1.Correlation research between single nucleotide polymorphism of methylenetetrahydrofolate reductase and Alzheimer′s disease in South China Han people

Objective To investigate the relationship between single nucleotide polymorphism (677C→T) of methylenetetrahydrofolate reductase and Alzheimer′s disease(AD) in south China Han people. Methods By applying polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP), MTHFR 677C→T mutation was detected in 45 AD patients and 48 healthy controls. Results The frequency of MTHFR 677C→T mutation of patients showed no significant difference to that of healthy controls (P > 0.05). There is no statistic significance between AD group and controls in C, T gene frequency(P>0.05). But T gene frequency is higher in AD group than in control group. Conclusion MTHFR C 677T is not the pathogenic factor for AD, but could have some effects on AD.

The rates of chronic obstructive pulmonary disease ( COPD) and lung cancer morbidity in Xuanwei city of Yunnan province are among the highest nationwide , and COPD is an major risk factor for lung cancer .The concur-rence of COPD and lung cancer is a common disease .There are common causative environmental risk factors and pathogenic processes between the two diseases , which provides new ideas for the research , prevention and treatment of both diseases .Additionally the discussion of the relationship between COPD and lung cancer can provide refer -ences for further studies of both diseases .
4.Investigation and analysis on professional identity of nursing students with different educational backgrounds

Objective To investigate and analyze the status quo of professional identity of nursing students with different educational backgrounds.Methods With stratified-cluster random sampling,635 students were surveyed using the nursing students' professional identity questionnaire.Results The total score of professional identity of junior college nursing students was significantly higher than that of undergraduate students.Junior college nursing students had a higher score in professional knowledge dimension than undergraduate and graduate students,while the score of nursing undergraduate students was higher than that of graduate students.Junior college nursing students had a higher score in professional will dimension than undergraduate and graduate students.Nursing graduate students had a higher score in professional expectation dimension than junior college and undergraduate students,while the score of undergraduate students was higher than that of junior college students.Whatever the educational background,professional skill and professional expectation were the top two dimensions,while professional knowledge and professional emotion were the last two dimensions.The difference was statistically significant between the answers of nursing students with different educational backgrounds to the following questions,such as which is the most probable factor affecting your learning enthusiasm,who has the greatest impact on your professional thought about nursing and which nursing field are you most willing to go in for.There was significant difference in professional identity score between different grades of junior college and undergraduate nursing students,while there is no significant grade difference in graduate students.Conclusions There were some differences of professional identity of nursing students with different educational backgrounds.Nursing educators should carry out the professional education according to the student's educational backgrounds.
5."Study on ""Different Diseases of the Same Syndrome"" Damp-heat Syndrome based on Metabonomics"

This study was aimed to reveal the material basis on different diseases of the same syndrome damp-heat syndrome from the level of metabonomics.The typical damp-heat syndrome patients diagnosed as chronic viral hepatitis B,non-alcoholic fatty liver disease,or chronic glomerulonephritis were included,with 30 cases in each disease.There were 30 healthy volunteers in the control group.The serum samples were detected by UPLC-QTOFMS and GC-TOFMS.And then,the results were analyzed by variance analysis in order to find out the generality and specificity of metabolic material in three different diseases with damp-heat syndrome.The results showed that through comparisons of different diseases with damp-heat syndrome,as well as the healthy group as control,it was revealed that inosine,uridine,aspartic acid,oleic acid glyceride and lactate were the same substances of three diseases of damp-heat syndrome.It was concluded that based on metabonomics,as for three different diseases with damp-heat syndrome,there were different substances among different diseases,but common substances related to damp-heat syndrome.Thus,it provided objective evidences for the theory of different diseases of the same syndrome in traditional Chinese medicine (TCM) from the level of metabonomics.
6.Comprehensive rehabilitation is superior to surgical intervention for many patients with recurrent lumbar disc herniation

Objective To compare the effectiveness of comprehensive rehabilitation therapy with that of surgical intervention for patients with recurrent lumbar disc herniation (LDH).Methods A total of 124 LDH patients were recruited antd randomly divided into an experimental group and a control group,each of 62.The experimental group was given comprehensive rehabilitation consisting of medicine,physical therapy,and spinal stabilization exercises,while the control group received lumbar spine surgery.The patients' back and leg pain scores and their Oswestry Disability Index (ODI) scores were compared at admission and discharge,as well as 3 and 12 months after discharge.Anxiety and depression were also assessed at admission and 3 months after discharge.Results At discharge and 3 and 12 months later the average back and leg pain scores and ODI ratings were significantly lower than that at admisshin for both groups.There were no significant inter-group differences.In terms of anxiety and depression,however,these had decreased signifieantly in both groups,but the experimental group's average rating was significantly better at diseharge.Conclusions Comprehensive rehabilitation is superior to surgical intervention in relieving the depression of patients with recurrent LDH.Comprehensive rehabilitation should be considered as the first ehoiee for patients with rccurrent LDH,and only supplemented by surgical intervention if it is not effective.
7.Preparation of standard molecular weight fragment mixture based on molecular cloning

Objective To prepare the standard molecular weight fragment mixtures. Methods Primers were designed to prepare clones which contained different sizes of standard molecular weight fragments. The template used for amplification of insert fragments was the pMD18-T vector. Bacteria culture and plasmid extraction were used to obtain abundant target fragment. Unlabeled DNA fragments were prepared by double digestion of the recombinant plasmids, and the fluorescent adaptor was prepared by annealing with two partial reverse complimentary DNA fragments. The unlabeled fragments and fluorescent adaptor were connected by DNA ligation reaction assisted with T4 DNA ligase. In this way, different sizes of standard molecular weight fragments were prepared. Standard molecular weight fragment mixture was finally prepared by mixing all the fragments together before purification. Results Ten standard molecular weight fragments of different sizes were prepared. The sizes of each fragment are 80bp, 124bp, 194bp, 224bp, 254bp, 304bp, 349bp, 399bp, 424bp and 454bp. The internal standard could accurately determine the size of PCR products amplified with the DNATyper15 kit. Conclusion Using this method, the standard molecular weight fragment mixture which meet the requirements of research and laboratory use was prepared, perfectly providing a new method for preparation of the DNA molecular weight standards. The peaks and the size of the prepared DNA internal lane standard are correct, which can be used to calculate the DNA fragments size in capillary electrophoresis.
8.Effect of Early Rehabilitation on Neurological Function and Cerebral Blood Flow of Patients with Acute Cerebral Infarction

Objective To investigate the effect of early rehabilitation on neurological function and cerebral blood flow of patients with acute cerebral infarction.Methods100 cases of acute cerebral infarction were randomly divided into the rehabilitation group(53 cases) and control group(47 cases).Both groups received the routine treatment;besides,patients in the rehabilitation group were treated with early rehabilitation training.The treatment efficacy was assessed by neurological function deficit evaluation,and the cerebral blood flow was measured by transcranial Doppler(TCD) before and 30 days after treatment.ResultsThe scores of neurological function deficit evaluation of all patients in each group decreased after treatment,but the scores of the rehabilitation group were obviously lower than that of the control group(P<0.01);and the results of TCD of the rehabilitation group were also superior to that of the control group(P<0.01).ConclusionEarly rehabilitation training may obviously improve neurological function and cerebral blood flow of patients with acute cerebral infarction.
9.Expression and correlation of Fascin-1 and EGFR in hormone receptor-positive breast cancer

Objective To explore the expression and correlation of Fascin-1 and epidermal growth factor receptor (EGFR) in hormone receptor-positive breast cancer.Methods The immunohistochemical technique,EnVision method,was used to evaluate the expression of Fascin-1 and EGFR in 294 cases of hormone receptor-positive breast cancer,which contains 290 cases of estrogen receptor (ER) positive and 244 cases of progestrone receptor (PR) positive.According to ER,PR,Epidermal growth factor receptor 2 (HER2),and Ki-67 status,all cases of hormone receptor-positive breast cancer were categorized into 2 subtypes:160 cases of luminal A and 134 cases of luminal B.Results Fascin-1 and EGFR protein positive rates in hormone receptor-positive breast cancer was 13.9% (41/294) and 30.6% (90/294),respectively.Fascin-1 positive rate was significantly higher in EGFR positive cases (30.0%,27/90) than in EGFR negative cases (6.9%,14/204) (x2 =27.857,P =0.000).In the ER positive and PR positive cases,Fascin-1 positive rates were both significantly higher in EGFR positive cases than in EGFR negative cases (x2 =29.23,P =0.000;x2 =27.596,P =0.000,respectively).In the Luminal A and Luminal B subtype,Fascin-1 positive rates were also both significantly higher in EGFR positive cases than in EGFR negative cases (x2 =23.247,P=0.000;x2 =5.325,P=0.021,respectively).Conclusions EGFR signal pathway may positive regulate Fascin-1 expression in hormone receptor-positive breast cancer.
10.Mathematical cognitive function in children with sleep disordered breathing based on event-related potential

Objective:To study the characteristics of mathematical cognitive function in children with sleep-disordered breathing(SDB) by event-related potential (ERP).Methods:From October 2020 to October 2022, totally 22 cases of SDB children and 22 cases of normal children aged 8-11 were selected.All subjects performed mathematic tasks including calculating and deciding.The EEG changes and behavioral data of children with SDB and normal children were recorded.The latency and amplitude of N1, P2, N2, P3 in leads Fz were measured and compared by Matlab. SPSS 23.0 software was used for statistical analysis, and t test or Mann Whitney U test were used for two independent sample data. Results:(1)Behavior test: the interaction effect between group and type, the group main effect, and the type main effect in accuracy between SDB group and normal group were not significant ( F=0.470, 3.590, 0.003, all P>0.05). The group main effect and interaction effect between group and type in reaction time between SDB group and normal group were not significant ( F=0.465, 1.991, both P>0.05), while the type main effect was significant ( F=18.010, P<0.05). (2)ERP test: the N2, P3 latencies for addition in children with SDB were longer than those in normal group(N2: (371.38±34.23)ms vs (348.12±26.34)ms; P3: (610.72±64.78)ms vs (529.05±30.25)ms)( t=2.526, 5.358, both P<0.05). There was no significant difference between SDB group and normal group in ERP latency and amplitude for subtraction(both P>0.05). The N2, P3 latencies for multiplication in children with SDB were longer than those in normal group(N2: (439.20±24.28)ms vs (351.14±25.26)ms; P3: (531.71±35.42)ms vs (415.23±19.01)ms)( t=11.792, 13.590, both P<0.05). The P3 amplitudes in children with SDB was higher than that in normal group(P3: (3.16±4.78)μV vs (0.38±3.27)μV)( t=2.248, P<0.05). The P3 latency for correct judgment in children with SDB was longer than that in normal group(P3: (420.38±34.79)ms vs (398.54±33.71)ms)( t=2.115, P<0.05). The P3 latency for wrong judgment in children with SDB was longer than that in normal group(P3: (475.25±51.11)ms vs (414.88±27.53)ms)( t=4.878, P<0.05). Conclusion:The latency of N2 and P3 in ERP of SDB children is prolonged, and P3 latency is more sensitive than N2, indicating that SDB children have impairment of mathematical cognitive function.The latency changes of N2 and P3 occurs earlier than the behavioral changes (reaction time and accuracy), which can be used as one of the electrophysiological indicators for early assessment of mathematical cognitive impairment in SDB children.
