1.Research progress of Manganese-enhanced MRI optic nerve tracing in vivo

Manganese-enhanced MRI (MEMRI) optic nerve tracing in vivo is a technology which can reflect the axonal transport status and visual pathway connection by utilizing divalent manganese ion's transportability in axons and its characteristics of paramagnetism,and such potential optic nerve tract-tracing technique has been widely used in ophthalmologic research.Mn2+ enters the neurons through the voltage-gating Ca2+ channel and is transported by microtubule system which is not depended on cellular electric activity.However,the trans-synaptic transport of Mn2+ relies on membrane potential.Taking advantages of MEMRI,optic nerve injury-related research could be implemented.Mn2+ is an intracellular contrast agent with metal toxicity,so low dose,fractionated,targeted delivery of Mn2+ is recommended to alleviate the adverse effects of Mn2+.Herein,the mechanism of divalent manganese ion optic nerve tracing,application of MEMRI in ophthalmologic research field,drug delivery approach of divalent Mn2+ and its toxicity were summarized.

Epigenetic modification processes involved in human disease include DNA methylation, histone acetylation and methylation, RNA-associated silencing.Epigenetic modification related with cell growth, differentiation, apoptosis, transformation and tumor progression-related gene transcription. There has been a long time that the gene mutations involved in tumor formation. In recent years.more and more evidences that genetic and epigenetic modifications leading to the formation of tumors. Epigenetic modifications has also play a very important role in the formation of tumors. This article discussed the DNA methylation.histone post-translational modification anomalies and small interference RNA impact on the formation of leukemia. It provides new prospects for the treatment of leukemia.
3.Diagnostic value of rifampin resistance real-time fluorescent quantitative nucleic acid amplification technology combined with T cell spot test of tuberculosis infection in tuberculous meningitis

Objective:To evaluate the diagnostic value of rifampin resistance real-time fluorescent quantitative nucleic acid amplification technology (GeneXpert) combined with T cell spot test of tuberculosis infection (T-SPOT.TB) in tuberculous meningitis (TBM).Methods:The clinical data of 70 TBM patients (TBM group) and 30 non-TBM patients (non-TBM group) in Shenyang Chest Hospital from March 2017 to December 2019 were retrospectively analyzed. All patients were detected by GeneXpert and T-SPOT.TB. The results of MGIT960 liquid culture of cerebrospinal fluid were used as the gold standard to evaluate the diagnosis of TBM by GeneXpert and T-SPOT.TB.Results:The positive rates of GeneXpert and T-SPOT.TB in parallel and in series in TBM group were significantly higher than those in non-TBM group: 27.14% (19/70) vs. 0, 72.86% (51/70) vs. 46.67% (14/30), 78.57% (55/70) vs. 46.67% (14/30) and 21.43% (15/70) vs. 0, and there were statistical differences ( P<0.01 or <0.05). The sensitivity and accuracy of two methods in parallel in TBM diagnosis were 78.57% and 71.00% respectively. The specificity and misdiagnosis rate of two methods in series in TBM diagnosis were 100.00% and 0 respectively. Conclusions:The sensitivity of GeneXpert and T-SPOT.TB in parallel is the highest in TBM diagnosis. The specificity of the two methods in series and GeneXpert alone is as high as 100.00%. The two methods can reduce the misdiagnosis rate of TBM in series and reduce the rate of missed diagnosis in parallel.

CYP2A6 is an important drug metabolic enzyme that plays a central role in pharmacology and toxicology.Coumarin and nicodine are mainly metabolized by it.CYP2A6 also have a close connection with diseases that related to smoking.While hnRNP A1 is a protein that shows an important regulative action on some specific genes at post-transcription level.Recently,researchers are paying more and more attention to the relationship between the activity of CYP2A6 and the expression of hnRNPA1.In order to master the change rule of CYP2A6 activity,we try to explain the regulative effect on CYP2A6 by hnRNP A1 at post-transcription level,which will bring an important effect on guiding clinical medicine treatments and smoking addictive therapy.
5.Observation on the effect of kangfu antiphlogistic suppository combined with Ceftriaxone on the treatment of chronic pelvic inflammatory

Objective To explore the effect of Kangfu antiphlogistic suppository combined with ceftriaxone on the treatment of chronic pelvic inflammation.MethodsFrom March 2015 to January 2017 in Deqing hospital of traditional Chinese medicine, 80 cases with chronic pelvic inflammatory as research object were divided into the observation group and the control group, 40 cases in each group.The control group were given Ceftriaxone, at this basis, the observation group were given Kangfu antiphlogistic suppository, The effect and the adverse reaction in the two groups were compared.ResultsThe effect was 92.5% in the observation group, which was higher than that in the control group 77.5%, the difference was statistically significant,P<0.05.There was no adverse reaction in the 2 groups.ConclusionThe effect is exactly that Kangfu antiphlogistic suppository combined with ceftriaxone sodium on the treatment of chronic pelvic inflammation, which is worthy of clinical use.
6.Clinical observation of the development of juvenile myopia wearing glasses with full correction and under-correction

AIM:To observe the effect of wearing glasses with full correction or under-correction on the development of juvenile myopia.
METHODS:This study included 132 cases ( 264 eyes ) from January 2008 to September 2012 who were collected from our clinic. They were divided into 2 groups, full correction and under- correction. Students in group 1 wore glasses with full correction, students in group 2 wore glasses with under-correction 0. 25D to 0. 5D lower than normal. Reexamination was done every 6mo. After 12mo, refractions were checked, the development of myopia was compared.
RESULTS: There were no statistically significant differences between two groups after 6mo ( P=0. 0693 );however, there were significant differences after 12mo( P=0. 0013).
CONCLUSION:The development of myopia is slow if students often wear glasses with full correction.
7.Auxiliary diagnostic value of fragmented QRS complex for myocardial infarction

Objective: To explore auxiliary diagnostic value of fragmented QRS complex on ECG for acute myocardial infarction (AMI).Methods: Clinical data of 102 AMI patients (AMI group) and 132 CHD patients (CHD) group, who were treated in our hospital from Apr 2013 to Apr 2014, were retrospectively studied, another 120 healthy subjects undergoing physical examination in our hospital during the same period were regarded as normal control group.AMI was diagnosed by myocardial perfusion tomography, all subjects received ECG examination, and incidence rates of fragmented QRS complex and pathological Q wave at different sites were recorded.Sensitivity, specificity and accuracy of fragmented QRS complex and pathological Q wave diagnosing AMI were compared.Results: Incidence rate of fragmented QRS complex was highest on inferior wall in AMI group, and that of pathological Q wave was highest on anterior wall in AMI group.Except fragmented QRS complex in sidewall, compared with normal control group, there were significant rise in incidence rates of fragmented QRS complex on anterior wall (0.8% vs.22.5%, 13.6%) and inferior wall (6.7% vs.26.5%, 17.4%);pathological Q wave on anterior wall (0 vs.41.2%, 29.5%), inferior wall (0.8% vs.40.2%, 35.6%) and side wall (0 vs.2%, 2.3%) in AMI group and CHD group, P<0.01 all.Compared with pathological Q wave, there was significant rise in diagnosing sensitivity for AMI (40.00% vs.89.47%), for CHD (41.94% vs.91.50%) of fragmented QRS complex (P<0.01 both), but there were no significant difference in specificity and accuracy between them (P>0.05 all).Conclusion: Fragmented QRS complex can be used as an important auxiliary index diagnosing acute myocardial infarction.When combined with pathological Q wave, it can further improve diagnostic value of ECG for acute myocardial infarction.
8.Effect of different depths of propofol anesthesia given by TCI on 5-HT metabolism in brain

Objective To investigate the effect of different levels of propofol anesthesia on 5-HT metabolism in brain. Methods Forty Japanese Long-ear rabbits of both sexes, aged 8 months-2yr, weighing 2-3 kg were used for study. Anesthesia was induced and maintained with propofol administered by a TCI system. Blood propofol concentration required for different depths of anesthesia, as indicated by BIS monitoring and loss of chewing reflex (light anesthesia) and loss of response to tail nipped (deep anesthesia), was determined by high-performance liquid chromatography(HPLC) in 10 rabbits. Thirty rabbits were randomly divided into 3 groups often animals in each group: (1) control group received no propofol; (2) light anesthesia group and (3) deep anesthesia group. Different depths of propofol anesthesia were maintained for 1 h. At the end of one hour propofol anesthesia the animals were decapitated. Brain was immediately removed and cerebral cortex, hippocampus and thalamus were separated on ice. Their contents of 5-HT and 5-hydroxyindole-3-acetic acid (5-HIAA) were measured by spectrophoto-fluorometer.Results 5-HT content of cerebral cortex was significantly increased after anesthesia. 5-HT content of hippocampus and 5-HIAA content of thalamus decreased significantly with increasing anesthetic depth. Conclusion Propofol improves 5-HT activity in cerebral cortex and thalamus and decreases 5-HT activity in hippocampus indicating that 5-HT metabolism may be involved in the mechanism of propofol anesthesia.
