Methods 2011 grade totally 86 Uygur clinical medical under-graduates in Xinjiang Medical University were enrolled as the research object and questionnaire was used to make satisfaction survey. Data descript and analysis was performed according to the percentage of each option. Results 86.9%students thought that topic seminar was good;100.0%students thought that semi-nar can help apply and mastery the knowledge;100.0%students considered that seminar conducted dur-ing extracurricular time was necessary to the growth of knowledge;97.8%students considered that search-ing for references in the spare time was helpful;100.0%students were interested in the content of the seminar and thought it was novel and practical;100.0%students thought that seminar can help broaden thinking way;43.5%students supported that contents exceeding the syllabus can be used in examination while 39.1%students opposed; preparation time for examination lasted for 5 to 7 days. Conclusions Seminar applied in biochemistry teaching is very necessary, which can mobilize students' initiatives to learn actively and help consolidate and understand knowledge.

Purpose To investigate the expression of Beclin1, LC3 and mTOR in colorectal cancer ( CRC) and their significance. Methods Immunohistochemistry, Western blot and real-time PCR were employed to detect the expression of Beclin1, LC3 and mTOR in CRC. Results The positive expression rate of Beclin1, LC3 and mTOR in 242 cases of CRC was 90. 50%, 87. 19% and 46. 28%, respectively, which were higher than that in adjacent tissues ( P<0. 05 ) . Moreover, the expression of LC3 in moderately and poorly differentiated CRC was higher than that in well differentiated CRC, and the positive rate of LC3 in CRC with lymph node metastasis was higher than that in CRC without lymph node metastasis. The overexpression of mTOR was related to lymph node metasta-sis (P<0. 05), but both differentiation degree and lymph node metastasis were not associated with Beclin1 (P>0. 05). The expres-sion of LC3 was positively correlated with Beclin1 and negatively correlated with mTOR in colorectal cancer (rs =0. 593, P<0. 01, rs= -0. 165, P<0. 01), and the expression of Beclin1 was not associated with mTOR (P>0. 05). Kaplan-Meier survival analysis re-vealed that the five-year survival rate of patients without nodal metastasis, positive expression of Beclin1, LC3 and negative expression of mTOR was higher than those with nodal metastasis, negative expression of Beclin1 and LC3, and positive expression of mTOR. Cox survival analysis results revealed that LC3, mTOR and lymphnode metastasis were independent prognostic factors. The results of IHC, real-time PCR and Western blot in fresh CRC tissues indicated that the expression of Beclin1, LC3 and mTOR in colorectal cancer was significantly higher than that in adjacent tissues (P<0. 05). Conclusions The aberrant expression of Beclin1, LC3 and mTOR may be associated with the development and progression of colorectal cancer. The simultaneous detection of Beclin1, LC3 and mTOR genes in colorectal cancer may be helpful for the evaluation of the progressive degree and the judgment of prognosis.

Purpose To detect the expression of Raptor,Rictor,angiogenesis-related factors HIF-lα,HIF-2α and VEGF and to investigate their relationship and significance in eolorectal cancer (CRC).Methods Immunohistochemistry,Western blot and RT-PCR were employed to detect the expression of Raptor,Rictor,HIF-lα,HIF-2α and VEGF in 120 cases of CRC and 60 cases of normal colorectal mucosa.CD34 labeled microvascular density (MVD) was also observed. The correlations between Raptor,Rictor,HIF-1α,HIF-2c,VEGF expression and the patients' clinicopathological features were analyzed. Results The positive rates of Raptor,Rictor,HIF-1c,HIF-2α and VEGF in CRC were significantly higher than those in normal colorectal mucosa (P < 0.05).Raptor and Rictor expression was correlated with the degree of tumor diffcrentiation and lymph node metastasis, respectively. The expression of HIF-1α,HIF2α and VEGF was higher in patients with lymph node metastasis than those in patients without lymph node metastasis (P <0.05).The MVD was higher in patients with Raptor or Rictor positive than that in patients with Raptor or Rictor negative (P <0.05).The expression of Raptor was positively correlated with HIF-1α and VEGF (P < 0.01),the expression of Rictor was positively correlated with HIF-2o and VEGF (P < 0.01),but the expression of Raptor was negatively correlated with Rictor (P<0.01).Conclusion The expression of mTOR core molecules Raptor and Rictor is related to the initiation and development of colorectal cancer and angiogenesis, and they promote angiogenesis in colorectal cancer by different ways.

Objective:To study the expressione of autophagy-related protein Beclin-1 and LC3 in cortex and hippocampus of rats after cerebral ischemia reperfusion injury and the efficacy of Edaravone. Methods:Sprague Dawley rats were randomly divided into sham group, model group and Edaravone group. The cerebral ischemia reperfusion model was induced via Zea Longa with blocking the middle cerebral artery of 2 h and reperfusing of 24 h.Animals assigned to sham group were only separated left common carotid artery.Edaravone was injected intraperitoneally at a dose of 1 0 mg/kg at 15 min before reperfusion. The condition of nerve injury of rats was conducted by Neurobehavioral score. The degree of brain injury and success of model were determined based on 2,3,5-triphenyltetrazolium chloride(TTC)staining. The changes of neuron stained by hematoxylin and eosin (HE) in cortex and hippocampus were observed. The expression of Beclin-1 and LC3 was measured by immunohistochemistry. Results:After cerebral ischemia reperfusion the neurobehavioral score of edaravone group was(2.00± 0.67),which was obcviously less than(2.50± 0.53) of model group(P<0.05).The infraction focus and the neuron injury in cortex and hippocampus neurons were also observed in model group, and the edaravone group reduced above expression. The positive rate of Beclin-1 of each group in cortex were (1 1.08± 0.85)%,(33.42± 1.57)% and (25.61± 1.39)%,there was significant difference between model group with sham group and edaravone group (P<0.05).The positive rate of Beclin-1 of each group in hippocampus were (10.34± 0.21)%,(31.82± 1.73)% and (22.74± 1.26)%,there was significant difference between model group with sham group and edaravone group(P < 0.05).The positive rate of LC3 of each group in cortex were (15.33± 0.47)%,(39.72± 1.73)% and (28.53± 1.61)%,there was significant difference between model group with sham group and edaravone group(P<0.05).The positive rate of LC3 of each group in hippocampus were (13.74± 0.37)%,(32.53± 1.43)% and(25.38± 1.23)%,there was significant difference between model group with sham group and edaravone group (P<0.05).Conclusion:The expression of Beclin-1 and LC3 was increased after cerebral ischemia reperfusion.Edaravone may reduce autophagy and brain injury through downregulation the expression of Beclin-1 and LC3.
