1.Effects of NEP1-40 on GAP-43 and RhoA expression in rats of cerebral ischemic-reperfusion model

Objective To study the effects of NEP1-40 on axon regeneration, motor function recovery of affected limbs and RhoA signal pathway in cerebral ischemia-reperfusion rats for exploring the possible mechanisms. Methods A total of 60 adult male Sprague-Dawley (SD) rats were equally divided into sham operation group (sham group), cerebral ischemia-reperfusion control group (control group), intra-lateral ventricle injection of PBS group (PBS group), and intra-lateral ventricle injection of NEP1-40 group (NEP1-40 group). The middle cerebral artery ischemia-reperfusion model (MCAI/R) was established by nylon monofilament occlusion method in rats. The changes in growth associated protein-43 (GAP-43) and RhoA expressions were determined by Western blotting technique. The motor function of affected limbs was tested by the "staircase test" of Montoya’s design. Results ① The expression of GAP-43 in NEP1-40 group was higher than that in the control and the PBS groups at 7 d and 14 d after MCAI/R, peaking at 7 d; ② The expression of RhoA in NEP1-40 group was significantly lower than that in the control and the PBS groups at each time point; ③ The results of "staircase test" in NEP1-40 group were much higher than those in the control group and the PBS groups at each time point after MCAI/R. Conclusion NEP1-40 can improve axon regeneration in cerebral ischemia-reperfusion rats and promote the recovery of motor function. The mechanism may be associated with the inhibition of RhoA signal pathway.
2.Advances in studies on the application of platelet-rich plasma in the field of orthopedicsand the underlying mechanisms

Platelet-rich plasma(PRP) is a highly-concentrated plasma enriched in platelets and various growth factors. These growth factors will be released abundantly by platelets when PRP is activated by thrombin, which plays a key role in accelerating wound healing, bone repair, and meanwhile, in controlling of inflammatory process. Biocompatibility and versatility of PRP have been confirmed in clinical studies as well as its excellent effects on tissue reparation. These have lead to the extensive use of PRP in many medical fields as a therapeutic alternative. Mechanism of PRP in bone and tissue repair the application of PRP in the field of orthopedics is reviewed in this article. In addition, attempt is made to discuss about some problems still existing in the clinical application of PRP.
3.Researches on the applications of bioglass in orthopaedics

Bioglass is one of the most important inorganic biomedical materials and is widely used in clinic. Orthopedists and researchers pay more and more attentions to bioglass due to its excellent biocompatibility and bioactivity. This paper introduces the current research status and development of bioglass in bone repairing,bone tissue engineering, anti-infection drug delivery material and interventional therapy. Brief review is given to the potential application of bioglass in orthopaedics.
4.Effect of Panax notoginseng on gastric mucosal injury, superoxide dismutase activity, malondialdehyde and nitric oxide contents in rats with water immersion restraint stress

BACKGROUND: Panax notoginseng is an effective medicine for curing gastric mucosal ulcer. There are many clinical reports that Panax notoginseng protects gastric mucosa.OBJECTIVE: To investigate the protective mechanism of Panax notoginseng on the gastric mucosal injury induced by water immersion restraint stress in rats.DESIGN: A randomized controlled animal trial.SETTING: School of Basic Medical Sciences, Daqing Branch of Harbin Medical University.MATERIALS: The study was conducted in the laboratory of Department of Pathophysiology of Harbin Medical University from September 2004 to October 2005. Forty-eight Wistar rats were used, either male or female, weighing 180-230 g.METHODS: The 48 Wistar rats were randomly divided into six groups with 8 rats in each group: normal control group, stress model group, cimetidine treatment group, Panax notoginseng of low, middle and high-dose groups (4, 8, 12 mg/time). In the cimetidine treatment group, cimetidine tablets were grinded into powders, then mixed with distilled water to prepare into suspension (1 tablet:10 mL), which was perfused intragastrically (5 mL), 3 times a day; In the Panax notoginseng groups, the powders in Panax notoginseng capsules were mixed with distilled water to prepare into suspension of corresponding concentrations (0.8, 1.6, 2.4 g/L), then administrated the same as those in the cimetidine treatment group. Stress models in rats were established by means of water immersion restraint stress. The gross and pathohistological changes of gastric mucosa were observed, and the activity of superoxide oxidase (SOD) and contents of malondialdehyde (MDA) and nitric oxide (NO) were determined.MAIN OUTCOME MEASURES:①Gross and pathohistological changes of gastric mucosa;②Changes of MDA and NO contents and SOD activity in the homogenate of gastric mucosa.RESULTS: All the 48 rats were involved in the analysis of results. The gastric mucosal hemorrhage and erosion in the cimetidine treatment group were reduced obviously as compared with those in the stress model group, SOD activity was obviously decreased [(12.61±0.87), (1.03±0.60) mkat/g], whereas the NO content was a little higher [(5.76±1.35), (0.97±0.58) nmol/g]. The MDA content was obviously higher in the stress model group than in the normal control group [(3.10±1.13), (0.09±0.02) μmol/g, P<0.01]. There were no obvious differences between the Panax notoginseng groups and the cimetidine treatment group except that the NO contents were decreased in the in Panax notoginseng groups.CONCLUSION: Gastric mucosal injury induced by water immersion restraint stress can be significantly protected by Panax notoginseng, which is not dose-dependent. The protective mechanism may be associated with that Panax notoginseng can eliminate the product of oxygen-derived free radicals, and it is not totally the same as that of cimetidine
5.THE MORPHOLOGICAL OBSERVATION OF THE OSTEOBLAST CULTURED WITH RECONSTRUCTED ACELLULAR EXTRACELLULAR MATRIX IN VITRO

Objective The construction of the reconstructed acellular extracellular matrix(REAECM) of bone and preliminary investigation of biocompatibility of REAECM was performed to supply the experimental basis for the extracellular scaffold of bone tissue engineering. Methods Applying the technique of culture in vitro, biocompatibility of the REAECM cultured with osteoblasts was observed using phase-contrast microscope and scanning eletron microscope at 1-4 weeks. Results The osteoblasts could attach and proliferate, even grow well into the foam of REAECM.Conclusion REAECM is an excellent scaffold material for bony tissue engineering construction, which pocesses a morphological structure of three-dimensional foam. The REAECM showed a good biocompatibility with osteoblasts co-cultured in vitro. There was prospective signs of REAECM to the attachment, proliferation of the osteoblasts

7.Allogeneic bone marrow mesenchymal stem cell transplantation for ventricular remodeling after myocardial infarction

BACKGROUND:Myocardial infarction leads to ischemic changes in the myocardium, triggering the emergence of ventricular remodeling, which is an important cause of death. Myocardial infarction is a common disease in the middle-aged and elderly population, but autologous bone marrow mesenchymal stem cells from these patients exhibit a weak ability of proliferation and differentiation. Therefore, a positive attempt of allogeneic stem cel transplantation is required in order to obtain better therapeutic outcomes. OBJECTIVE:To explore the effect of al ogeneic bone marrow mesenchymal stem cel s on ventricular remodeling after myocardial infarction. METHODS:Bone marrow mesenchymal stem cel s from 10 neonatal rats and 10 adult rats were isolated, cultured and identified. Another 40 rats were randomly assigned into four groups (n=10/group):model group, neonatal rat cel transplantation group, adult rat cel transplantation group, or sham group. Animal models of myocardial infarction were made in rats in the al groups except for the sham group in which the rats were given sham operation. Rats in the two cel transplantation groups were given the corresponding cel transplantation. Four weeks postoperatively, heart function of rats was detected in each group, and cardiac tissues were taken to detect changes in col agen formation and blood vessel density in the infarct area. RESULTS AND CONCLUSION:Four weeks after surgery, rats in the model group showed significant changes in cardiac function indexes as compared with the other groups (P<0.05), while compared with the model group, these cardiac function indexes improved in both two cel transplantation groups, but there was no significant difference between the two cell transplantation groups (P>0.05). Meanwhile, compared with the model group, significantly decreased collagen formation and increased blood vessel density were found in both two cell transplantation groups (P<0.05). Additionally, the vascular density of the infarct area was highest in the sham group (P<0.05). Experimental results show that both neonatal and adult rat bone marrow mesenchymal stem cel transplantation can improve cardiac function of rats, reduce the formation of collagen in the infarct area and delay ventricular remodeling after myocardial infarction.
8.Relationship between B-tupe natriuretic peptide and preeclampsia of hypertensive disorder complicating pregnancy as well as its significance

Objective To investigate and evaluate the correlation between brain natriuretic peptide (BNP) and gestational hypertension and preeclampsia of hypertensive disorder complicating pregnancy (HDCP).Methods Fifty cases with HDCP and 46 cases with mild and 83 cases with sever stage preeclampsia were selected as our subjects.And 33 cases with regular pregnancy and 31 with irregular pregnancy were served as control group.Plasma brain natriuretic peptide,urinary protein quantity(UBQ),24-hour urinary protein assay (UPA) were measured.The correlations of brain natriuretic peptide and UBQ,UPA,systolic pressure (SP),diastolic pressure (DP) were analyzed.Results The levels of brain natriuretic peptide in the group with gestational hypertension and mild,severe preeclampsia groups were (48.54± 18.27),(79.46± 32.18) and (292.24±213.08) ng/L,higher than that in normal pregnancy and non pregnant group ((27.84± 14.58) and (20.63± 8.28) n/L;F =49.583,P<0.05).While no significant difference exists between normal pregnancy group and non pregnant group.Grouped on the median values (199) of brain natriuretic peptide of the severe preeclampsia group,the levels of 24-hour UPA,systolic pressure and diastolic pressure were (5.46±2.68) g,(174.55± 13.58) mmHg,(113.74±9.91) mmHg in patients with brain natriuretic peptide ≥ 199 ng/L(n=42),significant higher than those in patients with brain natriuretic peptide < 199 ng/L(n =41;(4.34± 1.95)g,(165.31±11.12) mmHg,(106.05±8.02) mmHg;t=2.603,3.396,2.308;P=>0.010,0.001,0.024).The levels of 24-hour UPA,systolic pressure and diastolic pressure of patients with brain natriuretic peptide ≥ 86ng/L(n=20) in mild preeclampsia were (1.68±0.27) g,(163.69±8.29) mmHg,(105.45±6.71) mmHg,significant higher than those in patients with brain natriuretic peptide < 86 ng/L (n =26;(1.16 ± 0.31) g,(152.90±7.32) mmHg,(99.19 ± 5.25) mmHg;t =3.180,2.508,2.32;P =0.010,0.016,0.025).Brain natriuretic peptide was closely correlated with UPA,systolic pressure and diastolic pressure in hypertensive disorder complicating pregnancy (HDCP) (r =0.29,0.30;P < 0.01).Brain natriuretic peptide was closely correlated with UPA systolic pressure and diastolic pressure in mild preeclampsia (r =0.39,0.37,0.40;P <0.01).And correlation efficacy of brain natriuretic peptide with UPA,systolic pressure and diastolic pressure were 0.44,0.42 and 0.53 (P<0.01).Conclusion The level of brain natriuretic peptide is closely associated with the severity of gestational hypertension and preeclampsia of hypertensive disorder complicating pregnancy.Correlation of brain natriuretic peptide to the severity of gestational hypertension and preeclampsia is independent of urinary protein and hypertension.Brain natriuretic peptide is an important indicator for the severity of gestational hypertension and preeclampsia of hypertensive disorder complicating pregnancy.
9.Survey on inpatient medical experience at five public hospitals in Henan

Objective To learn the medical experience of inpatients at public hospitals in Henan and the influencing factors.Methods Inpatient experience questionnaire (IPEQ) was customized for a random sampling of 500 inpatients at five tertiary public hospitals in Henan.Results Overall satisfaction of inpatients experience scored 8.48,of which the satisfaction for technical competence was the highest (4.19)and that for emotional support the lowest(3.31).The correlation analysis revealed that the doctors' technical competence score the highest correlation with the experiences (0.652).Conclusion Overall satisfaction of inpatients experience at public hospitals in Henan was found high in general,but humanistic care and service flow at the hospitals require further improvement.

BACKGROUND:Exogenous basic fibroblast growth factor (bFGF) plays an important role in the ligament tissue healing process, and the use of transgenic methods to transfect exogenous genes into cells can promote the secretion of bFGF. OBJECTIVE:To observe phenotypic changes and the bFGF protein expression after bFGF recombinant adenovirus was used to transfect rabbit bone marrow mesenchymal stem cells (BMSCs). METHODS:Passage 2 BMSCs were divided into three groups:Ad.bFGF-eGFP group, Ad.eGFP group and control group. Under a phase contrast microscope we observed the changes in cellmorphology. The expression of bFGF protein in BMSCs was determined by enzyme-linked immunosorbent assay (ELISA). The proliferative curve was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT). RESULTS AND CONCLUSION:The transfected cells showed a uniform phenotype of fibroblasts. MTT colorimetric assay revealed that more proliferative activity of transfected BMSCs was shown in the Ad.bFGF-eGFP group than in the Ad.eGFP group and control group. ELISA results showed that expression of bFGF protein was higher in the Ad.bFGF-eGFP group than in the Ad.eGFP group and control group (P<0.05). BFGF recombinant adenovirus can induce the differentiation of BMSCs into fibroblasts, increase proliferative ability and promote the expression of bFGF protein.
