1.Studies on The Interactions Between NIRF and P53

HEK293 or HeLa cells were transfected by NIRF and, or P53, whole cell extracts andimmunoprecipitates were subjected to SDS-PAGE followed by Western blotting. GST pull-down was carried outto identify the interactions between NIRF and P53. In vitro ubiquitination reaction was carried out to identify P53ubquitinate by NIRF. The results suggested that NIRF could interact with P53 in vivo and in vitro. The results alsoshowed that NIRF could ubiquitinate P53 in vivo and in vitro. The results indicated that NIRF would be a newnegative regulator of P53.
2.Study on present situation and strategy study on graduate students' scientific ethics and academic atmosphere of 10 colleges and universities in Chongqing

Objective To understand the academic atmosphere current situation of graduate students and put forward relevant countermeasures to protect the academic integrity of graduate students.Methods The graduate student'scientific ethics and academic atmosphere construction situation of graduate students in 10 colleges and universities in Chongqing were investigated by questionnaire survey and analyzed.Results Through 1555 questionnaires,it was found that the average investment of time on scientific research in gruduate students was insufficient,the motivation of learning was complicated,and the situation of preventing academic misconduct was serious.Conclusion This study suggests that colleges and universities should establish an academic integrity security system for graduate students,including establishment of academic integrity system and specializing academic integrity supervision institutions,and strengthing academic integrity education.
3.Problems and countermeasures of clinical medical postgraduate of academic degree with the synergy of medical and education

Since the implementation of synergy of medical education, the education of clinical medi-cal postgraduate of academic degree inevitably has faced problems such as the decrease quality of the source of students, unclear training objectives and confused training model, and bad job prospects. Can these problems be solved effectively or not, which is directly related to the coordinated development of clinical medicine professional degree and academic degree. This study analyzes the problems faced by the graduate education of clinical medicine after the synergy of medical education and puts forward the corresponding countermeasures:firstly, to attract excellent clinical medical undergraduates and encourage medical students of related subjects or clinical doctors having working experience to apply for academic degree;secondly, the university, supervisors and graduates need to clear the training target and ensure the quality of graduate education; thirdly, employment guidance should be carried out throughout all the study stage of graduates, and the school should guide the graduates to form correct opinion of employment and to plan their career reasonably.
4.Research and approach on the quality management of clinical medical professional degree thesis

5.Analysis of mental health status and influencing factors of postgraduates in medical schools

Objective:To investigate the mental health status of postgraduates in a medical university in southwest China and analyze its influencing factors.Methods:A total of 622 clinical medical postgraduates from a medical university were investigated by basic information questionnaire and symptom Checklist-90 (SCL-90). Logistic regression was performed for data analysis. Excel 2010 was used for data entry, and SPSS 22.0 was used for statistical analysis.Results:The scores of each factor were as follows: somatization=1.24±0.44, compulsion=1.62±0.64, interpersonal sensitivity=1.4±0.54, depression=1.45± 0.64, anxiety=1.37±0.56, hostility=1.31±0.50, fear=1.18±0.42, paranoia=1.27±0.51, psychosis=1.25± 0.46, others=1.36±0.53. Besides, 169 (27.2%) people had positive mental health symptoms, and the top three symptoms were compulsion, depression and interpersonal sensitivity. The results of multivariate logistic regression analysis showed that grade, professional satisfaction, postgraduate study motivation and family education style were the influencing factors of mental health status of postgraduates in medical colleges and universities.Conclusion:The mental health status of postgraduates in medical colleges is worse than before. According to the influencing factors, targeted measures should be taken to improve the mental health level of postgraduates.
6.Practice and exploration of cultivating scientific research ability of postgraduates with professional master of clinical medicine

This investigation was conducted on clinical professional graduate students of Chongqing Medical University to analyze the problems and influencing factors in the process of scientific research ability training. We propose three measures to improve the model of training professional clinical graduate students and provide references for the cultivation of their scientific research ability. The measures are "clear orientation of clinical scientific research ability training", "constant pursuit of high-level original scientific research results" and "adherent implementation to strengthen training effect".

The cultivation of graduate students' scientific research ability is an important task of colleges and universities. The evaluation standards of cultivation achievements play an important guiding role. In recent years, quantitative academic evaluation based on the "five-only" has caused some negative effects on the cultivation of talents in colleges and universities. A new evaluation system needs to be established. During the transition period between the old and new academic evaluation systems, the evaluation criteria for the cultivation of graduate students' scientific research capabilities will inevitably need to be reformed. This study attempts to reform the existing evaluation standards by investigating the evaluation standards for the cultivation of scientific research capabilities in various colleges and universities. The aim is to improve the evaluation of the achievements in the cultivation of scientific research ability of graduate students and improve the quality of the cultivation of scientific research ability of graduate students.
8.Exploration of internal quality assurance system construction in Chinese-foreign joint education program of clinical medicine

To effectively introduce and utilize global advanced educational resources, the implementation of Chinese-foreign joint education program in medical universities and the establishment of an internal quality assurance system with Chinese characteristics and substantially equivalent to international standards of medical education is crucial for improving the quality of Chinese medical talent cultivation and achieving high-quality development of education opening up to the outside world in the new era. Using the Chinese-Foreign Joint Education Program jointly run by Chongqing Medical University and University of Leicester as an example, this paper proposes a plan for the development of an internal quality assurance system with three core indicators of management mechanism, team building, and student services. This plan provides experience and a reference for domestic universities to carry out cross-border medical education and improve the quality and efficiency of Chinese-foreign joint education programs.

UHRF2 is a ubiquitin-protein ligase E3 that regulates cell cycle, genomic stability and epigenetics. We conducted a co-immunoprecipitation assay and found that TIP60 and HDAC1 interact with UHRF2. We previously demonstrated that UHRF2 regulated H3K9ac and H3K14ac differentially in normal and cancer cells. However, the accurate signal transduction mechanisms were not clear. In this study, we found that TIP60 acted downstream of UHRF2 to regulate H3K9ac and H3K14ac expression. TIP60 is stabilized in normal cells by UHRF2 ubiquitination. However, TIP60 is destabilized in cancer cells. Depletion or inhibition of TIP60 disrupts the regulatory relationship between UHRF2, H3K9ac and H3K14ac. In summary, the findings suggest that UHRF2 mediated the post-translational modification of histones and the initiation and progression of cancer.

10.Advances in molecular function of p62 protein and its role in diseases.

Sequestosome 1 (SQSTM1/p62) is a selective autophagy adaptor protein that plays an important role in the clearance of proteins to be degraded as well as in the maintenance of cellular proteostasis. p62 protein has multiple functional domains, which interact with several downstream proteins to precisely regulate multiple signaling pathways, thereby linking p62 to oxidative defense systems, inflammatory responses and nutrient sensing. Studies have shown that mutation or abnormal expression of p62 is closely related to the occurrence and development of various diseases, including neurodegenerative diseases, tumors, infectious diseases, genetic diseases and chronic diseases. This review summarizes the structural features and molecular functions of p62. Moreover, we systematically introduce its multiple functions in protein homeostasis and regulation of signaling pathways. Furthermore, the complexity and versatility of p62 in the occurrence and development of diseases are summarized, with the aim to provide a reference for understanding the function of p62 protein and facilitating related disease research.
