Objective The purpose of this study was to investigate the effects of short-term and high-intensity training on the biomechanics characteristic of guinea pig's Achilles tendon, and thus to propose a new training mode for quickly enhancing the ability of withstand stress and anti-injury. Method Guinea pigs were assigned to different groups according to the training intensity they would undergo, i.e. high intensity group, medium intensity training for 8-weeks group and control group. The bilateral Achilles tendons were excised after different strength training. The variety of biomechanics characteristic of different intensity and different period were observed and compared respectively. Methods 40 patients with chronic instability of lateral ankle and 40 normal people were randomly selected. Bilateral ankles of each subject had two basic roentgenographic measurements named inversion stress anteroposterior roentgenogram and anterior drawer stress radiograph. Objective To investigate the effect of the spleen on the function of intestinal barrier during the course of acute pancreatitis in rats.Methods Rats were randomly divided into four groups: sham operation group; actue pancreatitis group; splenectomy group;splenectomy plus acute pancreatitis group.The serum levels of TNF-?,IL-1,IL-6 and IL-10 in each group were examined 24 hours after operation.Two days after operation ,the rate of bacterial translocation(BT) was determined and the terminal ileum was excised and examined by transmission electron microscopy to detect the injury of intestinal mucosa.Results The serum levels of TNF-?,IL-1?,IL-6 and IL-10 in splenectomy plus acute pancreatitis group were 3.06?3.61,16.46?5.52,19.90?6.89,6.94?3.93,and in acute pancreatitis group were 19.93?2.38, 42.79?4.31, 20.19?3.35, 39.28?12.69 respectively. Objective To study effects of recombinant ciliary neurotrophic factor(CNTF) on the changes of JAK\|STAT pathway and tyrosine phosphorylation in the injured neurons. Methods Sciatic nerve of rat was resected and sutured into silicone tube with local infusion of recombinant CNTF.The distribution and quantity of STAT3 and phosphotyrosine(PTyr) immunoreactivity in the neurons of L3\|L5 spinal anteriolateral nuclei and L5 spinal ganglion were observed and measured by immunohistochemical ABC method with computer image analysis. Results There was much STAT3 immunoreactivity in the neuron located in the nucleus of spinal anteriolateral nuclei.PTyr immunoreactivity showed higher in the cell membrane of spinal anteriolateral nuclei and in the cytoplasma and neucleus of spinal ganglion in CNTF group than that in SAL group.Conclusion\ The results suggest JAK\|STAT pathway in the injured motoneurons be activated and strengthened and tyrosine phosphorylation in the injured neurons be enhanced by recombinant CNTF.\;[

Methods Sciatic nerve of rat was resected and sutured into silicone tube with local infusion of recombinant CNTF.One or two weeks after nerve repaired,the distribution and quantity of S100 protein (S100),growth associated protein 43 (GAP-43), phosphotyrosine (PTyr) and signal transducer and activator of transcription 3(STAT3) immunoreactivity in the distal nerve of the injured sciatic nerve were observed and measured with immunohistochemical ABC method by computer image analysis. Results S100,GAP-43,PTyr and STAT3 immunoreactivity showed significantly higher in the distal nerve of the injured sciatic nerve in CNTF group than that in SAL group.Conclusion\ Recombinant CNTF could up-regulate the expressions of S100,GAP-43,PTyr and STAT3 in Schwann cells of the injured peripheral nerve.The results suggest that the JAK-STAT pathway can be strengthened,and the expressions of S100 and GAP-43 can be subsequently up-regulated by recombinant CNTF in Schwann cells.

Objective To study the platelet antibody screening and crossing match by surface plasmon resonance(SPR) ,and to find a new way for platelet compatibility testing .Methods The corresponding universal platelet antigen was fixed on the SPR chip surface by the amino coupling method .Platelet antibody positive and negative control serum were analysed by SPR micro-ar-ray ,the stability ,sensitivity and specificity of this technique were discussed ,and compared with MAIPA assay .Finally we used the SPR technology to cross match for ten cases of the platelet antibody positive patients before infusion ,and to evaluate the effect of platelet infusion .Results For the SPR technology ,the stability ,sensitivity and specificity of platelet antibody detection were all better ,106 cases of the repeated platelet transfusion samples were tested by SPR assay and MAIPA method ,there was no signifi-cant difference between them(chi-square=0 .333 ,P>0 .05) ,the total consistency was 97 .2% .The 10 cases of platelet antibody positive patients were crossed match before platelet transfusion by SPR technology ,the good results of 8 cases of them were found by the clinical tracking evaluation ,1 h CCI>7 .5 ,24 h CCI>4 .5 .Conclusion SPR technology for screening platelet antibody mat-ches with MAIPA method in basic quality ,but SPR assay is simple ,rapid ,reliable and intuitive ,label free ,which can satisfy the re-quirements for clinical rapid detection of platelet antibody screening and crossing match .

Objective To explore the role of Hippo pathway in the pathogenesis of autosomal dominant polycystic kidney disease (ADPKD),and find potential targets for drug therapy.Methods By means of immunofluorescence staining,Western blotting,Real-time PCR,the differences of sublocalization,expression and phosphorylation level about Hippo pathway molecules in Han:SPRD (cy/+) and ADPKD patients compared with the control were observed.Knockdown Yes kinaseassociated protein (YAP),transcriptional coactivator with PDZ binding motif (TAZ) and large tumor suppressor kinase1 (LATS1) in cystic lining epithelium cell line WT9-12 were took by siRNA interference,and then their effects on cell proliferation,apoptosis and cell cycle were assessed.Results In cystic lining epithelium of Han:SPRD(cy/+),decreased expression of LATS1 and increased expression of YAP were found compared with the control,and the immunofluorescence of YAP was distributed both in cytoplasm and nucleus,while distribution and expression level of TAZ were without significant variance.Abnormal mRNA expressions of Hippo pathway components in ADPKD patients were found (P ＜ 0.05).Down-regulation of LATS1 in WT9-12 cells could prohibit phosphorylation of YAP,and prompted proliferation and cell division.Knockdown YAP in WT9-12 cells could inhibited cell proliferation by arresting cell cycle in G0/G1 phase,but down-regulating TAZ showed no significant differences in proliferation and cell cycle.Conclusions Altered Hippo signaling exists in ADPKD,and YAP activation may be one leading cause of autosomal dominant polycystic kidney disease onset.In vitro,knockdown YAP in WT9-12 cells can inhibit cell proliferation by arresting cell cycle and depressing cell division,suggesting the expression level and activity of YAP are potential targets for ADPKD treatment.

In order to study the expression and the feasibility of scaled production of neuropeptide in the routine expression system such as E.coli with the pituitary adenylate cyclase activating polypep tide(PACAP) as an example, the following experiments were carried out. First, on the basis of the reported amino acid sequence of PACAP, DNA sequence of PACAP w as deduced and six partially complementary oligonucleotide fragments were design ed. The coding region of PACAP was obtained by renaturing the DNA fragments and ligation and identified by DNA sequencing. The coding region of PACAP was cloned into plasmid pGEX-4T-3 and transformed into E.coli BL21(DE3 ). An expression strain BLPACAP was selected. SDS-PAGE analysis revealed that t he GST-PACAP fusion protein was highly expressed and accumulated to about 30% o f the total bacterial proteins. By affinity chromatography, up to 90% GST-PACAP was purified by one step from bacterial lysate. The purified protein could prom ote neurite outgrowth of PC12 cells and the survival of spinal cord neurons.

AIM To study the influence of chronic stress on the level of CNTF and CNTF mRNA in hippocampal neurons of rats. METHODS The chronic stress model was established by chronic unpredictable mild stress, openfield test was performed to detect the behavior of rats. Immunohistochemistry and in situ hybridization were used to observe the level of CNTF and CNTF mRNA. RESULTS Compared to control group, the CNTF like immunoreactivity and signals of CNTF mRNA in situ hybridization in the hippocampal neurons of chronic stress group were significantly decreased. CONCLUSION These results show that chronic stress can significantly decrease the level of CNTF and CNTF mRNA in the hippocampal neurons of rats.

Objective:To elucidate the possible role of galanin in the development of experimental depression in rats. Methods:Openfield was performed to test the behavior of rats. The changes of the galanin level in different brain areas were determined by RIA. The effect of fluoxetine hydrochloride on galanin level were observed by intraperitoneal injection. Results:Compared to control group, the crossing times and rearing times decreased significantly in depressed rats, galanin level decreased remarkably in plasma, hypothalamus, hippocampus, forebrain, parietal lobe and temporal cortex of depressed rats. Intraperitoneal injection of fluoxetine hydrochloride obviously improved the depressed behavior in rats, increased the galanin level in the hippocampus and forebrain of depressed rats. Conclusion:Hippocampus and forebrain may be involved in the development of experimental depression and in the antidepressive effects of fluoxetine hydrochloride.
