1.Ginkgo-damole injection combined with bone marrow mesenchymal stem cell transplantation improves neurologic recovery from cerebral infarction

BACKGROUND:Reconstruction of damaged brain tissue through cel transplantation has become a new way to treat cerebral infarction. In recent years, bone marrow mesenchymal stem cels have become the new darling in cel transplantation therapy.
OBJECTIVE:To investigate the effect of ginkgo-damole injection combined with bone marrow mesenchymal stem cel transplantation to improve the neurological function of acute cerebral infarction rats and its mechanism.
METHODS:Animal models of middle cerebral artery occlusion were made in rats using suture method, and then 60 rat models were randomly divided into control group, cel transplantation group and combination group. The control group was given intravenous injection of PBSvia the tail vein; the cel transplantation group was given intravenous injection of bone marrow mesenchymal stem cel suspension (2.5×109/L) via the tail vein; the combination group was given intravenous injection of bone marrow mesenchymal stem cel suspension (2.5×109 /L) and ginkgo-damole injection (2 mL/kg, once a day, totaly 5 days)via the tail vein. Modified neurological severity scores were recorded at 1, 3 days and 1, 2 weeks after transplantation. At 2 weeks after transplantation, expressions of brain-derived neurotrophic factor and growth associated protein 43 in the brain were detected using RT-PCR; cel apoptosis detected using MTT assay; BrdU positive cels counted using
immunohistochemistry method.
RESULTS AND CONCLUSION:There were no differences in the modified neurologic severity scores among the three groups at 1, 3 days after transplantation (P > 0.05), but the modified neurological severity scores in the combination group were lower than those in the cel transplantation group and control group at 1, 2 weeks after transplantation (P < 0.05). The expressions of brain-derived neurotrophic factor and growth associated protein 43 in the brain were significantly higher in the combination group than the other two groups at 2 weeks after transplantation (P < 0.05); compared with the other two groups, the number of apoptotic cels was less but the number of BrdU positive cels was higher in the combination group (P < 0.05). These findings indicate that the combination of ginkgo-damole injection and bone marrow mesenchymal stem cel transplantation can increase the expressions of brain-derived neurotrophic factor and growth associated protein 43 in the brain, inhibit cel apoptosis and improve neurological function in rats with cerebral infarction.
2.The clinical characteristics of children with splenium of corpus callosum solitary lesions displayed on MRI

Objective To explore the prominent clinical features and the prognosis of corpus callosum solitary lesions cases in children.Methods There were 16 cases with corpus callosum solitary lesions receiving treatment at our hospital from January to December 2013.All of the clinical features were collected and retrospectively analyzed.Results The 16 cases aged 1 year to 13 years with an average age of (3.44 ± 3.65) years.The causes of the disease included the viral encephalitis,convulsions,and rotavirus infection.The most prevalent disorders were convulsions (10 cases),gastrointestinal symptoms (8 cases),fever (8 cases),positive cervical resistance (6 cases),fatigue (5 cases),altered states of consciousness (2 cases),headache (1 case),and dizziness (1 case).Corpus callosum lesions lasted for 13-50 days,with an average of (28.8 ± 10.2) days.Cranial magnetic resonance showed the splenium of the corpus callosum with isolated oval lesions,low signal on T1 weighted images and apparent diffusion coefficient images,high signal on T2 weighted images and diffusion weighted images.Conclusions The splenium of the corpus callosum lesions of children occur in a lower rate than adults with the etiology including intracranial infection,convulsions and rotavirus infection.Main clinical manifestations include fever,convulsions and gastrointestinal symptoms (nausea,diarrhea,etc).The cases followed-up showed the lesions and symptoms are reversible,and the prognosis is good.
3.Pathology of Glial Scar after Spinal Cord Injury

ObjectiveTo explore the time of chronic spinal cord injury and to determine the time and spatial distribution of glial scar after completely transected rat spinal cord injury. Methods24 adult female Wistar rats were completely transected spinal cord. The responses of activated glial fibrillary acidic protein (GFAP), NG2 and the length of the cavity on the 1st, 3rd, 7th day, 2nd week, 1st, 2nd, 3rd and 3.5th month after operation were examined with histochemistry and immunohistochemistry. ResultsThe length of the cavity and the expression of GFAP begun to be stable at the 2nd month post injury, and were fully stable at 3rd month. The expression of NG2 begun to be stable at the 1st month post injury, and fully stable at 3rd month. ConclusionThe injury begun to stabilize at 1st month post injury, and fully stabilized at 3rd month.
4.Meta Analysis of Relationship between Helicobacter pylori Infection and Autoimmune Thyroid Diseases

Objective To investigate the association between Helicobacter pylori(H. pylori) infection and Autoimmune thyroid diseases (ATDs). Methods The literatures on the association of H. pylori with ATDs were retrieved by searching databases from the inception of each da?tabase to October 2015. Data extraction and quality assessment were completed by two authors. Meta analysis was performed using RevMan 5.3 software,calculating the odds ratio and 95%confidence interval. Results Twelve papers were included for the meta?analysis. The total sample size was 1 615,with 918 cases and 697 controls,respectively. Compared with the controls,H.pyloriinfection significantly increased the risk of auto?immune thyroid diseases development with a pooled of[OR=1.86,95%CI(1.18,2.94)]. H.Pylori?CagA infection significantly increased the risk of autoimmune thyroid diseases development with a pooled of[OR=2.66,95%CI(1.61,4.41)]. H. pylori infection is associated with Graves dis?ease[OR=3.37,95%CI(1.90?5.97)]and Hashimoto′s thyroiditis[OR=1.83,95%CI(1.22,2.76)]. The results of publication bias and sensitivi?ty analysis confirmed the reliability and stability of this meta analysis. Conclusion H. pylori infection may be associated with an increased risk of developing the autoimmune thyroid diseases.
5.Degradable properties of biodegradable composite stents in the trachea

BACKGROUND:Through a ful investigation of biodegradable scaffolds, we propose a new self-expanding degradable poly-L-lactide coated endotracheal stent based on the design, production, experimental and clinical applications of nickel titanium memory alloy stent. OBJECTIVE:To design a kind of biodegradable endotracheal stent with poly-L-lactide and hydroxyapatite, and to test its mechanical properties, biocompatibility and biodegradation capacity. METHODS:With the technology of computer aided design, the stents were prepared with poly-L-lactide (Mr RESULTS AND CONCLUSION:The average radial supporting force of the tracheal stent was 7.8 kPa, the percentage of stent surface coverage was less than 20%, the stent expansion rate was≥4%, and the stent longitudinal shortening rate was≤9%, which reached the mechanical requirements for degradable endotracheal stents. After 4-16 weeks, there was no significant inflammatory response. The decline in molecular weight changes and weight loss ratio was higher for in vivo degradation than in vitro degradation at different time (P<0.05). These findings indicate that poly-L-lactide/hydroxyapatite composite stents have good mechanical properties, biocompatibility and biodegradability. 150 000) and hydroxyapatite materials, 20 mm to 26 mm in diameter. 6.Efficacy comparison between standard and reduced doses of bortezomib combination therapy in the treatment of multiple myeloma

Objective To compare the efficacy and safety of standard or reduced doses of bortezomib combined with adriamycin and dexamethasone in patients with multiple myeloma (MM).Methods Fifty-two newly diagnosed,refractory and relapsed patients received bortezomib [1.3 mg/m2 (standard dose group,26 patients) or 1.0-1.1 mg/m2 (reduced dose group,26 patients) on day 1st,4th,8th and 11 th],and adriamycin (10 mg/m2) plus dexamethasone (40 mg/d) on day 1 st-4th,and were treated for 1-6 courses.Adverse events were graded and compared.Results After treatment,the overall response rate of standard dose group [80.8%(21/26)] and reduced dose group [88.5%(23/26)] had no significant difference (P =0.739).The rate of neutropenia and thrombocytopenia in two groups had no significant difference[23.1%(6/26) vs.15.4%(4/26),P=0.281 ; 11.5%(3/26) vs.7.7%(2/26),P=0.620].The rate of Ⅲ-Ⅳ grade peripheral nerve disease,herpes zoster,lack of power and abdominal distension in standard dose group were significantly higher than those in reduced dose group [15.4%(4/26) vs.3.8%(1/26),P =0.038;26.9%(7/26) vs.7.7%(2/26),P =0.029;38.5%(10/26) vs.15.4%(4/26),P=0.045; 19.2%(5/26)vs.3.8% (1/26),P =0.028].Conclusion Reduced dose of bortezomib appears to result in similar overall response rate,but better tolerance and safety compared with standard dose.
7.Sunitinib malate-induced high expression of NKG2D ligands in nasopharyngeal carcinoma cell ABCG2~(high) CNE2/DDP

Objective:To investigate the inducing effects of sunitinib malate on expression of NKG2D ligands in nasopharyngeal carcinoma cell ABCG2high CNE2/DDP.Methods:ABCG2highCNE2/DDP cells and Allo-NK cells were isolated by magnetic activated cell sorting(MACS).Flow cytometry was used to evaluate the purity of isolated cells and the expression of NKG2D-ligands on target cells before and after incubation with sunitinib malate.Then the cytotoxic sensitivity of treated and un-treated ABCG2high CNE2/DDP cells to Allo-NK cells were measured by LDH releasing assay.Results:The positive rate of ABCG2 in ABCG2highCNE2/DDP cells was(91.40?2.32)%.More than 90% of isolated Allo-NK cells were proven to be CD3-CD16+CD56+ cells.The expression of MICA,MICB,ULBP1,ULBP2 and ULBP3 on ABCG2high CNE2/DDP cells incubated with sunitinib malate increased from(2.92?0.33)%,(4.27?0.33)%,(5.80?0.62)%,(11.10?3.15)%,and(7.75?1.14)% to(89.12?4.56)%,(66.10?2.22)%,(67.56?4.19)%,(69.37?8.83)%,and(63.28?3.31)%,respectively.At the E ∶T ratios of 10 ∶1 and 20 ∶1,the cytotoxic sensitivities of ABCG2high CNE2/DDP cells to Allo-NK cells increased from(15.32?13.86)% and(27.26?6.81)% to(41.12?4.12)% and(57.25?2.37)%,respectively,after treatment with sunitinib malate,with significantly difference found in the cytotoxic sensitivities of target cells in each group before and after sunitinib malate treatment(F=15.58,P=0.000).Conclusion:Sunitinib malate can up-regulate expression of NKG2D-ligands(MICA/B,ULBP1-3)in ABCG2high nasopharyngeal carcinoma cells,which results in higher cytotoxic sensitivity to Allo-NK cells.

Objective To provide reference of the general physiological index, blood physiology and biochemistry for middle and old-aged cynomolgus monkey.Methods 119 cynomolgus monkey aged from 11~16 (80 were males and 39 were females) were involved in the study.We detected the general physiological index( body weight, the crown-rump length and waist circumference ) , hematology and blood biochemistry parameters respectively and compared these parameters between males and females.Results Between males and females, there were significant differences in Body weight, the crown-rump length and Waist circumference of the general physiological index(P <0.01),NEUT#、LYMPH#、EO#、BASO#、NEUT%、LYMPH%、EO%、BASO%、MCV、MCH、RDW-CV、PCT、MONO%、HGB、HCT、MCHC、RDW-SD、MPV of hematology(P <0.01 or P <0.05)and TBIL、ALB、GLO、A/G、ALP、GLU、UREA、CREA、TCH、TG、CK、ALT、GGT of blood biochemistry ( P <0.01 or P <0.05 ) .There were no significant differences in other parameters ( P >0.05).Conclusion It provides reference of general physiological index, blood physiology and biochemistry parameters of cynomolgus monkey and basis for their application in biomedical domain.
10.Clinical analysis of superior mesenteric venous thrombosis

Objective To study the diagnosis and treatment of superior mesenteric venous thrombosis (MVT). Methods The clinical data of 13 MVT cases were retrospectively analyzed. Results The diagnosis of MVT in all 13 cases were comfirmed by surgery. Of them,diagnosis for 6 cases was made preoperatively, and misdiagnosed preoperatively in 7.11 patients were cured, and 2 died. Conclusions Because the symptom does not accord with physical sign in the early stage of MVT, the diagnosis of MVT is difficult . B US and CT are useful in the diagnosis of MTV.Intra and post operative coagulation treatment is the key to prevent the recurrence and to reduce the mortality.
