1.Cerebral vascular disease with combination of TCM and Western medicine treatment effectiveness evaluation

Objective To explore the combination of traditional Chinese and Western medicine therapy on stroke patients with therapeutic implications,in order to further improve the patients with acute cerebral vascular disease and provide scientific basis for treatment of.Methods The hospital admissions in patients with stroke were randomly selected a total of 104 cases as the object of study,in which 48 patients received combined treatment of TCM and Western medicine treatment,another 56 patients received conventional treatment.Results The combination of traditional Chinese and Western medicine treatment group after treated with NIHSS scores than in the conventional group decreased more obviously.The treatment group before treatment and after treatment of the various stages of NIHSS score had significant difference (all P < 0.01) ; integrated traditional Chinese and Western medicine treatment group and routine treatment group after treated with 90d Barthel index difference was statistically significant (P <0.05).Combination of traditional Chinese and Western medicine treatment group treatment effect was better than the conventional group,the difference was statistically significant (P < 0.05).Conclusion The combination of traditional Chinese and Western medicine therapy has affected stroke patients with postoperative quality of life.It can enable the patient to improve neurological function recovery,life skills,improve the quality of life.
2.Advances in mitochondrial function of airway epithelial cells in the bronchial asthma

Bronchial asthma is a chronic inflammatory disease of the airways, which is characterized by airway hyperresponsiveness(AHR), eosinophilia, elevated IgE, goblet cell metaplasia, airway remodeling and so on.Changes in airway epithelial structure and function are prominent features of asthma.Asthma airway epithelium has abnormal sensitivity to oxidative damage and apoptosis, and is more susceptible to reactive oxygen species(ROS)produced by infiltrating inflammatory cells.The effects of ROS accumulation and oxidative stress are the key links in the pathogenesis of asthma.Oxidants such as ROS can interfere with the structure of epithelial cells and cause tracheal remodeling.Conversely, tracheal remodeling can release inflammatory mediators and aggravate asthma.The main function of mitochondria is oxidative phosphorylation to synthesize ATP, and it is also an important source of ROS.Mitochondrial dysfunction includes energy metabolism conversion dysfunction, mitochondrial biological dysfunction, mitochondrial autophagy and kinetic abnormalities, and mitochondrial-dependent signaling pathway disorders.Mitochondrial dysfunction and cellular hypoxia play an extremely important role in the occurrence and development of bronchial asthma.This article reviews the changes in mitochondrial function of airway epithelial cells in asthma airways in recent years, in order to provide theoretical basis for mitochondria-targeted asthma treatment.
3.The influence of obstetric complications and pregnancy outcome because of prenatal anxiety and depression

Objective To investigate the influence of obstetric complications and pregnancy outcome because of prenatal anxiety and depression.Me~ods One hundred primipara were selected as the research objects,their emotions were evaluated by Self-Rating Anxiety Scale(SAS),Self-Rating depression Scale(SDS).The patients' scores of SAS ≥50points or SDS ≥ 53points were selected as the observation group,the patients'scores of SAS ≥ 50points and SDS≥53points were selected as the control group.The pregnant women childbirth way,preeclampsia,hyperemesis gravidarum,gestational diabetes,perinatal infection,hydramnios,oligohydramnios,premature rupture of membranes,postpartum hemorrhage,fetal distress,premature birth,low birth quality,neonatal asphyxia of two groups were compared.ResultsThe primipara cesarean delivery rate of the control group was lower than that of the observation group(4.00% vs 22.00%,x2 =7.161,P <0.05),the obstetric complications incidence of the observation group was higher than that of the control group (32.00% vs 2.00%,x2 =15.946,P < 0.05),the birth outcomes incidence of the observation group was lower than that of the control group (4.00% vs 20.00%,x2 =6.061,P < 0.05).Conclusion Prenatal anxiety and depression significantly increase pregnant women obstetric complications,increase the perinatal fetal intrauterine distress and the probability of premature birth,low birth quality and neonatal asphyxia.
4.Acute hypotensive episodes prediction based on non-linear chaotic analysis.

In intensive care units (ICU) , the occurrence of acute hypotensive episodes (AHE) is the key problem for the clinical research and it is meaningful for clinical care if we can use appropriate computational technologies to predict the AHE. In this study, based on the records of patients in ICU from the MIMIC II clinical data, the chaos signal analysis method was applied to the time series of mean artery pressure, and then the patient's Lyapunov exponent curve was drawn ultimately. The research showed that a curve mutation appeared before AHE symptoms took place. This is powerful and clear basis for AHE determination. It is also expected that this study may offer a reference to research of AHE theory and clinical application.

6.Studies of mutations and polymorphism of PINK1 gene in patients with Parkinson' s disease in littoral of Zhejiang Province

Objective To study deletion mutations and polymorphism of PINK1 gene exons in patients with Parkinson' s disease (PD) in the littoral of Zhejiang Province,and analyze the association between these changes and the etiology of PD.MethodsAll exons of PINK1 gene in 200 PD patients( 112early-onset PD and 88 late-onset PD) and 220 controls (68 young controls and 152 old controls ) were amplified by polymerase chain reaction (PCR). All the positive PCR products were sequenced,and genotypes were detected by dot blot allele and genotype frequencies of PINK1 were compared by the Chisquare test.ResultsNo deletion mutations of the exons were found in all patients and controls.However,a known heterozygote point mutation G12169A in exon 5 was identified in 2 patients with early-onset PD.And G12164A polymorphism and G12101A polymorphism were located on PINK1 gene of exon 5. There were G/G and G/A genotypes in G12164A polymorphism,no A/A genotype.There were G/A and A/A genotypes in G12101A polymorphism,no G/G genotype. The chain relation polymorphism A/A genotype frequency was significantly higher in the PD group 84/200 (42.0% ) than the control group (52/220,23.6%,x2 =4.034,P =0.045 ).The frequency was also significantly higher in the late-onset PD (40/88,45.5% ) than the old control (32/152,21.1%,x2 =3.951,P =0.047 ).There were no significant differences in alleles frequencies of other groups.ConclusionThe deletion mutation and point mutation are rare in PD patients in littoral of Zhejiang Province.Chain relation polymorphism at G12164A and G12101A in PINK1 gene might be a susceptible factor for PD patients.
7.Clinical research of the relationship between 5-lipoxygenase activating protein gene polymorphism and cerebral infarction susceptibility

Objective To investigate the expression of 5-lipoxygenase activating protein gene (ALOX5AP) polymorphism in the patients with cerebral infarction,and explore its relationship with cerebral infarction susceptibility.Methods Patients with cerebral infarction and healthy volunteers were selected for this study,whose venous blood was extracted and detected with polymerase chain reaction/restriction fragment length polymorphism (PCR-RFLP).Haplotype A (SG13S114T,SG13S89G,SG13S32A,SG13S25G),haplotype B (SG13S377A,SG13S114A,SG13S41A,SG13S35G),and their nucleotide polymorphism loci were observed.Results Single nucleotide polymorphism (SNP)-SG13S114,SNP-SG13S32 and HapA carrying rate were significantly different between patients with cerebral infarction and healthy volunteers (P <0.05).SNP-SG13S114 and SNP-SG13S32 were independent risk factors of cerebral infarction (OR > 1.0,P < 0.05).Conclusions The morbidity of cerebral infarction in Wenling City was influenced by SNP-SG13S114,SNP-SG13S32,and HapA carrying rate.
8.Accuracy evaluation of CyberKnife system

Objective To evalutate the accuracy of CyberKnife system and measure its relative output factor.Methods The accuracy of target localization of the system was measured with E2E test,film dosimetry,6D-skull tracking mode and fiducial tracking mode.Six kinds of detectors were used to detect the collimators of the system.Results The positioning errors were (0.531 ±0.293) and (0.482±0.297) mm respectively by 6D-skull tracking mode and fiducial tracking mode.The detectors proved the relative output factors of 5-mm collimator were 0.133 ±0.006,0.320±0.010,0.635 ±0.010,0.656±0.009,0.777± 0.013 and 0.680±0.014,and those of 35-mm collimator were 0.983±0.002,0.990±0.002,0.987±0.001,0.992±0.002,0.990± 0.010 and 0.981 ±0.010.Conclusion The total error of CyberKnife system was less than 1 mm.Large collimater has high consistencies between the output factors of the detectors,while there are significant differences between the output factors in case of the collimater with the diameter less than 15 mm.
9.Study on the Mechanism of Histone Deacetylase Inhibitor RGFP109 in Reversing Resistance of Glioma U251 Cells to Temozolomide

OBJECTIVE:To study the mechanism of histone deacetylase inhibitor RGFP109 in reversing resistance of glioma U251 cells. METHODS:TR/U251 cells resistance to temozolomide(TMZ)was extrablished. The test was divided into normal con-trol group,TMZ group(40 μmol/L)and TMZ(40 μmol/L)+RGFP109(0-120 μmol/L)different concentrations groups. After 24 h of adding into related medicines,CCK-8 was used to detect the cell survival rate and calculate the half inhibitory concentration (IC50). TUNEL and Annexin V/PI were used to detect the cell apoptosis in normal control group,TMZ group and TMZ+RGFP109 (42μmol/L)group. Immunoblotting was used to detect the O6-methyl guanine-DNA methyltransferase(MGMT),Survivin,B lym-phoma 2(Bcl-2),B lymphoma xL(Bcl-xL)protein expression;and gel migration test was used to detect the p65 acetylation level and its binding capacity with κB-DNA. RESULTS:Compared with normal control group,cell survival rate in TMZ+RGFP109 dif-ferent concentrations groups was obviously decreased (P<0.05),showing a concentration-dependent manner. When the RGFP109 concentration was 42 μmol/L,the sensitivity of TMZ to TR/U251 cells was the same with U251 cells. Compared with normal con-trol group,MGMT,Survivin,Bcl-2,Bcl-xL protein expressions in cells of TMZ groups were enhanced(P<0.01);p65 acetyla-tion level had no obvious changes,while the binding capacity of p65 and κB-DNA was strengthened (P<0.01). Compared with TMZ group,MGMT,Survivin,Bcl-2,Bcl-xL protein expressions in cells of TMZ groups were weakened(P<0.01);p65 acetyla-tion level was enhanced (P<0.01);and the binding capacity of p65 and κB-DNA was weakened (P<0.01). CONCLUSIONS:RGFP109 can reverse the resistance of U251 cells to TMZ by down-regulating the anti-apoptotic protein expressions adjusted by transcription factorκB(NF-κB)and weakening the binding of p65 andκB-DNA.
10.Effects of dexamethasone sodium phosphate on the proliferation of in vitro cultured rabbit articular chondrocytes

BACKGROUND: Studies have demonstrated that dexamethasone can reduce type Ⅱ collagen and increase type I collagen in articular cartilage surface matrix, but the action mechanism of glucocorticoid to articular chondrocyte proliferation remains unclear. OBJECTIVE: To investigate the effects of dexamethasone sodium phosphate on rabbit articular chondrocytes cultured in vitro. DESIGN: Comparative observation.MATERIALS: New Zealand rabbits, 1 month old, were used for chondrocyte isolation and culture.METHODS: Rabbit articular chondrocytes cultured in vitro were randomly divided into control and experimental groups. Cells of the control group were cultured in DMEM media without dexamethasone sodium phosphate. Cells of experimental groups were cultured in DMEM media with different concentrations of dexamethasone sodium phosphate (0.02, 0.1, 0.5 g/L), respectively. MAIN OUTCOME MEASURES: Primary and passage chondrocytes were observed. Flow cytometry and immunocytochemistry were used to observe the effect of dexamethasone sodium phosphate on cell proliferation and type Ⅱ collagen synthesis. The ultrastructural changes of cultured chondrocytes were observed by transmission electron microscopy. RESULTS: The attachment and proliferation of experimental group chondrocytes was slower than control group. There average gray scale values of the experimental groups were significantly greater than the control group (P < 0.05). The cellular proportions of S phase and G_2+M phase of the experimental groups decreased but the cellular proportions of G0/G1 period increased. Under transmission electron microscope the amount of rough endoplasmic reticulum and mitochondria of the experimental groups decreased. CONCLUSION: Dexamethasone sodium phosphate inhibited articular chondrocyte proliferation, possibly due to the decrease of type Ⅱcollagen and protein synthesis ability of chondrocyte.
