1.Effects of inactivated HIV-1 particles on human CD4~+T cell activation and Th1/Th2 cytokine secretion in whole blood

AIM:To investigate the effects of AT-2-inactivated HIV-1 particles on human CD4+T cell activation and cytokine secretion in whole blood (WB) in vitro. METHODS: HIV-1ⅢB particles were inactivated by AT-2 chemical and the concentration of p24 antigen was determined by p24 ELISA. AT-2-inactivated HIV-1ⅢB particles were added to human WB culture system in serial concentrations to stimulate the cells. PHA was used as positive control. After 24 h, all the cultural supernatants were harvested and the concentrations of Th1 (IL-2, IFN-? and TNF-?) and Th2 (IL-4, IL-6 and IL-10) cytokines released to the supernatants were detected by cytometric bead array (CBA). The percentage of CD69 expression on CD4+T cells from WB was detected by immuno-fluorescence staining plus flow cytometry. RESULTS: The concentration of p24 antigen in the AT-2-inactivated specimen was 85.5 ?g/L. 24 h later, the percentage of CD69 expression on CD4+T cells from control group was (1.62?0.63) %, whereas it was (38.82?6.00)%, (3.83?1.07)%, (5.94?0.85)% and (9.30?1.22)% in PHA group, HIV-1 (1/500) group, HIV-1 (1/50) group and HIV-1 (1/5) group, respectively. Cytokines secreted by WB in control group were mainly TNF-? and IL-6. However, all the six cytokines tested were strikingly increased in PHA group, as well as in HIV-1ⅢB groups. CONCLUSION: AT-2-inactivated HIV-1ⅢB particles activate CD4+T cells from WB, and up-regulate both Th1 and Th2 cytokine secretion in WB. Besides the effects of viral proteins, other mechanisms may be proposed that HIV-1 particles act as antigen presenting cell (APC) because many host-derived immune molecules are incorporated into HIV-1 envelop when it is released from infected cells by budding, and exert immune modulation.

AIM: To study the effects of [8-(diethylamino) octyl-3, 4, 5 -trimethoxybenzoate] (TMB-8), an intracellular Ca2+ antagonist, on the activation, proliferation and cell-cycle distribution of the mouse T lymphocytes stimulated by concanvalin A (Con A) in vitro. METHODS: After stimulated with Con A, T cells were treated with different concentrations of TMB-8 alone and its combination with cyclosporine A (CsA). The expression of CD69, the early marker of CD3+ T cell activation, was measured by FACS. The proliferation-related index was determined by carboxyl fluorescin diacetate succinmidyl ester (CFDA-SE) flow cytometry. Objective To investigate the T cell cytotoxicity induced by recombinant adenovirus carrying HIV-1 vpr gene.Methods C8166 cells infected with rAd-vpr or negative control rAd-vector,were analyzed for cell cycle distribution and cell death by flow cytometry.The discrimination of living cells,apoptotic and necrotic cells were differentiated with Hoechst-PI double staining under the confocal microscopy.Changes of mitochondrial membrane potential(△ψm)were monitored by JC-1 staining method.Results Annexin V-PI and Hoechst-PI staining indicated the death effects of HIV-1 Vpr on C8166 cells.PI flow cytometric analysis showed that cell cycle arrested in G2 phase.C8166 cell△ψm collapse mediated by Vpr was detected by JC-1 fluorescent staining.Conclusion The ability of recombinant adenovirus carrying HIV-1 vpr gene to induce mitochondria dysfunction,cell cycle G2 phase arrest and cell death was confirmed in C8166 cells.
4.Analysis of 22 cases of forensic psychiatric illness

Objective To explore the problems that should be paid attention to during the process of the identification of the mental illness of the judiciary.Methods The process and conclusion of 22 cases of forensic psychiatric expertise were reviewed and analyzed in the Department of psychiatric disorders,Beijing Anding Hospital,Capital Medical University from August 1,1982 to July 31,2016.The analysis indexes mainly include:(1) the time of the implementation of the identification;(2) the matters entrusted by the Commission;(3) the identification materials provided by the client;(4) the conclusion of the appraisal.Results (1) from July 31,1999 to August 1,1982,7 cases were identified from to August 1,1999,and 15 cases were identified from to July 31,2016.(2) from August 1,1982 to July 1999 31 during the day mainly identification issues for 6 cases (85.7%,6/7) mental state and the ability of criminal responsibility and 1 cases (14.3%,1/7) mental state and sexual self defensive ability,from August 1,1999 to July 2016 31 during the day in 15 cases of all matters for identification of mental state and civil capacity.(3) including the authentication materials provided by a client:Previous expert opinions and description of mental state of medical records,effective during the testimony of witnesses and / or detention,public security organs information records,audio-visual materials.(4) most of the identified people can be identified in the clinical diagnosis of mental illness and to determine the legal capacity,individual identification can not come to the conclusion.Conclusion The identification number and the difficulty of life;for his identification of civil capacity increased;should focus on ensuring the identification data of the authenticity,integrity,adequacy and validity;"conclusion" is also a kind of expert conclusion,it is suggested to use three cent method civil capacity rating was identified.
5.The application value of the combination of personality responsibility theory and motivation theory in the identification of the criminal responsibility ability of the patients with mental disorders

Objective To explore the value of the combination of personality responsibility theory and motivation theory in the evaluation of the ability of criminal responsibility of the patients with mental disorders. Methods A retrospective analysis of our hospital in 2013 and 2014 the implementation of identification of 101 cases of patients with mental disorders of the criminal responsibility, analysis indicators include: the reflection of the basic characteristics of personality; motive; identification of illegal behavior evaluation; the capacity of criminal responsibility. Results different symptoms reflects abnormal consistency and specificity of personality integrity; when the symptoms are not at the same time, with the implementation of the illegal behavior of the motivation also has realistic and non realistic differences; personality integrity can not damage the normal and correct evaluation of the illegal act, almost all favorable personality particularity of affected persons can recognize the illegality of the behavior; the integrality and consistency of the affected personality criminal responsibility were incomplete; special personality influence for full criminal responsibility. Conclusion The combination of personality responsibility theory and motivation theory is applied to the identification of the criminal responsibility ability of the patients with mental illness. To define the identity of the actor as the actor, to identify the patient's behavior is "not control" or "no control", it is worth promoting the application in the work of mental illness judicial identification.
