2.Construction of eukaryotic expression vector of pre-miR-15a and its inhibitory effect on Raji cells proliferation

Objective To construct an eukaryotic expression vector of pre-miR-15a,and to investigate the inhibitory effect of pre-miR-15a to Raji cells. Methods The pGCSIL-GFP vector encoding pre-miR-15a nucleotides was transfected into the bacterial competent cells,and then confirmed by PCR and sequencing analysis. The identified vector was transfected into Raji cells with oligofectamine 2000. The cells were divided into 3 groups,blank,negative control and pre-miR-15a group. Semi-quantitative RT-PCR was used to detect the expression of Bcl-2 mRNA,and immunofluorescence indirect for Bcl-2 protein expression. The growth of Raji cells was assayed by trypan blue dye exclusion method. Results PCR and sequences analysis indicated that the recombinant clones was identical with target sequences. Many green fluorescent cells were observed under fluorescent microscopy. The levels of Bcl-2 mRNA at every group had no obviously difference. Bcl-2 protein expression was obviously decreased at pre-miR-15a group compared with the other groups. Trypan blue dye exclusion method showed the cell growth was inhibited at 48 h and 72 h post-transfection. Conclusion We successfully construct the eukaryotic expression vector of pre-miR-15a,and it can inhibit the growth of Raji cells.
3.THE MACRO-MICROANATOMY OF THE VESSELS OF THE MEDICAL THORACIC FLAP

It is scanty in reports about the anatomy of the medial thoracic free flap, that which one of the perforating arteries of the internal thoracic artery is the main arterial source of this flap is still in dispute. This investigation was based on dissection of 50 sides of adult cadavers under the operating microscope.Among the perforating arteries of the internal thoracic artery in the upper four intercostal spaces, the second perforating artery presents a higher incidence of occurence, it also possesses a larger caliber and distributing area. So that it would be the first choice as a pedicle artery in the transplantation of the medial thoracic free flap. The next choice is the first perforating artery, because its distributing area is larger than that of the third as well as the fourth arteries.The second intercostal space is the widest among all other spaces. The internal thoarcic artery in this space is located more superficial and stands further apart from the lateral border of the sternum than that in the other three intercostal spaces. All these advantages are in favor for taking the second perforating artery as the pedicle artery of the free flap or even to take the internal thoracic artery of the same segment together with it. If a portion of the second costal cartilage isresected, the first two perforating arteries and their related portion of internal thoracic artery can be involved in the pedicle, and the area of the skin flap will be increased to a large extent.Some of the applied anatomy of the transplantation of the medial thoracic free flap has been discussed.
4.Liposuction combined with periareolar small incision to treat gynaecomastia

Objective To sum up the experience of liposuction combined with periareolar small incision to treat gynaecomastia.Methods 25 cases of gynaecomastia were treated by liposuction combined with periareolaf small incision within local swelling anesthesia from Jun.2012 to Jun.2015 in Department of Burn and Plastic Surgery.Results All the 25 cases were satisfied with the postoperative cosmetic effect,such as no obvious scar,flat chest,no obvious uneven feeling.Pathological results showed there were mammary gland hyperplasia,mam mary gland dilatation and fat and fibrous tissues hyperplasia.Conclusion Liposuction combined with periareolar small incision under local swelling anesthesia can achieve satisfied cosmetic effect in treating gynaecomastia.
5.Acitretin combined with total glucosides of peony for psoriasis:a Meta analysis

Objective To evaluate the clinical efficacy and adverse reaction of acitretin combined with total glucosides of peony for patients with psoriasis.Methods Clinical controlled trials that compared acitretin with combination therapy of acitretin and total glucosides of peony for patients with psoriasis were searched in the Cochrane Library,PubMed,Embase and so on(from establishment to October 2016).All extractive data analysis was performed by Review Manage version 5.3 after the data was extracted and the quality was assessed according to the inclusion criteria.Results A total of 8 articles were included.The results of meta-analysis showed that the total effective rate[OR=3.15,95%CI(2.04,4.86),P<0.01] and the psoriasis area and severity index(PASI)score[WMD=-2.05,95%CI(-2.74,-1.36),P<0.05] of treatment group were better than those of control group.The incidence rates of cheilosis,dryness of eye,dryness and itch of skin,dysfunction of liver,and hyperlipemia in treatment group were lower than those in control group(P<0.05).However,the incidence of rhinorrhagia was no statistically significant difference between the two groups(P>0.05).Conclusion Acitretin combined with total glucosides of peony for patients with psoriasis can improve the clinical efficacy and reduce the incidence of adverse reaction.
6.Study on the Antigens of Tumor FBL-3:I.Prediction of FBL-3 Antigen and Verification of Antigenicity of Synthetic Peptides

T lymphocytes play a very important role in tumor immunity, which recognize tumor antigenic peptide presented by MHC molecules on the surface of tumor cells through T cell receptor ( TCR) . Cytotoxic T lymphocytes kill tumor cells after recognizing antigenic peptide in the cleft of MHC class I molecules. It is now generally accepted that peptides naturally presented by a given MHC class I molecule have a specific motif which is referred to as MHC class I allele-specific consensus motif and that synthetic peptide corresponding to the identified naturally presented antigens exhibit remarkable activity in inducing T-cell responses. FBL-3 is a transplantable Friend virus-induced leukemia of B6 (H-2b) origin,which can induce the CTL against FBL-3 by immunizing B6 mice with atenuated FBL-3.Seven peptides were predicted as candidates of FBL-3 antigenic peptides in the light of H-2Db specific peptide binding motif and gag gene sequence of Friend virus. The synthetic peptides, named gagl to gag7, were obtainted. The results showed that CTL specific to gag3 could be induced by in vivo immunizing B6 mice with gag3 in IFA, but a primary T cell response could not be induced by in vitro immuniztion with the peptides. gag3 and gag5 could be recognized by specific CTL to FBL-3,because the CTLs obviously killed target cells (EL-4) pulsed with gag3 or gag5. Immunization with gag3 and gag5 could not induce an immune response to protect the subsequent challenge of FBL-3 in B6 mice which could be induced by immunizing B6 mice with parental FBL-3 tumor cells. This indicates these 7 synthetic peptides may not be the same antigens of the FBL-3.
7.Study on the Antigens of Tumor FBL-3: I . Analysis of Peptide Antigens from Peptides Pool of Tumor Cells by Acid Elution

T lymphocytes, which play a very important role in tumor immunity, recognize the tumor antigenic peptide presented by MHC molecules on the surface of tumor cells. FBL-3 is a transplantable Friend virus-induced leukemia of B6 (H-2~(b) ) origin, which can induce the CTL against FBL-3 by immunizing B6 mice with attenuated FBL-3. The present paper was to elute immunogenic peptides ( CD8+ T cell epitopes) from class I complexes at the cell surface of viable FBL-3 cells by acid treatment. The acid treated cells remained viable but lost sensitivity to be lysed by FBL-3 specific cytotoxic T lymphocytes (CTLs). The sensitivity of the acid-treated cell was restored to the FBL-3 specific CTLs by supplement of the acid-eluted cell-free supematants. Paptides were subsequently fractionated by reverse-phase high performance liquid chromatography (RP-HPLC) . The fractionated peptides in HPLC fractions 5~6 and 23 ~ 60 (tubes) showed the capacity to sensitize RAM-S cells, which could be lysised by FBL-3 specific CTLs. The experiment indicates that this method may be useful in the definition of petide epitopes relevant to tumor cells.
8.Compared the Efficacy of Circumcision Suture Surgery and Surgery Foreskin Cerclage Ring

Objective The curative effect comparison of circumcision suture surgery and surgery foreskin cerclage ring.Methods Selection in our department from July 2013 to October 2016 were selected from children with phimosis or re-dundant prepuce in children with a total of 562 cases as the research object , randomly divided into control group (182 cases) and observation group (380 cases).The observation group using disposable circumcision suture treatment , the control group using prepuce ring resection in the treatment of patients , observe the operation effect , two postoperative pain and complica-tions.Results The observation group operation success rate ( 100%) was significantly higher than the control group (94.09%), observation group operation time , intraoperative bleeding volume , postoperative 24h pain scores were signifi-cantly less than the control group , the adverse reaction rate of observation group ( 6.32%) was significantly lower than the control group (11.54%),the differences between groups were significantly P<0.05.Conclusion Ircumcision surgery su-ture is better than that of ring foreskin cerclage surgery , more safe and reliable .
9.Immunosuppresive effect of the anti—activated mouse lymphocyte antigen monoclonal antibody(2H_3)on the allograft rejection and its mechanism

To investigate the mechanism of inducing a prolongation of allograft survival by immunizing the syngeneic mouse with activated murine spleen cells,we used an anti—activated mouse Tlymphocyte antigen monoclonal antibody(2H_3)made in our laboratory to examine the effect of 2H3on allograft survival in vivo and on cell—mediated immune rection in vitro.The results showed that 2H3 could significantly prolong the allograft survival in vivo and obviously inhibit the proliferation in MLR and CTL generation in bulk MLR as well as the T lymphocyte prolifer-ation to ConA in vitro.These data indicate that one of the mechanisms of the prolongation of al-lograft survival by immunizing the syngeneic mouse with activated murine spleencells may be due to the generation of an anti—activated mouse lymphocyte antigen antibodies in immunized ani-mals which can block process of allorection.

Microarray technique, i. e. gene-chip technique includes gene-chip development, pretreatment of detected samples, probe hybridization, detection data analysis. The microarray technique in toxicology mainly was applied to identification of potential hazardous substances, screening for mechanism of action, dose-response relationship assessment, identification of interaction among chemical mixtures, biomarker of exposure and biomarker of susceptibility. Microarray technique has been used to study biomarker for human bladder cancer, to study fingerprints of gene regulation associated with cadmium chloride, benzo(a) pyrene and trichloroethylene. fn this paper, a new automated and multi-applied molecular biology workstation, NanoChip biomolecular system was introduced .
