Objective To study the mitochondrial cytochrome C oxidase 1(CO1) gene of Oncomelania snails from Miao River area in Hubei Province. Methods Oncomelania snails were collected from Miao River area, including upstream and downstream. Genomic DNA was extracted from the tissue of the snail. PCR was used to amplify a fragment of the CO1 gene. Sequences of the CO1 fragment were determined directly from the purified PCR products by an automated sequencer. Sequences for each individual were assembled and edited using ESEE 3 0 s. A distance matrix was computed using program DNADIST of PHYLIP(3 57). Unrooted maximum likelihood trees were calculated from program FITCH.Results The amplified CO1 gene of the snail was a fragment of 638 bp in length. Sequence analysis showed that the accumulated variable sites were significant different between upstream and downstream populations, being 29 and 46, respectively. From the number of variable sites in the gene, snails in this area were roughly separated into two groups. Each of them was a mixture of both upstream and downstream snails.Same haplotypes were confirmed to be present among the collected sites along the river. From the distance matrix of sequence divergence, the population upstream vs downstream differed by 0 0221?0 0105.Conclusion There were more variation in downstream population than that in upstream.Gene flow was identified in these populations. The phylogenetic trees suggest the existence of two groups, but all of them belong to O h hupensis .

BACKGROUND:Transplantation of autologous stem cells for treatment of liver cirrhosis has been widely reported. But up to now, there exist some concerns for clinical physicians, including relationship between stem cells and post-transplantation prognosis/turnover of liver cirrhosis, directed differentiation of stem cells in the impaired liver, and malignant phenotype.OBJECTIVE:To dynamically monitor the serum levels of alpha-fetoprotein(AFP)and AFP variants(AFP-L3)in decompensated cirrhosis patients following intrahepatic transplantation of peripheral blood stem calls via portal vein and evaluate the safety of this treatment method.METHODS:A total of 44 decompensated cirrhosis patients who underwent intrahepatic transplantation of peripheral blood stem cells via portal vein in the 309 Hospital of Chinese PLA in April 2007 were included in this study.Prior to and after surgery, serum levels of AFP and AFP-L3 were detected by chemiluminescence.Through the use of a positive criterion for liver cirrhosis,i.e.,the proportion of AFP-L3 in AFP[AFP-L3(%)]≥10%,and the relationship between decompensated cirrhosis treatment using stem cells transplantation and the malignant phenotype of liver cancer were analyzed.RESULTS AND CONCLUSION:At 2 months after surgery, serum level of AFP showed a transient increase. There was no significant difference in AFP-L3(%)between prior to and after surgery(P>0.05).No significant difference in AFP-L3-positve rate,as well as AFP-L3(%),existed among patients with different serum level of AFP.These findings indicate that clinical symptoms and liver function of decompensated cirrhosis patients recovered to some extent after transplantation of peripheral blood stem cells via portal vein. Results regarding serum level of AFP-L3,a serological marker of liver cancer, did not demonstrate the appearance of malignant biological phenotype.

ObjectiveTo investigate the effects of psychological intervention on patients with acute coronary syndrome (ACS) accompanied by psychogenic disturbance.Methods112 patients with ACS were evaluated with depression status inventory (DSI) and symptom checklist-90 (SCL-90). The cases accompanied with depression were randomly divided into the psychological intervention group (group A) and drug group (group B). Patients of group A were treated with psychological intervention besides routine therapy. The curative effects of the two groups were compared.Results79 cases (70.5%) had depression. Their scores of somatization, depression, anxiety, fear, paranoid ideation, and total score and positive symptom distress level of SCL-90 were significantly different compared with the normal group. After psychological intervention, to patients of the group A, the DSI score fell and symptoms improved significantly (P<0.01), the in-patient duration shorten (P<0.05).ConclusionPsychogenic factors is not neglectable in the process and prognosis of ACS. Effective psychological intervention can promote the patient's healing.

OBJECTIVETo explore the role of P38 signaling pathway in neonatal rat astrocyte swelling and the expression of aquaporin-4 (AQP4) after oxygen-glucose deprivation (OGD) and recovery (OGD/R).
METHODSPrimarily cultured neonatal rat astrocytes were subject to OGD for 5 h followed by oxygen-glucose recovery in the presence or absence of the P38 inhibitor SB203580 (10 µmol/L). The astrocytes were investigated at 0.5, 2, 8 and 24 h after oxygen-glucose recovery for morphological changes and cell injuries using lactate dehydrogenase (LDH) assay. The expressions of P38, P-P38, and AQP4 mRNAs and proteins in the astrocytes were detected using RT-PCR and Western blotting.
RESULTSOGD/R caused significantly enhanced expression of P-P38 protein, and this effect was blocked by SB203580. AQP4 mRNA and protein expression declined transiently at 0.5 h after OGD and increased gradually to reach the peak level at 8 h (P<0.05). Application of the SB203580 significantly lowered OGD-induced AQP4 mRNA and protein up-regulation (P<0.05). Astrocyte swelling occurred after OGD/R but was obviously lessened by SB203580. LDH release increased markedly after OGD/R, and was attenuated by treatment with SB203580 (P<0.01).
CONCLUSIONP38 signaling pathway participates in astrocyte swelling after OGD/R, and blocking this pathway can attenuate AQP4 up-regulation and ameliorate the cell swelling.
