3.Effects of basic fibroblast growth factor and sucralfate on healing of experimental gastric ulcers in rats

Objective To investigate the effects of basic fibroblast growth factor(bFGF)and sucralfate on healing of experimental gastic ulcer in rats. Methods Thirty-two famale Sprage-Dawley rats were divide into 4 groups,averagely.bFGF 100 ng?100 g -1 ?d -1 , sucralfate 40 mg?100 g -1 ?d -1 , bFGF 100 ng?100 g -1 ?d -1 + sucralfate 40 mg?100 g -1 ?d -1 and normal saline were used in experimental gastric ulcer rats induced by acetic acid for 21 days. Contents of gastric acid and pepsin were measured,and the healing rate and quality of gastric ulcer and the number of capillaries in ulcer bed were evaluated. Results The amount of gastric acid and pepsin in bFGF group had no different from those of normal saline group. The ulcer areas in bFGF group and sucralfate group were (2.9 ?0.8) mm 2 and (3.2?1.2) mm 2 respectively,which were significantly smaller than that of normal saline group (6.8? 1.4 ) mm 2, P

Tyrosine kinase inhibitor (TKI) may significantly improve the treatment outcome in chronic myeloid leukemia (CML).It is the most frequent question about whether the patients with durable complete molecular response (CMR) can safely discontinue TKI treatment without relapse.This has focused attention on the strategies to eradicate residual CML cells,especially the CML stem cells,which should result in long term leukemia-free survival and permanent cure.Here,the progress on discontinuation of TKI therapy and alternative approaches to eradicating CML stem cells in the 55th ASH annual meeting is reviewed.
5.Effects of POH in combination with STI571 on the proliferation and apoptosis of K562 cells.

The effects of monoterpene perilly alcohol (POH) alone or in combination with STI571 on the proliferation and apoptosis of the cell line K562 positive for Bcr/Abl were investigated. By using cell culture, the effect of the drugs on the proliferation of the cells was studied. TUNEL and flow cytometry assay of FITC-Annexin V and PI labeled cells were applied to detect the effects of the drugs on the apoptosis of the cells. The results showed that at 36 h, IC50 of POH on K562 positive for Bcr/Abl and HL-60 negative for Bcr/Abl were 81.0 +/- 11.3 micromol/L and 113.6 +/- 23.4 micromol/L respectively (P>0.05). POH could inhibit the proliferation of K562 in a time- and dose-dependent manner with the inhibitory rate of 100 micromol/L POH on K562 cells at 36 h being (53.2 +/- 3.65)%. K562 cells were more sensitive to STI571 than POH. IC50 of STI571 on K562 cells in 36 h was (0.256 +/- 0.054) micromol/L. In a time- and dose-dependent manner, POH induced the apoptosis of K562 cells with the percentage of apoptotic cells by 100 micromol/L POH at 40 h being (21.0 +/- 3.3)%. Both 100 micromo/L POH and 0.2 micromol/L STI571 had the same inhibitory effects on the K562 cells at 36 h. But at 12 and 24 h, the inhibitory rate of POH was significantly higher than that of STI571 (P<0.05) and the ability of STI571 inducing apoptosis at 36 h was greater than that of POH. 50 micromol/L, 100 micromol/L and 200 micromol/L POH in combination with 0.2 micromol/L STI571 could obviously increase the inhibitory effects on the cellular proliferation. Combined use of 50 micromol/L, 100 micromol/L, 200 micromol/L with 0.2 micromol/L STI571 could strongly induced apoptosis, especially 200 micromol/L POH in combination with 0.2 micromol/L STI571. It was concluded that the antileukemia effect of POH had no obvious Bcr/Abl positive selectivity. POH can inhibit the proliferation of K562 and induce the apoptosis in a time- and dose-dependent manner. K562 cells were more sensitive to STI571 than POH. POH in combination with STI571 could obviously enhance the abilities of STI571 inhibiting the proliferation and inducing apoptosis of K562 cells.

6.Microwave ablation for the treatment of hepatic cavernous hemangiomas:its serious complications and their prevention and treatment

Objective To analyze the complications caused by microwave ablation for hepatic cavernous hemangiomas, and to discuss the prevention and treatment of the complications. Methods During the period from July 2009 to May 2011 at authors’ hospital, a total of 34 times of microwave ablation procedure were carried out in 30 patients with hepatic cavernous hemangiomas. The microwave ablation-related complications were recorded and the prognosis was evaluated. Results Serious complications occurred in 2 patients (6.7%), including acute hemolysis leading to acute renal failure (n = 1) and abdominal wall abscess (n = 1). Postoperative fever was seen in 18 cases (60%), transient hemoglobinuria in 4 cases (13.3%), nausea with vomiting in 5 cases (16.7%), marked pain at the surgical area in 8 cases (26.7%), right pleural effusion in 4 cases (13.3%), hepatic dysfunction in 24 cases (80%) and leukocytosis in 11 cases (36.7%). No treatment-related death occurred. Conclusion For the treatment of hepatic cavernous hemangiomas, microwave ablation is minimally invasive with higher safety and satisfactory effect, although this technique carries somewhat higher risks when the hemangioma is larger or the location of the hemangioma is anatomically complicated. Some of these complications can be early detected and proper management should be actively adopted in order to avoid as well as to minimize the adverse results.
7.The surgical treatment of female stress urinary incontinence recurrence after operation

Objective To evaluate the effect and safety of transobturator inside-out tension-free urethral suspension (TVT-O) as the surgical treatment on post-operational recurrence of female stress urinary incontinence (SUI). Methods Eleven women with SUI recurrence who underwent TVT-O were selected in the research. Their incontinence recurred from 1 months to 8 years after the previous surgical treatment.Results The operative time was (28 ±9) min, the bleeding volume was (15 ±5) ml. Postoperative complications: urinary retention in 1 case, vaginal erosion in 1 case. Follow-up was enforced for 6-36months, median time was 16 months. Grouts-Blaivas analogue scale:among the 11 patients, 9 patients were cured, 2 patients were improved. ICI-Q-SF score: preoperative 13-21 ( 15.36 ± 2.34) scores compared to postoperative 0-9 (1.46 ±3.27) scores (t =5.850,P <0.01 ). I-QOL score: preoperative 23.33-36.67(29.85 ± 4.74) scores compared to postoperative 66.67-100.00 (95.15 ± 10.12) scores (t = 17.753,P <0.01 ). PISQ-12 score of 10 patients: preoperative 16-25 (20.10 ± 3.07) scores compared to postoperative 11-20( 14.00 ± 2.83 ) scores (t = 11.600, P < 0.01 ). Conclusion If conservative treatments fail, TVT-O is highly effective and safe for the postoperative recurrent SUI.
8.Prokaryotic clone, expression and immunological analysis of recombinant Schistosoma japonicum cyclophilin B

Objective To clone and express Schistosoma j aponicum cyclophilin B (SjCyPB) gene in E.coli,and to identify and analyze the immunity of recombinant proteins.Methods A pair of specific primers was designed according to GenBank of Schistosoma japonicum sequence.SjCyPB gene was amplify by PCR and then connected to pET28 vector.The recombinant plasmid pET28a (+)-SjCyPB was constructed and transformed into E.coli BL21 cell line,the recombinant plasmid was identified by double enzyme digestion and sequence analysis.After induced by isopropyl-B-D-thiogalaetoside (IPTG),the expressed recombinant proteinwas purified by affinity-chromatography,and then verified by Western blotting.Rats were immunized recombinant SjCyPB,andthe SjCyPB-specific IgG was detected by ELISA.Results SjCyPB gene was successfully inserted into pET28a(-) vecter which identified by double enzyme digestion and sequence analysis.Recombinant SjCyPB protein was highly expressed in E.coli.The Western Blotting analysis confirmed that the recombinant protein could specifically combine to S.japonicum-infected rabbit serum.Using recombinant protein to immunize rats,the SjCyPB-specific IgG antibody titer was 1 ∶ 51 200 detected by ELISA.Conclusion The recombinant SjCyPB is successfully constructed,and recombinant SjCyPB has immunogenicity and antigenicity.
9.The effect of EC on the ET-induced fever in rabbits

The effects of intravenous injection of purifed EC on the rectal tempera-ture in normal rabbits and febrile rabbits induced by ET and concentrations of Fe~(2+), Zn~(2+),Cu~(2+) in serum were observed. The results showed that EC could lower the rectal tempera-ture in normal rabbits and also the increased rectal temperature in febrile rabbits. Mean-while, EC could inhibit the formation of the second phase of ET-fever. But EC had noeffect on the changed concentrations of FC~(2+), Zn~(2+), Cu~(2+) in serum caused by ET. Thesefindings suggest that EC may particiate in the regulation of body temperarure under physio-logical and febrile conditions, but not of acute phase response produced by ET.
