1. Analysis on the characteristics of new occupational diseases in Changzhou City from 2006 to 2018

Objective:
To explore the characteristics of new occupational diseases in Changzhou City from 2006 to 2018, and provide scientific basis for relevant departments to formulate prevention and control strategies.
Methods:
In July 2019, the data of new occupational diseases in Changzhou City from 2006 to 2018 were collected, and the distribution of new occupational diseases, such as disease types, regions, population, industries, economic types and scale of enterprises were analyzed.
Results:
From 2006 to 2018, a total of 432 new cases of occupational diseases were reported in Changzhou, mainly including occupational pneumoconiosis and other respiratory diseases (175 cases, 40.51%) , occupational chemical poisoning (130 cases, 30.09%) , and occupational otorhinolaryngological and oral diseases (83 cases, 19.21%) . 2.Effect of Trimetazidine on Diabetic Patients with Coronary Heart Diseases: A Meta-Analysis of Randomized, Controlled Trials.

Objective To explore the therapeutic effects of trimetazidine (TMZ) on diabetic patients with coronary heart diseases.Methods We conducted a comprehensive electronic search of PubMed, EMBASE, and Cochrane databases between the inception dates of databases and May 2019 (last search conducted on 30 May 2019) to identify randomized controlled trials. The evaluation method recommended by Cochrane Collaboration for bias risk assessment was employed for quality assessment. Random or fixed models were used to investigate pooled mean differences in left ventricular function, serum glucose metabolism, serum lipid profile, myocardial ischemia episodes and exercise tolerance with effect size indicated by the 95% confidence interval ().Results Additional TMZ treatment contributed to considerable improvement of left ventricular ejection fraction (=4.39, 95%: 3.83, 4.95, <0.00001), left ventricular end diastolic diameter (=-3.17, 95%: -4.90, -1.44, =0.0003) and left ventricular end systolic diameter (=-4.69, 95%: -8.66, -0.72, =0.02). TMZ administration also significantly decreased fasting blood glucose (=-0.43, 95%: -0.70, -0.17, =0.001), glycosylated hemoglobin level (=-0.59, 95%: -0.95, -0.24, =0.001), serum level of total cholesterol (=-20.36, 95%: -39.80, -0.92, =0.04), low-density lipoprotein cholesterol (=-20.12, 95%: -32.95, -7.30, =0.002) and incidence of myocardial ischemia episodes (=-0.84, 95%: -1.50, -0.18, =0.01). However, there were no significant differences in serum triglyceride level, high-density lipoprotein cholesterol, exercise tolerance between the TMZ group and the control group. Conclusion TMZ treatment in diabetic patients with coronary heart disease is effective to improve cardiac function, serum glucose and lipid metabolism and clinical symptoms.

4.Two cases of fatal occupational acute severe toxic encephalopathy

Objective To analyze the onset and clinical treatment of two cases of occupational acute severe toxic encephalopathy deaths and to propose key strategies for prevention and treatment. Methods The basic information, on-site occupational health investigation and clinical data of two cases of occupational acute severe toxic encephalopathy deaths were retrospectively analyzed. Results Both cases involved a history of occupational short-term exposure to a large amount of chemicals, due to not wearing personal protective equipment (PPE) throughout the entire process or not wearing PPE at all. The clinical symptoms of patients were primarily related to neurological damage. Both patients developed severe consciousness disorders, leading to a clear clinical diagnosis of toxic encephalopathy during treatment. Combined with the results of cranial computed tomography, electroencephalogram, head magnetic resonance and other examinations and information from the on-site occupational health survey, both patients were diagnosed as occupational acute severe toxic encephalopathy after excluding similar diseases due to other causes. Multiple organ failure and toxic encephalopathy were the causes of death of these two patients. Conclusion Occupational acute severe toxic encephalopathy is characterized by a rapid onset, quick progression, and high mortality. Proper use of PPE is an effective measure to protect workers' health. Strengthening occupational health supervision in small and micro enterprises is necessary to prevent similar occupational chemical poisoning incidents.
5.Nur77 promotes invasion and migration of gastric cancer cells through the NF-κB/IL-6 pathway.

OBJECTIVE:
To analyze the association of Nur77 with overall survival of gastric cancer patients and investigate the role of Nur77 in invasion and migration of gastric cancer cells.
METHODS:
Oncomine database was used to analyze the expression of Nur77 in gastric cancer and gastric mucosa tissues, and the distribution characteristics of Nur77 protein between gastric cancer and normal tissues were compared using Human Protein Atlas. GEPIA2 was used to analyze the relationship of Nur77 expression and the patients' survival. The expression of Nur77 in gastric cancer cell lines GES-1, AGS and MKN-45 were detected by Western blotting. The regulatory interactions between IL-6 and Nur77 were verified by transfecting the cells with specific Nur-77 siRNA and Nur-77-overexpressing plasmid. The changes in migration ability of the cells following Nur-77 knockdown were assessed with scratch assay. The effect of Nur-77 overexpression or IL-6 knockdown, or their combination, on migration and invasion of the gastric cancer cells were examined using Transwell assay. The effect of Nur77 expression level on NF-κB/IL-6 pathway activation was analyzed using Western blotting.
RESULTS:
Oncomine database showed that gastric cancer tissues expressed a significantly higher level of Nur77 mRNA than normal tissues (P < 0.05). Nur77 expression was detected mostly in the nucleus, and a high Nur77 expression was associated with a poor survival outcome of the patients (P < 0.05). In gastric cancer cells, the high expression of Nur77 participated in the regulation of IL-6. Nur77 silencing significantly lowered the migration ability of the cells (P < 0.05), and IL-6 silencing significantly attenuated the enhanced migration caused by Nur77 overexpression (P < 0.05). Nur77 participates in the activation of NF-κB/IL-6 signaling pathway by regulating the expression of p-p65, p65, p-Stat3 and Stat3.
CONCLUSION
A high Nur77 expression is strongly correlated with a poor prognosis of gastric cancer patients. Nur77 promotes the invasion and migration of gastric cancer cells possibly by regulating the NF-κB/IL-6 signaling pathway.

6.Effectiveness of functional training and diet intervention on functional movement and physical health of obese male college students

Objective:
To reveal the effect of functional training & diet intervention on functional movement screen (FMS) and physical health of obese college male students.
Methods:
Totally 56 obese college male students were divided into experimental group and control group on average and the experimental group took functional training & diet intervention, 2 times per week, with a total of 15 weeks. The control group was only given routine physical exercise without diet intervention. Indexes including seven FMS indicators, seven body composition indicators and five physical fitness indicators were monitored before and after experiment.
Results:
The results showed that except for shoulder flexibility, other FMS indexes (vault, hurdle, straight lunge, trunk stability and trunk rotation stability) of experimental group significantly increased after functional training & diet intervention(t=-2.11, -2.27, -2.22, -2.08, -2.21, P<0.05). The total score of FMS increased significantly from 12.07 to 14.06(t=-4.22, P<0.05). The body composition index including fat content, body water, body fat ratio and BMI decreased to 23.97 kg, 38.83 kg, 26.34% and 27.79 kg/m2(t/χ2=-5.85, -8.30, 9.05, 7.23, P<0.05). Protein increased significantly to 19.00 kg(t=-5.75, P<0.05). The comprehensive score of body composition in the experimental group increased significantly from 64.00 to 66.71(t=-6.27, P<0.05). The scores of experimental group physical fitness indicators including 50 m, sit forward, standing long jump, pull up and 1 000 m increased to 8.24 s, 14.45 cm, 206.61 cm, 3.07 and 278.27 s(t=5.05, -3.96, -7.46, -3.62, 4.40, P<0.05).
Conclusion
The study indicates that 15 weeks of physical function training & dietary intervention could effectively improve the body shape, body composition and motor dysfunction of obese male college students, thus to reduce sports risk and improve physical health.
7.LncRNA FENDRR Affect Proliferation, Migration and Apoptosis of Lung Squamous Cell Carcinoma H226 Cells via ERK/MAPK Signaling Pathway

Objective To investigate the effects of lncRNA FENDRR on the proliferation, migration, invasion and apoptosis of LUSC H226 cells and its molecular mechanism. Methods The expression levels of FENDRR in normal lung epithelial cells BEAS, lung adenocarcinoma A549 and H1299 cells and LUSC H226 cells were detected by qRT-PCR. H226 cells were transfected with FENDRR-siRNA as the experimental group, or with FENDRR-siNC as a negative control group. Cell proliferation was detected by CCK-8 assay. Cell migration and invasion were detected by Transwell assay. Cell apoptosis was detected by flow cytometry. The protein expression levels of MEK, p-MEK, ERK and p-ERK were determined by Western blot. Results FENDRR levels in H226 cells were significantly lower than those in normal lung epithelial cells. Compared with the negative control group, the knockdown of FENDRR could significantly promote the proliferation, migration and invasion of H226 cells, inhibit the cell apoptosis, and increase the protein levels of p-MEK and p-ERK. The addition of ERK inhibitor U0126 rescued the effect of FENDRR knockdown on H226 cells. Conclusions The knockdown of lncRNA FENDRR can promote the proliferation, migration and inhibit the apoptosis of H226 cells through ERK/MAPK pathway.
8.Effect of sanguinarine on biomechanics of rat airway smooth muscle cells.

This study aimed to evaluate the effect of sanguinarine on biomechanical properties of rat airway smooth muscle cells (rASMCs) including stiffness, traction force and cytoskeletal stress fiber organization. To do so, rASMCs cultured were treated with sanguinarine solution at different concentrations (0.005~5 μmol/L) for 12 h, 24 h, 36 h, and 48 h, respectively. Subsequently, the cells were tested for their viability, stiffness, traction force, migration and microfilament distribution by using methylthiazolyldiphenyl-tetrazolium bromide assay, optical magnetic twisting cytometry, Fourier transform traction microscopy, scratch wound healing method, and immunofluorescence microscopy, respectively. The results showed that at concentration below 0.5 μmol/L sanguinarine had no effect on cell viability, but caused dose and time dependent effect on cell biomechanics. Specifically, rASMCs treated with sanguinarine at 0.05 μmol/L and 0.5 μmol/L for 12 and 24 h exhibited significant reduction in stiffness, traction force and migration speed, together with disorganization of the cytoskeletal stress fibers. Considering the essential role of airway smooth muscle cells (ASMCs) biomechanics in the airway hyperresponsiveness (AHR) of asthma, these findings suggest that sanguinarine may ameliorate AHR via alteration of ASMCs biomechanical properties, thus providing a novel approach for asthma drug development.
9. Event-related potential characteristics analysis on vigilant attention development of school-age children

10.Clinical phenotype and novel mutation in one of twins with glutaric acidemia type I.

OBJECTIVE:
To review the clinical features of a male twin affected with glutaric academia type I (GA-I) and analyze the variations of glutaryl-CoA dehydrogenase (GCDH) gene.
METHODS:
Clinical data of the pair of twins and their parents were collected. Genomic DNA was extracted from peripheral blood samples, and variants of GCDH genes were detected by capture sequencing using a customized panel. Variants of the twins and their parents were verified by Sanger sequencing.
RESULTS:
The level of glutaric acyl carnitine (C5DC + C6OH) was 3.26 μmol/L in the male twin. The relative level of glutaric acid in urine was 547.51 by gas chromatography mass spectrometry analysis. Cerebral ultrasonography showed that the patient had subependymal hemorrhage, but no serious clinical manifestation was noted. After treating with special formula milk powder and L-carnitine, the boy showed good growth and development. Two heterozygous variants of the GCDH gene were detected in the patient, among which c.416C>G was suspected to be pathogenic, while c.109_110delCA was unreported. The variants were respectively inherited from his parents. The twin girl only carried the c.416C>G variant.
CONCLUSION
GA-I can be diagnosed by mass spectrometry, urine gas chromatographic mass spectrometry, imaging as well as genetic diagnosis. Early diagnosis and intervention is important.
