To study the effect of Cu2+ and Zn2+ on amyloid-beta peptides (Abeta) aggregation, the morphology, size and cell toxicity of Abeta40 aggregates formed with the metal ions have been observed by the methods including ultraviolet spectroscopy, fluorescence spectroscopy and transmission electron microscopy. The results showed that Cu2+ and Zn2+ can accelerate Abeta40 aggregation, and both changed the morphology and size of Abeta40 aggregates. Zn2+ induced Abeta40 to form fibrous Abeta40 aggregates, while the amorphous and fibrous aggregates were produced by the interaction between Cu2+ and Abeta40. In addition, H2O2 was produced when Abeta40 reduced Cu2+. The relationship between metal ions and Abeta40 aggregates was analyzed, and the function of metal ions in Alzheimer's disease (AD) was illustrated in the research.

Objective To investigate the effects of a novel PPARγ agonist DH9 on Wntβ-catenin pathway in human polycystic kidney cystic-lining epithelial cells (WT9-12).Methods WT9-12 cells were treated with different concentrations of DH9 for 72 hours and the proliferation was assessed by MTT.WT9-12 cells were pretreated with SB216763 or GW9662 for two hours and then treated with DH9 for 72 hours.Western blotting was applied to detect the protein expression of β-catenin,phospho-β-catenin,GSK3β,phospho-GSK3β.Results DH9 could effectively inhibit the proliferation of the cells.60 μmol/L DH9 could facilitate β-catenin down-regulation (P<0.01) and phospho-β-catenin up-regulation (P<0.01).Inhibition of GSK3β by SB216763 could protect WT9-12 cells against DH9-facilitated β-catenin repression in a dose-dependent manner despite phosphorylating deactivation, but PPARγ inhibitor GW9662 couldn't.Conclusions DH9can effectively block the proliferation of WT9-12 cells. The effect may be mediated by facilitating the down-regulation of β-catenin via GSK3β-dependent mechanism.

The effects of Lipoid of Mujingzi〔LP,Fruits of vitex neguando L Var cannabifo- lia (Sieb et Zucc) Hand mazz〕 on Smooth muscles are reported. The results were as follows:① In the isolated perfused mice lung-bronchi, LP caused significant increase of perfusion medium dropping through the lung-trachea, and the effect of LP was dose-dependent; ACh (Acetyl-choline)-induced contraction of the isolated mice trachea smooth muscles could be relaxed by LP. ② When LP was given introperitoneally(100~ 300 mg?kg-1) into guinea pig, the incubationperiod of asthma induced by acetycholine together with histamine was prolonged. The optimum dilated effect of LP on smooth muscles of trachea in mice can be concluded as the results of synergistic effects by 4 compositions from LP (Oleic acid,Linoleic acid, Palmitic acid and Stearic acid).

Objective To study the factors affecting the activities of daily life (AOL) of patients after the first stroke of cerebral infarction in order to formulate the intervention strategy for improving the capability of ADL of patients. Method A total of 149 patients with the first stroke of cerebral infarction admitted from Oct. 2008 to Dec.2008 were enrolled in this study. The demographics of patients, cerebral infarction risk factors, apopletic score as per National Institute of Health stroke scale (NIHSS), white blood cell count (WBC) and plasma glucose (PG) were recorded on the first day of admission, and many other laboratory examinations were done on the next morning. The occurrences of infection in lung and urinary tract, and atrial fibrillation were recorded during hospi-talization. The NIHSS score and score of Glasgow Coma Scale (GCS) were taken within 24 hours after admission, on the 14th day and at the end of the third month after cerebral infarction respectively. Barthel index (BI) was taken in the second week as well as at the end of third month by follow-up in the OPD or by telephone. The correlation analysis and multiple linear stepwise regression analysis were used to find the risk factors. Results The PG level, WBC count and NIHSS score were independently associated with ADL in the second week as well as at the end of the third month after cerebral infarction. Besides, the urinary tract infection during hospital stay was also independently associated with ADL at the end of the third month after cerebral infarction. Conclusions The plasma glucose level, WBC count, NIHSS score and urinary tract infection are the risk factors. Positive measures should be taken to control these risk factors so as to improve the capability of ADL of the patients after cerebral infarction.

Objective To investigate the effects of Ganmaidazao Decoction (GMDZD), Sinisan(SNS) and Sijunzi Decoction (SJZD) recipe on endogenous nerve growth factor (NGF) of rat with chronic stress. Methods In order to establish the rat models with chronic stress, the feet of animals were stimulated with a pat-tern of electrical impulses (long term, high voltage and constant current). The alterations of behavior of animals were observed and quantified by open-field and tail-suspension test. Levels of NGF in pitttitary, hypetholamus and blood plasma were quantified using radio-immunoassay (RIA) method. Results 1) In comparision with the wild-type (WT) control rats, locomotor activity decreased significantly in the models and groups treated by Chinese medicinal herbs (P<0.01). In addition, the Chinese medicinal herbs treated groups exhibited increased locomotor activity than the model groups (P<0.01). 2) The cross time of the model groups decreased significantly compared with the WT controls (P < 0. 01). Meanwhile, three Chinese medicinal herbs treated groups displayed longer cross time than the model groups (P< 0. 01). 3)The vertical movement times of the model groups decreased significantly compared with the WT controls (P < 0.01). Meanwhile, three Chinese medicinal herbs treated groups displayed in-creased vertical movement times than the model groups (P <0.05). 4)In the tail-suspension test, the duration of immobility increased significantly in the model groups compared with the WT controls (P < 0. 01). However, the duration of immobility of three Chinese medicinal herbs treated groups were less than the model groups (P < 0.01). 5) Compared to the model groups, the level of NGF in pituitary of animals treated by GMDZD and SNS in-creased significantly (P<0.01). In the hypothalamus, the level of NGF in SJZD treated groups decreased in com-parision with the WT control and model groups (P<0.05). Whereas, GMDZD and SNS treated groups decreased significantly than WT control and model groups (P<0.01). In the blood plasma, the level of NGF showed a signif-icant reduction in the model groups compared with the WT control. Conclusion The results suggest that the GM-DZD,SNS and SJZD recipe have positive effects on rat with chronic stress through regulating the level of endogenous NGF.
