1.Effects of low dose Glibenclamide on secondary damage after acute spinal cord injury in rats

Objective To investigate the effects of Glibenclamide on reduction of secondary damage after acute spinal cord injury in rats.Methods Ninety rats were randomly divided into control group (laminectomy alone),spinal cord injury group(injury group),and treatment group(treated with Glibenclamide after spinal cord injury),with 30 rats in each group.The pathological morphology changes of injured spinal cord were observed by HE staining and electron microscope.The expressions of sulfonylurea receptor 1 (SUR1)were detected by immunohistochemical method at 45 min,6 h,24 h,3 d and 7 d after spinal cord injury,and IPP 6.0 software were used for quantitative analysis.The function recoveries of the hind limbs of rats were evaluated by BBB score.The blood sugar level was detected quantitatively.Results HE staining showed that tissue bleeding and microglia proliferation getting severe with time after spinal cord injury.Compared to the injury group,tissue bleeding,microglia proliferation and inflammatory cell invasion was less severe in treatment group.Showed by electron microscope,inflammatory cell invasion,myelin sheath layer structure damage and mitochondrial swelling were significantly reduced after Glibenclamide treatment.Detected by immunohistochemical staining,the expressions of SUR1 at all time points after injury,except for 45 min that there were no SUR1 expressions in all groups,were much weaker in the treatment group than in the injury group.The SUR1 expression reached the peak at 24 h after injury in injury group,and decreased gradually with time.Significant differences were found in the SUR1 expression among three groups by oneway ANOVE.The BBB scores of treatment group were significant higher than that of injury group.The blood sugar slightly decreased in the treatment group,while no significant difference was found among three groups.Conclusion Glibenclamide can significant reduced the secondary damage after acute spinal cord injury.The protection of Glibenclamide after spinal cord injury may relate to its suppression of SUR1.
2.Effects of rosiglitazone on the atherosclerosis and oxidative stress in diabetic rabbits

Rosiglitazone was used for intervention of atherosclerosis in diabetic rabbits.The results showed that the intima/medium thickness ratio,cross section area of plaque,and the expressions of NADPH oxidase p22phox,gp91 phox were decreased;while the total anti-oxidative ability was increased after administration of rosiglitazone as compared with the non-intervention group(all P<0.05).Compared with rosiglitazone treatment group,serum hepatocuprein leveI in rosiglitazone prevention group was increased,while serum malonaldehyde level decreased(both P<0.05).This study suggests that rosiglitazone may have the effect of reducing the oxidative stress and the formation of atherosclerotic plaque in diabetics.
3.Effects of fluvastatin on the expression of connective tissue growth factor and type Ⅳ collagen in mesangial cells induced by transforming growth factor-?1

Aim To investigate the effects of fluvastatin on the cellular proliferation and the expression of connective tissue growth factor(CTGF)and type Ⅳ collagen(Col Ⅳ)in rat mesangial cells(MCs)induced by transforming growth factor-?1(TGF-?1).Methods MCs are divided into the following groups according to different factors:control group,TGF-?1 group,TGF-?1 plus fluvastatin(Flu,different concentrations)groups.The influence of Flu on rat proliferation of MCs was detected by MTT.The expression of CTGF was measured with RT-PCR and Western blot respectively.The secretion of Col Ⅳ protein was quantitated by ELISA.Results 5 ?g?L-1 TGF-?1 could stimulate proliferation of MCs and the expression of CTGF.Col Ⅳ in MCs significantly.Flu could inhibit proliferation of MCs and the expression of CTGF,Col Ⅳ in MCs induced by TGF-?1 in a dose-dependent manner:1 ?mol?L-1 Flu could suppress proliferation of MCs and downregulate the expression of CTGF,Col Ⅳ significantly,while 10 ?mol?L-1 Flu could just suppress the expression of CTGF protein significantly.Simultaneously exogenous CTGF could promote the expression of Col Ⅳ in a dose-dependent manner:2.5 ?g?L-1 CTGF could just upregulate the expression of Col Ⅳ.Conclusion Fluvastatin could inhibit proliferation of MCs and expression of CTGF-mediated Col Ⅳ in MCs.

Objective To explore the inhibition effect of recombinant mutant type(mt) p27 gene adenovirus (Ad-mt p27) transfection on the proliferation of colorectal cancer cell line SW480. Methods The recombinant mt p27 adenovirus vector was transfected into SW480 cells, western blotting analysis was applied to determine the p27 expression, and cell cycle distribution was analyzed using flow cytometry, and growth curve was drawn to observe the cell growth. Results There was high expression of p27 protein in the transfected SW480 cells. 77 9% cells were blocked in G 1/G 0 phase in the transfected group, which the percentage of the G 1/G 0 phase of Ad-LacZ transfected group and normal control group were 27 5% and 25 29% respectively. Ad-mt p27 transfection markedly inhibited SW480 cell growth in 24h and the inhibition effects was maintained for 7 days. Conclusion Recombinant adenovirus could efficaciously trasfect mt p27 gene into SW480 cells, and obtain high level mt p27 exprerssion. mt p27 markedly bolcked SW480 cell cycle progression and prominently inhibited SW480 cell growth. The mutant p27 gene transfection may be a new way of gene therapy for colorectal cancer.
7.Atrial septal puncture guided by right anterior oblique 45 degree projection

Objective To introduce a simple and reliable way for atrial septal puncture. Methods Atrial septal puncture had been carried out in 627 patients with various underlying heart diseases, and the patients aged between 4-78 years old. Three protocols for precise location of atrial septal puncture points were recommended, 1) Site of atrial septal puncture point in cranial-caudal direction was determined under posterior-anterior projection, which was confined to the cranial side of the inferior margin of left atrium silhouette at a distance of 1 cone body height along the midline of spine. If the inferior margin of left atrium silhouette was unclear, angiogram of pulmonary artery to display left atrium and placement of coronary sinus electrodes could be done to verify it. 2) Under right anterior oblique 45 degree view, the puncture point was located between one cone body height anterior to posterior margin of left atrium silhouette and the isometric line between the posterior margin of left atrium silhouette and atria-ventricular suculus. 3) The arch feature of puncture needle and distal part of sheath turned into a straight line under 45 degree of right anterior oblique view. Results The success rate of atrial septal puncture was 99.8% (626/627), and that with only one try was 71.93% (451/627), the rate of tamponade was 0.32% (2/627). No death occurred. Conclusion Simple, reliable and safe, atrial septal puncture under 45 degree of right anterior oblique view is an easily-mastered approach.
8.MRI qualitative estimation of cerebrospinal fluid flow in normal volunteers

Objective To evaluate the feasibility of the cerebrospinal fluid (CSF) flow measurement by cine phase contrast MRI. Methods Evaluation of intracranial and intraspinal CSF flow was accomplished by the use of periphery gated cine phase contrast MRI technique. Normal patterns of pulsatile flow within the ventricles, cisterns and cervical subarachnoid space were established by this technique. Results CSF flow within ventricle, cistern and subarachnoid space could be clearly displayed, and the relationships between the cardiac cycle and the CSF pulsations were demonstrated on both magnitude reconstruction and phase reconstruction MR images. Conclusions Cine phase contrast MRI is capable of demonstrating intracranial and intraspinal CSF flow. As a new noninvasive technique, it is really superior than other modalities in studying the flow pattern and direction of CSF.
9.Retinoid X receptor agonist inhibits TGF-β1-induced collagen synthesis in cardiac fibroblasts by repressing Smad2 activation

AIM:To investigate the effect of activation of retinoid X receptor (RXR) on transforming growth factor β1 (TGF-β1) induced collagen synthesis under hypoxic environment in rat cardiac fibroblasts (CFs) and underlying molecular mechanisms .METHODS: CFs were cultured using myocardial tissue with dry method .Hypoxic environment was established for CFs by continuous nitrogen supplement .Type I and type III collagens in supernatants were detected by ELISA.Nuclear and cytoplasmic extractions were prepared using NE-PER nuclear and cytoplasmic extraction reagents .The protein levels of Smad2 and p-Smad2 were determined by Western blot and immunocytochemical staining .RESULTS:Un-der hypoxic condition , TGF-β1 (0.01~10 μg/L) increased the synthesis of type I and type III collagens in a dose-de-pendent manner in the CFs .At the concentration of 5μg/L, the synthesis of collagen I and III was significantly increased as compared with control group (P<0.01).RXR agonist 9-cis-retinoic acid (9-cis-RA;10 -9 ~10 -6 mol/L) decreased TGF-β1 (5μg/L)-induced synthesis of type I and III collagens in a dose-dependent manner in the CFs under hypoxic con-dition.The synthesis of type I and type III collagens was significantly inhibited by 9-cis-RA (P<0.01).Smad2 inhibitor ( 20 nmol/L) showed similar inhibitory effect on the synthesis of type I and III collagens induced by TGF -β1 under hypoxic condition.Compared with TGF-β1 intervention group, the cytoplasmic level of p-Smad2 in the CFs was significantly in-creased in TGF-β1+9-cis-RA group, but the nuclear p-Smad2 level was significantly decreased (P<0.05).CONCLU-SION:Retinoid X receptor agonist 9-cis-RA inhibits TGF-β1-induced synthesis of type I and type III collagens in the CFs by repressing p-Smad2 nuclear translocation under hypoxic condition .
10.Clinical application of double incision in phacoemulsification combined with trabeculectomy

Objective Conducting phacoemulsification combined with trabeculectomy through double incision to treat glaucoma and cat-aract,and observing the visual acuity,intraocular pressure before and after the operation and complications of this therapy. Methods Totally 60 patients (78 eyes) with glaucoma and cataract from January 2010 to December 2013 in our hospital were observed. 32 patients (40 eyes) who were given phacoemulsification combined with trabeculectomy through double incision were regarded as the observation group,and the other 28 patients (38 eyes) who were merely given phacoemulsification were regarded as the control group. The visual acuity,intraocular pres-sure and anterior chamber of the two groups were observed and recorded,and they were followed up for 36 months. Results Visual acuity and intraocular pressure of the two groups were improved after operation,but the observation group achieved a better result which showed a significant difference (P< 0. 05). Anterior chamber inflammation occurred in patients of the observation group,and 2 cases were of anterior chamber exudation. The control group,by contrast,has less anterior chamber inflammation. There was no hyphema,shallow anterior chamber, choroidal detachment occurred in both of the two groups. Conclusion Using phacoemulsification combined with trabeculectomy through double incision to treat glaucoma and cataract could receive a good recovery of visual acuity and intraocular pressure. Although,more severe anterior chamber inflammation occurred after operation compared with the control group,it can be alleviated soon after medication. This meth-od is a good therapy which can reduce pain and economic burden of patients from multiple operations. But it requires a strict selection of ap-propriate case.
