1.EXPERIENCES IN RE -OPERATION OF LIVER RESECTION OF PRIMARY LIVER CANCER

From January, 1960 to April. 1983, 292 cases of primary liver cancer were treated by liver resection. Of them, 21 cases underwent re-exploratory operation for relapsed reason and 14 tumours were removed with a resectability rate of 66.7%.In this series recurred tumours in 3 patients were resected by a two-stage operation with hepatic artery ligation or embolizaton as the first step.Following-up results: 7 out of 14 resected cases survivors, 2 survived above one year, 4 more than half a year and 1 less than half a year; in another 7 cases died post-reoperativelly, 1 above one year, 4 more than half a year, 1 less than half a year.As for AFP detection 4 patients were negative and in 1 patient the AFP titre decreased to 100 ug/ml on the 33rd post-reoperative day. From the above data the authors believe that re-operation may be considered as a most effective method if it is performed at a proper time and radiotherapy, hepatic artery ligation or embolization are of great value in treating recurred cases. For the improvment of the surgical results of primary liver cancer,a preliminary scheme for investion and management procedure for such patients is suggested by the authors as well.
2.Optimization of fermentation conditions of molluscicidal endophyte LL3026 from Buddleia lindleyana

Objective To optimize the fermentation conditions of molluscicidal endophyte LL3026 from Buddleia lindleyana. Method The medium composition and cultivation conditions were optimized by orthogonal and single factor experiments. Re-sults The experiments showed that the conditions of initial pH 3,fermentation temperature 30℃,volume of liquid 100 ml(250 ml Erlenmeyer flask),and 3D-xylitol 0.5 g/L were optimum,and the molluscicidal activity of the fermentation filtrate reached 95%. After three hatches of cultivation,the predicted values were verified by validation experiments. Conclusion Endophyte LL3026 from Buddleia lindleyana has a good molluscicidal activity after the optimization.
3.The treatment of oral powered microdedrider adenoidectomy with nasal endoscope in children with secretory otitis media

ObjectiveTo study the effect of of oral powered microdedrider adenoidectomy with nasal endoscope in children with secretory otitis media. Methods74 children with secrctory otitis media treated with adenoidectomy with nasal endoscope randomly divided into group A 37 cases and group B 37 cases,the group A were treated with trans-oral adenoidectomy under endoscope, the group B were treated with trans-nasal adenoidectomy under endoscope,then the total effective rate,operation time, blood loss,incidence of complications and serum IgE, NO, IL-6,TNF-αand IL-2 of the two groups before and after the treatment were compared. ResultsThe cure, effective, and ineffective of group A were 34 cases ,3cases and 0 cases ,the total effective rate was 100.0%, the cure, effective,and ineffective of group B were 26 cases,7 cases and 4 cases,the total effective rate was 89.2% ,the total effective rate of group A was significantly higher than that of group B( x2 =6. 869 ,P <0.05 ) ,the operation time of group A was( 8.6± 1.6) min, blood loss was( 12.3 ± 4. 1 ) ml, incidence of complications was 2.7% ( 1 case postoperative hemorrhage), operation time of group B was ( 14.2 ± 2.1 ) min, blood loss was( 17.8 ± 3.5 ) ml, incidence of complications was 10.8% (2 cases nasal adhesion and 2 cases postoperative hemorrhage) ,there were significant differences between the two groups ( t =5. 967,6.553, x2 =7. 142, all P < 0.05 ), serum IgE, NO, IL-6,TNF-αand IL-2of the group A after the treatment were all lower than those of group B, ( t =5. 557,6. 348,5. 968,6. 976,5. 884, all P < 0. 05 ).ConclusionThe effect of oral powered microdedrider adenoidectomy with nasal endoscope in children with secretory otitis media better,and it was worthy of popularization and application.
4.Relationship between high risk human papilloma virus infection and p53 polymorphisms and esophageal cancer

ObjectiveTo study the relationship between human papilloma virus(HPV) infections,p53 polymorphisms and esophageal cancer.MethodsA case-control study of 204 esophageal cancer cases (esophageal cancer group) and 102 healthy control cases (control group) biopsies were conducted to extract DNA,detected the HPV by PCR method and analyzed p53 polymorphisms by PCR-RFLP method,then analyzed the relationship between them and esophageal cancer incidence rate.ResultsThe positive rate of HPV16 in esophageal cancer group[59.3%(121/204)] was higher than that in control group [11.8% ( 12/102 ) ] (P < 0.05 ).And the frequency of Arg/Arg genttype of p53 in esophageal cancer group [ 51.5% (105/204) ] was higher than that in control group [ 19.6% (20/102) ] (P < 0.05 ).The frequency of Arg/Arg genotypo in HPV16 positive patients [66.9%(81/121 )] was higher than that in HPV16 negative patients [ 28.9% (24/83)] (P < 0.05).ConclusionsHigh risk HPV phys an important role in development of esophageal cancer,p53 Arg/Arg genotype is one of high risk genetic factors for HPV-associated esophageal cancer.
5.Construction of recombinant vectors carrying antisense RNA to dualˉtarget BAGˉ1 and Bclˉ2 and its effect on proliferation of SGCˉ7901 cell

Objective To construct the recombinant co-expression vector carrying antisense RNA to dual-target BAG-1 and Bcl-2 genes and recombinant single expression vector carrying antisense RNA to target BAG-1 and Bcl-2 gene respectively,then to pre-liminarily investigate their effect on the proliferation of gastric cancer cell SGC-7901 in order to lay the foundation for further study the effect of this recombinant vector on the tumor cells.Methods RT-PCR was used to amplify the full length of BAG-1 and Bcl-2 cDNA from total RNA of gastric cancer cell line SGC-7901.The BAG-1 cDNA fragment and the Bcl-2 cDNA fragment were insert-ed into pMD18-T simple vector respectively.The pMD18-T-BAG-1 was digested with BamH Ⅰ and Cla Ⅰ and the pMD18-T-Bcl-2 was digested with EcoR Ⅰ and Nhe Ⅰ.Then the BAG-1 cDNA fragment and the Bcl-2 cDNA fragment were inserted into the mcs1 and mcs2 of the eukaryotic co-expression vector pVITRO2 in the antisense orientation respectively.The construction of the single expression vector pVITRO2-AsBAG-1 and pVITRO2-AsBcl-2 was confirmed by restriction endonuclease treatment and se-quence identification.Then the Bcl-2 cDNA fragment was inserted into the mcs2 of the recombinant vector pVITRO2-AsBAG-1 in the antisense orientation to construct the co-expression vector pVITRO2-AsBAG-1-Bcl-2,and the recombinant vector was also iden-tified by restriction endonucleases digestion and sequence identification.Then the recombinant vector was transfected into SGC-7901 cell respectively.The proliferation of the cell was determined by the MTT assay.The level change of BAG-1 and Bcl-2 mRNA in SGC-7901 cell was detected by the semi quantitative RT-PCR and the change situation of cell cycle was detected by the flow cytom-etry(FCM).Results The restriction endonucleases digestion and sequencing identification indicated that the eukaryotic co-expres-sion vector pVITRO2-AsBAG-1-Bcl-2 and the single gene expression vector pVITRO2-AsBAG-1 and pVITRO2-AsBcl-2 were con-structed successfully.The MTT assay method demonstrated that compared with the control group,the recombinant vector could in-hibit the proliferation of the cells in time dependent manner,and the inhibiting effect was most notably in the 72 h transfection group(P <0.01);inhibition ratio of recombinant vector groups was significantly higher than that of control group and pVITRO2 group(P <0.01).BAG-1 and Bcl-2 mRNA expression in recombinant vector groups were significantly decreased compared with that in the control group and pVITRO2 group(P <0.01),The RT-PCR results showed that the expression level of BAG-1 mRNA and Bcl-2 mRNA was significantly decreased(P <0.01),moreover the co-expression vector group was more notably than the single expression vector groups(P <0.05),but the pVITRO2-AsBcl-2 had no obvious effect on the expression of BAG-1 mRNA(P >0. 05);the FCM detection results showed that the apoptosis rate of the recombinant vector groups was significantly higher than that of the control group and the pVITRO2 group with statistical difference(P <0.01),and the co-expression vector group was more nota-bly than single expression vector groups(P <0.01).In addition,the effect of the co-expression recombinant vector group was more significant than that of the single expression co-expression vector(P <0.01 ),the apoptosis rate was increased from 0.57% to 15.75%.Conclusion The co-expression recombinant vector pVITRO2-AsBAG-1-Bcl-2 and single expression vector pVITRO2-As-BAG-1 and pVITRO2-AsBcl-2 are successfully constructed and they can inhibit the proliferation of SGC-7901 cell and induce cell apoptosis,moreover the pVITRO2-AsBAG-1-Bcl-2 vector is most notably.
6.Simulating calculation of passenger brain tissue deformation during vehicle crash accident using finite element method

Objective To establish the plane strain model of brain tissues on mid-sagittal plane and discuss the relation between the brain tissue deformation and brain injury. Methods By referring to the computerized tomography pictures of the human body and using plane strain and finite element method (FEM), we applied plane strain hypothesis to establish a model for calculation of the brain tissue deformation caused by rotational inertia loading during vehicle crash accident. Results At the 12th ms, the shear strain field adjacent to parietal lobe reached 0.148 and the maximum positive shear stress located at white material of the cerebellum was up to 8 088.0 Pa. At the 28th ms, the maximum shear stress located at the cerebellum reached 900.9 Pa. Great shear strain existed in the cerebrospinal fluid region all the time and shear stress with large absolute value presented on the boundary of white matter and gray matter. Conclusions Under rotational inertia loading, the shear strain (stress) in brain tissues can lead to diffuse axonal injury. The uneven strain (stress) may cause serious avulsion damage in brain tissues.
7.Cellular signal transduction of the hypoxia response

Hypoxia is a common physiological and pathological stimulus in many human diseases .The cellular oxygen sensors and the following activation of multiple cellular signal transduction pathways involved in hypoxia responses can regulate cell survival as well as respiration , blood circulation , metabolism and so forth .The cell response to hypoxia has a complex diversity .Hypoxia-induc-ible factor ( HIF) pathway in an oxygen dependent manner plays a central role during the hypoxia response .The HIF-independent path-ways are equally important under hypoxic conditions which can maintain the oxygen balance and metabolism balance .
8.The role of TRPM7 ion channel in the proliferation and migration of oral squamous cancer cells

Objective:To investigate the role of the transient receptor potential melastatin subfamily member 7 (TRPM7) ion channel in the proliferation and migration of oral squamous cancer cells OC2.Methods:The expression of TRPM7 in OC2 cells was examined by western blotting,RT-PCR and indirect immunofluorescence assay.The influence of TRPM7 on cell proliferation and migration under the inhibition of TRPM7 ion channel with 2-APB or siRNA TRPM7 in OC2 cells was examined by MTT and Transwell respectively.The influence of PI3K/AKT signal pathway on the expression of TRPM7 in OC2 cells was tested by Western blotting.The change of positive ion current when the OC2 cells were treated with TRPM7 inhibitor and activator was explored with patch clamp technique.Results:TRPM7 overexpression in OC2 cells mainly in the cytoplasm was observed,siRNA TRPM7 downregulated the expression of TRPM7.Inhibition of TRPM7 (2-APB or siRNA TRPM7) inhibited the growth and proliferation of OC2 cells in a time-and dose-dependent manner.Blockage of TRPM7 ion channel inhibited the migration capacity of OC2 cells.Inhibition of TRPM7 downregulated the expression of phosphorylated AKT and phosphorylated ERK.Patch clamp assay showed the activation of TRPM7-like current in OC2 cells.2-APB could weaken this current while Bradykinin could enhance the current.Conclusion:TRPM7 may regulate the proliferation and migration of oral cancer cells by the regulation of PI3K/AKT and MAPK/ERK signal pathways and cell positive current.
9.Design of Wireless Data Transmission ECG Holter Based on MC35I

Objective To design an ECG Holter system based on wireless data communication. Methods The system utilized low power microcontroller MSP430F149 to achieve wireless data transmission by module MC35I. ECG data which stored temporarily in SD card were transmitted in the form of SMS through wireless networks. The received messages were converted into intact files of ECG by data integration. Results It realizes ECG playback, analysis and can send messages automatically to inform relatives and doctors. Conclusion The preliminary clinical application indicates that the Holter has stable performance and strong anti-interference ability, and especially suitable for critical patient care.
