Objective: [WT5”,6BZ]To investigate the anti tumor effects of melittin in vitro . Methods: Melittin was isolated and purified from bee venom by gel filtration. The cytotoxicity of melittin against established cell lines(SMMC 7721,BEL 7402 and Hep 3B cell) was determined using by MTT assays. Mitomycin, vincristine and cinobu facini were used as control. The time effect relationship of melittin against SMMC 7721 cell line was studied. Results: Melittin showed the anti tumor effect in a dose dependent matter for tumor cells in vitro ,but there was no time effect relationship. The anti tumor effect of melittin was better than the control. Conclusion: Melittin has marked and quick anti tumor activity in vitro against established cell lines. [

Objective To investigate the change of renal 1-alpha hydroxylase and its effect on bone mineral density in the old rats with type 2 diabetic nephropathy. Methods Forty Wistar rats of 18 months old were randomly divided into normal control (N), diabetes (D), diabetes treated with vitamin D_ 3 (T1) and diabetes treated with 1-?(OH)D_ 3 (T2) groups, respectively. Ten rats in each group. Dual energy X-ray absorption (DEXA) was used to determine bone mineral density (BMD) of lumbar spines and femoral bone. 24 h urinary protein excretion, serum 25(OH)D_ 3 and 1,25(OH)_ 2 D_ 3 were measured by radioimmunoassay. Results Compared with controls, 24 h urinary protein excretion increased remarkably D, T1, T2 groups, while BMD greatly decreased, much lower in D group and T1 group than T2 group (P0.05). Objective:To isolate and purify melittin from bee venom and to establish an SDS PAGE and HPLC method for the determination of melittin content.Methods:Melittin was isolated and purified from bee venom by gel filtration on Sephadex G 25,Sephadex G 50 and Sephadex G 75.Homogenicity and molecular weight of melittin were determined by SDS PAGE and HPLC.Results:The sample was a single peptide chain with a molecular mass of about 3 000 dalton as revealed by SDS PAGE.The average recovery was 95.97% and RSD was 1.07%.Conclusion:Melittin prepared by a combination of gel filtration has high purity.The method for the determination of melittin with SDS PAGE and HPLC is accurate,reliable and simple. [

Objective To investigate the relationship between lipid abnormality and benign prostatic hyperplasia (BPH) in men receiving physical examination. MethodsFour hundred and one people who participated health examination in our hospital in May 2008 were included in the study. They were divided into two groups according to history of disease, IPSS score, the results of digital rectal examination and transrectal ultrasound: BPH group (192 cases) and no BPH group (209 cases). The blood lipid level and risk factors were compared in two groups. ResultsThere were no statistical differences in blood lipid level and risk stratification of cardiovascular disease between the two groups (TG:P=0. 698;TC:P=0. 654;LDL-C:P=0. 880; HDL-C:P=0. 531; risk stratification: P= 0. 251). Objective To explore how does chymase affect diabetic cardiomyopathy by investigating the gene expression of chymase and the relationship between the gene expression of chymase and the angiotensin Ⅱ in the cardiomyopathy of streptozotocin-induced diabetic hamasters. Methods Diabetic hamsters were induced by intraperitoneal injection of STZ 40 mg/kg once a day for 3 days. After stabilization of diabetic state for 18 weeks, the myocardial ultrastructure was observed with electron microscope and pathologic changes were observed by light microscopy. Immunohistochemistry was used to measure the level of expression of type Ⅰ and type Ⅲ collagen in diabetic and normal hamster hearts. Level of blood glucose, lipoprotein was determined using biocehemical methods. Apoptosis of cardiomyocyte was measured using TUNEL methods. Radioimmunuoassay method was used to determine the level of angiotensin Ⅱ. RT-PCR was used to determine chymase gene expression (corrected by?-actin). ResultsComparing with the control group, levels of serum glucose, TG, TC, LDL in DM group were much higher. Concentrations of collagen Ⅰ, Ⅲ and angiotensin Ⅱ [(95.8?16.0)?g/kg tissue vs (51.1?20.8)?g/kg tissue] in myocardial tissue in DM group were much higher than those in control group. RT-PCR result showed: Comparing with the control group, the mRNA expression of chymase in DM group was promoted significantly (0.810?0.026 vs 0.490?0.087). OBJECTIVE: To investigate the anti-tumor effect of bufalin and its regulation on Bcl-2 and Bax proteins in orthotopically transplanted tumor of human hepatocellular carcinoma in nude mice. METHODS: Orthotopically transplanted tumor of human hepatocellular carcinoma was established in nude mice. The mice were randomly divided into five groups: high-dose bufalin-treated group (1.5 mg/kg), medium-dose bufalin-treated group (1 mg/kg), low-dose bufalin-treated group (0.5 mg/kg), adriamycin-treated group (8.0 mg/kg), and normal saline-treated group. After 25 days, mice were sacrificed. The tumor volume was measured, and the pathological changes of tumor tissues were detected by HE staining to observe the tumor necrosis degree. Cell morphological changes were also observed by an electron microscopy. Label index of tumor cell apoptosis was assessed by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL), and the expressions of Bcl-2 and Bax proteins were determined by immunohistochemical method. RESULTS: The tumor volume in the bufalin-treated groups was shrunk significantly compared with that in the normal saline-treated group (P<0.01). The survival time of the bufalin-treated groups was prolonged compared with that of the adriamycin-treated group and the normal saline-treated group P<0.05. Apoptotic characteristics could be seen in tumor tissues of the bufalin-treated groups. The label index of tumor cell apoptosis in the bufalin-treated groups (5.87+/-2.13, 8.86+/-2.96 and 10.60+/-3.42 in low-, medium- and high-dose groups respectively) was higher than that in the adriamycin-treated group (3.28+/-0.98) (P<0.05, P<0.01). The expression of Bax was up-regulated, while no changes were detected as to Bcl-2 protein in tumors of the bufalin-treated groups. CONCLUSION: Bufalin has significant anti-tumor effect on the orthotopically transplanted tumor of human hepatocellular carcinoma in nude mice. Its effect might be related to up-regulation of Bax protein and inducement of the tumor cell apoptosis.
