1.Effects of cyclooxygenase-2 on expression of vascular endothelial growth factor

Objective It has been known that cyclooxygenase-2(COX-2) acts as a tumor promoter in rodent models for colorectal cancer, but its precise role in the processes of carcinogenesis remains unclear. The study was designed to observe the relationship between expression of COX and the expression of vascular endothelial growth factor (VEGF) in the mouse embryo fibroblast (MEF) with knock out of COX-1 gene (COX-1 -/- ) or COX-2 gene (COX-2 -/- ) and wild MEF cells (COX-1 +/+ /COX-2 +/+ ). Methods We cultured the mouse embryo fibroblasts, measured the VEGF levels in the culture medium of these cells using ELISA, and extracted mRNA from these cells to identify the expressions of VEGF isoforms by RT-PCR. Results VEGF level could hardly be measured in the COX 2-deficient cells (COX-2 -/- ), however, the VEGF level was significantly increased in the cells with COX-2 gene (COX-2 +/+ ) and decreased by celecoxib, a COX-2 inhibitor. The level of VEGF was not associated with COX-1 expression. COX-2 inhibited the expressions of three isoforms of VEGF at mRNA level. Conclusions COX-2 plays an important role in the VEGF secretion and synthesis and therefore, it has an effect on the angiogenesis and tumor growth.
2.Influence of Ratios of Drug to Lipid on the Characteristics of Solid Lipid Nanoparticles Containing Mifepristone

AIM:The effective abortifacient drug,mifepristone,was incorporated in the solid lipid nanoparticles (SLNs). The aim of this paper was to study the influence of ratios of drug to lipid on the characteristics of SLNs.METHOD:The physicochemical properties of the fine dispersed systems,such as the size distribution,Zeta potential had been analyzed by laser diffractometry(LD).The measurements of entrapment efficiency(EE)and thermal analysis of DSC were performed as well.RESULT:It was showed if the weight of lipid remained unchanged,the mean particle size of SLNs increased with the increase of drug amount.Drug entrapment efficiency was the highest while about 50 mg drug was loaded on weight 1 g lipid.Simultaneously,the charge of Zeta potential agreed well with the amount of free drug in the colloidal system.DSC analysis results showed that the melting peak of mifepristone at approx.190 ℃ disappeared.CONCLUSION:It was evidence that the drug incorporation would affect the average particle size and Zeta potential of the colloidal systems and the physical state of crystalline drug mifepristone in SLNs was present in the amorphous form or molecularly dispersed at even so high adding amount of 250 mg/g(lipid),which there was no report about the physical state of the drug and SLN with so high drug-loading.It was suggested that the model drug mifepristone could influence the property of nanodispersion system and the crystalline character of the drug was altered by the nanometer carrier system vice versa.

4.Establishment and Application of Hospital ADR Monitoring System

OBJECTIVE:To improve the quality and efficiency of ADR monitoring in hospital for the convenience of reporters and ADR monitoring personnel.METHODS:Combined with the characteristics of hospital information system,computer technology such as network,ASP.NET programming language,SQL server 2005 database and data transformation were used to develop and apply ADR monitoring system based on hospital local area network.RESULTS:The ADR reporting quality has been improved and the quantity of ADR reports increased markedly and was kept stable.CONCLUSION:The system contributed to a informationalized and networking administration of ADR reporting,and it improved ADR reporting level thus deserving to be recommended.
5.The genetic susceptibility of HLA-DRB1,-DQB1 alleles in esophageal squamous cell carcinoma

Objective To probe into the association between the polymorphisms of HLA DRB1, DQB1 alleles and esophageal squamous cell carcinomas and then to further examine the features of their immunogenetics. Methods The polymorphisms of HLA DRB1, DQB1 alleles were typed by sequence specific primer based on polymerase chain reaction in 42 patients with esophageal squamous cell cancer and 136 normal control subjects. Results Allele frequency of HLA DRB1 *0901 allele was significantly higher in esophageal cancer patients than that in normal controls ( 0.250 0 vs 0.139 7 , P = 0.028 , odd ratio= 2.053 , etiologic fraction= 0.128 2 ) and so was allele frequency of HLA DQB1 * 0301 ( 0.297 6 vs 0.187 5 , P = 0.046 , odd ratio 1.835 , etiologic fraction 0.135 4 ). There was no association between the rested alleles of HLA DRB1 and DQB1 alleles in the patients. Conclusions Individuals carrying HLA DRB1 * 0901 and/or DQB1 * 0301 may be susceptible to esophageal squamous cell carcinoma, and nucleotide sequences of these two alleles were in accordance with the corresponded allele sequence (exon 2) of genebank.

Objective To investigate the expression of inducible nitric oxide synthase (iNOS) in human gastric carcinoma and its relationship with angiogenesis, lymph nodes metastasi s and clinical staging of gastric carcinoma. Objective To evaluate the role of CREB-binding protein (CBP) in cyclooxygenase 2 (COX-2) expression in spinal cord in a rat model of neuropathic pain (NP).Methods Forty male SD rats weighing 220-250 g in which intrathecal catheter was successfully implanted were randomly divided into 4 groups (n=10 each):sham operation group (group Sham),group NP,negative control group (group NC) and group CBP.NP was induced by CCI at 5 days after successful implantation of the intrathecal catheter.Normal saline,negative lentivirus vector (RNAi-NC-LV) and CBP shRNA lentivirus vector (CBP RNAi-LV) 20 μl were injected intrathecally at 7 days after CCI in groups NP,NC and CBP,respectively.Paw withdrawal threshold to mechanical stimuli (MWT)and paw withdrawal latency to thermal stimulus (TWL) were measured at 1 day before operation and at 3,5,7,10,12 and 14 days after operation.The rats were sacrificed after the measurement of MWT and TWL at 14 days after operation and the lumbar segment of the spinal cord was then removed for determination of the expression of CBP,acetylated histone H3/H4 (Ac-H3,Ac-H4) and COX-2 protein (by immunohistochemistry),and CBP and COX-2 mRNA (by RF-PCR).Results Compared with group Sham,MWT and TWL were significantly decreased at different time points after operation in groups NP,NCand CBP,and the expression of CBP,Ac-H3,Ac-H4 and COX-2 protein,and CBP and COX-2 mRNA was up-regulated at 14 days after operation in groups NP and NC (P < 0.05).Compared with group NP,MWT and TWL were significantly increased at 10,12 and 14 days after operation and the expression of CBP,Ac-H3,Ac-H4 and COX-2 protein,and CBP and COX-2 mRNA was downregulated at 14 days after operation in group CBP (P < 0.05).Conclusion CBP is involved in the development of NP by up-regulating COX-2 expression and increasing histone H3 and H4 acetylation in spinal cord.

9.Factors affecting shoulder joint functions following artificial humeral head replacement: 22 cases analysis

OBJECTIVE: To investigate the factors affecting shoulder joint functions following artificial humeral head replacement. METHODS: A total of 22 patients with complex fracture of proximal humerus, managed by humeral head replacement in Changhai Hospital of The Second Military Medical University from June 2006 to December 2008 were collected, including 12 males and 10 females, with average age of 67 years (52-86 years). During the procedure, soft tissue damage was minimized, the bone of the tuberosity was reserved as much as possible, appropriate matching size of humerus head prosthesis was selected, and early staging functional rehabilitation was performed. Neer's scoring system was used to evaluate the treatment results after replacement. RESULTS: The 22 patients were followed up for 10-30 months, mean 22.6 months. The shoulder joint functions of 14 cases were recovered more quickly through early post-operative staging rehabilitation, No shoulder joint unstable and stiff occurred. The shoulder joint functions of 8 cases were recovered slowly as they complained pain and did not perform early physical therapy after surgery, shoulder joint of 2 cases presented little unstable and 1 case presented stiff. Only 4 cases of all complained occasionally slight shoulder pain, but could continue daily work. The mean movement ranges of shoulder were as follow: mean flexion for 96°(80°-103°), mean extension for 38° (32°-45°), mean abduction for 86°(80°-110°), mean adduction for 31°(30°-35°), mean external rotation for 32°(30°-37°) and internal rotation for 42° (10°-75°). No vasculardamage, nerve damage, lessening prosthesis or prosthesis dislocation was found in all cases. Based on Neer's scoring system, 5 cases were rated as excellent, 11 as good and 4 as fair, with excellent rate of 82% and case satisfaction rate of 87%. CONCLUSION: In order to recover maximally the shoulder joint functions for post-artificial humeral head replacement, we must try our best to avoid soft tissue damage, reserve the bone of the tuberosity, select appropriate humerus head prosthesis, perform early post-operative staging rehabilitation, as well as maintain a normal retroversion of the prosthesis.
10.Correlation study of electrocardiography and risk stratification in patients with early repolarization syndrome

Objective To explore the value of electrocardiagraphy and risk stratification in patients with early repolarization syndrome( ERS ). Methods All of 271 eases were divided into three groups: 7 ERS patients with cardiac events (syncope,clinical sudden death or incompleted, ventricular fibrillation and induced ventricular fibrillation during electrophysiological), 82 ERS patients without cardiac events, and 182 cases without heart disease who were matched for age and sex. QRS interval,Tpeak-Tend (TpTe) interval,corrected QT interval(QTc) and QT deviation degree (QTd) in surface ECG were compared among the three groups. Results TpTe interval in ERS patients with cardiac events was significantly longer than that in ERS patients without cardiac events and the control subjects [ ( 123.3±22.4 ) ms vs (87.7±15.5 ) ms; ( 123.3±22.4) ms vs (83.5±15.1 ) ms, P < 0.05 ]. There was no significant difference in QRS interval, QTc and QTd among three groups. Conclusion TpTe interval in ECG might be useful in risk stratification with ERS, but further research is needed.
