1.Research progress of Nrf2/ARE pathway regulating mechanism

Nuclear factor E2 related factor Nrf2 is a nuclear transcription factors involved in a variety of protein expression. As a center of oxidative stress regulation, it combines with antioxidant components (antioxidant responsive element, ARE) and activates downstream multiple anti-oxidation, anti-inflammatory and detoxifying enzyme protein expression. This signaling pathway is involved in the development of inflammation, tumor and other pathological process. This review describes the basic structure, biological effects and signaling pathways of Nrf2, summarizes the latest progress about mechanisms of factors, which are involved in the positive and negative regulations of signal pathway, providing a new target for anti-inflammatory, antioxidant, and antitumor biochemical treatment. Based on these, the paper also looks forward to applicating bioinformatics technology and providing better prospects for the development of target intervention.
2.Advances on the relationship between thrombin-activatable fibrinolysis inhibitor (TAFI) and recurrent pregnancy loss

Recurrent pregnancy loss (RPL) is a common complication of pregnancy, which affects 2%fertile women. A recent research has found that high level of thrombin-activatable fibrinolysis inhibitor (TAFI) can reduce the occurrence risk of early RPL. TAFI is one kind of carboxypeptidase, which can be activated as TAFIa. TAFIa can make the fibrinolysin lose its working site, which can interact with the fibrin to play a role in the regulation of fibrinolysis and the inhibition of throm?bus formation. The damage of fibrinolytic system is one of the risk factors for the occurrence of RPL in pregnant women, which has become one of the hotspots in the medical profession. In this paper, recent literature on TAFI and its relationship with recurrent pregnancy loss has been reviewed, hoping for new ways and clues in clinical treatment and prevention of RPL.
3. In vivo cell tracking

Owning to the progression of silence and technology, increasing techniques for in vivo cell tracking have been applied in medical research; they include radionuclide imaging, magnetic resonance imaging, optical imaging, etc. These imaging methods have their own characteristics in specificity, sensitivity and manipulation mode. Researchers should choose the appropriate method while taking the following into consideration requirement of the image, object of the image, the aim of the image, and the characteristics of each imaging method. In this paper we summarize the techniques of cell tracking in vivo.
4.Correlation between polymorphism of CYP2D6 gene with effect of tamoxifen in patients with breast cancer

Objective To investigate the correlation of CYP2D6 gene polymorphism and serum tamoxifen (TAM) and its active metabolites (4-hydroxy tamoxifen,4-OH-TAM) in terms of prognosis of breast cancer patients.Methods The oral mucosas and serum were obtained from 200 breast cancer patients receiving oral TAM from Jan 2008 to Oct 2010.Real-time RT-PCR was used to determine CYP2D6* 10 gene polymorphism.The relationship between CYP2D6* 10 gene polymorphism and clinicopathological features and prognosis was assessed by Chi-square test and Cox proportional model.Serum TAM and 4-OH-TAM concentration were determined by liquid chromatography-trap mass spectrometry (LC-MS).Results Of 200 breast cancer patients,CYP2D6 * 10/* 10 homozygotes were found in 94 cases (47%),CYP2D6 wt/wt wild type in 48 cases (24%) and CYP2D6 wt/* 10 heterozygote in 58 cases (29%).The concentration of serum 4-OH-TAM in patients with CYP2D6 wt/wt wild type and wt/* 10 heterozygote was significantly higher than that in patients with CYP2D6 * 10/ * 10 homozygote (F =4.31,P =0.01).CYP2D6 gene polymorphism was not related to clinicopathological features (P > 0.05).Logrank test showed that patient's disease-free survival with mutational CYP2D6 (mean,47.2 months) was significantly shorter than that in patients without mutational CYP2D6 (mean,51.2 months) (P =0.018).Cox proportional model showed that CYP2D6 genotype was related to disease-free survival (HR =2.755,95%CI:1.230 ~6.173,P =0.014).Conclusions CYP2D6 *10/* 10 genotype is related to curative effect of TAM on breast cancer patients,detection of CYP2D6 * 10/* 10 genotype helps improve the choice of TAM therapy.
5.Results of monitoring indicators of iodine deficiency disorders in Guangming District of Shenzhen City in 2010-2012

Objective To know the current situation of prevention and control of iodine deficiency disorders in Guangming District of Shenzhen through analyzing related monitoring indicators from 2010-2012.Methods According to the National Iodized Salt Monitoring Program,in Guangming District of Shenzhen,2 Street Offices were chosen,then 4 Neighborhood Committees were chosen in each Street Office randomly,15 household salt samples were selected randomly in each Neighborhood Committee; 5 primary schools were chosen in this district,and 20 urine samples were selected from 8-10 years old children in each school in 2011,one source water and one tap water sample were collected of all the water supply companies in this district in 2012.Salt iodine was determined by direct titration method; urinary iodine was determined by As3--Ce4+ catalytic spectrophotometry method; water iodine was determined by sulfate Ce catalytic spectrophotometry of drinking water standard test method.Results Salt iodine were 27.13 and 21.23 mg/kg in 2010 and 2012,respectively.The rates of qualified iodized salt in 2010 and 2012 were 93.33% (112/120) and 90.00% (108/120),respectively.The median concentration of urinary iodine of 8-10 years old children in 2011 was 208.19 μg/L.The median concentration of water iodine in 2012 was 31.60 μg/L.Conclusions The district isn't an iodine excess.The rates of qualified iodized salt in resent years are in line with national standards.There is no iodine deficiency in children and additional supplementation of iodine is not necessary.But relevant monitoring still needs to be improved.

Sinus histiocytosis with massive lymphadenopathy( SHML) is also called Rosai-Dorfman disease.It is a kind of benign lymphoid tissue proliferative diseases with unknown etiology.SHML appeared mostly in children and adolescent.It has diverse clinical manifestations accompanied with multiple organ inju-ry,and no clear laboratory indicators could support the disease,being a rare disease in pediatrics,easyot miss diagnosis.Thsi article reviewde the latest progress on diagnostis and treatment of SHML,to improve teh un-derstanding of the disease.
7.Reactivation of human herpesvirus 8 by hepatocyte growth factor/scatter factor similar to that produced by HIV-1-infected T cells via ORF50 promoter of HHV-8

Objective To investigate a potential role for ORF50 promoter of human herpesvirus 8(HHV 8)in reactivation of HHV 8 in primary effusion lymphoma(PEL)BC 3 cells by HIV 1 related inflammatory cytokine,hepatocyte growth factor/scatter factor(HGF/SF). Methods The persistent stimulation of BC 3 was performed by recombinant HGF/SF and treated BC 3 cells were collected at the 3 rd and 7 th day after persistent stimulation respectively. Northern blot, quantitative PCR(real time PCR), and electron microscopy(EM)were carried out to detect the expression of mRNA of minor capsid protein ORF26 and the presence of viral particles of HHV 8 from treated BC 3 cells. Co transfection of BC 3 and human umbilical veil endothelial cells (HUVECs) with HHV 8 ORF50 promoter driven luciferase construct built previously by us and recombinant expression plasmid named pEV containing HIV 1 Tat coding gene was performed and stimulation of co transfected cells with recombinant HGF/SF and/or HIV 1 recombinant gp120 was conducted at 24 hours after transfection to induce luciferase gene expression. Then relative luciferase activity unit(RLU) was calculated at 24 hours after stimulation. In the meantime, stimulation of co transfected cells with 12 O tetradecanoylphorbol 13 acetate(TPA) was performed to be used as positive control. Results It was found that HGF/SF induced an 4.1 folds increase in mRNA expression of ORF26 when it was added individually to BC 3 cells at the 7 th day after persistent stimulation. Viral particles of HHV 8 were readily identified in BC 3 cells stimulated with HGF/SF at the 7 th day with EM analysis. Recombinant HGF/SF could upregulate promoter activity of HHV 8 ORF50 promoter in BC 3 and HUVECs cells and however, HIV 1 Tat and gp120 failed to act synergistically with HGF/SF to enhance ORF50 promoter activity in HUVECs. Conclusions The results show that HIV 1 related inflammatory cytokine HGF/SF is responsible for the induction of HHV 8 lytic cycle replication and the induction may be, at least partly mediated by ORF50 promoter of HHV 8.
8.Observation of the clinical effect of sodium hyaluronate combined with glucocorticoid on the treatment of periarthritis of shoulder

Objective To investigate the clinical effect of sodium hyaluronate combined with glucocorticoid on the treatment of periarthritis of shoulder.Methods106 patients in department of orthopedics, the second hospital of Ningbo with periarthritis of shoulder were randomly divided into 2 groups (n=53).The control group were treated with anti-inflammatory and analgesic treatment, at this basis, the study group were treated with sodium hyaluronate combined with Glucocorticoid.5 weeks for a course of treatment.Levels of serum inflammation, laboratory-related indicators were compared.ResultsCompared with before treatment, shoulder joint activity in the two groups increased, VAS score decreased, the scores of strength of shoulder joint, range of motion and daily activities increased, levels of serum IL-6, IL-10, TNF-α decreased(P<0.05).Compared with the control group, scores of activity in the study group were higher, VAS scores were lower, scores of strength of shoulder joint, range of motion and daily activities were higher, levels of serum IL-6, IL-10, TNF-α were lower(P<0.05).The effective rate in the control group(73.59%) was lower than the study group(90.57%), but there was no statistical difference.ConclusionThe effect is accurate which sodium hyaluronate combined with glucocorticoid was used in the treatment of periarthritis of shoulder, and it can reduce the inflammation index and improve the joint activity.
10.Clinical research on micro-movement sensitive mattress sleep monitoring system in children with obstructive sleep apnea hypopnea syndrome.

OBJECTIVETo evaluate the diagnostic value of micro-movement sensitive mattress sleep monitoring system (MSMSMS) in children with obstructive sleep apnea hypopnea syndrome.
METHODSSixty-five children with the complaint of sleep disorder were collected and detected by polysomnography and micro-sensitive mattress sleep monitoring system overnight at least seven hours. The date from two instruments were analyzed by computer automatically and then modified by two professional staff double-blind separately. The data about the diagnosis of sleep breath disorder and other sleep physiology information were compared between the two groups.
RESULTSSixty-one cases finished the study finally. The mean age of these children was (7.0 ± 2.7) and the youngest was 3 years old, maximum was 13 years old, male children 46 cases, female patients 15 cases, body mass index (BMI) median [25 quantile; 75 quantile] 16.00 [14.80;17.5]kg/m2. Mean PSG measured apnea hypopnea index (AHI) was 4.50 [2.30;10.15] times/h, and mean MSMSMS AHI was 3.63 [2.56; 6.43] times/h. There was a significant correlation between PSG AHI and AHI by MSMSMS (r = 0.935, P < 0.01). A Bland-Altamplot of PSG AHI and MSMSMS AHI was also used to assess the accuracy of MSMSMS. 95. 1% of the data was fallen in the 95% consistency areas. For AHI--5 times/h and nighttime minimum oxygen < 91% or obstructive apnea index( OAT) > 1 time/h and nighttime minimum oxygen saturation < 91 % as threshold value, the MSMSMS diagnosing sensitivity and specificity were 82.9% and 92.3%. The misdiagnosis rate and missed diagnosis rate were 7.7% and 17.1%.
CONCLUSIONSThe MSMSMS offer a simple and comfortable method to monitor children's sleep. It improves the compliance in the process of sleep monitoring. Besides, as a diagnostic tool in the diagnosis of OSAHS on children has a high credibility in AHI.
