1.Effect of continuous passive motion on the healing of experimental full thickness defects of articular cartilage in knee of rabbit

Objective To study the effect of continuous passive motion (CPM) on the healing of man made full thickness defects of articular cartilage in the knee of rabbit. Methods A diameter of 3.2mm full thickness defect of articular cartilage in the facies patellaris of femur were made in 60 knees of rabbits. Then the rabbits were randomly divided into groups A,B and C. The animals in groups A and B were treated with CPM for 8 hours and 2 hours daily, respectively, for 4 weeks, while those in group C kept in cage for free activity only. Half of the animals of every group were sacrificed at the end of the 4th and 12th weeks. The H.E. staining, safranin O staining and immunohistochemical analysis of collagens I and II were conducted, the gross appearance and histological change were observed and scored. Results There was significant difference with regard to the score of gross appearance and histological change among groups A, B and C. Conclusion The healing of full thickness defects of articular cartilage in rabbits treated by CPM is better than that in rabbits without CPM treatment. CPM can promote tissue repair and stimulate synthesis of collagenⅡand the glycosaminoglycan.
2.The experimental studies of the effects of As_2O_3 on the expression of telomerase hTERT-mRNA and apoptosis of HL-60 cells

AIM To investigate the effects of apoptosis and the expression of telomerase hTERT mRNA activity of HL-60 cells treated with As 2O 3 in different concentrations. METHODS HL-60 cells was cultured with As 2O 3 in different concentrations for 48 hours, then, cell apoptosis was detected with flow cytometry and the expression of telomerase hTERT mRNA was determined with RT-PCR respectively. RESULTS The percentage of the apoptosis in HL-60 cells were 2 28%?0 40%?2 55%?0 22% and 47 57%?2 79% respectively, as the cells were treated with 0 1 ?mol?L -1,1 0 ?mol?L -1,5 0 ?mol?L -1 As 2O 3 for 48 hours. 3.Effect of berberine on proliferation and apoptosis of MCF-7 cells

Objective To investigate the effect and mechanisms of the Berberine on the human breast adenocarci-noma cell line MCF-7 . Methods MTT method was used to evaluate the proliferation effect of MCF-7 and the per-centage of apoptotic cells were determined by flow cytometric analysis. The expressions of JAK2, p-JAK2, p-STAT3, STAT3, Bax, Bcl-2, Cleavage-PARP and Cleavage-Caspase3 were detected by Western blotting. Results The results showed that BBR treatment decreased the activation of JAK2/STAT3 signal pathway and up-regulated the expression of Bax, Caspase3 and PARP activation with the decrease of the expression of Bcl-2. Wherefore, ex-pression of the constitutively active form of STAT3 could attenuate the effect of BBR on the MCF-7 cell. Conclusion Berberine can induces apoptosis and proliferation inhibition of breast adenocarcinoma cell lines of MCF-7 through inhibition of the JAK2/STAT3 signaling pathway.
4.Study on the Blood Concentration of Fluorouracil Administered into Guinea Pigs by Arterial&Intravenous Infusion and Celiac Infusion

OBJECTIVE:To compare and analyze blood concentration changes of fluorouracil administered to guinea pigs by different paths.METHODS:The peripheral vein and portal vein blood samples of guinea pigs were taken at different time after administered with fluorouracil in3different ways:group A(by arterial infusion),group B(by intravenous infusion)and group C(by celiac infusion).The blood concentration changes were determined and analyzed by HPLC.RESULTS:The portal vein and peripheral vein blood concentrations in group A and B reached peak value5min after administration with flu?orouracil while group C reached peak at10min;the portal vein blood concentrations in group A reached the highest in the first20min while group C showed the highest value in the last30min.CONCLUSION:Arterial infusion combining celiac infusion path was recommended for local chemotherapy;arterial infusion combining intravenous infusion or intravenous infusion com?bining celiac infusion was recommended for the local therapy while giving consideration to systemic chemotherapy.

Objective To test the antiproliferative and proapoptotic effect of Survivin small interfering RNA(siRNA) expression vector on lung adenocarcinoma cell A549 in vitro.Methods The Survivin-siRNA expression vector was constructed and conformed by sequencing.The inhibitory effect of Survivin-siRNA was tested by fluorescent quantitative reverse transcription polymerase chain reaction(FQ RT-PCR),Western blot and immunohistochemistry.The cell proliferation and apoptotic rate were assayed by tetrazolium bromide(MTT)colorimetry and flow cytometry.Results Survivin-siRNA expression vector was constructed and transfected into A549 cell.It effectively reduced mRNA and protein level of Survivin.Immunohistochemistry also showed lower expression of Survivin.The A549 cells transfected with Survivin-siRNA had a lower cellular growth rate than that of control group(P

As a rehabilitation strategy in the project of community development, community-based rehabilitation (CBR) can improve quality of life effectively in the community disabled, possessing the characteristics of wide coverage, convenience, being economical and effectiveness, so it has been developed and promoted in our country. At present, there are various problems in CBR, which could not meet the diverse demand of people with disablities in community. As a result, it is important to propagandize rehabilitation concept, built rehabilitation team, strengthen multidisciplinary cooperation and emphasize the development of the disabled rehabilitation system in rural community.
9.A Research on Dactylogram of She Nationality Juveniles

Objective To find out the distribution of the normal rating dactylogram of the juveniles of the She Nationality. Methods According to living body measure need, Prussian blue was used. Result Each type of dactylogram in number was as follows: LU>WS>WD>AT>AS. Ridgs count of each finger in quantitative order:thumb>ring finger>middle finger>index finger>little finger. Conclusion The rate of appearance of normal finger prints in various nationalities and regions has obvious difference, the normal rating of the dactylogram of the juveniles of the She Nationality differs from that of the local juveniles of the Han nationality.
10.Effect of continuous passive motion in prevention of osteoarthritis induced by experimental articular cartilage injury

Objective To study whether the early continuous passive motion (CPM) can prevent the degeneration of articulation after the articular cartilage was experimentally defected, and the effect of the different durations of continuous passive motion therapy. Methods Fifteen New Zealand rabbits were used to establish the model of full-thickness defects of bilateral knee articular cartilage by operation. Then all the animals were divided into groups A, B and C: Group A served as the control group, and groups B and C served as the observation groups and were administered with 2 hours of continuous passive motion a day (group B) and 8 hours of continuous passive motion a day, respectively. All rabbits were sacrificed at the 12th week after operation, and their articular cartilage were observed. The cartilage and synovium were obtained to have the hematoxylin and eosin stain and immunohistochemistry stain. After that, the pathological assessment and calculation of the ratio of MMP-3 positive cells were conducted and compared among groups.
