1.Effects of TGF-?_(1,2) on basal and FSH-stimulated production of estradiol and progesterone by human granulosa cells in vitro

Objective To investigate the possibility that TGF ? may also influence the basal and FSH stimulated production of estradiol and progesterone by granulosa cells in vitro. Methods Granulosa cells were obtained from infertile patients undergoing in vitro fertilization and embryo transfer treatment and cultured with serum free supplemented HAM′s F 10 medium(control) or increasing concentrations of TGF ? 1,2 in the absence or presence of FSH (96 U/ml). The media were collected in 24, 48, 72 and 96 h intervals and assayed for estradiol and progesterone. Results Increasing amounts of TGF ? had a significant effect on estradiol production, especially in the presence of FSH,The estradiol value was evaluated from (12 077.9?5 835.3) pmol/L and (13 656.1? 8 404 3) pmol/L to (16 606.8?7 993.3) pmol/L and (19 480.4?913.8) pmol/L. TGF ? inhibited FSH stimulated progesterone but not unstimulated progesterone release ( P

Ankle syndesmosis injury is one of the most common ankle fractures complicated with injury,but there is certain challenges with diagnosis and treatment.The treatment of lower tibia and fibula injury is the main surgical treatment,the key is anatomical reduction.Along with some new treatment methods,the treatment effect of the lower tibia and fibula screws has been questioned more and more,And there is still a controversy on whether the removal of the lower tibia and fibula screws.Now for the ankle syndesmosis injury treatment were summarized.
3.Verification of Flow Field Performance of Air Dynamic Wind Tunnel

Objective To verify the flow field performance of the air dynamic wind tunnel. Methods The performance of the flow field was evaluated by testing the flow speed of sample-point on the sample-transect of the tunnel. Results Among the flow range of the tunnel, the relative deviations of the speeds of each sample-point on the sample-transect were less than 10%. When the flow rate was set, the relative deviations of the sample-transect average speed in different test period were less than 10%. The ratio of average wind velocity of upper reach to down reach of the tunnel was between 0.92 and 1.03. Conclusion The flow field of the air dynamic wind tunnel is even, stable and coincident.
4.Quantification of expression of leukotriene B4 inducing tumor necrosis factor-? and interleukin-1? at mRNA level in synovial membrane cells of rheumatoid arthritis by real-time quantitative PCR

Objective: To investigate quantification of expression of LTB4 inducing IL-1? and TNF-? at mRNA level in synovial membrane cells of rheumatoid arthritis. Methods: Primary cultured synovial cells from RA patients were treated with exogenous LTB4,MK-886(inhibitor of 5-lipoxygenase activating protein) and Bestatin(inhibitor of leukotriene A4 hydrolase) in the presence of LIT respectively, expressions of TNF-? and IL-1? were detected at mRNA level by Real-time Quantitative PCR. Results: Expressions of basic TNF-?(TNF-?/GAPDH) and IL-?(IL-?/GAPDH) at mRNA level in primary cultured synovial cells were 0.02?0.00 and 0.16?0.01 respectively. LTB4(10~ -9 mol/L-10~ -8 mol/L) was shown to induce dose-dependent increase of mRNA expression of TNF-?.(7-15 times) and IL-1? (1 time) , endogenous product of LTB4 by LIT significantly increased mRNA expressions of TNF-? (145 times) and IL-1?(12 times) respectively. LIT-treated synoviocytes with addition of MK-886 (5-LOX exciting protein FLAP inhibitor) (1-10 ?mol/L) were inhibited to secrete LTB4 dose-dependently, following the markedly down-regulated expressions of TNF-? (15%-66%) and IL-1? (41%-71%) at mRNA level . Bestatin(100 mg/L) could also remarkably diminish LTB4-induced mRNA expressions of TNF-?(86%) and IL-1?(79%). Conclusion: LTB4 of synovial membrance cells in rheumatoid arthritis could induce expressions of TNF-? and IL-1? at mRNA level, and their expression at mRNA level had been quantified successfully. It is a beneficial help to quantify all kinds of cytokines in methodology.
5.Effects of parecoxib sodium on laparoscopic cholecystectomy patients after remifentanil fast track anesthesia

Objective To observe the effects of parecoxib sodium on laparoscopic cholecystectomy ( LC ) patients after remifentanil fast track anesthesia.Methods 120 patients underwent general anesthesia for laparoscopic cholecystectomy were divided into two groups using random number table,60 cases in each group.The observation group was injected parecoxib sodium 40mg+saline solution 5 mL,while the control group was injected only saline so-lution 5mL prior to induction of anesthesia.The heart rate,mean arterial pressure,awaked time,pain score,agitation score and adverse reaction were compared.Results 5, 15, 30min after extubation, heart rate were ( 75.7 ± 4.8)times/min,(79.3 ±6.2)times/min,(74.2 ±5.2)times/min in the observation group and (85.2 ±3.8)times/min, (88.2 ±5.9)times/min,(86.1 ±6.2)times/min in the control group,and the mean arterial pressure were (103.5 ± 11.3)mmHg,(104.7 ±12.6)mmHg,(108.4 ±12.7)mmHg in the observation group and (132.3 ±16.3)mmHg, (135.2 ±14.6) mmHg,(133.6 ±11.8) mmHg in the control group,there were statistifically significant differences (t=4.722,4.357,4.236,4.732,4.367,4.063,all P <0.05).The pain score,agitation score were (2.5 ± 0.9)points,(1.4 ±0.8) points in the observation group,which were lower than (4.2 ±1.2) points,(3.2 ± 1.2)points in the control group(t =4.232,5.234,all P<0.05).Conclusion Parecoxib sodium can reduce the fast-track anesthesia agitation and pain,improve hemodynamics and stress response in LC patients.
6.Analysis of the risk factors of 471 patients with stroke in Huainan area

Objective To explore the composition situation of the risk factors of patients with stroke in Huainan area. Methods 471 continuous registration and hospitalized patients with stroke in Huainan were selected,of which,362 cases were ischemic stroke,109 cases were hemorrhagic stroke,and the known risk factors were statistically analyzed. Results Of all patients,the incidence of hypertension was first(69. 3% was in ischemic stroke, 77. 9% was in hemorrhagic stroke) ,and hypertension was the most important risk factors. The composition of risk factors in stroke patients of different gender were different,and the smoking,drinking problem was more prominent in male patients (51.1% ,33.2%respectively)than that in female patients(5.1% ,2.0% respectively) (x2 =111-81,67.62, all P<0.01) ;The incidence of diabetes, heart disease, atrial fibrillation was higher in female patients (37. 6% ,31. 9% ,19.3%respectively) than that in male patients (22.3% ,20.4% ,8.8% respectively) (x2 = 13. 12,8.09,11. 12,all P<0.01). Hypertension,diabetes accounted for only 40.8% and 47.4% of patients taking regular medication. Atrial fibrillation was more common in patients with hemorrhagic stroke. Conclusion Controlling blood pressure and blood sugar was still important in the primary prevention and secondary prevention of stroke in the region. People of different gender should be targeted for individual health education, and promoting a healthy lifestyle should be paid attention, so as to the occurrence of stroke could be effectively prevented.

10.Comparative study on the characteristics of bone metabolism in forelimb malformation WHBE rabbits via the OPG/RANK/RANKL system

Objective To comparatively investigate the characteristics of bone metabolism in forelimb malformation WHBE rabbits based on the OPG/RANK/RANKL system.Methods Healthy male 2-2.5-month old WHBE rabbit ( HWR) , Healthy Japanese rabbit ( HJR) and forelimb malformation WHBE rabbit ( FMWR) were used in this study and divided into 3 groups, HWR, HJR and FMWR groups, with 10 rabbits in each group.The shape of forelimb and mean gray value observed from X-ray film were examined, and the bone tissue micro-morphology was analyzed using HE staining.Re-al-time fluorescence quantitative PCR was used to determine the expression of osteoprotegerin ( OPG) and receptor activator of NF-KB ligand ( RANKL) mRNA.The expressions of OPG, receptor activator of NF-KB ( RANK) and RANKL protein in serum and bone tissue were assayed by enzyme-linked immunoassay ( ELISA) and immunohistochemistry, respectively. Results Compared with the HWR and HJR groups, rabbits in the FMWR group appeared abnormal in toutuous forelimb and thinner bone cortex.The mean gray values of X-ray in the FMWR group were lower than that in the HWR group ( P<0.05).There were significant differences between the FMWR and healthy rabbits (HWR and HJR) in the following pa-rameters:RANKL mRNA expression and RANKL/OPG mRNA ratio in the liver(P<0.05, P<0.01), serum protein expression of RANK and RANKL and RANKL/OPG ratio (P<0.05, P<0.01), and positive index of expression of RANKL protein and RANKL/OPG ratio in the bone tissue(P<0.05, P<0.01).Furthermore, the gene expression lev-els of OPG and RANKL of HWR were significantly higher than that of HJR(P<0.05, P<0.01).Conclusions The FMWR show some abnormal symptoms in bone metabolism as well as decrease of bone quality and histological changes of bone micro-structure, due to the significant increase of RANKL/OPG ratio of FMWR.The breed differences between WHBE and Japanese rabbits in RANKL mRNA expression level may be one of factors inducing limb malformation in the WHBE rabbits.
