1.The Value of Transrectal Echography in the Diagnosis of Peri-rectal Lesions

Objective To assess the value of transrectal echography(TRE) in diagnosing peri-rectal lesions. Methods The echography images of 287 cases of peri-rectal lesions were retrospectively analyzed,and contrasted with the results of postoperative pathologic examination and the final clinical diagnosis. Results 83 cases of peri-rectal recess lesions were found by TRE, and the occurate rate of diagnosis was 91%. 123 cases of peri-rectal viscera lesions were found by TRE, and the accurate rate of diagnosis was 94%. Conclusion TRE plays an role in the diagnosis of peri-rectal recess and viscera lesions, and in the dynamic and real-time detection of the obstructive causes of bladder orifice.
2.DETECTION OF CAg IN CSF OF 231 CEREBRAL CYSTICERCOSIS PATIENTS

The results of detection of circulating antigen(CAg) in cerebrospinal fluids(CSF) of 231 cerebral cysticercosis patients with McAb(4F8)-based ELISA were reported and compared with the case histories , clinical manifestations and CT scans. No relationship was found between CAg detected in CSF of these cases and history of taeniasis. However, the positive rate of CAg in cerebral cysticercosis patients with subcutaneous nodules was found significantly higher than those in cases with simple cerebral cysticercosis and in cases with subcutaneous nodules disappeared after anti-Cysticercus therapy before CSF collection. The results of CAg detection in CSF were related with the CT findings as well as the stage of the disease. It is thus indicated that MeAb(4F8)-based ELISA might be useful not only for diagnosis of neurocysticercosis, but also for the evaluation of efficacy of anti-Cysticercus therapy.
3.Effects of rosuvastatin on endothdial function during myocardial ischemia-reperfusion injury in rabbits

Objective To study the change of endothelial function during myocardial ischemia-reperfusion injury in rabbits and the effect of Rasuvastatin. Method Sixteen New Zealand rabbits were randomLy divided into two groups: ischemia/reporfusian injury group (control group) and Resuvastatin group(drug group). The myocar-dial ischemia-reperfusion model was established by occlusion of left anterior descending coronary artery for 40 min-utes evidenced by the elevation of the ST segment≥0.2 my on ECG waveform, and after release of ooclusion, the ST segment of ECG retttmed to 1/2 or more of the normal wavefonn, which was the evidence of successful reperfu-sion. The rabbit serum nitric oxide (NO) and plasma endothelia-1 (ET-1) content were assayed before occlusion, 40 minutes, 60 minutes and 180 minutes after reperfusion. SPSS11.5 software was used for ANOVA(Repested Measurement designs). P<0.05 was considered as statistically significant. Results There were no significant differences in serum NO and plasma ET-1 between two groups bsfore isehemia, but 40 minutes, 60 minutes and 180 minutes after reperfusion, the levels of serum NO in drug group were higher than those in control group [(82.000±13.825), (63.375±17.541), (50.250±18.987)μmol/L vs. (63.125±18.962), (43.500± 16.518), (29.625±14.162) μmol/L, P<0.05], and the levels of plasma ET-1 content in drug group were lower than those in the control group [(282.541±38.928), (315.152±55.263), (377.795±60.427) pg/mL vs. (331.785±35.341),(375.914±5.204),(459.829±70.110) pg/mL, P<0.05]. Conclusions By the means of increasing serum NO and decreasing plasma ET-1, rosuvastatin can improve the endothelial function in rabbits with isehemia-reperfusion injury.
4.Comparison of different analyzing methods for detection of free amino acids in plasma

Objective To evaluate the accuracy of different methods in detecting free amino acids in plasma.Method 40 blood samples from healthy volunteers were analyzed by an automatic amino acid analyzer ( Li+system) and the results compared with previous reports using other analyzers.Results The results ob-tained by this analyzer for major amino acids [asparagine (Asn), glutamic acid (Glu), glutamine (Gln), valine (Val), tyrosine (Tyr), phenylalanine (Phe), ornithine (Orn), arginine (Arg)] were similar with those previously reported using other amino acid analyzers ( all P>0.05 ) and liquid chromatography tandem-mass spectrometry (LC-MS/MS) (all P>0.05).Except for Glu and Tyr, the results for major amino acids showed large deviation compared to the results from high performance liquid chromatography ( HPLC) ( all P<0.05).Conclusion The amino acid analyzer (Li+) and LC-MS/MS (iTRAQ kit) could accurately detect free amino acids in plasma.
5.Clinical and genetic analysis of a pedigree with acne inversa

Objective To detect γ-secretase gene mutations in a large Chinese pedigree with acne inversa (AI).Methods Clinical evaluation was carried out in a large pedigree with AI through field investigation.Peripheral blood samples were obtained from 17 family members (11 affected and 6 unaffected) and 100 unrelated healthy human controls.DNA was extracted from the blood samples,and PCR was performed to amplify all the coding regions of PSEN 1,PSENEN and NCSTN genes followed by DNA sequencing analysis.Results There were 67 members over 5 generations in this family,of whom,25 (13 males and 12 females) were affected by AI.AI was inherited in an autosomal dominant manner in this family.Skin lesions were mainly distributed on the neck,back,chest and buttocks,and occasionally in subaxillary regions.DNA sequencing revealed a novel missense mutation,c.1258C> T (p.Q420XP),in the exon 11 of the NCSTN gene in 11 affected family members,which leads to a substitution of glutamine by a premature termination codon at amino acid 420 (p.Q420X).The mutation was undetected in either the unaffected members or the unrelated healthy controls,and had not been registered in the single nucleotide polymorphism (SNP) database in National Center for Biotechnology Information.Conclusions There is a novel heterozygous missense mutation,c.1258C > T in the exon 11 of the NCSTN gene,which may be the molecular basis of pathogenesis of AI in this family.
6.Effect of recombinant immunodominant major surface antigen of Echinococcus multilocularis in immunological assay for diagnosis of echinococcosis

Objective To evaluate effect of the non-fused recombinant antigen in immuno diagnosis of echinococcosis. Methods The coding sequence of the target gene was cloned into prokaryotic non-fusion expression vector pQE30 ( + ) and expressed in E. coli. The recombinant protein was identified by SDS-PAGE and Western Blot, purified with affinity chromatography and used in ELISA to detect specific antibody in patients' sera. Results SDS-PAGE and Western blot demonstrated that Echinococcus multilocularis immunodominant major surface antigen gene was highly expressed in E. coli. The result of ELISA showed that out of 68 echinococcosis sera, 66 samples (97.1% ) were detected positively by the assay. Only 2 samples (2% ) were positive in 100 sera from cases with other infective diseases including cysticercosis or healthy persons. The sensitivity and specificity of the assay were 97.1% and 98% respectively. Conclusion Echinococcus multilocularis immunodominant major surface antigen gene has been highly expressed in E. coli in non-fusion form. ELISA with the recombinant protein is highly sensitive and genus-specific for echinococcosis diagnosis.
7.ECM1 Gene Mutations in a Family with Lipoid Proteinosis

Objective To study mutations in the extracellular matrix protein 1 (ECM1) gene in a family with lipoid proteinosis (LP). Methods Bi-directional DNA sequencing was used to detect the ECM1 gene in patients with LP as well as among their parents, siblings and children. Results Sequencing from the affected individuals revealed a new compound heterozygote of missense/nonsense mutations, C220G/R476X, which were not found in the control group. The father was a carrier of the missense mutation C220G and the mother was a carrier of the nonsense mutation R476X, both on one allele. The other siblings, the patients' children and their siblings' children were carriers of either C220G or R476X mutations. Conclusion A new compound heterozygous mutation of ECM1 gene was identified in this LP family.
8.Effect of Intravenous Infusion of Vasostatin-2 on Hemodynamics in Experimental Rats With Spontaneous Hypertension

Objective: To observe the effect of the intravenous infusion of vasostatin-2 (VS-2) on hemodynamics in experimental rats with spontaneous hypertension (SH).
Methods: A total of 36 (14-16) weeks male SH rats with the mean body weight at (160-250) g were randomly divided into 6 groups:①Control group, the rats received normal saline (100μl/kg),②Catestatin (20μg/kg) group,③VS-2 (5μg/kg) group,④VS-2 (10μg/kg) group,⑤VS-2 (20μg/kg) group and⑥VS-2 (40μg/kg) group. n=6 in each group. The average blood pressure (BP), heart rate (HR) and barorelfex sensitivity (BRS) were monitored and compared upon VS-2 treatment and between VS-2 and catestatin treatments in conscious and freelance rats.
Results: Compared with prior treatment, VS-2 (20μg/kg) and VS-2 (40μg/kg) could obviously decrease the HR, BP and BRS in SH rats. In VS-2 (20μg/kg) group, HR by bpm was (341.3 ± 19.3) vs (365.5 ± 25.5), BP by mmHg was (133.0 ± 8.9) vs (147.5 ± 11.2) and BRS by ms/mmHg was (0.52 ± 0.18) vs (0.37 ± 0.12);in VS-2 (40μg/kg) group, HR was (348.8 ± 30.8) vs (374.5 ± 34.8), BP was (131.5 ± 9.3) vs (151.7 ± 10.8) and BRS was (0.53 ± 0.05) vs (0.38 ± 0.03), all P<0.01. Catestatin treatment could also decrease the HR as (318.7 ± 13.4) vs (365.5 ± 25.5), BP as (119.7 ± 7.3) vs (147.5 ± 11.2) and BRS as (0.58 ± 0.15) vs (0.35 ± 0.11), all P<0.01. Compared with catestatin (20μg/kg), the rats received VS-2 (20μg/kg) had the weaker reduction of HR as (318.7 ± 13.4) vs (341.3 ± 19.3), BP as (119.7 ± 7.3) vs (133.0 ± 8.9), all P<0.01, while BRS was similar as (0.58 ± 0.15) vs (0.52 ± 0.18), P>0.05.
Conclusion: Intravenous infusion of VS-2 may obviously affect HR, BP and BRS in experimental SH rats;compared with the same dosage of catestatin, VS-2 had the weaker reduction of HR, BP and BRS.
9.Investigation of overall Wiener spectrum measurement in a computed radiography system

Objective To study the effect of image processing on overall Wiener spectrum (WS) in a computed radiography system. Methods Imaging plate was exposed using conventional X-ray equipment. Overall WS sampling films were made by changing 3 parameters of spatial frequency processing, including frequency rang(RN), frequency type(RT), and degree of enhancement(RE), and by changing 1 of 4 parameters of gradation processing(rotation amount,GA).WS values were calculated by scanning on micro-densimeter for every film proceeded as discrete signal using fast Fourier transform. Results (1) The WS value of subscan direction was 7.26?10 -5 mm 2 when the spatial frequency was 1.0 LP/mm, it was 1.57 times larger than that of scan direction. The WS had peaks under high spatial frequency at sunscan direction. (2) The WS value of RN with 0 at scan direction was 7.90?10 -5 mm 2 when the spatial frequency was 1.0 LP/mm, it was 1.70 times larger than that of RN with 5, and 3.30 times larger than that of RN with 9. (3) The WS value of RE with 1.5 at scan direction was 1.05?10 -4 mm 2when the spatial frequency was 1.0 LP/mm, it was 2.27 times larger than that of RE with 1.0, and 2.92 times larger than that of RE with 0.5. (4) The WS value of GA with 1.5 at scan direction was 1.25?10 -4 mm 2 when the spatial frequency is 1.0 LP/mm, it was 2.70 times larger than that of GA with 0.8. (5) The WS value of RT with T at subscan direction was 9.80?10 -5 mm 2 when the spatial frequency was 1.0 LP/mm, it was 1.02 times larger than that of RT with F, and 1.35 times larger than that of RT with R, and also 1.55 times larger than that of RT with P. Conclusions RN, RE, and GA contributed strongly to Wiener spectra, but the contribution of RT was small. When processing the clinical images, adequate processing parameters should be chosen to reduce the image noise, so as to meet the clinical diagnostic need.
10.Receiver operating characteristic curve evaluation on computed radiography: an experimental study

Objective To get the maximum information from computed radiography (CR) output images by changing post-processing parameters. Methods Six experimental photos of polypropylene balls of 2.0 mm in diameter were taken by means of one time X-ray exposing on an imaging plate (IP) with different post-processing parameters including rotation amount (GA), gray gradation type (GT), rotation center (GC), shifting amount (GS), frequency rank (RN), frequency type (RT), and degree of enhancement (RE). 6 photos were viewed by three students and one radiologist on a 6 000 lx illuminance viewbox. Receiver operating characteristic (ROC) curves were made by means of 5-value-differentiation method. Results The largest mean area value (Az) below ROC curves of a low contrast experimental photo with post-processing parameters GA=1.0,GT=A, GC=1.6, GS=0.3, RN=4.0, RT=R and RE=3.0 was 0.96, and the maximum information was obtained. The smallest mean area value (Az) was 0.78 with changed post-processing parameters GA=0.8, GS=-0.2 and RE=0.5 while other parameters were not changed. The minimum information was obtained from this photo. Conclusion In order to get the maximum information from a CR output image, the post-processing parameters should be suitably selected.
