3.Role of Toll-like receptor 3 in the immunoregulatory function of embryonic bone marrow mesenchymal stem cells on Th17 cells
Chinese Journal of Tissue Engineering Research 2014;(45):7217-7221
BACKGROUND:Previous studies have found that embryonic bone marrow mesenchymal stem cel s can promote human Th17 cel proliferation, but the inherent regulatory mechanisms stil need to be elucidated. OBJECTIVE:To investigate the role of Tol-like receptor 3 in the immunoregulation of Th17 cel s by mesenchymal stem cel s.
METHODS:Human CD4+T cel s from healthy donors were isolated by immunomagnetic bead method, and then cultured alone or co-cultured with embryonic bone marrow mesenchymal stem cel s for 4 days. The mRNA expression level of interleukin-17, Tol-like receptor 3 and MyD88 was detected by real-time PCR.
RESULTS AND CONCLUSION:Compared with CD4+T cel cultured alone group, the mRNA level of interleukin-17 was significantly higher in the co-culture group (3.59±0.11 vs. 1.14± 0.08, P<0.01). Consistent with the expression of interleukin-17 mRNA, increased level of Tol-like receptor 3 mRNA was detected in the co-culture group compared with the CD4+T cel cultured alone group (3.10±1.60 vs. 0.94± 0.01, P<0.05). Furthermore, MyD88 in the co-culture group was significantly higher than that in CD4+T cel cultured alone group (2.29±0.05 vs. 1.85±0.31, P<0.01). Tol-like receptor 3 may be involved in the immunoregulation of Th17 cel s by embryonic bone marrow mesenchymal stem cel s, which provides experimental evidence for potential cel therapeutic strategy.

Another 30 healthy people were used as controls. The concentrations of plasma GH , insulin growth factor 1 (IGF-1) and insulin growth factor binding protein 3 (IGFBP-3) were measured by radioimmunoassay, and growth hormone sensitivity was assessed by log(IGF-1 /GH). Results Plasma concentration of GHC[(5. 83 ? 1. 41) mlU/L vs (1. 56 ? 0. 97) mlU/L were higher in grade Ⅳ group than those in normal subjects ( P
5.Advancement on the use of percutaneous mechanical circulatory support devices in severe cardiovascular diseases
Chinese Critical Care Medicine 2016;28(12):1161-1164
Percutaneous mechanical circulatory support (PMCS) devices are effective in the treatment of patients with cardiogenic shock and acute decompensated heart failure, as a bridge to the recovery of heart function or further treatment. The intra-aortic balloon pump (IABP) is now the most widely used PMCS. New PMCS devices including Transseptal device of Tandem-Heart, transaortic valve device of Impella and extracorporeal membrane oxygenation (ECMO) might provide more effective hemodynamic supports. Doctors should choose appropriate PMCS devices and their working modes, according to patient's clinical conditions, based on the working principles and hemodynamic effects of the devices, in order to achieve the best effects and help patients live through the crisis of diseases.

Their ages ranged from 34 to 62 yr and weights from 48 to 70 kg. The patients were premedicated with intramuscular morphine 0.2 mg?kg-1 and scopolamine 0.3mg. Radial artery was cannukted for direct BP monitoring and Swan-Ganz catheter was placed via right internal jugular vein for hemodynamic monitoring before induction of anesthesia.TOF,BIS,ECG and SpO2 were also monitored throughout anesthesia. Anesthesia was induced with midazolam 0.05-0.1 mg?g-1 and fentanyl 10-15?g?kg-1. The patients were randomized to receive either rocuronium 0.6 mg?kg-1 (group Ⅰ,n=10) or vecuronium 0.1 mg?kg-1(group Ⅱ,n=10) to facilitate tracheal intubation when BIS value dropped to 60%.The patients were mechanically ventilated (VT 8-10 ml?kg-1 ,RR 10-12 bpm).Systolic arterial pressure (SAP),MAP,HR,cardiac output (CO), PCWP, CVP, mixed venous blood O2 saturation (SvO2), BIS and TOF were recorded and stroke volume (SV), stroke index (SI), LVSWI and rate-pressure product (RPP) were calculated before anesthesia (To), 1 min after administration of muscle relaxant (T1), when TOF reached 0 (T2) and 1,2,3,4,5,7,10,15,20,25,30 min after tracheal intubation (T3-13).Results The demographic data including age, sex and body weight were comparable between the two groups. In rocuronium group HR increased by 17.43% -7.54%,SAP increased by 16.94% - 12.3% and RPP increased by 13.96% - 22.67% respectively during 1-7 min after intubation (T3-8) as compared with the baseline values (To), significantly higher than those in vecuronium group (P
8.Experimental Study on Mizolastine inhibiting Allergic Contact Dermatitis in Murine Model
Chinese Journal of Dermatology 1994;0(06):-
Objective To investigate the mechanism of mizolastine inhibiting allergic contact dermatitis (ACD) in murine model. Methods The murine model of allergic contact dermatitis by topical DNFB was used. Using this model murine inhibition of ear swelling was observed after oral administration of mizolastine in different doses. The levels of IFN-?, TNF-? and IL-4 in the sera of these mice were detected by enzyme-linked immunosorbent assay (ELISA). Results Murine ear swellings were markedly suppressed in each dose group of mizolastine (P 0.05). Different doses of mizolastine inhibited the expression of these three cytokines to different degrees. Conclusion The therapeutic effect of mizolastine on murine ACD may be played by inhibiting the expression of some cytokines.
9.Effects of hemin on the expression of hemeoxygenase in human umbilical vein endothelial cells
Journal of Chinese Physician 2001;0(02):-
Objective To observe the effects of hemin on the expression of hemeoxygenase(HO) on the human umbilical vein endothelial cells. Methods The hemin and zinc protoporphyrin-Ⅸ were added in the cultured endothelial cells to induce and inhibit the expression of HO-1,respectively.The expressions of HO-1 and HO-2 mRNA were detected using reverse transcription polymerase chain(RT-PCR) and the relative amount of CO released into the media was quantitated as carboxyhemoglobin(COHb) by enzyme linked immunosorbent assay(ELISA).The proliferation of endothelial cells was detected by methyl thiazolyl tetrazolium test(MTT).Results The expression of HO protein was mainly detected in the cytoplasm of endothelial cell. The expression of HO-1 mRNA in endothelial cell and the amount of COHb in the media increased significantly with the treatment of hemin. Conclusion The expression of HO-1 and the increase of endogenous CO are the molecular bases of the proliferation of endothelial cells, and the HO/CO system may play an important role in the development of cardiovascular diseases.
