2.Biological effects of nonthermal plasma combined with X-ray irradiation on human hepatocellular carcinoma in nude mice
Journal of International Oncology 2017;44(4):257-261
Objective To investigate the biological effects of nonthermal plasma (NTP) combined with X-ray irradiation on human hepatocellular carcinoma in nude mice. Methods By subcutaneous inoculation, tumor models of human hepatocellular carcinoma HepG2 in nude mice were established, and these mice were divided into control group, NTP treatment group, X-ray irradiation group, combined treatment group. The changes of tumor microenvironment were observed using hematoxylin-eosin (HE) staining. The terminal deoxynucleotidyl transferase-mediated nick end labeling (TUNEL) was used to determine the level of neuronal apoptosis in tumor tissues. The expression of metal matrix proteinase-2 (MMP-2) was detected by immunohistochemical assay.Transmission electron microscope was used to observe cell structure changes. Results The final tumor volumes of control group, NTP treatment group, X-ray irradiation group, combined treatment group were (543.96±108.45), (436.54±65.49), (351.66±56.68), (281.97±35.60)mm3, with a statistically significant difference (F=9.63, P=0.01), and the difference between X-ray radiation group and the combination treatment group was significant (P=0.05).HE staining showed that there was a larger area of necrosis in the combined treatment group compared with the other groups.TUNEL showed that the apoptotic indexes were (0.95±0.13)%, (5.82±0.26)%, (7.53±0.43)%, (11.07±0.35)% respectively, with a statistically significant difference (F=547.76, P=0.00), and the difference between X-ray radiation group and the combination treatment group was statistically significant (P=0.00).The immune scores of each group were 12, 9, 9, 2.Electron microscopic observation showed that there were more apoptotic bodies in the combined treatment group than those in the other groups, accompanied by mitochondrial edema. Conclusion NTP and X-ray irradiation therapy in the treatment of human hepatocellular carcinoma in nude mice model has a synergistic effect.Probably, it can be a new type of treatment in curing cancers.
4.A novel classification-coding scheme for medical image retrieval
Chinese Medical Equipment Journal 2003;0(10):-
A novel mono-hierarchical muti-axiel classification coding scheme for medical image retrieval is proposed. The so-called MOAB coding scheme consists of four axes with three to four positions ranging from0to9,from A to Z.In particular, the modality code M describes imaging modality and relevant technical detail, and the orientation code O models examined body orientation. The anatomy code A refers to the body region examined and the biology code B describes the biological system examined. The MOAB classification-coding scheme enables a unique classifi-cation of medical images so that medical image retrieval can be efficient. The code is flexible and easy to be ex-tended.
5.ULTRASTRUCTURE STUDIES ON ERYTHROID LINE IN THALSSEMAIA
Journal of Chongqing Medical University 1987;0(01):-
Ultrastructure varities in erythroid line of 6 patients with Thalassemia were studied by electron microscopy. A number of electron-dense inclusions (alpha or beta chains precipitation) were noted in cytoplasm or nuclei of polychromatic erythroblasts, normoblasts, reticulocytes and erythrocytes. The nuclear membranes showed absence or duplication and the space between two layers of nuclear membranes dilated. Autophage vacuoles and vacuolation were easily observed within the early and late stages of erythroid series. The figure of red cells was markedly abnormal, the characteristic ropalocytes might be found. In addition, the various forms of squeezed and partly destructed RBC which had been sequestrated in the minute sinuses of spleen were observed. The observations above provide direct morpdological evidences of the pathogenesis in Thalassemia
6.Anti-hyperuricemic activity of bergenin and its mechanism
Acta Universitatis Medicinalis Anhui 2014;(1):63-66,67
Objective To explore the anti-hyperuricemia activity of bergenin in the model of hyperuricemic mice induced by potassium oxonate. Methods 60 Kunming male mice were divided randomly into six groups, which were normal control group;hyperuricemic model group;and hyperuricemic groups with 20 , 40 , 60 mg/kg berge-nin, and 5 mg/kg allopurinol. Mice were orally administered once daily with 250 mg/kg potassium oxonate for 7 continuous days to create the model, and then three doses of bergenin and allopurinol were orally initiated on the day 1 h after potassium oxonate was given, separately. Serum uric acid, creatinine and urea nitrogon levels, as well as urinary uric acid and creatinine levels were measured. mRNA and protein expression levels of mouse kidney u-rate transporter 1(URAT1), and glucose transporter 9(GLUT9) were also determined. Results Compared with hyperuricemic model group, bergenin significantly reduced serum uric acid, creatinine and urea nitrogon levels, in-creased 24 h uric acid and creatinine excretion, and fractional excretion of uric acid in hyperuricemic mice;mRNA and protein levels of mURAT1 and mGLUT9 were also markedly down-regulated. Conclusion Anti-hyperuricemia effect of bergenin is attributed to the enhancement of uric acid excretion and reversion of mouse urate transporters o-ver-expression.
7.Reproduction of an animal model simulating acute kidney injury caused by high intensity military training
Medical Journal of Chinese People's Liberation Army 2001;0(12):-
Objective To reproduce an animal model simulating acute kidney injury caused high intensity military training in rats by forced exhausting running on the treadmill, that could be used for the investigation of problems arising from military training. Methods 40 male SD rats were randomly divided into three groups:control group(n=8),ordinary exercise group(n=16)and forced exercise group(n=16).The animals in ordinary exercise group were commonly fed.The animals in the other two groups were forced to take on one-time exhausting running exercises on the treadmill by different ways. The rats in forced exercise group were forced to run on the treadmill till exhaustion to reproduce the animal model of overtraining. The indexes of urine, serum and kidney tissues were observed and analyzed shortly after training and 24 hours later. Results After training, the values of urine protein, urinary albumin, urease, serum urea nitrogen, creatinine, angiotensin Ⅱ in serum and kidney tissues of the animals in forced exercise group increased obviously compared with those in control group and ordinary exercise group. Those indexes in ordinary exercise group were higher than in control group(P
8.hTERT antisense oligonucleotides enhance apoptosis induced by cisplatin in HeLa cell lines
Chinese Pharmacological Bulletin 2003;0(12):-
Aim To investigate the effects of human telomerase reverse transcriptase(hTERT)antisense phosphorothioate oligodeoxynucleotide(ASODN)on telomerase activity and apoptosis induced by crisplatin(DDP)in HeLa cell lines. Methods HeLa cells were collected after transfection of 0.05,0.1,0.2 ?mol?L-1of ASODN and the total DNA was extracted.Telomeric repeat amplification protocol(TRAP)-AgNO3 stain was used to detect the activity of telomerase.Apoptosis of HeLa cells was detected by AO staining morphological observation and flow cytometry(FCM)analysis technology. Results The telomerase activity of HeLa cells was expressed in high level, which was significantly reduced dose-dependently and time-dependently after treatment with ASODN of hTERT.Two days after transfection of ASODN of 0.05 ?mol?L-1,0.1 ?mol?L-1,0.2 ?mol?L-1,the telomerase activity decreased by 21.77%,52.42% and 71.10% respectively. After treatment with ASODN for 24,48 h and 72 h,the telomerase activity decreased by 12.48%,38.27% and 71.10% respectively. In morphological observation using acridine orange(AO)staining method,HeLa cells displayed typical apoptotic figure after treated with DDP plus ASODN of hTERT.The percentage of apoptosis of HeLa cells treated with DDP of 1.5 mg?L-1,3.0 mg?L-1,for 48 hours after 24 hours exposure to ASODN of 0.2 ?mol?L-1(50.35%,29.67%)was significantly higher than that treated with DDP alone(19.67%,11.38%,P
9.On Thought of Globus Hystericus Treated by Basing on Differentiation of Signs
Journal of Zhejiang Chinese Medical University 2013;(12):1461-1462,1468
[Objective]To discuss the treatment based on syndrome differentiation of globus hystericus.[Methods]We combine consulting historical literatures with clinical experience and completely summarize the syndrome differentiation of globus hystericus.[Results]More pathogenesis can lead to globus hysteri-cus from clinical practice, such as insufficiency of heart yang ,insufficient kidney yang and exogenous pathogenic factors. In the principle of syndrome dif-ferentiation, we must base on the different pathogenesis to treat globus hystericus. [Conclusion]Because of its complexity, we can not rapidly draw a con-clusion without a careful analysis when facing globus hystericus and its like, which leads us to neglect the potential dangerous factors.
