1.Low-frequency transcranial magnetic stimulation and the expression of KCa1.1, NaV1.6, NMDAR1 and GAD65 proteins in the CA3 region of the hippocampus

Objective To investigate whether repeated low-frequency transcranial magnetic stimulation (rTMS) can affect the expression of Kca1.1,Nav1.6,NMDAR1 and/or GAD65 protein in the pyramid layer of the CA3 region of the hippocampus.Methods Fifty rats were randomly divided into an experimental group and a sham group.The former was administered low-frequency rTMS on 14 consecutive days ; the latter were given sham stimulation for 14 days.After the protocols were completed,each group was sub-divided into 6 h,24 h,1 week,3 week and 6 week sub-groups.The rats of each sub-group were sacrificed at the corresponding time points and the expression of KCa1.1,Nav1.6,NMDAR1 and GAD65 in the CA3 region of the hippocampus was examined using immunohistochemical techniques.Results Compared with the sham group,KCa1.1-positive neuron densities increased significantly in the period 6 h to 3 weeks after rTMS.GAD65-positive neuron densities were also elevated significantly from 6 h to 3 weeks.NaV1.6-and NMDAR1-positive neuron densities decreased transiently at 6 h after the completion of the rTMS protocol.Conclusions These results show that low-frequency rTMS can upregulate the expression of KCa1.1 and GAD65,and that the effect lasts for at least 3 weeks.It transiently downregulates the expression of NaV 1.6 and NMDAR1 in the hippocampal CA3 region,at least in rats.These changes may be one of its anti-epileptic mechanisms.
2.Investigation and Rationality Evaluation of Proton Pump Inhibitors Use in Our Hospital during Perioperative Period

OBJECTIVE:To understand the rationality of proton pump inhibitors (PPI) in our hospital during perioperative period,in order to provide reference for rational use of drugs in clinical practice. METHODS:Through randomly collecting discharge cases of surgical system of May and Dec. in 2015 from Hospital Information System,the use of PPI during perioperative period was investigated retrospectively,and the rationality of drug use was evaluated. RESULTS:Among 522 cases,418 cases were given PPI for preventing stress ulcer during perioperative period,with utilization rate of 80.1%. Among them,the top three were neurosurgery,orthopedics and general surgery (100%,92.6% and 92.3% respectively). Pantoprazole and lansoprazole (72.5%,14.8%,respectively) were the top two categories of the use of PPI. The irrational utilization rate of PPI was 70.8%during perioperative period. In the top three,the number of drugs was exceeded,the drug was not used,course of treatment was too long (73.7%,64.1% and 62.4% respectively). CONCLUSIONS:The utilization rate of PPI prevention is higher in the department in our hospital during perioperative period,and there is unreasonable situation in some preventive drugs,and the measures need to be implemented and strengthened.
3.Effects of Aerobic Exercise on Plasma Metabolites in Prediabetes Subjects

Objective To examine the effect of aerobic exercise training on plasma metabolites in pre-diabetes mellitus(PDM)subjects.Methods Twenty-five PDM subjects were selected,and each 10 age-and sex-matched with normoglycemia and new-onset T2DM subjects were enrolled.PDM subjects underwent aerobic exercise(n=13)or health education (n=12) for 12 weeks.The aerobic exercise was con ducted 3 times/week,50 min/time at 40%-60% of oxygen uptake reserve.Liquid chromatography-mass spectrometry(LC-MS)was used to analyze the spectrum of plasma metabolites.Principal component analysis was employed to study the differences in plasma metabolite profiles between subjects with different glycemic level and between PDM subjects before and after aerobic exercise.Results After training,the body fat percentage,2-hour plasma glucose,and low density lipoprotein cholesterol of PDM subjects reduced significantly(by 4.6%,16.22% and 9.27% on average).Comparison of plasma metabolizers in PDM subjects before and after exercise revealed 31 differential metabolites,with 25 metabolites such as phosphatidylcholine,lysophosphatidylcholines,sphingomyelin and betaine increasing,and 6 metabolites,such as oleic acid and cytosine decreasing.Conclusion Aerobic exercise has a marked effect on the plasma metabolites in PDM subjects.Phosphatidylcholine,lysophosphatidylcholines,sphingomyelin and oleic acid may be the potential biomarkers of aerobic exercise improving the glycolipid metabolism in PDM subjects.These findings can help us better understand the mechanism of aerobic exercise preventing T2DM diabetes.
