Mitochondria play a central role in the regulation of energy metabolism and signal transduction in eukaryotic cells. Although many fluorescent labeling strategies have been developed for mitochondrial studies, the methods that enable labeling efficiency assessment at the single-mitochondrion level are still lacking. By employing the unique advantages of high sensitivity flow cytometry ( HSFCM ) in the sensitive, rapid, and quantitative multiparameter analysis of individual mitochondria, here we examined the performance of several different mitochondrial labeling strategies from the perspectives of brightness, labeling ratio, and stability. Mitochondria isolated from HeLa cells transfected with pAcGFP1-Mito plasmid upon transient or stable transfections, and mitochondria directly labeled with MitoTracker Green or SYTO 62 were analyzed by a laboratory-built three-channel HSFCM. Upon the quantitative measurement of fluorescence brightness, it was found that the fluorescence intensity of green fluorescent protein ( GFP ) in mitochondria isolated from cells with stable transfection was about 17. 7-fold higher than the transient transfection ones, and was approximately two orders of magnitude brighter than mitochondria labeled with MitoTracker Green. On the other hand, the fluorescence signal of SYTO 62 labeling decreased upon washing, indicating its rapid dissociation rate. The strong fluorescence intensity and good labeling stability make stable transfection an efficient method to label mitochondria. The experimental results demonstrates that HSFCM provides a powerful analytical tool to assess the performance of mitochondrial fluorescence labeling via high throughput single mitochondria analysis.

This article reported a case of fetal giant hepatic hemangioma with cardiomegaly managed with intrauterine treatment. At 23 weeks of gestation, the patient was referred to Guangdong Women and Children Hospital due to abnormal abdominal echogenicity of the fetus, which was suspected to be a hepatic hemangioma or a hepatic arteriovenous fistula. The prenatal ultrasound at 26 weeks of gestation revealed an enlarged fetal hepatic hemangioma of 45 mm×35 mm×42 mm and an enlarged heart (cardiothoracic area ratio of 0.50). So, with the patient's informed consent, the fetus was treated with intrauterine administration of propranolol and dexamethasone and closely monitored by ultrasound. The volume of the lump still increased at the beginning of the medication, but started to shrink in the 7th week. Besides, the fetal cardiac load was reduced and the condition was controlled. The patient delivered at 37 weeks of gestation. The baby received a CT examination on the fourth day after birth which revealed an abdominal mass of 40 mm×30 mm×44 mm requiring no treatment, and no abnormalities were reported during a one-year follow-up.

OBJECTIVE:To investigate the correlation of storage life and effective composition content with color value of Carthamus tinctorius ,and to provide reference for the quality evaluation of C. tinctorius with different years of storage. METHODS:Using 24 batches of C. tinctorius from same place of production with different years of storage (0,1,2 years,8 batches each type )as samples ,the contents of hydroxysafflor yellow A (HSYA)and kaempferol were determined by HPLC. Color value [lightness value (L*),red-green value (a*),yellow-blue value (b*)] were determined by spectrophotometer. SPSS 19.0 statistical software was used to analyze the correlation of storage life and effective composition content with color value. RESULTS:Kaempferol content was still high after 1 year or 2 years of storage (0.161%,0.061%,respectively). However ,the content of HSYA decreased with the prolongation of the storage life (the average content of HSYA were 2.46%,1.58%,and 1.51% after storage 0,1 and 2 years, respectively),and the color of the drug became darker (a* value decreased ). Results of correlation analysis showed that the content of HSYA was positively associated with color value L*,a*(r=0.430,0.781,P<0.05 or P<0.01);the content of HSAY was negatively associated with storage life (r=-0.777,P<0.01). There was no correlation between the remaining variables (P>0.05). CONCLUSIONS :The longer the storage life ,the darker the color and the lower the content of HSYA ,so it is not suitable for over year and multiyear preservation.
