepatocellular carcinoma (PHC) 70 cases were examined by immune-spresding assay,APAAP immune-bridge assay and so on in the study,the changes of serumal IgG,IgM,IgA,IgD,Ige,complement C_3(C_3),total complment (CH50),circulatory,immune complex (CIC),E-rosetter formation rate (E-RFC) and T-lymphocyte subsets in the group of NC,ASC,CH,LC and PHC were investigated respectively. Results:Compared to contrals,the humoral immune markers of all groups of liver disease changed markedly,Ige increased changed apparently and IgG increased.Secondly from ASC,CH,LC, to PHC,mean while,C_3 and CIC increased obviously and E-RFC decreased obviously in the group of PHC.Compared to the group of other blood type incresed markedly (P
4.Progress in intervention procedure of Achalasia

Achalasia is a primary esophageal motility disorder, with the main symptom of dysphagia; and caused by the tonus increase and abnormal relaxation of the lower esophageal sphincter(LES). The etiology remains unclear, the objective of the current treatment approaches for achalasia containing the reduction of the LES tone and obstruction to relieve the patients' symptoms; including pharmacologic treatment, botulinum toxin treatment, surgical myotomy, pneumatic dilatation and cardia stent dilatation. The temporary cardia stent dilatation possesses some better advantages and effects; and ought to be the first choice of minimal invasive interventional management for achalasia.
5.Nucleic acid detection technology used in blood screening of blood donors

Objective To evaluate the necessity and feasibility of nucleic acid test for donors blood screening .Methods From July 1 ,2011 to December 31 ,2014 ,a total of 170 316 blood samples which were negative in enzyme-linked immunosorbent assay (ELISA)and qualified in aianine aminotransferase detection ,were selected in this study stochastically .All the samples were detected hepatitis B virus(HBV) ,hepatitis C virus(HCV) ,human immunodeficiency virus(HIV) by nucleic acid amplification technology (NAT) .NAT positive samples were reconfirmed in National Center for Clinical Laboratories(NCCL) .Results A total of 160 cases of nucleic acid reactive samples were found out ,the total response rate was 0 .09% ,The response rate of Roche nucleic acid detec-tion system was 0 .10% ,response rate of David nucleic acid detection system was 0 .08% ,there was no significant difference be-tween the two methods(P>0 .05) .In 27 cases of specimens ,14 cases were confirmed as HBV DNA positive ,no HCV RNA and HIV RNA were detected ,the confirmed positive rate was 51 .85% .There were 2 samples detected by chemiluminescence HBsAg reactivity .Conclusion ELISA screening of blood donors has missing phenomenon ,nucleic acid detection method could be used as an effective supplement of the ELISA ,could improve the safety of blood for clinical use ,detection sensitivity is better than ELISA .

9. Enhancing effect of negative electret on percutaneous drug absorption of meloxicam patch in SD rats

Objective: To determine meloxicam concentrations in both plasma and extract solution of tested-skin in SD rats and the fluorescence intensity, location of fluorescein sodium salt (NaFI) in striped rat skin within 4 h after transdermal administration, so as to investigate the enhancing effect of negative electret on percutaneous absorption of meloxicam and percutaneous absorption route of NaFI. Methods: Pharmaceutical method and grid-controlled constant corona charge technique were used to prepare electret meloxicam patch. High performance liquid chromatography (HPLC) method was employed to determine meloxicam concentration after transdermal administration. NaFI was used as probe to determine the localization and percutaneous absorption route of NaFI by using laser scanning confocal microscopy (LSCM). Results: (DNegative electret and its polypropylene(PP) electret meloxicam patch exhibited a good charge storage stability. (2)The results of HPLC demonstrated that electret had a remarkable enhancing effect on percutaneous absorption of meloxicam after application for 1-4 h. (3) LSCM further proved the enhancing effect of negative electret on percutaneous absorption of small molecules. We also found that the stratum corneum and the hair follicle areas were the two main pathways for the enhancing effect of the electret. Conclusion: Negative electret can be used as an enhancer for transdermal permeation of meloxicam.
10. HPLC determination of CPT-11 and its metabolite 7-ethyl-10 HCPT in incubation system of rat liver microsomes

OBJECTIVE: To establish an HPLC method for determing irinotecan (CPT-11) and its metabolite 7-ethyl-10 HCPT in rat liver microsome incubation system, and to optimize the incubation conditions. METHODS: CPT-11and SN-38 were determined by HPLC. Single factor design was used to optimize the incubation conditions. RESULTS: The linear range of CPT-11 and 7-ethyl-10 HCPT in rat liver microsome incubation system were 20-4000 ng · mL-1 and 2-400 ng · mL-1, respectively. The optimal incubation conditions were as follows: 10 μmol · L-1 CPT-11, 0.02 mg liver microsomes and incubation for 15 min. CONCLUSION: The HPLC method is accurate and suitable for the determination of CPT-11and 7-ethyl-10 HCPT in rat liver microsomes. The incubation condition can be applied in drug interaction studies of irinotecan.
