1.Development of the 2148hk/rr Gene Knock-out Mutant of Streptococcus suis Type 2

To construct gene knock-out mutant of a two-component signal transduction system named 2148hk/rr in Streptococcus suis type 2 virulent strain 05ZYH33.Recombinant gene knock-out vector was constructed consisting of Spc~r cassette with flanking homology regions to the target genes,the isogenic 2148hk/rr-deficient mutant was screened by allelic replacement.PCR analysis and Southern hybridization confirmed that the coding genes of 2148hk/rr were replaced completely by spc~r cassette.Conclusion The mutant of 05ZYH33 2148hk/rr system was successfully constructed,which laid the foundation for farther research on the role of this two-component signal transduction system during infection.
2.Advance research of primary gallbladder cancer

Primary gallbladder cancer is a relatively common biliarv malignant tumor but with a high mortality rate.Most patients with this disease reach an advanced stage when late onset symptoms occtur, and the prognosis tends to be poor even after radical operation.Searching for new diagnostic approaches of primary gallbladder, standardizing treatment strategy, conducting multieenter clinical trials of new drug, developing preventive measures according to different situations are significant methodls to improve the treatment effect prognosis.In this article, we reviewed the research progress of high risk factors, diagnostics, and treatment strategies of primary gallbladder cancer.

4.Resveratrol neuroprotection in cerebral ischemia

Resveratrol is a polyphenolic compounds in many plants.It has many effects inchuding antioxidation,anti-inflammation,inhibiting apoptosis,and scavenging free radicals.Resveratrol preconditioning has neuroprotective effect in ischemia-reperfusion in rats by activating silent information regulator 2 homolog 1.This effect is similar to brain ischemic preconditioning.In addition,resveratrol can also attenuate the brain infraction volume of cerebral ischemia/reperfusion in mice and improve neurological function.This article reviews the neuroprotective effect of resveratrol and its mechanisms in cerebral ischemia.
5.Artificial Neural Network Method for Quality Estimation of Traditional Chinese Medicine

An artificial neural network method for quality estimation of traditional Chinese medicine was suggested, and quality of Hou-Po was estimated by the proposed method in comparison with the analytical results of gas-liquid chromatography. The successful rate reached 100%. The results showed that the neural network method is reliable, and therefore may be referred to as an effective technique for the quality estimation of traditional Chinese medicine.
6.Inhibitory kinetics of paeonol on the activity of mushroom tyrosinase oxidizing L-Dopa

Aim To evaluate the effect of paeonol on the activity of tyrosinase and provide experimental evidence for the treatment of hyperpigmentation disorders. Methods Tyrosinase activity was estimated by measuring the oxidation rate of L-3,4-dihydroxyphenylalanine (L-Dopa). The inhibitory effects of paeonol on the activity of mushroom tyrosinase and Michaelis-Menten kinetics were deduced from the Lineweaver-Burk plots. Results The inhibitory concentration of paeonol leading to 50% enzyme Paeonol is a potential mixed inhibitor of mushroom tyrosinase. The mixed inhibition function may originate from its ability to form a Schiff base with a primary amino group and to chelate copper at the active site of tyrosinase.
7.Roles of AMP-activated protein kinase in cerebral ischemia

Adenosine monophosphate-activated protein kinase (AMPK) is a serine/threonine protein kinase,which is a energy regulation switch in the cells of the body.In the case of nutritional deficiencies and ischemia,AMPK system is activated as the metabolism and stress signal transduction component to regulate the expression of downstream target proteins.After acute ischemic stroke,AMPK is activated and aggravates neuronal apoptosis,and giving AMPK inhibitor may reduce cerebral ischemic injury.The activation of AMPK after stroke may result in the upregulation of brain-derived neurotrophic factor expression and the activation of endothelial nitric oxide synthase,and they play the protective roles for neuronal regeneration and repair.This article reviews the advances in research on the roles of AMPK in experimental cerebral ischemia.
8.Stent insertion combined with high-intensity focused ultrasound ablation for malignant obstructive jaundice caused by pancreatic carcinoma

Objective To investigate the safety and efficacy of stent insertion combined with high-intensity focused ultrasound(HIFU) ablation for malignant obstructive jaundice caused by pancreatic carcinoma.Methods From January 2013 to February 2017,a total of 80 patients with malignant obstructive jaundice caused by pancreatic carcinoma underwent biliary stent insertion in our center,who were divided into combined treatment(stent insertion with HIFU ablation) group(n=38) and solitary stent insertion group(n=42).The stent patency time and survival time were compared between the two groups.Results Relief of jaundice were archived in all patients in both groups.Cholangitis was observed in 7 cases in both two groups.Reobstruction of stent was observed in 15 patients in both two groups.The median stent patency time was 179 days in combined treatment group and 119 days in solitary stent insertion group(P<0.01);the median survival time was 209 days in combined treatment group and 145 days in solitary stent insertion group,the differences were significant(P<0.01).Based on the univariate and multivariate Cox regression analyses,the predictors of prolonging survival included subsequent HIFU ablation after stent insertion and earlier tumor stage(P<0.05).Conclusion Combined biliary stent insertion and HIFU ablation is a safe and effective method for malignant obstructive jaundice caused by pancreatic carcinoma,and seems to extend stent patency time and survival when compared with biliary stent only.
9.The experience of diagnosis and treatment for pancreatic cystic neoplasms

Objective To explore the diagnosis and treatment for pancreatic cystic neoplasms.Methods The clinical data of 10 cases with pancrestic cystic neoplasms treated from 2004 to 2008 was reviewed and both domestic and internal pertinent literatures were summarized.Results The pancreatic cystic neoplasms mostly happened in middle-aged women.The clinical manifestations showed no specificity.Ultrasonography, CT and MRI could offer important sign for diagnosis.The rate of surgical resection was high and the prognosis was satis-factory.Conclusions The pancreatic cystic neoplasms is being muh with the development of imagings.It is dif-ficuh to distinguish the feature of the cystic neoplasms before surgery, so surgerical resection should be done for all cystic tumors found.
10.EFFECT OF STROMAL CELL INFUSION ON HEMATOPOIETIC RECOVERY AFTER PERIPHERAL BLOOD STEM CELL TRANSPLANTATION IN MICE

The present experiment was to study the effect of stromal cell infusion on hematopoietic recovery after peripheral blood stem cell transplantation in mice. BALB/c mice pre conditioned with high dose chemotherapy/radiotherapy were infused with peripheral blood stem cells (PBSCT group) or a combination of peripheral blood stem cells and bone marrow stromal cells (experimental group). Four week survival rate, white blood cell (WBC) count, bone marrow nucleated cells (BMNC), granulocyte macrophage colony forming units (GM CFU), fibroblast colony forming units (F CFU) were determined.
