Objective To observe the effects of mouse indoleamine 2,3-dioxygenase (IDO) gene on CD4+ T cells by transfecting an eukaryotic expression vector containing mouse IDO gene fused with enhanced green fluorescent protein (pEGFP-N1). Methods The immature dendritic cells (imDCs) derived from mouse bone marrow were cultured in vitro and morphologically identified by transmission electron microscope, scanning electron microscope and flow cytometry. The immature dendritic cells were transfected by DOTAP liposome, and the expression of green fluorescence protein was confirmed with inverted fluorescence microscope. The spleen-derived CD4+ T lymphocytes from mice were isolated and purified in vitro, and then carried to mixed lymphocyte reaction (MLR) with 3H-TdR incorporation. The effect of IDO gene on the proliferation of spleen-derived CD4+ T lymphocytes of the allergized mouse challenged by ovalbumin was assayed by 3H-TdR MLR incorporation, and the apoptosis of spleen-derived CD4+ T lymphocytes was assayed by TUNEL. 9.Effects of Aminophylline on expression of Fas mRNA and apoptosis in eosinophils

AIM To investigate the effects of aminophylline (AM) on expression of Fas mRNA and apoptosis in eosinophils(Eos) of bronchalveolar lavage fluid(BALF) in asthmatic guinea pig, and the mechanism of AM promoting Eos apoptosis. METHODS After guinea pigs were stimulated by ovalbumin(OVA) 48 h, hypodense Eos(HEos) and normodense Eos(NEos) were purified from BALF by gradients of percoll. Purified Eos were incubated with different concentration of AM (10 -6 ~10 -3 mol?L -1 ) in RPMI 1640 medium 24 h, the mRNA of Fas in Eos was measured by hybridization and apoptosis was detected by TdT mediated dUTP nick end labeling. RESULT Eos cultured under the condition of AM administration in vitro, apoptotic Eos increased and the expression of Fas mRNA increased in a dose dependent manner. The expression of Fas mRNA was correlated with Eos apoptosis. CONCLUSION AM promote Eos apoptosis and expression of Fas in dose dependent manner in vitro. Fas might be one of mechanism that AM promoting apoptosis of Eos.
10. Total body γ-irradiation decreases wound breaking strength during wound healing in rats

Objective: To investigate the influence of total body irradiation with different doses of 60Coγ on wound breaking strength during wound healing in rats. Methods: Rats were exposed to 60Coγ radiation at dosages of 4,6 ,and 8 Gy. Within 30 min after irradiation, full thickness skin wounds were made on the shaved back of rats to establish animal models of irradiated-trauma injury plus skin wounds (n=20), and non-irradiated rats with pure incision injury were used as controls (n=10). The rats were sacrificed at day 10 after treatment, and the full thickness skin wounds were harvested. Bio-mechanics method and histopathology examination were used to evaluate the wound breaking strength and histological features after healing. Results: The wound breaking strength of model groups were greatly retarded with the increase of irradiation doses. Statistical results showed that on day 10 the wound breaking strength values were significantly different between 4 Gy group ([114.26±0.29] g) and control group ([117.12±1.86] g, P>0.05), and the wound breaking strength values of 6 Gy group ([91.87±1.96] g) and 8 Gy group ([55.26±2.64] g) were significantly lower than that of the control group (P<0.05). H-E staining showed that the wounds in the irradiation trauma groups had looser and disorder collagenous fiber and less fibroblast proliferation than control group. Conclusion: Radiation injury may result in delayed wound healing, and the wound breaking strength decreases with the increase of irradiation dose in model of irradiation injury plus full thickness skin wounds.
